Morphometric analysis of the inter-mastoid triangle for sex determination: Application of statistical shape analysis

  • Purpose: Sex determination can be done by morphological analysis of different parts of the body. The mastoid region, with its anatomical location at the skull base, is ideal for sex identification. Statistical shape analysis provides a simultaneous comparison of geometric information on different shapes in terms of size and shape features. This study aimed to investigate the geometric morphometry of the inter-mastoid triangle as a tool for sex determination in the Iranian population. Materials and Methods: The coordinates of 5 landmarks on the mastoid process on the 80 cone-beam computed tomographic images(from individuals aged 17-70 years, 52.5% female) were registered and digitalized. The Cartesian x-y coordinates were acquired for all landmarks, and the shape information was extracted from the principal component scores of generalized Procrustes fit. The t-test was used to compare centroid size. Cross-validated discriminant analysis was used for sex determination. The significance level for all tests was set at 0.05. Results: There was a significant difference in the mastoid size and shape between males and females(P<0.05). The first 2 components of the Procrustes shape coordinates explained 91.3% of the shape variation between the sexes. The accuracy of the discriminant model for sex determination was 88.8%. Conclusion: The application of morphometric geometric techniques will significantly impact forensic studies by providing a comprehensive analysis of differences in biological forms. The results demonstrated that statistical shape analysis can be used as a powerful tool for sex determination based on a morphometric analysis of the inter-mastoid triangle.

SRY와 Amelogenin gene의 증폭에 의한 말의 성(sex) 결정 예 (A case of sex determination by amplification of SRY and Amelogenin gene in horse)

  • The objective of present study was to ascertain sex determination for individual identification, parentage control, and sex chromosome anomalies in horse. PCR amplification products of the equine sex determining region of the Y chromosome gene (SRY) and amelogenin gene (AMEL) were detected by using agarose gel electrophoresis. A normal sire and foal II showed 1 SRY band (430 bp) and 3 AMEL (AMELX, AMELY, and AMELX/Y) band, 175 bp, 160 bp, 190 bp, respectively, and a normal dam and foal I showed a single AMELX band (175 bp). These results enables a quick diagnosis for sex determination prior to cytogenetic analysis.

  • Silver barb (Barbonymus gonionotus) is among the most economically important freshwater fish species in Thailand. It ranks fourth in economic value and third in production weight for fisheries and culture in Thailand. An XX/XY sex-determination system based on gynogenesis was previously reported for this fish. In this study, the molecular basis underlying the sex-determination system was further investigated. Genome-wide single-nucleotide polymorphism data were generated for 32 captive-bred silver barb individuals, previously scored by phenotypic sex, to identify sex-linked regions associated with sex determination. Sixty-three male-linked loci, indicating putative XY chromosomes, were identified. Male-specific loci were not observed, which indicates that the putative Y chromosome is young and the sex determination region is cryptic. A homology search revealed that most male-linked loci were homologous to the Mariner/Tc1 and Gypsy transposable elements and are probably the remnants of an initial accumulation of repeats on the Y chromosome from the early stages of sex chromosome differentiation. This research provides convincing insights into the mechanism of sex determination and reveals the potential sex determination regions in silver barb. The study provides the basic data necessary for increasing the commercial value of silver barbs through genetic improvements.

  • 사람에게서 유래된 DNA시료의 X,Y 특이의 alphoid gene을 PCR법으로 분석하면 성별을 확인할 수 가 있다. PCR법으로 alphoid gene을 분석한바 매우 예민도가 높아 genomic DNA 약 60pg까지 성별을 분석할 수 있었다. 그리고 성별이 혼합되어 있는 DNA에서 female DNA의 1/10비 까지는 male DNA를 분석할 수 있었다. 따라서 이 결과는 혼합된 DNA에서 X,Y 특이의 alphoid gene을 분석하는데 기준으로 활용할 수가 있다.

  • The effects of rearing temperature on the sex determination of zebrafish were examined to address the questions, "Is it possible to control phenotypic sex artificially; and what genes may be involved in that process\ulcorner". At higher temperature(${32-34}^{\circ}C vs 28.5^{\circ}$C), a dramatic shift of the sex ratio(female 1 : male 9) was found. with DD-PCR cloning, several candidate genes were cloned and analyzed. Further analysis should help to understand sex-determining mechanisms involved in the artificial induction.induction.

사람치아에서 성별감정시 RPS4Y 유전자의 유용성 (Usefulness of RPS4Y Gene on Sex Determination in Human Teeth)

  • 사람 Y염색체상에 존재하는 RPS4Y(Ribosomal Protein S4Y) 유전자는 성별감정시 유용한 유전자로 규명되어 유전질환의 조기 발견이나 예방 및 태아의 성별판정 등에 응용되고 있다. 신원이 불분명한 사체에서 성별감정시, 기존의 성별감정에 이용되고 있는 다른 유전자와 함께 RPS4Y 유전자를 검색함으로써 성별감정의 신뢰도를 높힐 수 있을 것으로 사료된다. 또한 사체의 손상이 심할 때 유전자를 이용한 개인식별은 제한을 받게 된다. 이때 치아는 인체의 기관 중 가장 견고한 구조로 구성되어 있어 외부 환경에 대한 물리적, 화학적 저항성이 높아 법의치과학적 개인식별에 널리 이용되므로, 본 연구에서는 사람 치아에서 중합효소연쇄반응법을 이용한 RPS4Y유전자를 검출하여 법의학적 성별감정에 응용하고자 하였다. 남녀 각각 10개의 치아에서 치수와 상아질을 분리한 후 DNA를 추출하여 중합효소연쇄반응을 시행하였다. RPS4Y 유전자를 검출한 결과, 남자에게서만 특이적으로 유전자가 검출되었으며, 이는 사람 치아에서 RPS4Y 유전자를 이용한 성별감정이 법의학적 개인식별의 성별감정 실무에 있어서 다른 유전자와 함께 유용하게 사용될 수 있을 것으로 사료된다.

  • The human genomic deoxyribonucleic acid(DNA) was extracted from the pulp, dentin of 22 teeth by clelex, phenol methods. Samples of the tooth-derived DNA were amplified by polymerase chain reaction(PCR), electrophosed for sex determination by detection of X-Y homologus amelogenin gene and D1S80 locus detection The following results have been achieved. 1. Chelex and phenol method are effective to sex determination in the pulp and dentin 2. Chelex method is not suitable for detection of D1S80 locus. 3. Concentration and purity of DNA for teeth using chelex method is lower than using phenol method. From the above investigation, chelex method is simple, rapid for sex determination, but it is not suitable for detection of VNTRs.

Identification of Sperm mRNA Biomarkers Associated with Sex-Determination in Korean Native Cows

  • This study was conducted to analyze the specific genes associated with sex-determination in Korean native cow. The highly organized spermatogenesis requires accurate spatial and temporal regulation of gene expression, which is governed by transcriptional, post-transcriptional, and epigenetic processes. Recently, farmers have been interested in determining the sexual identity of the calves in their farm. We analyzed the sperm of Korean native and Holstein cows, which were supplied from Hanwoo Improvement Center. We evaluated sperm motility and expression of sperm-specific genes after treating semen with both male- and female reagents. Sperm motility in Korean native cows decreased by approximately 10% in the first 30 minutes after treatment with sex-determination reagent. However, sperm motility of Holstein cows decreased to 60-70% after 15 minutes and to 20-30% after 30 minutes. We selected six specific genes expressing in the spermatozoa to analysis the gene expression level. The Real-time PCR results suggest that the selected genes (Gimap4, Tmeff1, Rac2, Abi2, Rac1, and Clu) were highly expressed in the group treated with the male reagent compared to the group treated the female reagent and to the untreated-group (control). In the present study, we suggest that the selected genes play a pivotal role in sex-determination.

Sex determination from lateral cephalometric radiographs using an automated deep learning convolutional neural network

  • Purpose: Despite the proliferation of numerous morphometric and anthropometric methods for sex identification based on linear, angular, and regional measurements of various parts of the body, these methods are subject to error due to the observer's knowledge and expertise. This study aimed to explore the possibility of automated sex determination using convolutional neural networks(CNNs) based on lateral cephalometric radiographs. Materials and Methods: Lateral cephalometric radiographs of 1,476 Iranian subjects (794 women and 682 men) from 18 to 49 years of age were included. Lateral cephalometric radiographs were considered as a network input and output layer including 2 classes(male and female). Eighty percent of the data was used as a training set and the rest as a test set. Hyperparameter tuning of each network was done after preprocessing and data augmentation steps. The predictive performance of different architectures (DenseNet, ResNet, and VGG) was evaluated based on their accuracy in test sets. Results: The CNN based on the DenseNet121 architecture, with an overall accuracy of 90%, had the best predictive power in sex determination. The prediction accuracy of this model was almost equal for men and women. Furthermore, with all architectures, the use of transfer learning improved predictive performance. Conclusion: The results confirmed that a CNN could predict a person's sex with high accuracy. This prediction was independent of human bias because feature extraction was done automatically. However, for more accurate sex determination on a wider scale, further studies with larger sample sizes are desirable.

Sex determination by radiographic localization of the inferior alveolar canal using cone-beam computed tomography in an Egyptian population

  • Purpose: The purpose of this study was to evaluate possible differences in the location of the inferior alveolar canal in male and female Egyptians. Materials and Methods: This cross-sectional retrospective study involved the evaluation of 210 CBCT scans of Egyptian individuals (18-70 years old). The inferior alveolar canal was localized by measuring 8 linear dimensions: 2 for the vertical localization of the mental foramen (superior and inferior to the mental foramen), 4 at the first molar bifurcation for the vertical and horizontal localization of the inferior alveolar canal (superior, inferior, buccal, and lingual to the inferior alveolar canal), and 2 for the horizontal localization of the mandibular foramen (anterior and posterior to the mandibular foramen). The measurements were statistically analyzed via comparative analysis, stepwise logistic regression, and receiver operating characteristic (ROC) curve analysis. Results: Six of the 8 measured distances differed to a statistically significant extent between the sexes. Regression analysis suggested a logistic function with a concordance index of 84%. The diagnostic accuracy capabilities of the linear measurements as sex predictors were calculated using ROC analysis, and the 6 best predictors for sex determination were selected and ranked from highest to lowest predictive power. Moreover, combining these 6 predictors increased the predictive power to 84%. Conclusion: The location of the inferior alveolar canal in the Egyptian population varies significantly by sex; accordingly, this anatomic landmark could be used as a reliable indicator of sexual dimorphism.
