Antidepressant Effects of JG02 on Chronic Restraint Stress Animal Model (만성구속스트레스 동물모델에 대한 JG02의 항우울 효과)

  • Objectives: As a general emotion, everyone can temporarily experience depression, but depressive disorder is a disease that excessively affects daily life. Among the various causes of depression, the deficiency of monoamine-based neurotransmitters such as serotonin and epinephrine are considered significant. Thus, antidepressants that target monoamines are used frequently. However, side effects such as nausea, vomiting, insomnia, anxiety, and sexual dysfunction are observed. Thus, it is necessary to develop a new therapeutic agent with fewer side effects. In this study, we investigated the antidepressant effect of JG02, used to treat depression by normalizing the flow of qi (氣) in Korean medicine. Methods: C57BL/6 mice were selected and randomly divided into six groups: normal, control, amitriptyline, and JG02 (50, 125, 250 mg/kg), respectively. Except for normal, depression was induced by applying restraint stress at the same time for six hours daily for 14 consecutive days. Saline, amitriptyline or JG02 samples were orally administered two hours before applying the stress. After that, a forced swimming test and an open field test were performed. Additionally, serum corticosterone, serotonin mRNA, BDNF mRNA, and protein in the hippocampal region were measured and compared. Results: JG02 decreased immobility time rate in the FST and increased the zone transition number and travel distance in the OFT. Also, JG02 inhibited the release of serum corticosterone, and increased serotonin, BDNF gene expression, and BDNF protein in the hippocampus. Conclusions: In this study, JG02 showed significant antidepressant effects on the chronic restraint stress mice model. When further research is performed based on JG02, the development of a new antidepressant is considered highly possible.

In Vivo Artificial Parthenogenetic Treatments on Live Silkworm Moth, Bombyx mori Can Induce Higher Parthenogenesis (살아있는 누에 나방(Bombyx mori)에 대한 인공적 단위 발생 처리의 단위 발생란 유발 촉진 효과)

  • The silkworm performs sexual reproduction for the production of its healthy offsprings from generations to generations. Parthenogenesis in the silkworm, Bombyx mori acquires immense use in the development of outstanding homozygouse lines with higher viability, hybrid vigour, combining ability and less phenotypic variability, and it can serve as a powerful tool in controlling sex of the offsprings as well as a useful tool in selection of breeding schemes. However, naturally occuring parthenogenesis in silkworm could not be found so far. Fortunately, artificial induction of parthenogenesis is possible in silkworm. So, it is very important to find out novel methods for induction of parthenogenesis. We investigated to attempt to get a novel parthenogenetic method. Accordingly, parthenogenetic studies on between unfertilized in vivo ovarian eggs of live silkworm moth(novel) and unfertilized in vitro ovarian eggs(conventional) taken out from live silkworm moth were investigated by hot water ($46^{\circ}C$), hot air ($46^{\circ}C$) and low temperature ($0^{\circ}C$ and $-20^{\circ}C$) treatments. The best ratio of parthenogenetic eggs was obtained with in vivo ovarian eggs of live silkworm moth rather than with in vitro ovarian eggs taken out from live silkworm moth in all the treatments. The optimum exposure time absolutely depended upon the temperatures of treatments and the forms of in vivo or in vitro ovarian eggs. From these results, we expect that in vivo artificial parthenogenetic treatments on live silkworm moth will be useful for the higher induction of parthenogenesis in the silkworm, B. mori.

Development of PCR-based markers specific to Solanum brevicaule by using the complete chloroplast genome sequences of Solanum species (엽록체 전장유전체 비교를 통한 PCR 기반의 Solanum brevicaule 특이적 분자마커 개발)

  • Solanum brevicaule is one of the tuber-bearing wild Solanum species. Because of its resistance to several important pathogens infecting potatoes during cultivation, it can be used for potato breeding. However, the fact that S. brevicaule used in this study has an EBN value of two causes the sexual reproduction barriers between the species and cultivated potatoes. In this study, specific markers for discriminating S. brevicaule from other Solanum species were developed on the basis of the results of sequence alignments with the whole chloroplast genomes of S. brevicaule and seven other Solanum species. The chloroplast genome of S. brevicaule was completed by next-generation sequencing technology described in other recent studies. The total sequence length of the chloroplast genome of S. brevicaule is 155,531 bp. Its structure and gene composition are similar to those of other Solanum species. Phylogenetic analysis revealed that S. brevicaule was closely grouped with other Solanum species. BLASTN search showed that its genome sequence had 99.99% and 99.89% identity with those of S. spegazzinii (MH021562) and S. kurtzianum (MH021495), respectively. Sequence alignment identified 27 SNPs that were specific to S. brevicaule. Thus, three PCR-based CAPS markers specific to S. brevicaule were developed on the basis of these SNPs. This study will facilitate in further studies on evolutionary and breeding aspects in Solanum species.

PCR-based markers to select plastid genotypes of Solanum acaule (Solanum acaule 색소체 유전자형 선발을 위한 특이적 분자마커 개발)

  • The tetraploid Solanum acaule is a wild potato species from Bolivia widely used for potato breeding because of its diverse attractive traits, including resistance to frost, late blight, potato virus X, potato virus Y, potato leafroll virus, potato spindle tuber viroid, and cyst nematode. However, the introgression of useful traits into cultivated potatoes via crossing has been limited by differences in endosperm balance number between species. Somatic fusion could be used to overcome sexual reproduction barriers and the development of molecular markers is essential to select proper fusion products. The chloroplast genome of S. acaule was sequenced using next-generation sequencing technology and specific markers for S. acaule were developed by comparing the obtained sequence with those of seven other Solanum species. The total length of the chloroplast genome is 155,570 bp, and 158 genes were annotated. Structure and gene content were very similar to other Solanum species and maximum likelihood phylogenetic analysis with 12 other species belonging to the Solanaceae family revealed that S. acaule is very closely related to other Solanum species. Sequence alignment with the chloroplast genome of seven other Solanum species revealed four InDels and 79 SNPs specific to S. acaule. Based on these InDel and SNP regions, one SCAR marker and one CAPS marker were developed to discriminate S. acaule from other Solanum species. These results will aid in exploring evolutionary aspects of Solanum species and accelerating potato breeding using S. acaule.

  • With the growth of Chinese manufacturing industry and strengthening of manufacturing industry in advanced countries, the competitiveness of Korean manufacturing industry has become weak. Accordingly, it is time when energy for new growth is required very strongly. This research is the basic research to construct a cognitive and emotional information system to support sound design in manufacturing industry, and aims to develop AUI (Auditory User Interface) guideline. To fulfill research aims, this research conducted interviews to persons belonging to the manufacturing industry, and analyzed contexts and major characteristics related with AUI design. In addition, this research conducted survey on 269 persons on user preference on representative function of products. By analyzing survey data, this research examined the most suitable melody method for each sound, and difference in user preference between men and women. Based on beep sound, it performed analysis focusing on the number of sounds, melody types, and code. It also extracted AUI guidelines usable when users of the product into male and female. This research has significance in the sense that it built the basic guide data which can support AUI design in manufacturing industry.

Utilization of Hypothalamic Hormones for Maturational Induction in Seawater Chum Salmon, Oncorhynchus keta (해면연어의 성숙을 유도하기 위한 시상하부호르몬의 이용)

  • Freshwater-adapted maturing chum salmon (Oncorhynchus keta) involuntarily captured by stationary nets in Yang-yang seashore areas were transferred to freshwater in an outdoor raceway tank at Yeongdong Inland Fisheries Research Institute, NFRDI, Yang-yang, Gangwon, Korea and kept over 1 day until the start of the experiments. The freshwater-adapted females were single-injected intraperitoneally with gonadotropin-releasing hormone analogue, (GnRH-a: $70\;{\mu}g/kg$ body weight, BW) alone or combined with a dopamine receptor antagonist, pimozide($700\;{\mu}g/kg$ BW). Although gonadosomatic indices [GSI, (gonad weight/BW)${\times}100$] did not show significant changes in both 2004 and 2005, GSI of GnRH-a-injected fish during the 2005 trial slightly increased on the 5th and 7th days post-injection compared to those of vehicle treated fish. Hepatosomatic indices [HSI, (liver weight/BW)${\times}100$] of fish injected with GnRH-a alone and combined with pimozide decreased significantly on the 7th day post-injection in 2004(P<0.05). In 2005 trials, HSI was significantly reduced in GnRH-a treated fish on the 7th day post-injection (P<0.05). Pimozide-injected fish showed a pattern with increase of GSI and decrease of HSI, without significant differences. Taken together, these results suggest that at least in part hypothalamic hormones and dopamine receptor antagonist may induce sexual maturation in freshwater-adapted maturing chum salmon. It remains to evaluate these preliminary results by further researches.

  • With the development of cyberspace and its increasing interaction with our daily lives, cybercrime has been steadily increasing in recent years and has become more prominent as a serious social problem. Notably, the "four major malicious cybercrimes" - cyber fraud, cyber financial crime, cyber sexual violence, and cyber gambling - have drawn significant attention. In order to minimize the damage of cybercrime, it's crucial to delve into the specifics of each crime and develop targeted prevention and intervention strategies. Yet, most existing research relies on indirect data sources like statistics, victim testimonials, and public opinion. This study seeks to uncover the characteristics and factors of cybercrime by directly interviewing suspects involved in 'copyright infringement', 'gambling' related to illicit online content, and 'pornography crime'. Through coding analysis and text mining, the study aims to offer a more in-depth understanding of cybercrime dynamics. Furthermore, by suggesting preventative and remedial measures, the research aims to equip policymakers with vital information to reduce the repercussions of this escalating digital threat.

Development of PCR-based markers for selecting plastid genotypes of Solanum hjertingii (Solanum hjertingii 색소체 유전자형 선발을 위한 PCR 기반 분자마커 개발)

  • The tetraploid Solanum hjertingii, a wild tuber-bearing species from Mexico is a relative of potato, S. tuberosum. The species has been identified as a potential source of resistance to blackening for potato breeding. It does not exhibit enzymatic browning nor blackspot which are physiological disorders. However, due to their sexual incompatibility, somatic hybridization between S. hjertingii and S. tuberosum must be used to introduce various traits from this wild species into potato. After somatic hybridization, molecular markers are essential for selecting fusion products. In this study, the chloroplast genome of S. hjertingii was sequenced by next-generation sequencing technology and compared with those of other Solanum species to develop specific markers for S. hjertingii. The chloroplast genome has a total sequence length of 155,545 bp, and its size, gene content, order and orientation are similar to those of the other Solanum species. Phylogenic analysis including 15 other Solanaceae species grouped S. hjertingii with S. demissum, S. hougasii, and S. stoloniferum. After detailed comparisons of the chloroplast genome sequence with eight other Solanum species, we identified one InDel and seven SNPs specific to S. hjertingii. Based on these, five PCR-based markers were developed for discriminating S. hjertingii from other Solanum species. The results obtained in this study will aid in exploring the evolutionary aspects of Solanum species and accelerating breeding using S. hjertingii.

Factors Affecting Female College Students' Reproductive Health Information Seeking Behaviors on the Internet (여대생의 인터넷 생식건강정보 탐색에 영향을 미치는 요인 연구)

  • The purpose of this study is to identify the factors affecting female college students' behaviors in seeking reproductive health information on the Internet and to explore the relationships among these factors. Based on the Health Belief Model(HBM) and the Theory of Planned Behavior(TPB), perceived sensitivity, perceived severity, perceived benefit, perceived barriers, subjective norms, perceived behavioral control, and affective evaluation were defined as key factors, and the study was designed accordingly. An online survey was distributed to female college students in Seoul through the university student's online community, 'Everytime.' The results showed that the intention of female college students to seek reproductive health information via the Internet was associated with higher perceived sensitivity, perceived benefit, and subjective norms, and lower perceived barriers. There were statistically significant differences between groups in terms of sexual experiences, experience with reproductive system disorders, and the level of health interest. We believe that this research outcome will contribute to assessing the level of awareness regarding reproductive health among female college students, thereby aiding in the development of online health information literacy education or related service programs by university libraries, health institutions, and similar entities targeting female college students.

Reproductive Ecology and Spawning Behavior of the Natural Monumental and Endangered Species, Cobitis choii (Pisces: Cobitidae) in Geumgang (River), Korea (천연기념물이며 멸종위기종인 미호종개 Cobitis choii (Pisces: Cobitidae)의 번식생태 및 산란행동)

  • A survey was conducted in 2011 to investigate the reproductive ecology and spawning behavior of the natural monumental and endangered species, Cobitis choii. C. choii exhibits sexual dimorphism with the development of lamina circularis on the second pectoral fin ray in males at one year old, facilitating easy differentiation between males and females. A total of 1,288 females and 881 males were collected from January to December, resulting in a sex ratio of 1 : 0.68. The spawning season was estimated to be from late June to early July (spawning peak from late June, with water temperatures ranging from 22 to 25℃), coinciding with a sharp decrease in the condition index. Gonadal maturity was observed with maturity indices of 17.5±2.11% for females and 2.9±0.46% for males, with fecundity of 2,444±838 eggs and egg size of 0.75±0.05 mm. Spawning behavior was induced by hormone injection (Human Chorionic Gonadotropin, HCG) in laboratory conditions. Experimental results revealed that males and females paired in a 1 : 1 ratio before spawning. Males pressed the abdomen of females using their lamina circularis to induce spawning, followed by immediate fertilization. Spawning behavior was repeated 15 to 25 (average 20) times per pair of males and females, and 80 to 120 eggs were spawned per time, and the total amount of spawned eggs were 2,500±250 on average.
