Enhancement of Biocontrol Activity of Serratia plymuthica A21 -4 Toward Phytophthora Blight of Pepper by Amendment of Nutritional Condition

  • Serratia plymuthim A21-4 strongly inhibits the mycelial growth, zoospore formation, and cystospore germination of Phytophthor spp and Pythium species. The bacterial isolate produced antifungal substance and chitinase. The bacteria also enhanced to plant growth remarkably in low nutritional condition. The application of cell suspension of A21-4 to pepper seedlings in greenhouse experiments and soil drenching in farmer's field was proved successfully to control the phythophthora blight of pepper. For the effective control, however, relatively high density of cell number(10$\^$9/cfu/$m\ell$) is required. Density effect was similar in plant growth promoting activity of A21-4. Though this investigation we improved the problem with changes of culture condition of bacteria and some nutritional amendment.

Control of the Root-Knot Nematode (Meloidogyne spp.) on Cucumber by a Liquid Bio-Formulation Containing Chitinolytic Bacteria, Chitin and Their Products (키틴분해세균, 키틴 및 그들의 산물이 함유된 미생물제에 의한 오이의 뿌리혹선충(Meloidogyne spp.) 방제)

  • A liquid bio-formulation containing chitinolytic bacteria, chitin and their products was assessed for its potential biological control against root-knot nematodes on cucumber. The bio-formulation was prepared by cultures of three chitinolytic bacteria, Chromobacterium sp. strain C-61, Lysobacter engymogenes and Serratia plymuthica in minimal medium supplemented with chitin. Under pot conditions, the bio-formulation showed better growth of cucumber plants, and less root galls and population density of Meloidogyne spp. than control media without the bio-formulation. In a greenhouse, 75-fold diluted bio-formulations were treated instead of water around cucumber plants through hoses for drip irrigation six times at 5-day intervals from the transplanting date. After 30 and 60 days, the treatment provided about 7% and 10% enhancement in the plant height and about 78% and 69% reduction in population density of Meloidogyne spp. in the rhizosphere, respectively. In addition, the experiments showed that the control effects occurred only in the soils contacted with the bio-formulation. Undiluted bio-formulations were drenched three times at 10-day intervals around cucumber plants severely infested with Meloidogyne spp. The treatment showed about 37% plant enhancement without dead plants compared with 37% death in the untreated control, and about 82% nematode reduction. These results suggest that the bio-formulation can be practically used to control the root-knot nematode on cucumber.

Distribution and Biodegradation of Crude oil-Degrading Bacteria in P'ohang Coastal Area (포항근해 원유분해세균의 분포 및 원유분해능)

  • Seawater samples were collected from P'ohang coastal area during April 1995 - January 1996. The distribution of total heterotrophic bacteria and crude oil-degrading bacteria (CDB) were studied. In addition, biodegradation of crude oil was investigated through mono and mixed culture. The heterotrophic bacterial distribution was in the range of 4.1 $\times$ $10^4$- 1.2 $\times$ $10^5$ CFU/$m\ell$, respectively. The percent of crude oil-degrading bacteria against total heterotrophic bacteria was 0.05-0.54% which was lower than other marine samples reported. Therefore it could be suggested that the distribution of crude oil-degrading bacteria in the seawater of P'ohang coastal area was highly affected by presence of petroleum hydrocarbon. Taxonomical characteristics of 26 isolates were investigated. The results of identification were showed 7 genera which were Acinetobacter spp., Bacillus spp., Citrobacter spp., Micrococcus spp., Moraxella spp., Rhodococcus spp., and Serratia spp. Appearance of Enterobacteriaceae indicated that the seawater was polluted with wastewater. Also genus of Bacillus had predominant in CDB on P'ohang coastal area. In flask culture, biodegradation of crude oil was enhanced by addition of mixed culture of CDB.

  • The in vitro and in vivo antibacterial activities of a new fluoroquinolone, DWP20364(1-cyclopropyl-5-amino-6,8-difluoro-7-(2,7-diazabicyclo[3.3.0]oto-4-ene-7-yl)-1 ,4-di-hydro-4-oxoquinoline-3-carboxylic acid) were evaluated in comparison with those of ciprofloxacin(CPFX), sparfloxacin(SPFX) and ofloxacin(OFLX). DWP20364 was more potent than CPFX and OFLX against Staphylococcus spp., Streptococcus spp. and Enterococcus faecium MD8b and it was similarly or slightly less active than CPFX against Escherichia spp. and Pseudomonas spp.. For MRSA and OFLX resistant strains (Staphylococcus spp.(14),Enterococcus spp.(4), Acinetobacter spp.(2), Pseudomonas spp.(9), Klebsiella spp.(2) and Serratia spp.(6)),DWP20364(MICs for 90% of strains,0.025 and 12.5${\mu}$g/ml, respectively) was 4 to 32 folds more potent than SPFX and CPFX. The activity of DWP20364 decreased moderately in the presence of 5mM $Mg^{2+}$. However, various pHs and the concentrations of various serum had no effect on the activity of DWP20364. DWP20364 possessed a bacteriocidal effect at the 1MIC against gram positive and gram negative strains. The protective effect of DWP20364 against systemic infections in mice caused by S. aureus Smith or S. aureus L2379 was superior to that of CPFX and SPFX but it was less active than that of CPFX against infection by P. aeruginosa E-2.

Antimicrobial Resistance Characteristics of Gram-Negative Bacteria Isolated from Inland Pollution Sources in the Drainage Basin of Iwon-myeon (Taean-gun), South Korea (태안군 이원면 육상오염원 배출수에서 분리한 그람음성균의 항생제 내성 특성)

  • Fecal contamination levels of discharge water from inland pollution sources were investigated in Iwon-myeon (Taean-gun), South Korea. Gram-negative bacteria were isolated during the investigation and the antimicrobial resistance patterns of the isolates were examined to estimate their impact on the coastal environment. The ranges of total coliform and fecal coliform of 12 samples from four major inland pollution sources were 79-490,000 MPN/100 mL and 2.0-490,000 MPN/100 mL, respectively, with the highest level of fecal contamination at Station No. 3. A total of 137 strains (14 genus) were isolated, of which 86 strains (62.8%) were Enterobacteriaceae. The identified isolates were as follows: Pseudomonas spp. (35 strains), Klebsiella spp. (20 strains), Serratia spp. (20 strains), and Escherichia spp. (19 strains). The isolated Gram-negative bacteria showed the highest antimicrobial resistance to ampicillin (81.8%), followed by amoxicillin/clavulanic acid (64.2%), ceftiofur (61.3%), and cefoxitin (59.1%). Antimicrobials in which less than 10% of isolates showed antimicrobial resistance were ciprofloxacin (3.6%) and gentamicin (2.2%). Resistance to one or more antimicrobials was observed in 121 strains (88.3%) and 84 strains (61.3%) showed a tendency for multiple antimicrobial resistance.

Evaluation of Characteristics of Microorganisms Isolated from Public Drinking Water Facilities in Gwangju City (광주지역 먹는물 공동시설의 미생물 특성 및 분포조사)

  • Objective: This study was performed to detect indicator bacteria in drinking spring water samples in Gwangju City and to identify their genus using the VITEK-II system. Methods: The subjects were ten drinking spring water sites in Gwangju. Samples of spring water were taken every month from September 2019 to August 2020. We analyzed for the indicator bacteria Yersinia and microorganisms isolated from the spring water. Result: According to the research results on indicator bacteria, general bacteria in st1-st7 with sterilization facilities in the spring and summer were investigated in the range of 0-2 CFU/mL and 0-12 CFU/mL. In st9, where a sterilization facility was not installed, the most general bacteria were detected (160 CFU/mL). Total coliform and fecal coliform showed unsatisfied rates of 16.7 and 11.1% in spring and 14.7 and 11.8% in summer, respectively. The unsatisfied rates of total coliform for the designated and non-designated spring water facilities were 3.8 and 47.1%, respectively, and for the fecal coliform group they were 2.5 and 35.3%. The difference was confirmed according to the presence of a sterilization facility. Yersinia spp. was not detected in all drinking spring water. Forty-one strains in 25 species were isolated from ten sites. The results classified as major dominant species are Pseudomonas spp. 14.6%, Pantoea spp. 9.8%, Serratia spp. 9.8%, Acinetobacter spp. 9.8%, Citrobacter spp. 7.3%, Bordetella spp. 7.3%, Delftia spp. 4.9%, and Enterobacter spp. 4.9%. Conclusions: Based on the result that various species derived from fecal pollution and artificial pollutants were detected in the non-specified public spring water facilities that many people use, the facilities need institutional complements such as continuous management or complete shutdowns.

Biological Control of Blue Mold of Apples by Bacillus spp. and Serratia marcescens (Bacillus spp. 및 Serratia marcescens에 의한 사과 푸른곰팡이병의 생물적 방제)

  • The 1080 epiphytic bacteria obtained from 370 samples of pome and stone fruits including apple, pear, peach, grape, apricot and Chinese quince were screened for antagonistic activity against postharvest pathogens, Penicillium expansum, Alternaria alternata and Botrytis cinerea. Among tested antagonistic bacteria, eight bacterial isolates inhibited mycelial growth of the postharvest pathogens and were identified as Bacillus amyloliquefaciens (three strains), B. megaterium, B. subtilis var. gladioli, B. licheniformis, B. pumilus and Serratia marcescens based on biochemical characteristics and utility of carbon and nitrogen compounds (Biolog system). Eight carbohydrates were evaluated for their effect on mycelial growth and germination of the postharvest pathogen, P. expansum to select nutrients for enhancing bio-control efficacy. The growth of four selected antagonists, B. amyloliquefaciens P43-2, B. amyloliquefaciens A71-2, B. licheniformis P94-1, and S. marcescens P76-9 were also tested. As a result, 1% glucose (w/v) strongly stimulated growth of the antagonists, suppressed mycelial growth of the postharvest pathogen, and had a little comparatively stimulatory effect on germination of the the postharvest pathogen. It was confirmed that the addition of 1% glucose (w/v) greatly enhanced biocontrol effect of B. amyloliquefaciens P43-2, B. licheniformis P94-1, and S. marcescens P76-9. Application of B. amyloliquefaciens P43-2, B. licheniformis P94-1, and S. marcescens P76-9 with the addition of 1% glucose (w/v) increased the control efficacy up to 48%, 46%, 14% compared with those of the antagonists without glucose, respectively. When the antagonists were applied to control postharvest disease caused by P. expansum in apple wounds, the population of B. amyloliquefaciens P43-2 and B. licheniformis P94-1 increased until 4 days after inoculation (DAI) of the antagonists and then decreased from 10 DAI. Meanwhile the population of S. marcescens P76-9 decreased at early stage (4 DAI), but increased from 7 DAI, and finally maintained constantly until 10 DAI in apple wounds.

  • Antagonistic microorganisms from rhizosphere soil were isolated, identified, and applied successfully as the biocontrol agents of damping-off caused by Rhizoctonia spp. Rhizosphere antagonists isolated from rhizosphere soil were identified as Trichoderma viride, T. harzianum, T. hamatum, T. polysporum, Gliocladium sp., Pseudomonas fluorescence, P. stutzeri, P. cepacia, Enterobacter sp., Serratia sp. and Erwinia herbicola. Of these, the most promising ones in vitro were T. virdie, T. harzianum, Gliocladium sp., Serratia sp., P. stutzeri, and P. cepacia. These above six antagonists were efficient in reducing disease incidence to $40{\sim}70%$ when the reselected rhizosphere antagonists preparations were applied to the soil at $10^6$ propagules per gram. Among six antagonists, T. viride was the most promising biocontrol agents against R. solani isolates in soil. The suppressive effect was more evident in steam-sterilized soil than in non-sterilized field soil.

Antimicrobial susceptibility and prevalence of gram-negative bacteria isolated from bovine mastitis (젖소 유방염으로부터 분리한 그람음성균의 분포 및 항생제 감수성)

  • Environmental mastitis has increased particularly in well-managed or low somatic cell countherds that have successfully controled contagious pathogens. Major pathogens of environmental mastitisare Escherichia coli (E. coli) and Streptococcus uberis. The present study was conducted to investigate1,865 quaters of 241 Korean dairy farms from 2001 to 2004. Prevalence of major gram-negative bacteriaisolated from mastitis milk were E. coli (22.7%) and Enterobacter spp. (16.3%) in coliforms and Pseudomoassp. (10.3%) and Serratia spp. (7.9%) in non-coliforms. The results on antibiotic susceptibility by agardifusion test against these pathogens were 86.7% in piperaciliin, 94.6% in cefepime, 85.5% in amikacin,87.7% in gentamicin and so on. In contrast, the susceptibility against ampicillin (41.9%), cephalothin (9.9%),streptomycin (39.9%) and tetracycline (46.7%) appeared to be below 50%. Gram-negative bacteria showed(96.8%). Acording to year, distribution of high $256{\sim}64{\mu}g/ml$ on cephalothin get increased, but the othersare diferent. These findings demonstrate that major gram-negative bacteria were E. coli and Enterobacterspp. isolates, and often encountered the diverse antibiotic resistant patterns.

Characterization of Phosphate-solubilizing Microorganisms in Upland and Plastic Film House Soils (밭과 시설재배지 토양의 인산가용화 미생물의 특성)

  • With the aim to explore the possible role of phosphate-solubilizing bacteria in soil, we conducted a survey of phosphate-solubilizing microorganisms colonizing in upland and plastic film house soils. Soil EC, pH, organic matter, available phosphate, exchangeable cation such as potassium, calcium and magnesium, and total P of plastic film house soils were higher than those of upland soils. Phosphate-solubilizing bacteria population was higher in plastic film house soils than upland soils, but species of phosphate-solubilizing bacteria was more diverse in the upland soils than the plastic film house soils. There was significant positive correlation between phosphate solubilization and phosphate-solubilizing bacteria in soils. Bacillus, Cedecea, Brevibacillus, Paenibacillus, Pseudomonas, Serratia spp. were isolated from upland soils and Bacillus and Cellulomonas spp. were from plastic film house soils.
