돼지 폐렴병소로부터 분리한 Pasteurella multocida의 capsular serogroup과 toxA gene의 분포 (Capsular serogroups and toxA gene of Pasteurella multocida isolated from Pneumonic Lung Lesions of Swine)

  • The present study was conducted to investigate the capsular serogroups and distribution of toxA gene of Pasteurella (P.) multocida isolated from pneumonic lung lesions of swine in Korea. A total number of 91 (36.3%) P. multocida isolated from 251 lung lesions. P. multocida isolates were typed for capsular serogroup and toxA gene by polymerase chain reaction. Of the 91 strains, serogroup A and D were 69 strains (75.8%) and 22 strains (24.2%), respectively. Sixty one strains (67.0%) out of 91 strains were detected as toxA gene, and 47 strains (77.0%) and 14 strains (23.0%) belongs to serogroup A and D, respectively.

  • The Salmonella rfb gene encoding for the biosynthesis of the oligosaccharide-repeating units of the O-antigenic determinants was cloned and sequenced. A set of nucleotide primers(a forward and reverse) was selected to target a defined region of the guanosine diphospho-mannose(GDP-Man) pyrophosphorylase synthase gene : rfbM of Salmonella C serogroup. The primer set was used to develop a PCR-based rapid and specific detection system for Salmonella C1 serogroup. Amplification bands of predicted size(1,422bp) were generated from 11 different Salmonella C1 isolates. The bands were verified to be specific for the C1 serogroup by Southern blot analysis using reference homologous DNA specificity was further confirmed by the lack of reactivity with heterologous DNA derived from non-salmonella members of the family enterobacteriaeceae. A specificity of 100% was deduced along with a very high sensitivity shown by a detection limit of 1fg of a purified DNA template. The isolated DNA sequence was found to be 99.8% homologous to S montevideo but the related primers amplified with the predicted band sizes with all the Salmonella C1 serogroups tested. It is concluded that the PCR protocol based on the rfbM gene from S cholerasuis is optimal fast and specific for the detection of Salmonella C1 serogroup and also the corresponding probe is suitable for rapid detection of all Salmonella C1 serogroup DNA tested. This technology should facilitate the identification of contaminated pig products and for any other products contaminated with the Salmonalla C1 serogroup. The immediate impact of this developed method will be in the area of food safety of pig products with the potential prospect for adaptation to other food inspection technologies.

제한효소 DNA 분석법에 의한 국내분리 렙토스피라균의 동정 (Restriction Endonuclease DNA Analysis of Leptospiral Field Isolates from Korea)

  • The genomes of leptospiral field isolates from Korea belonging to serogroup Icterohaemorrhagiae (21 strains) and serogroup Canicola (1 strain) were analysed and compared by restriction enzyme analysis with EcoRI and HindIII as digesting enzymes. One isolate belonging to serogroup Canicola showed the same pattern as serovar portlandvere. All 21 isolates belonging to serogroup Icterohaemorrhagiae showed almost same patterns as Leptospira serovar lai from China, But with very slight differences 21 isolates could be classified into 8 subtypes and these grouping seems to reflect the differences in epidemiological niche. And also the geographical data consisted with the grouping into 8 subtypes. According to our results, we concluded that the restriction endonuclease analysis of chromosomal DNA will be an accurate and reliable method to compare and classify pathogenic leptospires.

SefA 유전자 PCR에 의한 Salmonella serogroup D1의 특이적 검출 (Specific detection of Salmonella serogroup D1 by polymerase chain reaction(PCR) for sefA gene)

  • Sal enteritidis thin fimbriae, SEF14, were found to be restricted to the predominantly poultry-associated members of the Salmonella serogroup D1 that are considered as the important pathogens in poultry industry. SefA together with sefB and sefC encode the proteins involved in SEF14 biosynthesis. In order to develop the rapid and specific detection methods for Salmonella serogroup D1, a PCR technique for the amplification of sefA gene was established, and its specificity and sensitivity were investigated with various microorganisms. The bacterial genomic DNA was extracted by colony-picking and rapid boiled-lysate technique. In comparison of Sef I and Sef II primers used in the PCR, Sef I primer for sefA gene of 513bp showed higher specificity than that of Sef II. The established PCR was as sensitive as to detect 1pg of Sal enteritidis DNA. When 73 strains in 28 genera including the reference strains and the field isolates of various Salmonella serotypes, Bacillus subtilis, Bordetella bronchisepdca, E coli, Listeria spp., Micrococcus luteus, Rhodococcus equi, Staphylococcus spp., Streptococcus spp., Vibrio parahemolyticus, Yersinia spp. were studied, the established PCR yielded specifically positive results with only Salmonella serogroup D1. The results suggested that the PCR for sefA gene could be a potential candidate among the specific detection methods for Salmonella serogroup D1.

  • The aim of this study was to characterize the molecular features of serogroup C Neisseria meningitidis strains circulating in Beijing, China. Twenty out of 23 strains belonged to ST 4821. The causative serosubtype for meningococcal meningitis was P1.12-1,16-8. All of the strains expressed class 3 PorB protein. Among the five pulsed-field gel electrophoresis patterns observed, pattern III predominated.

  • 콩의 공중질소이용도(空中窒素利用度) 향상에 바람직한 우수근류균(優秀根瘤菌) 선발(選拔)의 기초자료를 얻고자 우리나라 토착(土着) B. japonicum 의 혈청형별(血淸型別) 콩 공생효과(共生效果)와 그 유의성(有意性)을 분석한 결과는 다음과 같았다. 1. 근류균(根瘤菌)의 근류형성정도(根瘤形成程度)는 USDA 123혈청형(血淸型) 균주(菌株)를 접종한 구에서 가장 높고 다음으로 YCK 150혈청형(血淸型) > YCK 117혈청형(血淸型) > YCK 141혈청형(血淸型) > USDA 110 혈청형(血淸型) 순위의 경향이었는데, 콩 품종별(品種別) 혈청형(血淸型)에 따라서 상이하였다. 2. 콩 건물생산(乾物生産)에 있어서 근류균(根瘤菌) 접종효과(接種效果)는 콩 품종별로 상이하였으나 평균적으로 볼 때, USDA 110혈청형(血淸型)과 YCK 141혈청형(血淸型) 균주(菌株)의 접종구에서 우수하였고 USDA 123 혈청형(血淸型) 균주(菌株)의 접종구(接種區)에서는 저조하였다. 그리고 균주접종(菌株接種)에 따른 건물생산량(乾物生産量)이 가장 높았던 콩은 방사(放射)콩이었다. 3. 혈청형내(血淸型內) 균주(菌株)들의 공생효과(共生效果) 유사성(類似性)(n=20)은 근류수에서 100%, 질소고정력에서 90%, 콩 건물중에서 80% 수준(水準)이었으며, 균주(菌株)의 공생관계(共生關係) 변이분석(變異分析)에서는 또 각 혈청형간(血淸型間), 콩 품종간(品種間)에 근류수와 근류무게, 질소고정력, 콩 건물중 모두 고도(高度)의 유의성(有意性)($$P{\leq_-}0.01$$)을 보였다. 그러므로 B. japonicum 의 면역혈청학적(免疫血淸學的) 특성은 콩의 공중질소이용도(空中窒素利用度) 개선연구(改善硏究)에 이용가치가 높은 것으로 판단되었다.

한국에서 분리된 Vibrio cholerae non-O1의 혈청형의 분포 (The Distribution of Serogroup of Vibrio cholerae non-O1 Isolated in Korea)

  • 우리나라 연안해역의 해수나 어패류에서 V. cholerae non-O1의 분포를 밝힘과 동시에 1996년 4월부터 동년 11월까지 분리된 환경분리주와 1984년부터 1996년까지 서울 및 부산소재 각 대학병원에서 환자로부터 분리된 총 55균주를 대상으로 생화학적 특성과 혈청형을 분석하였다. V. cholerae non-O1은 한국의 연안에서 206거체중 21검체(10%)에서 분리되었으나 V. cholerae O1 및 O139는 검출되지 않았다. 지역별로는 군산에서 16%로 검출률이 제일 높았다. 환경이나 환자에서 분리된 V. cholerae non-O1 55 균주에 대한 혈청형 분석결과 15종류의 혈청형이 검출되었는데 환경분리주 12종류, 환자분리주에서는 6종류이였다. 그런데 환경분리주에서는 O14가 68.3%(28/41)이였고, 환자분리주에서는 O8이 약 36%(5/14)로 많았고 그다음으로 O2와 O10등의 순이었다. 그리고 O2, O10와 O39는 환경 및 환자분리주에서 동시에 검출되었다.

Bactericidal Antibody Responses to Meningococcal Recombinant Outer Membrane Proteins

  • Secretin PilQ is an antigenically conserved outer membrane protein that is present in most meningococci and PorA is a major protein that elicits bactericidal immune response in humans following natural disease and immunization. In the present study, BALB/c mice were immunized subcutaneously with rPilQ406-770 or rPorA together with Freund's adjuvant (FA). Serum antibody responses to serogroup A and B Neisseria meningitides whole cells or purified proteins and functional activity of antibodies were determined by ELISA and serum bactericidal assay (SBA), respectively. Serum IgG responses were significantly increased in the immunized group with rPilQ406-770 or rPorA together with FA compared to control groups. IgG antibody response of mice immunized with rPilQ406-770 was significantly more than mice immunized with rPorA (OD at 450 nm was 1.6 versus 0.83). The booster injections were effective in increasing the responses of antirPilQ406-770 or anti-rPorA IgG significantly. Antisera produced against rPilQ406-770 or rPorA demonstrated strong surface reactivity to serogroup B N. meningitides in comparison with control groups. Antisera raised against rPorA or rPilQ406-770 and FA demonstrated SBA titers from 1/1024 to 1/2048 against serogroup B. The strongest bactericidal activity was detected in sera from mice immunized with rPilQ406-770 mixed with FA. These results suggest that rPilQ406-770 is a potential vaccine candidate for serogroup B N. meningitidis.

Ampicillin과 Chloramphenicol 내성 Salmonella typhimurium 분리의 증가 (Increased Isolation of Ampicillin and Chloramphenicol Resistant Salmonella typhimurium)

  • Salmonella typhi infection, which was the most frequent enteric infection in Korea, has been decreasing, while the infection of other serogroups of Salmonella has been increasing since the later part of 1970s. In 1986, the number of serogroup B isolated by us increased to 46, which corresponds 21.1% of all enteropathogenic bacteria isolated from stool specimens. Salmonella isolates resistant to antimicrobial agents were extremly rare in Korea, in the 1970s. However, 7 of 13 serogroup B isolates showed resistance to ampicillin or to chloramphenicol in 1984. Among the serogroup B isolates in 1986, 71.7% and 69.6% were resistant to ampicillin and to chloramphenicol respectively. The minimum inhibitory concentrations of ampicillin and chloramphenicol against these isolates were >$128\;{\mu}g/ml$ and $128\;{\mu}g/ml$ respectively.

Neonatal Sepsis and Meningitis Caused by Neisseria meningitidis Serogroup B: a Case Report

  • 수막구균 감염은 신생아에서 높은 사망률을 보이지만 매우 드물고, 단체 생활하는 연장아나 청소년에서 더 흔하다고 알려져 있다. 이 증례에서 소개된 생후 16일 신생아는 수막구균 B 혈청군 패혈증과 수막염이 진단되었지만 조기 항생제 치료를 통해 합병증 없이 건강하게 퇴원하였다. 이 증례 보고의 목적은 신생아에서 수막구균 감염의 발생률과 중요성을 상기시키고 노출된 가족들과 의료진에 대한 조사와 예방 요법의 필요성을 강조하기 위함이다. 한국에서는 많은 신생아가 출생 후 산후조리원 시설에서 지내내므로 감염 발생과 전파에 단체 생활이 영향을 미칠 수도 있음을 생각해 볼 필요가 있다. 질병의 조기 발견, 격리, 그리고 경험적 치료가 치명적인 질병의 전파를 예방하기 위해 매우 중요하다.
