Effects of $\alpha$-Tocopherol and Perilla oil on the Toxicity of Polychlorinated biphenyl in Rat (랏트에 대한 Polychlorinated Biphenyl의 독성에 미치는 $\alpha$-Tocopherol과 Perilla oil의 효과)

  • Effects of ${\alpha}$-tocopherol and perilla oil on the toxicity of polychlorinated biphenyls (PCB) in male rat were studied. Rats were fed ad libitum for 6 weeks with the animal diet which contains PCB 30 ppm and 100 ppm. Perilla oil (0.5 g/kg body weight) and ${\alpha}$-tocopherol (30 mg/kg body weight) were administered intraperitoneally twice a week for 6 weeks. Rats fed with PCB showed enlargement of liver and spleen, increase in aspartate aminotransferase, alkaline phosphatase, sereum lipid and cytochrome P 450 and decrease in body weight and glutathione. When perilla oil was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were significantly augmented, compared to rats fed with PCB alone. This means that perilla oil potentiates the toxicity of PCB. On the other hand when ${\alpha}$-tocopherol was administered to rats fed with PCB increase in aspartate aminotransferase, alkaline phosphatase, serum lipid and cytochrome P45O and decrease in body weight and glutathione were signigicantly reduced, compared to rats fed with PCB alone. This means that u-tocopherol reduces the toxicity of PCB. From the above results, it may be concluded that PCB is metabolized by microsomal mixed function oxidase and the metabolite causes the toxicity and microsomal glutathione plays a role of protection on the toxicity of PCB.

  • Rice, the staple food in Korea, is deficient to some extent in protein, lipid and vitamins. This study was carried out in order to investigate the effects of dietary supplementation to the rice diet of cellulose, ginseng, and ${\alpha}$-tocopherol on lead toxicity in rats. Using male rats fed the rice diet with the distilled drinking water containing 750mg of lead as nitrate per liter, for 11 weeks, organ weights, hemoglobin levels, serum glutamic pyruvic transaminase activity and accumulation of lead in liver, blood and kidney were observed. Supplementation of cellulose, ginseng and ${\alpha}$-tocopherol to the lead groups showed the protective effect significantly in the weight of liver but no influence in hemoglobin levels. Ginseng especially decreased the serum glutamic pyruvic transaminase activity to normal level. The three supplemented diets reduced the lead accumulation in kidney and blood, but not in liver.

Effects of Paprika (Capsicum annuum L.) on Serum Lipid Profile and $\alpha$-Tocopherol Concentration in Rats Fed a High-Cholesterol Diet (파프리카 급여가 고콜레스테롤 식이 흰쥐의 혈청 지질과 $\alpha$-Tocopherol 농도 변화에 미치는 영향)

  • In conclusion, rats were fed diets containing either NC, HC, HC-FDP, HC-WEP, or HC-EEP for 6 weeks. At the 6th week, increases in body weight and visceral fat were lower in the paprika fed groups as compared to the HC group. Serum levels of triglycerides, total cholesterol, low density lipoprotein cholesterol, and atherogenic index values were significantly lower in the paprika diet fed groups than the HC group (p<0.05). In particular, the lipid lowering effects in the HC-EEP group were superior among the paprika fed groups. Also, serum $\alpha$-tocopherol levels were lower in the control group compared to the paprika fed groups. The supplementation of paprika may exert lipid lowering effects and saving effect of $\alpha$-tocopherol in the serum of high-cholesterol diet fed rats. However, it should be noted that the results are based on very small sample numbers and a short experimental period.

High Serum Level of Retinol and α-Tocopherol Affords Protection Against Oral Cancer in a Multiethnic Population

  • Background: A comparative cross-sectional study involving oral cancer patients and healthy individuals was designed to investigate associations between retinol, ${\alpha}$-tocopherol and ${\beta}$-carotene with the risk of oral cancer. Materials and Methods: This study included a total of 240 matched cases and controls where subjects were selected from the Malaysian Oral Cancer Database and Tissue Bank System (MOCDTBS). Retinol, ${\alpha}$-tocopherol and ${\beta}$-carotene levels and intake were examined by high-performance liquid chromatography (HPLC) and food frequency questionnaire (FFQ) respectively. Results: It was found that results from the two methods applied did not correlate, so that further analysis was done using the HPLC method utilising blood serum. Serum levels of retinol and ${\alpha}$-tocopherol among cases ($0.177{\pm}0.081$, $1.649{\pm}1.670{\mu}g/ml$) were significantly lower than in controls ($0.264{\pm}0.137$, $3.225{\pm}2.054{\mu}g/ml$) (p<0.005). Although serum level of ${\beta}$-carotene among cases ($0.106{\pm}0.159{\mu}g/ml$) were lower compared to controls ($0.134{\pm}0.131{\mu}g/ml$), statistical significance was not observed. Logistic regression analysis showed that high serum level of retinol (OR=0.501, 95% CI=0.254-0.992, p<0.05) and ${\alpha}$-tocopherol (OR=0.184, 95% CI=0.091-0.370, p<0.05) was significantly related to lower risk of oral cancer, whereas no relationship was observed between ${\beta}$-carotene and oral cancer risk. Conclusions: High serum levels of retinol and ${\alpha}$-tocopherol confer protection against oral cancer risk.

  • High-fat diet up-regulates either insulin resistance or triglycerides, which is assumed to be related to the expression of peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ and PPAR-${\gamma}$. The beneficial effects of vitamin E on insulin resistance are well known; however, it is not clear if vitamin E with a high-fat diet alters the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$. We investigated the effects of d-${\alpha}$-tocopherol supplementation on insulin sensitivity, blood lipid profiles, lipid peroxidation, and the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat (HF) diet-fed male C57BL/6J model of insulin resistance. The animals were given a regular diet (CON; 10% fat), a HF diet containing 45% fat, or a HF diet plus d-${\alpha}$-tocopherol (HF-E) for a period of 20 weeks. The results showed that the HF diet induced insulin resistance and altered the lipid profile, specifically the triglyceride (TG) and total cholesterol (TC) levels (P < 0.05). In this animal model, supplementation with d-${\alpha}$-tocopherol improved insulin resistance as well as the serum levels of TG and very-low-density lipoprotein-cholesterol (VLDL-C) (P < 0.05). Moreover, the treatment decreased the levels of malondialdehyde (MDA) in the serum and liver while increasing hepatic PPAR-${\alpha}$ expression and decreasing PPAR-${\gamma}$ expression. In conclusion, the oral administration of d-${\alpha}$-tocopherol with a high-fat diet had positive effects on insulin resistance, lipid profiles, and oxidative stress through the expression of PPAR-${\alpha}$ and PPAR-${\gamma}$ in a high-fat diet-fed male mice.

Effects of the P/S Ratio of Dietary Lipids and Antioxidant Vitamin Supplements on the Level of Serum Lipids and Liver. Lipid Peroxidation in Rats Treated with DMBA

  • This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. In vivo Antioxidant Effects of Aralia elata Seemann Ethanol Extract Administered with Benzo($\alpha$) pyrene

  • This study has examined the effects of Aralia elata Seemann ethanol extract on antioxidant enzyme systems inrats along with benzo($\alpha$) pyrene(B(a)P) administration . The ethanol extract of Aralia elata Seemann (50mg/kg body wt.) was fed to rats for 4 weeks by stomach tubing. The extract administration increased antioxidant activities of glutathione sulfur transferase(GST) comparing to the control. also total superoxide dismutase(SOD) and Cu, Zn-SOD activities were stimulated. Catalase activities were increased by 50% with the extract feeding compared to the control . Combined administration of B($\alpha$)P and the extract increased GST activity in B($\alpha$) P group. Although total SOD acitivity was decreased , Cu, Zn-SOD was greately increased from 0.10unit to 0.18 unit and catalase activity also was increased compared to the group of B($\alpha$) P. GST activity in CLE group was 1.32 unit, increased by 33% comparing to the group CL of 0.99unit. Cu, Zn-SOD and catalase activities in thegroup fed high fat and ethanol extracts were increased by 25% and 39%, respectivley comparing to the group of high fat. In addition , total SOD was decreased but, Cu, Zn-SOD acitivity was increased from 0.09 unit to 0.18unit. Catalase activity was 76.05 unit in the group of B($\alpha$) P and extract comparing to 65.26 units in B($\alpha$)P group. Serum $\alpha$-tocopherol of rat was markedly increased by theextract. Administration of B9$\alpha$)P reduced $\alpha$-tocopherol levels in the serum, on the other hand, lard in the diet increased $\alpha$-tocopherol levels in the serum. The above results indicate that Aralia bud exerts antioxidant functions in vivo against B($\alpha$)P. Further research may be necessary for the identification fo the biologically active material.

  • Ascorbate-$Fe^3+$-EDTA에 의해 생성된 hydroxyl radical (HO.)을 이용하여 단백질과 지질의 산화적 손상을 유도하였으며, 이 손상에 대한 멜라토닌(melatonin), glutathione, $\alpha$-tocopherol의 방어효과를 분석하였다. 단백질로는 bovine serum albumin (BSA), 지질로는 phosphatidyl-choline (PC) liposome을 이용하였다. 그리고 BSA와 PC liposome의 혼합계에서도 각 항산화제의 효능을 분석하였다. 단백질의 산화적 손상은 carbonyl기의 생성 및 단백질 절단을, 지질산화는 lipid peroxidation (LPO, malondialdehyde 와 4-hydroxyalkenal) 분석을 통하여 조사하였다. 단백질의 산화는 멜라토닌과 glutathione이, 지질산화는 멜라토닌과 $\alpha$-tocopherol이 효과적으로 억제하였다. 그러나 glutathione은 지질, $\alpha$-tocopherol은 단백질의 산화를 억제하지 못하였다. BSA와 PC liposome이 동시에 존재하는 혼합물에서는 수용성과 지용성 특성을 모두 지닌 멜라토닌은 단백질, 지질 모두에 대해 산화방지효과가 있으나 수용성인 glutathione은 단백질에, 지용성인 $\alpha$-tocopherol은 지질에만 주로 효과가 있는 것으로 조사되었다.

Effect of Supplementation of Antioxidant Nutrient Against Oxidant Stress during Exercise

  • This study was undertaken to evaluate the effect of 4 weeks of $\alpha$-tocopherol(800 I.U./d) supplementation on oxidant stress of eleven female aerobic -majoring students during rest and exercise. Changes in the activity of the antioxidant enzyme glutathione peroxidase were also studied. Serum $\alpha$-tocopherol concentration was significantly increased with vitamin E supplementation(710.1$\pm$113.8$\mu\textrm{g}$/dl vs. 1,485,8$\pm$105.2$\mu\textrm{g}$/dl). In addition, serum MDA concentration, an index of lipid peroxidation, significantly decreased after vitamin E supplementation. However, MDA values after exercise increased to pre-supplementation levels. Serum glutathione peroxidase activity significantly increased with vitamin E supplementation. The enzyme activity showed a trend toward decrease after exercise. Serum cholesterol values were not significantly affected by vitamin E supplementation. However, serum triglycerides significantly increased after supplementation against oxidative stress during resting periods. These supplements appraently work by decreasing lipid peroxidation and increasing glutathione peroxidase activity. However, vitamin E supplementation did not prevent exercise-induced increases in lipid peroxidation.

The Comparison of Blood Retinol and $\alpha$-Tocopherol Concentrations between Diarrhea and Normal Calves (정상과 설사증 송아지에서 혈중 retinol과 $\alpha$-tocopherol의 비교)

  • The purpose of this study is to examine and compare the concentrations of blood retinol and $\alpha$-tocopherol in normal and diarrhea calves. The subjects of the experiments are from three groups. Each group consists of 5 calves(diarrhea, treatment and control group). The CBC, serum chemistry, serum retinol and a-tocopherol concentrations were estimated in each group. Blood retinol concentration was $13.3{\pm}7.0{\mu}g/100ml$ in diarrhea group, $31.5{\pm}6.9{\mu}g/100ml$ in treatment group and $28.1{\pm}11.8{\mu}g/100ml$ in the control group. The blood concentration in diarrhea group was significantly lower than that of the control group in the case of retinol(p<0.05). However, there were no significance between the treatment poop and the control group. The $\alpha$-tocopherol concentration in blood was $266.0{\pm}127.6{\mu}g/100ml$ in diarrhea group, $432.2{\pm}172.7{\mu}g/100ml$ in the treated group and $579.3{\pm}145.8{\mu}g/100ml$ in the control group. In the case of $\alpha$-tocopherol, the laboratory group were significantly lower than the control group(p<0.05), except for the treatment group. As in retinol concentration there were no significance between treatment group and the control group. In the test of CBC, PCV was significantly lower in the group with diarrhea than the control group(p<0.05). Fibrinogen concentrations in diarrhea calves were significantly higher than the treatment and control group. In conclusion, the blood retinol and $\alpha$-tocopherol concentration in diarrhea calves are lower than normal calves. Medication of retinol and $\alpha$-tocopherol on calves with diarrhea is recommended.
