The Effect of Dietary Fat Levels and Sources of Dietary Fiber on Serum and Liver Lipids of Rats (지방의 섭취량과 첨가된 섬유소의 종류가 흰쥐의 체내 지질 수준에 미치는 영향)

  • The effect of fiber sources on lipid metabolism was investigated in relation to the level of dietary fat. After rats were fed each diet for 4 weeks, all animals were sacrificed to collect the liver, and blood samples by heart puncture. Total lipid, total cholesterol, phospholipid and HDL- cholesterol were determined in serum. Total lipid and total cholesterol were also determined in liver. These results were compared with the histochemical appearances by the method of Oil red-O staining. Higher level of fat in the diet appeared to increase the total lipid levels in serum and more fat in the liver cells were shown from the high fat diet judging from the E.M. pictures. Phospholipid concentration in serum was greater in the rats of high fat groups than those of the control or non-fat groups. High fat level caused to decrease the HDL- cholesterol concentration in serum. Pectin decreased serum and liver lipids and the deposit of fat in the liver cells. HDL-cholesterol levels in serum was increased by pectin. Whereas the cellulose supplementation had no effect to reduce serum and liver total lipid levels, and cellulose did not decreased the deposit of fat in the liver cells. From these results, it is concluded that dietary fat levels led to changes in the lipid metabolism of rats and pectin may exhibit a lowering total lipid contents in serum and liver. Cellulose may slightly elevate serum and liver levels, or have no lowering effect.

Correlations between the Fatty Acid Composition of Serum Phospholipids and Blood Pressure

  • Studies of the relationship between the composition of serum fatty acids and blood pressure are complex and controversial. Fatty acids, important constituents of biological membranes, could potentially affect vasoreactivities including blood pressure. In this study the compositions of fatty acids in serum phospholipids were compared between three types of hypertensive subjects (men, pre-menopausal women, and post-menopausal women) and their respective nrmotensive controls. Serum lipids were extracted and phospholipids were separated by thin layer chromatography. The percentage of palmitic acid (16 : 0) in serum phospholipids was significantly higher and the percentage of stearic acid (18 : 0) was significantly lower in all three hypertensive groups, compared with their corresponding control groups. Only in the group of post-menopausal women, palmitic acid was closely associated wish increases in both systolic (SBP) and diastolic blood pressure (DBP), while stearic acid was associated with decreases in both SBP and DBP. The polyunsaturated fatty acids in serum phospholipids behaved differently from saturated fatty acids. The ratios of products / precursor fatty acids, such as $\sumLCPUFA\omega6/18 : 2\omega$6, 20 : 4$\omega$6/18 : 2$\omega$6, ∑LCPUFA$\omega$3/18 : 3$\omega$3 and 22 : 6$\omega$3/20 : 5$\omega$3, were all clearly associated with both SBP and DBP in hypertensive, post-menopausal women. Desaturation and elongation in fatty acid metabolism could affect the bioavailability of eicosanoid precursors. Changes in the constituent fatty acids of phospholipids and eicosanoid precursors may also influence fluidity, ionic transport, hormone receptors and enzyme activities in biological membranes. In conclusion, both systolic and diastolic blood pressure in post-menopausal women was positively associated with the level of palmitic acid, and negatively associated with the level of stearic acid, in serum phospholipids. The relationships between serum phospholipid-$\omega$6 and $\omega$3 series fatty acids and blood pressure in women, especially in post-menopausal women, require further investigation by taking into consideration hormonal status and eicosanoid metabolism. Funker study is needed to determine the value of dietary manipulation of fatty acid constituents of serum phospholipids, relating to hypertension in women.

A Clinical Study on the Association between Sasangin and Serum Leptin Level (사상인과 혈청 렙틴 농도의 연관성에 관한 임상연구)

  • 1. Objectives: Leptin is ob-gene originated hormone that is concerned with energy metabolism, obesity and asthma which features are supposed to related with Sasangin's 4 viscera physiological theory of Sasang constitutional medicine(SCM). This study aims to investigate whether serum leptin concentration is associated with Sasangin. 2. Methods: Two Way ANOVA analysis was used on the constitution and serum leptin concentration corresponding with BMI among 1054 cases drawn from the Constitutional Information Bank, which was a database with clinically confirmed constitution cases collected from 21 oriental medical institutes in Korea starting from November 2007 to May 2009. The measurement of serum leptin concentration is implemented through radioimmunoassay by request to SCL. 3. Results: Male subjects showed notable difference(p=0.033) in the comparison between constitution and serum leptin concentration corresponding to BMI levels. Tae-eumins showed the highest serum leptin concentration levels in the normal and overweight groups, followed by Soyangin and Soeumin. As the groups changed from overweight to obese, there was an interaction phenomenon in the order of serum leptin concentration levels(p=0.040). As a result, Soeumins showed the highest serum leptin concentration levels and Tae-eumins and Soyangins followed in place. Female subjects did not show statistically notable differences in comparison between constitution and serum leptin concentration corresponding to BMI levels(p=0.239). Serum leptin concentration levels among overweight and obese groups, resulted in the order of Soeumins, Tae-eumins, and Soyangins. But in the as groups changed from overweight to normal the order resulted in Tae-eumin, Soyangin, and Soeumin(p=0.660). 4. Conclusion: Due to a number of limitations in this study, the examination of the relation between constitution and leptin, a main hormone responsible for the digestion, appetite, and energy metabolism, resulted to be uncertain. But statistically notable differences were confirmed in male Tae-eumins with the consideration of BMI. Further examination of the relation between constitution and leptin is necessary through prospective studies with the restriction of confusion variables in the future.

The Effects of Natto Mucilage on the Components of Serum Lipid in Rats (낫도(Natto) 점질물이 흰쥐의 혈청 지질성분에 미치는 영향)

  • Natto is a Japanese traditional food made from whole soybeans by the fermentation of Bacillus natto. The purpose of this study was to investigate the effect of Natto mucilage on lipid metabolism in rats. The levels of total cholesterol, LDL-cholesterol and triglyceride in serum of rats fed Natto mucilage diets were lower than those in control rats, but serum HDL-cholesterol and phospholipid were higher than those in control rats. The results indicate that Natto mucilage may have more beneficial roles in lipid metablism because it decreased serum cholesterol and increased bilirubin excretion.

Hepatoprotective activity of methanol extracts of Glinus oppositifolius and Trianthema decandra against paracetamol induced liver damage

  • The plants, Trianthema decandra and Glinus oppositifolius are commonly used by tribal people in India for the treatment of liver diseases. Hepatoprotective activity of methanol extracts of Glinus oppositifolius and Trianthema decandra at the dose of 250 and 500 mg/kg body weight administered orally was evaluated against paracetamol induced liver damage in rats. Biochemical parameters such as serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase, serum alkaline phosphatase, bilirubin, total serum protein, lipid peroxide and glutathione content of the liver were estimated to determine liver function and metabolism. From the biochemical observations, it was concluded that methanol extracts of Glinus oppositifolius and Trianthema decandra significantly restored the altered biochemical parameters towards normal condition in paracetamol induced liver damage.

  • Alkaloidal fraction, a principle of ginseng extract was administered to rats orally in the amount of 2.5 mg per day per 100 gm body weight continuously for eight weeks to assess the lipid content of the serum and liver tissue. The findings can be summarized as follows: 1. Total cholesterol contents of serum and liver tissue were found to decrease with the administration of ginseng alkaloidal fraction. 2. Phospholipid contents increased in the serum and decreased in the liver tissue with the administration of ginseng alkaloidal fraction. 3. Triglyceride contents of the serum as well as liver tissue increased with the administration of ginseng alkaloidal fraction during the first 4 weeks but decreased in the second 4 weeks to reach a level below that of the pre-experimental period.

Brown Rice Phytosterol Improves Hypertension and Lipid Metabolism in Spontaneously Hypertensive Rats (본태성 고혈압쥐(SHR)에서 현미 식물성스테롤의 혈압 및 지질 대사 개선 효과)

  • This study was conducted to investigate the effects of brown rice and brown rice phytosterol on blood pressure and lipid levels in spontaneously hypertension rats (SHR). SHR were grouped according to blood pressure and fed either a control diet or experimental diets containing 50% brown rice powder or 5% brown rice phytosterol for 3 weeks. Body weight gain and epididymal fat weight were significantly reduced in the brown rice powder and brown rice phytosterol groups compared to control. Brown rice and brown rice phytosterol diets suppressed age-dependent increases in systolic blood pressure compared to control. In addition, brown rice and brown rice phytosterol diets decreased total lipid, triglyceride, and cholesterol levels in the liver, whereas serum HDL cholesterol increased. Lastly, brown rice phytosterol reduced TBARS contents in the kidney. These results suggest that brown rice and brown rice phytosterol exert antihypertensive effects that improve lipid metabolism in SHR.

Metabolomics reveals potential plateau adaptability by regulating inflammatory response and oxidative stress-related metabolism and energy metabolism pathways in yak

  • Species are facing strong selection pressures to adapt to inhospitable high-altitude environments. Yaks are a valuable species and an iconic symbol of the Qinghai-Tibet Plateau. Extensive studies of high-altitude adaptation have been conducted, but few have focused on metabolism. In the present study, we determined the differences in the serum metabolomics between yaks and the closely related species of low-altitude yellow cattle and dairy cows. We generated high-quality metabolite profiling data for 36 samples derived from the three species, and a clear separation trend was obtained between yaks and the other animals from principal component analysis. In addition, we identified a total of 63 differentially expressed metabolites among the three species. Functional analysis revealed that differentially expressed metabolites were related to the innate immune activation, oxidative stress-related metabolism, and energy metabolism in yaks, which indicates the important roles of metabolites in high-altitude adaptation in yaks. The results provide new insights into the mechanism of adaptation or acclimatization to high-altitude environments in yaks and hypoxia-related diseases in humans.

Vitamin C Intake and Serum Leverls in Smoking College Students (흡연대학생의 비타민 C 섭취량과 혈청수준)

  • As ciarette smoking adversely affects vitamin C metabolism in humans, smokers need substantially more vitamin C intake than do non-smokers to achieve similar serum vitamin C concentration. To provide the basic information currently available for the determination of vitamin C requirement for Korean smokers, we investigated the differences the serum vitamin C values between smokers(n=53) and non-smokers(n=62) in relation to their intake of the vitamin through diet in 115 male college students, who had not been using vitamin C supplements. Dietary intakes of vitamin C were determined by a 24-hour recall, and serum vitamin C was determined using the 2, 4-dinitrophenylhydrazine method. The mean vitamin C intakes of smokers and non-smokers were consuming less than 75% of the Koeran RDA for vitamin c in their diet. Smokers consumed yellow and green leafy vegetables more often (P=0.02) and fresh fruits less often(P=0.006) than non-smokers. The mean serum vitamin C concentration of smokers consuming the same amounts of vitamine C as non-smokers, 64.3 umol/1, were 20% lower than for non-smokers, 80.1 umol/l(P<0.05). The risk of low serum vitamin C concentration (LoC) among smokers were 3.8% compared with 1.6% in non-smokers, and the odd ratio for LoC risk was 2.43. There were no correlations between dietary and serum vitamin C for smokers and non-smokers. It was concluded that smokers might require at least 20% more vitamin C to reach the same concentration comparable to non-smokers.

Effects of Ganoderma lucidum on Lipid Metabolism in Dietary Hypercholesterolemic Rats (영지 열수추출액이 식이성 고콜레스테롤 혈증 흰쥐의 지질대사에 미치는 영향)

  • For the investigation of the effects of G. lucidum on prevention and attention of hypercholeste-rolemia and atherosclerosis dietary hypercholesterolemic rats were fed with 2.0g% G. lucidum extracts for 4 weeks. And then concentrations of total cholesterol triglyceride phospholipid in serum and liver and those of HDL-cholesterol lipid peroxide glucose in serum were analyzed. Concentration of total cholesterol in serum was the lowest in the control group(basal diet+ wa-ter) and HDL-cholesterol in serum were significantly higher in groups of control 2(hyperchole-sterolemic ratslongrightarrowbasal+water) and 4(hypercholesterolemic ratslongrightarrowbasal+G.l. extract) The concentration of triglyceride in serum were signficantly lower in groups 4 and 5(hypercholeste-rolemic ratslongrightarrowcholesterol+G. l. extract) than in the control group. Phospholipid content in serum were not significantly different among all groups. Ttal cholesterol in liver was significa-ntly higher in hypercholesterolemic rats than in the control group and triglyceride concentration were signficantly higher in groups of 3(hypercholesterolemic ratslongrightarrowcholesterol+water) and 5 than others. Phospholipid in liver was significantly higher in group 3 than in the control group but groups of 4 and 5 were lower. As for lipid peroxide in serum was the lowest in group 4 and glucose concentration was lowest in group 5 than in other groups.
