The 29 May 2004 Offshore Southeast Coast of Korea Earthquake Sequence: Shallow Earthquakes in the Ulleung Back-arc basin, East Sea (Sea of Japan)

  • The 29 May 2004 offshore Uljin, Korea earthquake was predominantly thrust-faulting at a depth of approximately 12 (±2) km. The mainshock attained the seismic moment of M0 =5.41 (±1.87)  1016 N m (Mw = 5.1). The focal mechanism indicates a subhorizontal P-axis trending 264° and plunging 2°. The orientation of P- and T-axis is consistent with the direction of absolute plate motion generally observed within the plates, hence the cause of the May 29 shock is the broad-scale stress pattern from the forces acting on the downgoing slab along the Japan trench and inhibiting forces balancing it. The 29 May 2004 earthquake occurred along a deep seated (~12 km), pre-existing feature that is expressed on the surface as the basement escarpment along the western and southern slopes of the Ulleung basin. The concentrated seismicity along this basement escarpment suggests that this feature may qualify as a seismic zone - the Ulleung basement escarpment seismic zone (UBESZ).

Reduction of TNE ${\alpha}-induced$ Oxidative DNA Damage Product, 8-Hydroxy-2'-Deoxyguanosine, in L929 Cells Stably Transfected with Small Heat Shock Protein

  • Previous studies have demonstrated that oxidative stress involving generation of reactive oxygen species (ROS) is responsible for the cytotoxic action of $TNF{\alpha}$. Protective effect of small heat shock proteins (small HSP) against diverse oxidative stress conditions has been suggeted. Although overexpression of small hsp was shown to provide an enhanced survival of $TNF{\alpha}$-sensitive cells when challenged with $TNF{\alpha}$, neither the nature of $TNF{\alpha}$-induced cytotoxicity nor the protective mechanism of small HSP has not been completely understood. In this study, we have attempted to determine whether $TNF{\alpha}$ induces oxidative DNA damage in $TNF{\alpha}$-sensitive L929 cells. We chose to measure the level of 8-hydroxy-2'-deoxyguanosine (8 ohdG), which has been increasingly recognized as one of the most sensitive markers of oxidative DNA damage. Our results clearly demonstrated that the level of 8 ohdG increased in L929 cells in a $TNF{\alpha}$ dose-dependent manner. Subsequently, we asked whether small HSP has a protective effect on $TNF{\alpha}$-induced oxidative DNA damage. To accomplish this goal, we have stably transfected L929 cells with mouse small hsp cDNA (hsp25) since these cells are devoid of endogenous small hsps. We found that $TNF{\alpha}$-induced 8 ohdG was decreased in cells overexpressing exogenous small hsp. We also found that the cell killing activity of $TNF{\alpha}$ was decreased in these cells as measured by clonogenic survival. Taken together, results from the current study show that cytotoxic mechanism of $TNF{\alpha}$ involves oxidative damage of DNA and that overexpression of the small hsp reduces this oxidative damage. We suggest that the reduction of oxidative DNA damage is one of the most important protective mechanisms of small HSP against $TNF{\alpha}$.

나사잭 메커니즘을 이용한 비폭발식 분리장치 (Non-explosive separation device using screw jack mechanism)

  • 본 논문에서는 나사잭의 메커니즘을 이용한 인공위성의 비폭발식 분리장치를 설계 및 제작하였다. 분리장치의 구동기로는 정격토크가 $1.7kgf{\cdot}cm$의 성능을 가진 회전형 피에조 모터를 사용하여 분리장치의 안정적인 작동이 이뤄지도록 하였다. 또한 인공위성의 분리장치로의 성능을 검증하기 위하여 반응속도 실험, 준정적 하중실험, 충격실험을 수행하였다. 실험을 수행한 결과, 반응속도는 약 1.172초로 측정되었고, 45kgf의 하중에서도 안정적으로 견딜 수 있음을 확인하였다. 최대 충격 가속도는 18G가 측정이 되었는데 이것은 폭발식 분리장치에 비해 매우 작은 값이다. 우리는 이러한 실험을 통하여 제안한 분리장치의 신뢰성을 확인하였으며, 인공위성 분리장치로써의 가능성을 제시하였다.

종양 세포 용해액에 따른 수지상세포 유도 항원 특이 면역반응 차이의 기전 연구 (Mechanism of Differential Ag-specific Immune Induction by Different Tumor Cell Lysate Pulsed DC)

  • Background: Tumor cell lysate has been considered as a preferential antigen source for the therapeutic dendritic cell pulsing. Our experiences with in vivo study with animal tumor model indicate the tumor cell lysate dependent differential effect of DC therapy. Our previous data show that MC38 lysate pulsed-DC induced stronger ag-specific immunity than CT26 lysate pulsed-DC in vitro. In this study we tried to reveal the mechanism for differential induction of ag-specific immunity of different colon cancer cell lysate pulsed-DCs. Methods: MC38 and CT26 cell lines were prepared as lysate by freezing-thawing procedure. Tumor cell antigenicity was confirmed by detecting the surface expression of MHC I/II & B7.1/2 molecules. IL-10, IL-12 and TGF-beta in the tumor cell lysate were detected by ELISA and the presence of heat shock proteins were analysed by western blotting. Results: The secretion of IL-10, a immune-inhibitory cytokine was about 470% higher in CT26 lysate than in MC38. Hsp 70 was detected only in the MC38 lysate but not in the CT26. On the other hand, Hsp 60 and 90 expression were not different in two colon cancer cell lysates. Conclusion: In two different colon cancer cell lysate, immune inhibitory IL-10 (higher in CT26) and Hsp70 (MC38 superiority) were differentially expressed. These data indicate that higher agspecific immunity induction by MC38 lysate pulsed-DC may due to the expression of hsp70 and lower secretion of IL-10, a immune-inhibitory cytokine than CT26 lysate. The significance of other cytokine and the surface marker expression will be discussed.

Molecular cloning of metal-responsive transcription factor-1 (MTF-1) and transcriptional responses to metal and heat stresses in Pacific abalone, Haliotis discus hannai

  • Background: Metal-responsive transcription factor-1 (MTF-1) is a key transcriptional regulator playing crucial roles in metal homeostasis and cellular adaptation to diverse oxidative stresses. In order to understand cellular pathways associated with metal regulation and stress responses in Pacific abalone (Haliotis discus hannai), this study was aimed to isolate the genetic determinant of abalone MTF-1 and to examine its expression characteristics under basal and experimentally stimulated conditions. Results: The abalone MTF-1 shared conserved features in zinc-finger DNA binding domain with its orthologs; however, it represented a non-conservative shape in presumed transactivation domain region with the lack of typical motifs for nuclear export signal (NES) and Cys-cluster. Abalone MTF-1 promoter exhibited various transcription factor binding motifs that would be potentially related with metal regulation, stress responses, and development. The highest messenger RNA (mRNA) expression level of MTF-1 was observed in the testes, and MTF-1 transcripts were detected during the entire period of embryonic and early ontogenic developments. Abalone MTF-1 was found to be Cd inducible and highly modulated by heat shock treatment. Conclusion: Abalone MTF-1 possesses a non-consensus structure of activation domains and represents distinct features for its activation mechanism in response to metal overload and heat stress. The activation mechanism of abalone MTF-1 might include both indirect zinc sensing and direct de novo synthesis of transcripts. Taken together, results from this study could be a useful basis for future researches on stress physiology of this abalone species, particularly with regard to heavy metal detoxification and thermal adaptation.

X-Ray, UV and Optical Observations of Classical Cepheids: New Insights into Cepheid Evolution, and the Heating and Dynamics of Their Atmospheres

  • To broaden the understanding of classical Cepheid structure, evolution and atmospheres, we have extended our continuing secret lives of Cepheids program by obtaining XMM/Chandra X-ray observations, and Hubble space telescope (HST) / cosmic origins spectrograph (COS) FUV-UV spectra of the bright, nearby Cepheids Polaris, ${\delta}$ Cep and ${\beta}$ Dor. Previous studies made with the international ultraviolet explorer (IUE) showed a limited number of UV emission lines in Cepheids. The well-known problem presented by scattered light contamination in IUE spectra for bright stars, along with the excellent sensitivity & resolution combination offered by HST/COS, motivated this study, and the spectra obtained were much more rich and complex than we had ever anticipated. Numerous emission lines, indicating $10^4$ K up to ${\sim}3{\times}10^5$ K plasmas, have been observed, showing Cepheids to have complex, dynamic outer atmospheres that also vary with the photospheric pulsation period. The FUV line emissions peak in the phase range ${\varphi}{\approx}0.8-1.0$ and vary by factors as large as $10{\times}$. A more complete picture of Cepheid outer atmospheres is accomplished when the HST/COS results are combined with X-ray observations that we have obtained of the same stars with XMM-Newton & Chandra. The Cepheids detected to date have X-ray luminosities of log $L_X{\approx}28.5-29.1$ ergs/sec, and plasma temperatures in the $2-8{\times}106$ K range. Given the phase-timing of the enhanced emissions, the most plausible explanation is the formation of a pulsation-induced shocks that excite (and heat) the atmospheric plasmas surrounding the photosphere. A pulsation-driven ${\alpha}^2$ equivalent dynamo mechanism is also a viable and interesting alternative. However, the tight phase-space of enhanced emission (peaking near 0.8-1.0 ${\varphi}$) favor the shock heating mechanism hypothesis.

Sensitivity of a Hyperactivated Ras Mutant in Response to Hydrogen Peroxide, Menadione and Paraquat

  • We have explored the impact of altering the Ras-cAMP pathway on cell survival upon oxidative exposures. A hyperactivated Ras mutant of Saccharomyces cerevisiae, intrinsically more sensitive to heat shock than the wild type, was investigated with regard to oxidative stress. In this paper we report that the response of iral, ira2-deleted mutant (IR2.53) to an oxidant, such as hydrogen peroxide (H2O2) or menadione is more sensitive than that of the wild type. IR2.53 showed a dramatic decrease in survival rate when challenged with 0.1 mM H2O2 for 30 min. The greater sensitivity of IR2.53 was also noticed with treatment of 0.01 mM menadione. Prior to oxidative stresses by these oxidants, both the wild type and the mutant were preconditioned with a mild heat shock (37 ℃, 30 min), resulting in improved survivals against oxidative stresses. Rescue of IR2.53 from menadione stress by heat pretreatment was more clearly demonstrated than that from H2O2 treatment. On the other hand, no significant difference was observed between the wild type and the IR2.53 mutant in their survival rates upon paraquat treatments. These findings imply that the mechanism by which H2O2 and menadione put forth their oxidative effects may be closely associated with the cAMP-Ras pathway whereas that of paraquat is independent of the Ras pathway. Finally, the level of glutathione (GSH) was measured enzymatically as an indicator of antioxidation and compared with the survival rate. Taken all these together, this study provides an insight into a mechanism of the Ras pathway regulated by several oxidants and suggests that the Ras pathway plays a crucial role in protection of cell damage following oxidative stress.

VCM 액추에이터의 전자기력을 이용한 HDD 래치 설계 (A HDD Latch Design Using Electro-magnetic Force of VCM Actuators)

  • Various types of latch designs for hard disk drives using load/unload mechanism have been introduced to protect undesired release motions of a voice coil motor(VCM) actuator from sudden disturbances. Recently, various inertia-type latches have been widely used because locking performance is better than that of other types of latch. However there has been a limit in the inertia type in order to guarantee perfect latch and unlatch operations because of changes in latch/unlatch conditions due to mechanical tolerance and temperature-dependent friction. In this paper, a reliable and robust magnetic latch mechanism is proposed through only simple modifications of coil and yoke shapes in order to overcome the mechanical limit of current inertia-type latches. This new magnetic latch does not have only a simple structure but it also ensures reliable operations and anti-shock performance. The operating mechanism of the proposed latch is theoretically analyzed and optimally designed using an electromagnetic simulation.

PFC2D에서의 발파에 의한 파괴 메커니즘의 수치적 모델링 (Numerical Simulation of Fracture Mechanism by Blasting using PFC2D)

  • 발파에 의한 암반의 손상이나 파쇄는 폭약의 폭굉 과정에서 발생하는 충격파와 가스팽창의 영향에 의해 야기된다. 발파에 의한 파괴 메커니즘을 완전히 이해하기 위해서는 두 메커니즘을 같이 연구해야한다. 본 연구에서는 개별 요소법에 기초한 수치해석 프로그램인 PFC2D를 이용하여 발파공 벽면에 작용하는 폭굉압과 가스압을 동시에 모델링 할 수 있고 이에 따른 암반 내 균열 발생을 확인할 수 있는 알고리즘을 개발하였다. 또한 시멘트-모르타르 블록에서의 모형 발파시험을 수치해석을 수행함으로써, 개발된 알고리즘을 검증하였다.

초기 초신성 잔해의 비열적 전파복사 : 약한 자기장 근사 (NONTHERMAL RADIO EMISSION FROM SNR IN THE PRE-SEDOV STAGE OF EVOLUTION : WEAK MAGNETIC APPROXIMATION)

  • It has been recognized that the morphologies of the SNRs from the radio observation are "barrel shaped". To interpret the mechanism of the radiation and the physical state of the environments, we have analytically calculated the dynamical structure of the interacting region in the case where the ejectum has a steep power-law density profile($\rho{\sim}r^{-n}$) and the ambient medium has a shallow power-law density profile($\rho{\sim}r^{-s}$), assuming that the cosmic rays are isotropically accelerated in the shock wave and the magnetic fields are very weak. The calculated synchrotron radio maps show that the emission from the equator is intense and the emissions from the central and polar regions are less intense. Also the thicknesses of the shell are strongly dependent on s and weakly on n. The azimuthal intensity ratio $\alpha$ increases as the efficiency of the cosmic ray acceleration increases and s decreases. We compared the results with the morphology of the SNR A. D. 1006(type I SNR). It does agree with the case of s = 0, w = 0.3 - 0.5. This value for w is consistent with the results by Eichler(1979). It provides us the evidence of the cosmic ray acceleration in the shock wave.
