The Gut-Heart Axis: Updated Review for The Roles of Microbiome in Cardiovascular Health

  • Cardiovascular diseases (CVDs), including coronary artery disease, stroke, heart failure, and hypertension, are the global leading causes of death, accounting for more than 30% of deaths worldwide. Although the risk factors of CVDs have been well understood and various treatment and preventive measures have been established, the mortality rate and the financial burden of CVDs are expected to grow exponentially over time due to the changes in lifestyles and increasing life expectancies of the present generation. Recent advancements in metagenomics and metabolomics analysis have identified gut microbiome and its associated metabolites as potential risk factors for CVDs, suggesting the possibility of developing more effective novel therapeutic strategies against CVD. In addition, increasing evidence has demonstrated the alterations in the ratio of Firmicutes to Bacteroidetes and the imbalance of microbial-dependent metabolites, including short-chain fatty acids and trimethylamine N-oxide, play a crucial role in the pathogenesis of CVD. However, the exact mechanism of action remains undefined to this day. In this review, we focus on the compositional changes in the gut microbiome and its related metabolites in various CVDs. Moreover, the potential treatment and preventive strategies targeting the gut microbiome and its metabolites are discussed.

  • Around the world, fermentation of foods has been adopted over many generations, primarily due to their commercial significance with enriched flavors and high-profile nutrients. The increasing application of fermented foods is further promoted by recent evidence on their health benefits, beyond the traditionally recognized effects on the digestive system. With recent advances in the understanding of gut-brain interactions, there have also been reports suggesting the fermented food's efficacy, particularly for cognitive function improvements. These results are strengthened by the proposed biological effects of fermented foods, including neuroprotection against neurotoxicity and reactive oxygen species. This paper reviews the beneficial health effects of fermented foods with particular emphasis on cognitive enhancement and neuroprotective effects. With an extensive review of fermented foods and their potential cognitive benefits, this paper may promote commercially feasible applications of fermented foods as natural remedies to cognitive problems.

Perspectives on Bovine Milk-Derived Extracellular Vesicles for Therapeutic Applications in Gut Health

  • Extracellular vesicles (EVs) are nanosized vesicles secreted from cells into the extracellular environment and are composed of a lipid bilayer that contains cargos with biological activity, such as lipids, proteins, mRNAs, and noncoding microRNAs (miRNAs). Due to their biological activity and their role in cell-to-cell communication, interest in EVs is rapidly increasing. Bovine milk is a food consumed by people of all ages around the world that contains not only a significant amount of nutrients but also EVs. Milk-derived EVs also exhibit biological activity similar to other source-derived EVs, and studies on bovine milk EVs have been conducted in various research fields regarding sufficient milk production. In particular, not only are the effects of milk EVs themselves being studied, but the possibility of using them as drug carriers or biomarkers is also being studied. In this review, the characteristics and cargo of milk EVs are summarized, as well as their uptake and stability, efficacy and biological effects as carriers, and future research directions are presented.

장관내(腸管內) 칼슘흡수 실험방법에 관한 연구 (An Experimental Method of Calcium Absorption in Rat Small Intestine - in situ Closed Gut Segment Technique -)

  • 본 연구는 식후(食後) 단시간내(短時間內)에 장내용물(腸內容物)이 존재(存在)하고 있는 장(腸)loop내(內)에 $^{45}Ca$을 직접 주입(注入)한 후, 장(腸)loop로부터의 $^{40}Ca$$^{45}Ca$의 소실량(消失量), 또는 문맥혈중(門脈血中) $^{45}Ca$의 출현량(出現量)에 의해 Ca흡수량을 측정 비교할 수 있는 in situ closed gut segment법(法)의 하나를 확립(確立)하고져 행하여 졌다. 성장기 흰쥐에게 casein식(食)을 급여하고 검토한 결과, in situ closed gut segment법(法)의 실험 최적조건(最適條件)은 ligation부위는 회장중앙부 15-20cm, 주입액(注入液)의 조성(組成)은 $0.2{\sim}0.5{\mu}Ci/5{\mu}gCa/0.5ml$, pH 7.0, $37^{\circ}C$, 장(腸)loop로부터 $^{40}Ca$$^{45}Ca$의 소실량(消失量) 측정(測定)은 $^{45}Ca$주입후(注入後) 30분간, 문맥혈중(門脈血中)의 $^{45}Ca$농도 측정은 15분간이었다. 이상의 실험조건에서 흰쥐에게 casein식(食)과 egg albumin식(食)을 급여하고 Ca흡수량을 비교한 결과, casein식(食)에 의해 2~5 배의 높은 Ca흡수효과를 보였으며, 문맥혈중 $^{45}Ca$농도로부터의 평가는 장(腸)loop로 부터의 평가보다 과소평가 되지만 비교연구에는 충분히 유효함을 보였다.

Symbiotic Bacterial Flora Changes in Response to Low Temperature in Reticulitermes speratus KMT001

  • Lower termites require symbiotic microbes in their gut. The microbial communities in the termites must adapt to the termite temperature. Reticulitermes speratus KMT001 from Bukhan Mountain in Seoul may require a special symbiotic microorganisms for growth in low temperature Korean habitat. A metagenomics analysis showed a dramatic change in the symbiotic bacterial flora in the gut of R. speratus KMT001 in response to low temperatures of $4^{\circ}C$ or $10^{\circ}C$. Elusimicrobia, which are endosymbionts of flagellate protists, is the dominant phylum in the termite gut at ${\geq}15^{\circ}C$ but its population decreased drastically at low temperature. Four representative bacterial strains isolated from R. speratus KMT001 in a previous study produced maximum ${\beta}$-glucosidase levels within the temperature range of $10^{\circ}C-30^{\circ}C$. Elizabethkingia sp. BM10 produced ${\beta}$-glucosidase specifically at $10^{\circ}C$. This strain supported the existence of symbiotic bacteria for the low temperature habitat of the termite. This identified bacterium will be a resource for studying low temperature adaptation of termites, studying the gene expression at low temperatures, and developing an industrial cellulase at low temperature.

Interactions between NCR+ILC3s and the Microbiome in the Airways Shape Asthma Severity

  • Asthma is a heterogeneous disease whose development is shaped by a variety of environmental and genetic factors. While several recent studies suggest that microbial dysbiosis in the gut may promote asthma, little is known about the relationship between the recently discovered lung microbiome and asthma. Innate lymphoid cells (ILCs) have also been shown recently to participate in asthma. To investigate the relationship between the lung microbiome, ILCs, and asthma, we recruited 23 healthy controls (HC), 42 patients with non-severe asthma, and 32 patients with severe asthma. Flow cytometry analysis showed severe asthma associated with fewer natural cytotoxicity receptor (NCR)+ILC3s in the lung. Similar changes in other ILC subsets, macrophages, and monocytes were not observed. The asthma patients did not differ from the HC in terms of the alpha and beta-diversity of the lung and gut microbiomes. However, lung function correlated positively with both NCR+ILC3 frequencies and microbial diversity in the lung. Sputum NCR+ILC3 frequencies correlated positively with lung microbiome diversity in the HC, but this relationship was inversed in severe asthma. Together, these data suggest that airway NCR+ILC3s may contribute to a healthy commensal diversity and normal lung function.

서울 진오기굿의 재차구성과 의미 (The Composition and Principles of Seoul Jinogigut (Shamanistic Ritual))

  • 이 글은 서울지역에서 행해지는 진오기굿의 구성 원리를 도출하고 이를 바탕으로 의미를 규명한 글이다. 진오기굿은 신과 인간이 복합적으로 연결되어 있으면서, 이승과 저승이 혼재되어 있는 세계이고, 인간의 세계에 있던 망자가 신의 세계로 들어가는 과정을 보여주는 의례이다. 진오기굿은 망자를 저승으로 인도하는 과정을 차례로 보여준다. 먼저 신 중심의 거리가 연행되는데, 신이 어떤 과정을 통해 망자를 저승으로 인도할지를 보여준다. 다음으로는 신과 인간이 중심이 되는 거리로 사자를 따라 저승으로 가는 망자의 모습을 보다 구체적으로 보여준다. 마지막으로 인간 중심의 구조로 인간이 어떻게 저승으로 들어가는지를 보여준다. 이러한 반복을 통해 망자가 마침내 저승에 좌정했음을 드러낸다. 신 중심의 굿거리가 신과 인간의 만남을 거쳐 인간 중심으로 연행되는 것이 진오기굿 후반부 저승굿의 짜임이다. <도령돌기> 이후에 연행되는 <베가르기>, <뒷영실>은 망자가 온전하게 저승에 들어갔음을 확인하는 의례이다. 베를 갈라 망자가 저승으로 들어가는 것이 완벽하게 이루어졌음을 보여주고, <뒷영실>에서 망자는 다시 한 번 굿청에 들어와 굿을 해 준 것에 대한 감사함과 함께 죽음의 세계에 안착했음을 알려준다. 저승에 좌정한 조상신에게 비로소 제사를 올리는 <상식거리>는 진오기굿을 통해 인간이었던 망자가 조상신으로 변화했음을 알려준다. 진오기굿은 갑작스러운 죽음을 겪은 재가집을 위로해주는 기능도 수행한다. 재가집의 위로를 위해 가장 필요한 것은 망자가 무사히 저승으로 갔음을 보여주는 것이다. 진오기굿은 그러한 과정을 삼중구조를 통해 보여준다. 신들의 세계에서 망자가 어떻게 천도될 것인가를 보여주고, 저승에서 이승으로 나온 사재를 따라 망자가 무사히 저승으로 들어감을 보여준다. 다시 바리공주 뒤를 따라 저승으로 들어가는 망자의 모습을 보여주어 남은 가족의 마음을 달래준다.

Intestinal absorption of aloin, aloe-emodin, and aloesin; A comparative study using two in vitro absorption models

  • Aloe products are one of the top selling health-functional foods in Korea, however the adequate level of intake to achieve desirable effects are not well understood. The objective of this study was to determine the intestinal uptake and metabolism of physiologically active aloe components using in vitro intestinal absorption model. The Caco-2 cell monolayer and the everted gut sac were incubated with $5-50{\mu}M$ of aloin, aloe-emodin, and aloesin. The basolateral appearance of test compounds and their glucuronosyl or sulfated forms were quantified using HPLC. The % absorption of aloin, aloe-emodin, and aloesin was ranged from 5.51% to 6.60%, 6.60% to 11.32%, and 7.61% to 13.64%, respectively. Up to 18.15%, 18.18%, and 38.86% of aloin, aloe-emodin, and aloesin, respectively, was absorbed as glucuronidated or sulfated form. These results suggest that a significant amount is transformed during absorption. The absorption rate of test compounds except aloesin was similar in two models; more aloesin was absorbed in the everted gut sac than in the Caco-2 monolayer. These results provide information to establish adequate intake level of aloe supplements to maintain effective plasma level.
