Purpose: The arterial base deficit and the serum lactate level are widely recognized indicators of injury severity, adequacy of resuscitation and outcome. The purpose of this study is to evaluate the usefulness of the arterial base deficit as an injury-severity indicator in multiple-trauma patients with head injuries. Methods: Data were retrospectively collected from trauma patients over 18 years of age who had been admitted at the emergency center between October 2005 and July 2006. The patients were divided into head-injury and non-head-injury groups. These patients were then sub-divided into minor ($$ISS{\leq_-}15$$)-injury and major ($$ISS{\geq_-}16$$)-injury groups according to their injury severity scores (ISS). We analyzed the differences in the base deficits and the serum lactate levels between the major- and the minor-injury sub-groups in both the head-injury and the non-head-injury groups. Results: In the non-head-injury group, we found statistically significant differences in the arterial base deficit between the major-injury and the minor-injury sub-groups($-6.86{\pm}2.40mmol/L$ vs. $-1.37{\pm}0.73mmol/L$, p=0.010). In the head-injury group, no significant differences were noted between the two sub-groups($-2.50{\pm}1.28mmol/L$ vs. $-1.51{\pm}0.74mmol/L$, p=0.897). Moreover, the differences in arterial base deficit between the major-injury and the minor-injury sub-groups were not significant both for either single-head-trauma or multiple-head-trauma patients (p=0.643 vs. p=0.832). Conclusion: We conclude that neither the arterial base deficit nor the serum lactate level can be used to predict injury severity in multiple-trauma patients with head injuries.
Objective: In this study we aimed to evaluate the effect of dietary live yeast supplementation on ruminal pH pattern, fermentation characteristics and associated bacteria in beef cattle. Methods: This work comprised of in vitro and in vivo experiments. In vitro fermentation was conducted by incubating 0%, 0.05%, 0.075%, 0.1%, 0.125%, and 0.15% active dried yeast (Saccharomyces cerevisiae, ADY) with total mixed ration substrate to determine its dose effect. According to in vitro results, 0.1% ADY inclusion level was assigned in in vivo study for continuously monitoring ruminal fermentation characteristics and microbes. Six ruminally cannulated steers were randomly assigned to 2 treatments (Control and ADY supplementation) as two-period crossover design (30-day). Blood samples were harvested before-feeding and rumen fluid was sampled at 0, 3, 6, 9, and 12 h post-feeding on 30 d. Results: After 24 h in vitro fermentation, pH and gas production were increased at 0.1% ADY where ammonia nitrogen and microbial crude protein also displayed lowest and peak values, respectively. Acetate, butyrate and total volatile fatty acids concentrations heightened with increasing ADY doses and plateaued at high levels, while acetate to propionate ratio was decreased accordingly. In in vivo study, ruminal pH was increased with ADY supplementation that also elevated acetate and propionate. Conversely, ADY reduced lactate level by dampening Streptococcus bovis and inducing greater Selenomonas ruminantium and Megasphaera elsdenii populations involved in lactate utilization. The serum urea nitrogen decreased, whereas glucose, albumin and total protein concentrations were increased with ADY supplementation. Conclusion: The results demonstrated dietary ADY improved ruminal fermentation dose-dependently. The ruminal lactate reduction through modification of lactate metabolic bacteria could be an important reason for rumen pH stabilization induced by ADY. ADY supplementation offered a complementary probiotics strategy in improving gluconeogenesis and nitrogen metabolism of beef cattle, potentially resulted from optimized rumen pH and fermentation.

BACKGROUND/OBJECTIVES: The effectiveness of natural compounds in improving athletic ability has attracted attention in both sports and research. Gynostemma pentaphyllum (Thunb.) leaves are used to make traditional herbal medicines in Asia. The active components of G. pentaphyllum, dammarane saponins, or gypenosides, possess a range of biological activities. On the other hand, the anti-fatigue effects from G. pentaphyllum extract (GPE) and its effective compound, gypenoside L (GL), remain to be determined. MATERIALS/METHODS: This study examined the effects of GPE on fatigue and exercise performance in ICR mice. GPE was administered orally to mice for 6 weeks, with or without treadmill training. The biochemical analysis in serum, glycogen content, mRNA, and protein expressions of the liver and muscle were analyzed. RESULTS: The ExGPE (exercise with 300 mg/kg body weight/day of GPE) mice decreased the fat mass percentage significantly compared to the ExC mice, while the ExGPE showed the greatest lean mass percentage compared to the ExC group. The administration of GPE improved the exercise endurance and capacity in treadmill-trained mice, increased glucose and triglycerides, and decreased the serum creatine kinase and lactate levels after intensive exercise. The muscle glycogen levels were higher in the ExGPE group than the ExC group. GPE increased the level of mitochondrial biogenesis by enhancing the phosphorylation of peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) protein and the mRNA expression of nuclear respiratory factor 1, mitochondrial DNA, peroxisome proliferator-activated receptor-δ, superoxide dismutase 2, and by decreasing the lactate dehydrogenase B level in the soleus muscle (SOL). GPE also improved PGC-1α activation in the SOL significantly through AMPK/p38 phosphorylation. CONCLUSIONS: These results showed that GPE supplementation enhances exercise performance and has anti-fatigue activity. In addition, the underlying molecular mechanism was elucidated. Therefore, GPE is a promising candidate for developing functional foods and enhancing the exercise capacity and anti-fatigue activity.
After pretreatment with ginseng followed by induction of acute intoxication of alcohol, the activities of alcohol dehydrogenase (ADH), microsomal ethanol-oxidizing system (MEOS) and aldehyde dehydrogenase(Ald DH) increased respectively compared to the groups treated with alcohol alone. In case that ginseng was given to rats fed with 5% alcohol instead of water for 60 days, the activities of ADH and MEOS increased compared to the groups treated. On the contrary, the activity of Ald DH in mitochondrial fraction decreased to an extent of about 35% in chronic alcoholism, but after pretreatment of ginseng the activity was restored to the control level. On the other hand, the catalase activity was not significantly affected by either treatment. Ginseng butanol fraction significantly increased the serum isocitrate dehydrogenase activity which is inhibited by alcohol-treated in rat. Alcohol-induced lactate dehydrogenase activity was decreased to control level in liver by ginseng treatment. And the serum level of lactic acid also decreased by ginseng treatment in alcohol-intoxicated rat. Ginseng butanol fraction markedly decreased the xanthine oxidase activity in the ethanol-treated rat liver. It was also observed that ginseng reduced the blood concentration of uric acid on experimentally reduced hyperuricemia by alcohol treatment. Uricase activity was not affected by either treatment. Ginseng butanol fraction decreased the hepatic aniline hydroxylase activity which was induced by alcohol-treated rat. These results suggest that the treatment with ginseng can be promoted the recovery from alcohol intoxication and some therapeutic effect on alcoholinduced metabolic disease.
To investigate the effects of yellow soybean powder on serum lipid level and enzyme activities, male Sprague-Dawley(SD) rats were given hyperlipidemic diets for 6 weeks. Rats(n=8) were divided into three groups consisting the normal group(N), hyperlipidemic diets group(C) and hyperlipidemic diets and yellow soybean powder group(YSP). YSP group lowered triglyceride, total lipid, phospholpid, ester-cholesterol and free-cholesterl than control group. YSP group lowered total-cholesterol, VLDL-cholesterol and LDL-cholesterol but increased HDL-cholesterol of the serum compared to the control group. Athergenic Index(AI) and LHR(LDL-cholesterol/HDL-cholesterol ratio) of YSP group was decreased by control group (p<0.05). On the other hand, glutamic oxaloacetate transaminase(GOT), glutamic pyruvate transaminase (GTP), lactate dehydrogenase(LDHase) and alkaline phosphatase(ALP) enzymes activities were decreased by hyperlipidemic diets group (C). These results suggest that yellow soybean powder fed a lipid lowering effect in serum of rats.
The present study was undertaken to find a protective effect of Coptis Rhizoma Extract on liver injury induced by benzo(a)pyrene in rats. This crude drug significantly suppressed the increase of several biochemical parameters such as transaminase, lactate dehydrogenase and alkaline phosphatase in serum and liver and total cholesterol. But there were little changes in elevated serum phospholipid level. The results suggested that Coptis Rhizoma prevent bezo(a)pyrene-induced hepatic injury.
This study was performed to investigate the effect of chongkukjang intake on lipid metabolism and liver function in ethanol consumed rats. Twenty one Sprague-Dawley male rats aging 4 weeks old were used as experimental animals, which were divided into three dietary groups; casein diet(CA), soybean diet(SB) and chongkukjang diet(CJ). Alcohol was consumed with water as 25%(v/v) ethanol solution. After 4 weeks of experimental period, rats were sacrificed to get blood and liver samples for analysis of lipids, lipid peroxides, antioxidative enzymes and biochemical indices of liver function. The mean body weight, food intake and liver index were not significantly different among three groups. Serum level of total lipid, triglyceride and total cholesterol of chongkukjang diet group was the lowest among three groups although the difference was not significant. HDL-cholesterol level was significantly(p<0.05) higher in chongkukjang diet group than that of casein diet group. LDL-cholesterol level of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. Liver TBARS of chongkukjang and soybean diet group was significantly(p<0.05) lower than that of casein diet group respectively. The superoxide dismutase activity of chongkukjang diet group was significantly(p<0.05) higher than that of casein diet group. Catalase activity was not significantly different among three groups. As indices of liver function, glutamic oxaloacetic transminase(GOT), glutamic pyruvic transminase(GPT), $\gamma$-glutamyl transpeptidase($\gamma$-GTP) and lactate dehydrogenase(LDH) were not significantly different among three groups. Serum alcohol concentration and activities of alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) were not significantly different among three groups. The chongkukjang diet seems to give a beneficial effect for improving lipid metabolism by increasing HDL-cholesterol level and SOD activity while reducing liver TBARS level. However, effect on liver function has to be investigated further.
In order to assess the correlation of the myocardial damage and the duration of cardiopulmonary bypass, measurement of creatine kinase [CK], lactate dehydrogenase [LDH], asparatate aminotransferase [AST], and MB band of CK [CK-MB] were carried out on the first, third, fifth, seventh, and ninth day in 44 patients following open heart surgery [POD 1,3,5,7,9]. And the patients were divided into three groups according to the duration of aortic cross clamp time [ACT]: Group I [ACT< 60 minutes. n=19], Group II [60 minutes < ACT< 90 minutes, n=7] and Group III[90 minutes > ACT, n=18]. 1. The leakage of CK in total patients increased to the highest level at POD 1, with rapid decrease and recovery at POD 7. The leakage of CK in Group III were greater than in Group I from POD 1 to POD 3 [P < 0.01]. The recovery time of CK level was shorter in Group I [POD 3] than in Group II and III [POD 7]. 2. The serum levels of LDH in total patients increased to the highest level at POD 1, with slow recovery until POD 9. The levels of LDH in Group III were higher than in Group I until POD 9 [P < 0.005]. The levels of LDH in Group I and II recovered but not in Group III. 3. The serum levels of AST in total patients increased to the highest level at POD 1, with rapid decrease and recovery at POD 7. The levels of AST in Group III were greater than in Group I from POD 1 to POD 5 [P < 0.05]. The recovery time of AST level was shorter in Group I and II [POD 5] than in Group III [POD 7]. 4. The positive cases for CK-MB in 36 patients were 22 [61.1 %] as a whole, 5[41.6%] in Group I, 4[57.1 %] in Group II, 13[76.4 %] in Group III at POD 1, and a case in each group at POD 3, and only a case in Group Ill at POD 5. It is concluded that the myocardial injury was closely related with the duration of cardiopulmonary bypass in open heart surgery.

This study was carried out to evaluate Effects of Cheongyoung-tang on CHT-Induced Brain Edema and Neurological Disturbance Symptom in Rats. we make a comparative study of the such parameters as neurological severity score(NSS) and changes in water content, lactate, glucose and hematological substance. 1. ΔNSS increased in CYT1 and CYT2 as compared with the control group, which showed no efficacy. 2. In water content of both hemispheres, CYT1 and CYT2 as compared with the control group have no difference. 3. Water content of the left hemisphere decreased in CYT1 and CYT2 as compared with the control group. 4. The lactate level in serum decreased in CYT1 and CYT2 as compared with the control group. 5. In hematological changes, RBC, PLT and MCHC increased in CYT1 and CYT2 as compared with the control group, but MCV decreased. According to these results, CYT has an effect on CHT-induced brain edema and neurological symptom.
Objectives : Bambusae Caulis in Liquamen (BCL) has been used for stamina fortification in Oriental Medicine for thousand years. The goal of the present study was to investigate the muscle antifatigue effect of BCL. Methods : The mice were divided into three groups. The control group was allowed to swim for 10 minutes without BCL infusion. For the preventive effect of BCL, another group referred to BCL-1 was fed with BCL an hour before the swimming exercise. BCL-2 group for the reversion effect was forced to swim for 10 minutes at first, then fed with BCL followed by half an hour rest and another 10 minute swimming exercise. All the serum samples collected by heart puncture were used for the measuring biochemical factors such as LDH, CPK, glucose, lactate, triglycerides and total cholesterol. Results : In the BCL-2 group, lactate and LDH were significantly reduced compared to the control. BCL infusion during the exercise was effective in metabolizing LDH, reducing the conversion ratio of pyruvate into lactate. In the BCL-1 group, it was not effective in antifatigue function. The level of glucose was significantly reduced in BCL-1 group compared to the control. When the BCL was infused before the exercise, it is assumed that glycogenolysis seen during the exercise was prevented. When the BCL was infused during the exercise, the triglycerides and total cholesterol were increased significantly compared to the control. BCL seems to cause the increase of lipolysis to utilize the fat as an energy source. Unlike other parameters, CPK did not show any changes from BCL infusion. Conclusions : Based on the above results, BCL is found to be involved in energy metabolism. Further studies are needed to find out the underlying mechanism of BCL for its effect on stamina fortification.
