Background: To reveal establishment strategy of Sparganium erectum, we tried to find realized niche of adults through field survey and effects of water level on the establishment process through mesocosm experiments. Results: In the field survey, the height and coverage of community living in deeper water were greater than those of community living in shallow water. There was no statistically significant difference (p > 0.05) in the means of water and soil properties between the two communities. In mesocosm experiments, we found no correlation between water levels and germination rates, but S. erectum seedlings have characteristics of post germination seedling buoyancy when S. erectum seeds germinated in inundation conditions. Shoot height, total leaf length, and survival rates of sinking seedlings in shallow water levels at -5, 0, and 5 cm were higher than those in deeper water levels at 10 and 20 cm. Floating seedlings established in water levels of 3 and 6 cm only. The seedlings could live up to 6 weeks in floating state but died if they were unable to establish. Conclusions: The water level around adult S. erectum communities in the field were different from the water level at which S. erectum seedlings can survive in the mesocosm experiments. The findings provided not only understanding of S. erectum habitat characteristics but also evidence to connect historical links between the early seedlings stage and adult habitat conditions. We suggested the logical establishment strategy of S. erectum based on the data.
Enzyme activities and phenolic compound were compared to investigate the physiological characteristics during shoot formation from diploid and haploid of Nicotiana tabacum w BY4. The Nakata medium with 1.0 mg/L IAA, 0.5 mg/L Kinetin and 3 g/L activie carbon was excellent to induce the haploid plants from the middle size anther within 30 days after culture. The MS medium with 0.5 mg/L 2,4-D was good for callus induction from leaf explants of diploid and haploid, and a lot of plane were regenerated from calli on the MS medium supplemented with 2.0 mg/L BAE Activities of peroxidase for both of diploid and haploid plane were the highest at 2.0mg/L BAP Activities of IAA oxidase and catalase of haploid Plants were high or than those of diploid plants. On the other hand, activity of peroxidase of haploid plants were lower than those of diploid plants.
Embryogenic calli of Codonopsis lanceolata were cultured on MS agar medium containing various concentrations of sucrose as a carbon source. Upon transfer to MS basal medium, somatic embryos of cotyledonary stage converted to plantlets. When sucrose was added with greater than 4%, the number of shoots and roots regenerated from somatic embryo increased. However, the growth of shoots and roots was retarded in agar medium with more than 2% sucrose, but promoted in medium with lower concentration of sucrose. Saponin contents of shoots regenerated from somatic embryos, embryogenic calli, non-embryogenic calli, and native roots were determined by HPLC. Saponin contents of native root was variable, depending on regenerant, embryogenic calli, and cotyledonary embryos. The saponin contents of regenerated roots in medium with high sucrose was similar to native roots. Saponins content based on cell differentiation to shoot and root was dramatically decreased. This results could be effectively controlled for the production of useful secondary metabolites.

Brassinosteroids are steroidal plant hormones and are known to modulate physiological and cellular response in a wide range of plant species. Considerable insights has been achieved of the physiological role of brassinosteroid in Brassica species in the past few years, but their effect on direct organogenesis has not been extensively studied. In this sense, under optimal basal media and growth conditions we tested the cellular and organogenic response of 24-epibrassinolide (EBL) in a variable concentration (0.1 to $5.0\;{\mu}M$) and Zeatin (Z) (1.0 to $100\;{\mu}M$) and their synergic effect on hypocotyl explants of cauliflower and broccoli. The isolated EBL accelerated cell elongation and promotes direct organogenesis. One micromolar EBL + $10\;{\mu}M$ of Z was the most efficient combination for cell elongation, cell differentiation as well as for organogenesis. A suppressing effect on root induction was confirmed for all the tested hormone levels. The general results indicate a synergic effect of EBL-Z and EBL potentates Zeatin activity, at least in certain tissues. Besides de genetic factors, we can speculate that the natural hormone concentration in the explants might affect the responses by application of exogenous growth regulators. Experiments with new plant growth regulators, like brassinolide, are important aiming to maximize or accelerate plant regeneration for in vitro multiplication or for genetic transformation.
Immature flower buds of 'Desio' carnation were cultured on MS agar medium supplemented with 1 ㎎/L 2,L-D. Embryogenic calli were formed from 5-10% of the buds less than 20 ㎜ in length, but only non-embryogenic calli were produced from explants of shoot apex leaf, internode, and flowere buds larger than 20 ㎜. The same method was applied to 16 cultivars of cut Sower carnation and embryogenic calli were obtained in 7 cultivars. Several embryogenic callus lines were selected and maintained through subcultures over 120 weeks without loss of embryogenic competence. The embryogenic cultures were also proliferated rapidly in liquid agitation cultures using MS medium supplemented with 1mg/L 2,4-D. Numerous embryos were formed on the periphery of the cell aggregates upon transfer to auxin-free MS agar medium. Plantlets were transplanted in potting soil and grown to bloom in six months.

When root initiation ratio of cut Birdsfoot trefoil (Lotus corniculatus L.) stems was examined in several medium conditions containing different IBA concentration, the higher IBA contration of medium was elucidated superior then lower IBA concentration. Highest root initiation ratio was confirmed at 2.5 mg/$\ell$ of IBA and the ratio was 90~95%. When the stems from regenerated shoots from callus were treated at 8 kinds of medium for 12 days, the root iniation result was 9 (45%) at 1/2 SH-0 medium, 10 (50%) at SH-0 medium, 10 (50%) at SH-0.5IBA medium, 10 (50%) at SH-1.0IBA medium, 13 (65%) at SH-1.5IBA medium, 14 (70%) at SH-2.0IBA medium, 19 (95%) at SH-2.5IBA medium and 14 (70%) at SH-3.0IBA medium.

When the stems from regenerated shoots from callus were treated at 8 kinds of medium for 10 days, the root iniation result was 10 (50%) at 1/2 SH-0 medium, 10 (50%) at SH-0 medium, 12 (60%) at SH-0.5IBA medium, 15 (75%) at SH-1.0IBA medium, 15 (75%) at SH-1.5IBA medium. 10 (50%) at SH-2.0IBA medium, 9 (45%) at SH-2.5IBA medium and 8 (40%) at SH-3.0IBA medium.
Optimal regeneration conditions of ginseng transformants were studied. It has been known that Ferritin Light Heavy Chain (FLHC) gene remove the several heavy metal by combination, store and transport. To obtain the ginseng tolerant to heavy metal, binary vector was introduced in Agrobacterium by tri-parental mating and then Agrobacterium tumefaciens MP90/FLHC was selected on the AB media and MS media containing kanamycin. Explants were co-cultured with Agrobacterium tumefaciens MP90/FLHC, which contained NPT II as a selectable marker, tadpole ferritin heavy chain (FLHC) gene and human ferritin light chain gene and then a number of embryos were induced. The induced embryo transferred to shooting media consisting of MS medium supplemented with GA 10 mg/L. As a result of examination that induced the normal growth of transfomants, transformants showed the equivalent growth in both root and shoot on the media containing the 1/3 MS.
Nitrate reductase deficient (NR) mutant lines were selected indirectly by their resistance to 100mM chlorate in cell cultures of P. parviflora. A total of 585 chlorate resistant lines were confirmed by a second passage on a high concentration of chlorate. Frequency of spontaneous mutation was $9.7{\times}10^{-7}$ in 3 month old suspension-cultured cells, and in non-selective media containing amino acids as sole nitrogen source. The frequency of mutation could be increased up to 11-fold by culture for 12 months. Out of 40 randomly selected calli, 22 were fully deficient in NR. The rest of the clones contained a decreased level of NR activity. Further characterization was carried out in 13 mutant lines which were fully deficient in NR and in 5 mutant lines containing residual (0-7.0%) NR activity, as compared to wild-type cells cultured on the same medium. The $NR^-$ mutants were tentatively classified as defective in the NR apoenzyme (nia-type; 11 mutant lines including the 5 with residual NR activity) or in the molybdenum cofactor (cnx-type; 7 mutant lines) by the XDH activity. The cnx-type could be further classified into two groups. In one group (5 mutant lines) of these, the NR activity could be partially restored by nonphysiologically high (1.0mM) molybdate in the culture medium. Both types of $NR^-$ mutants were unable to grow on minimal medium containing nitrate as sole nitrogen source, but grew well on amino acids. They also proved to be extremely sensitive to the standard medium ($MSP_1$) containing nitrate and ammonium. Shoot regeneration was obtained only in the $NR^-$ mutants, which contained residual NR activity, but they so far have failed to grow into plants.

Aralia elata is found in mountain areas all over Korean peninsula. Aralia elata is the scientific name for Japanese angelica tree. The tree belongs to the family Araliaceae, commonly known as ginseng family. Bud sprouts from apical shoot tip of the plants are rich in flavor and thus mainly used for both folk medicine and vegetable. The stalks with apical buds are gathered in the early spring and planted in sandy soil or water in the greenhouse. The sprouting buds are then collected and sold as fresh vegetable. Although the plants have been used for food, they have been cultivated in a very small scale. In spring, local farmers just go around mountain areas to search the trees and gather the stalks as much as they get and sell them to the market. No conservation efforts have been made to stop the exploitation or to save the dwindling population. We tried to provide local farmers with the plants that may be used as an alternative to stalks from wild populations. This will bel! p conserve the wild populations. However, it is hard to propagate them either by conventional cuttings or by seed germination in a short period of time. Mass propagation using tissue culture systems have shown a great promise with several woody plants. Recently we developed a mass propagation technique via somatic embryogenesis system using mature and/or juvenile explants for Aralia elata. Several factors affecting somatic embryogenesis system including SE(somatic embryo) induction, embryogenic callus proliferation, SE germination, plant regeneration and transplanting to field frill be presented. And some problems arising for the somatic embryogenesis system will be also discussed.
