Three-dimensional evaluation of the relationship between dental and basal arch forms in normal occlusion (정상교합자에서 치열궁과 기저궁 관계의 삼차원적 평가)

  • Objective: The purposes of this study were to evaluate the relationship between the dental and basal arch forms; to analyze their differences in the tapered, ovoid, and square arch forms in normal occlusion by using three-dimensional (3D) virtual models; and to test the hypothesis that the overjet and maxillomandibular basal arch width difference have a significantly positive correlation. Methods: Seventy-seven normal occlusion plaster casts were examined by 3D scanning. Facial axis (FA) and WALA points were digitized using the Rapidform 2006 software. The dimensions of the dental and basal arches and the overjet were measured. The samples were classified into 3 groups according to arch forms: tapered (n = 20), ovoid (n = 20), and square (n = 37). Analysis of variance (ANOVA) was used to compare the dental and basal arch dimensions. The Pearson correlation coefficients between the intercanine as well as the intermolar widths at the FA and WALA points were calculated. Results: With regard to the basal arch dimensions, the tapered arch form showed a larger mandibular intermolar depth than the ovoid. Strong correlations were noted between the basal and dental intermolar widths in both the upper and lower arches (r = 0.83 and 0.85, respectively). Moderate correlation was found between the upper and lower intercanine widths (r = 0.65 and 0.48, respectively). Conclusions: The 3 dental arch form groups differed only in some dimensions of the skeletal arch. Moderate correlations were found between the basal and dental intercanine widths. These findings suggest that the basal arch may not be a principle factor in determining the dental arch form.

Early bone healing onto implant surface treated by fibronectin/oxysterol for cell adhesion/osteogenic differentiation: in vivo experimental study in dogs

  • Purpose: This study aimed to evaluate the effects of fibronectin and oxysterol immobilized on machined-surface dental implants for the enhancement of cell attachment and osteogenic differentiation, on peri-implant bone healing in the early healing phase using an experimental model in dogs. Methods: Five types of dental implants were installed at a healed alveolar ridge in five dogs: a machined-surface implant (MI), apatite-coated MI (AMI), fibronectin-loaded AMI (FAMI), oxysterol-loaded AMI (OAMI), and sand-blasted, large-grit, acid-etched surface implant (SLAI). A randomly selected unilateral ridge was observed for 2 weeks, and the contralateral ridge for a 4-week period. Histologic and histometric analyses were performed for the bone-to-implant contact proportion (BIC) and bone density around the dental implant surface. Results: Different bone healing patterns were observed according to the type of implant surface 2 weeks after installation; newly formed bone continuously lined the entire surfaces in specimens of the FAMI and SLAI groups, whereas bony trabecula from adjacent bone tissue appeared with minimal new bone lining onto the surface in the MI, AMI, and OAMI groups. Histometric results revealed a significant reduction in the BIC in MI, AMI, and OAMI compared to SLAI, but FAMI demonstrated a comparable BIC with SLAI. Although both the BIC and bone density increased from a 2- to 4-week healing period, bone density showed no significant difference among any of the experimental and control groups. Conclusions: A fibronectin-coated implant surface designed for cell adhesion could increase contact osteogenesis in the early bone healing phase, but an oxysterol-coated implant surface designed for osteoinductivity could not modify early bone healing around implants in normal bone physiology.

  • The stress concentration factors and stress intensity factors for a simple beam and a cantilever are analyzed by using finite element method and phtoelasticity. Using the analyzed results, the estimated graphs on stress concentration factors and stress intensity factors are obtained. To analyze stress concentration factors of notch, the dimensionless notch length H(height of specimen)/h=1.1~2 and dimensionless gap space r(radius at the notch tip)/h=0.1~0.5 are used. where h=H-c and c is the notch length. As the notch gap length increases and the gap decreases, the stress concentration factors increase. Stress concentration factors of a simple beam are greater than those of a cantilever beam. However, actually, the maximum stress values under a load, a notch length and a gap occur more greatly in the cantilever beam than in the simple beam. To analyze stress intensity factors, the normalized crack length a(crack length)/H=0.2~0.5 is used. As the length of the crack increases, the normalized stress intensity factors increase. The stress intensity factors under a constant load and a crack length occur more greatly in the cantilever beam than in the simple beam.

Seroprevalence of Toxoplasmosis with ELISA and Rapid Diagnostic Test among Residents in Gyodong-do, Inchon city, Korea: A Four-Year Follow-up

  • ELISA has been used for the diagnosis of toxoplasmosis, but it is being gradually replaced by a rapid diagnostic test (RDT). We compared and analyzed ELISA and RDT results using the sera collected during 4 consecutive years from residents of Gyodong-do (Island), Incheon-city, Korea. Sera from 921, 993, 940, and 838 adult residents were collected on a yearly basis (2010-2013). ELISA was performed by using a crude extract of T. gondii RH strain antigen and IgG/IgM RDT mounted with recombinant fragment of major surface antigen (SAG1), GST-linker-SAG1A, were applied to the sera. Comparison between groups was analyzed by the Student's t-test. The positive seroprevalence surged from 14.7% (135/921, 2010), 23.1% (231/993, 2011), 23.6% (222/940, 2012), and 32.1% (269/838, 2013) by ELISA. In contrast, RDT showed a more moderate increasing trend from 21.7% (200/921, 2010), 25.5% (253/993, 2011), 28.9% (272/940, 2012) and 33.1% (277/838, 2013). Discrepancies between ELISA and RDT were noted near the cut-off value. At the OD 0.15-0.24 range, RDT could detect 16.1% (169/1051) more positives, which suggests an early or acute toxoplasmosis, but at the OD 0.25-0.34 range, ELISA could detect 35.9% (92/256) more positives of possible chronic infections. Over the OD > 0.35 ELISA and RDT agreed in the majority of the cases. This surge in seroprevalence may be caused by the organic agriculture in addition to eating behavior or increase in pets among Koreans. These facts may be applied on a full-scale national survey using RDT to supplement ELISA to define the characteristics of the infection.

Comparative Analysis Study of Introduction to Industry textbook for Technical High School (전문계 고등학교의 현행 공업입문 교과서 비교연구)

  • The major purposes of this study were 1) to present a desirable guideline for writers who would develop the new Introduction-to-Industry textbook through the comparative analysis of 4 different kinds of current Introduction-to-Industry textbooks, 2) to help educators, who make national college entrance exam in the vocational inquiry area, make good exam items with the results of the study, and 3) to offer effective methods of teaching and learning in the field of industrial education with them. Research method used in this study was mainly literature review with the scrutinization of the literature related to the Introduction-to-Industry textbooks. Results of this study showed that all textbooks compared do mostly comply with the national writer's specific guidelines for the Introduction-to-Industry textbooks. However there were varieties for the all textbooks in a sense of deployment balance for the contents of the textbook chapters. There were also considerable differences for the deployment of supplementary materials, evaluative materials, visual materials and etc. according to the characteristics of each textbook. The degree of the connectivity between the textbooks and the national college entrance exam in the area of the vocational inquiry was similar for the each textbook. However, it was not good enough for the students to study one textbook for the preparation of the exam in the vocational inquiry area because the problems in the exam were set in the range of covering the all different kinds of textbooks by the examiners.

Improvement of Cellobiose Dehydrogenase(CDH) and $\beta$-Glucosidase Activity by Phanerochaete chrysosporium Mutant (Phanerochaete chrysosporium 변이주에서의 Cellobiose Dehydrogenase(CDH)와 $\beta$-Glucosidase 활성 향상)

  • Cellobiose dehydrogenase(CDH) as a hemoflavoenzyme is secreted out of cell in the cellulose degradation. As CDH strongly bound to amorphous cellulose, it helps cellulose hydrolysis by cellulase. CDH may have an important role of saccharification process for bioethanol production. In this study, Phanerochaete chrysosporium ATCC 32629 was selected for the production of CDH among other strains tested. The optimal temperature and pH of CDH produced by P. chrysosporium ATCC 32629 were ${55^{\circ}C}$ and 4, respectively. To improve the activity of CDH, the mutation of P. chrysosporium was performed using proton beam that has high energy level partially. As a result, P. chrysosporium mutant with the high activity was selected at 1.2 kGy in a range of 99.9% lethal rate. The CDH and $\beta$-glucosidase activities of mutant were 1.4 fold and 20 fold higher than those of wild strain. Therefore, P. chrysosporium mutant with the high activities of CDH and $\beta$-glucosidase was obtained from mutation by proton beam irradiation.

An Analytical Study on Generation of Pore-Water Pressures Induced by Flow and Waves in Seabed, and Resulting Liquefaction (흐름과 파에 의한 해저지반내 간극수압의 발생과 액상화에 관한 해석적인 연구)

  • Analytical solutions for interaction between seabed and waves such as progressive wave or partial standing wave with arbitrary reflection ratio or standing wave have been developed by many researchers including Lee et al.(2014; 2015a; 2015b; 2015c; 2015d) and Yamamoto et al.(1978). They handled the pore-water pressure as oscillating pore-water pressure and residual pore-water pressure separately and discussed the seabed response on each pore-water pressure. However, based on field observations and laboratory experiments, the oscillating and residual pore-water pressures in the seabed do occur not separately but together at the same time. Therefore, the pore-water pressure should be investigated from a total pore-water pressure point of view. Thus, in this paper, the wave-induced seabed response including liquefaction depth was discussed among oscillating, residual, and total pore-water pressures' point of view according to the variation of wave, seabed, and flow conditions. From the results, in the field of flow with the same direction of progressive wave, the following seabed response has been identified; with increase of flow velocity, the dimensionless oscillating pore-water pressure increases, but the dimensionless residual pore-water pressure decreases, and consequently the dimensionless total pore-water pressure and the dimensionless liquefaction depth decrease.

Optimization of a Virus-Induced Gene Silencing System with Soybean yellow common mosaic virus for Gene Function Studies in Soybeans

  • Virus-induced gene silencing (VIGS) is an effective tool for the study of soybean gene function. Successful VIGS depends on the interaction between virus spread and plant growth, which can be influenced by environmental conditions. Recently, we developed a new VIGS system derived from the Soybean yellow common mosaic virus (SYCMV). Here, we investigated several environmental and developmental factors to improve the efficiency of a SYCMV-based VIGS system to optimize the functional analysis of the soybean. Following SYCMV: Glycine max-phytoene desaturase (GmPDS) infiltration, we investigated the effect of photoperiod, inoculation time, concentration of Agrobacterium inoculm, and growth temperature on VIGS efficiency. In addition, the relative expression of GmPDS between non-silenced and silenced plants was measured by qRT-PCR. We found that gene silencing efficiency was highest at a photoperiod of 16/8 h (light/dark) at a growth temperature of approximately $27^{\circ}C$ following syringe infiltration to unrolled unifoliolate leaves in cotyledon stage with a final SYCMV:GmPDS optimal density $(OD)_{600}$ of 2.0. Using this optimized protocol, we achieved high efficiency of GmPDS-silencing in various soybean germplasms including cultivated and wild soybeans. We also confirmed that VIGS occurred in the entire plant, including the root, stem, leaves, and flowers, and could transmit GmPDS to other soybean germplasms via mechanical inoculation. This optimized protocol using a SYCMV-based VIGS system in the soybean should provide a fast and effective method to elucidate gene functions and for use in large-scale screening experiments.

Spatial-temporal Variations of Nitrate Levels in Groundwater of Jeju Island, Korea: Evaluation of Long-term (1993-2015) Monitoring Data (제주도 지하수질산염 농도의 시·공간적변화 특성: 장기(1993-2015) 모니터링 자료의 평가)

  • The spatio-temporal variations of nitrate concentrations in groundwater of Jeju Island were evaluated by an analysis of time series groundwater quality data (N = 21,568) that were collected from regional groundwater monitoring (number of wells = 4,835) for up to 20 years between 1993 and 2015. The median concentration of $NO_3-N$ is 2.5 mg/L, which is slightly higher than those reported from regional surveys in other countries. Nitrate concentrations of groundwater in wells tend to significantly vary according to different water usage (of the well), administrative districts, and topographic elevations: nitrate level is higher in low-lying agricultural and residential areas than those in high mountainous areas. The Mann-Kendall trend test and Sen's slope analysis show that nitrate concentration in mid-mountainous areas tends to increase, possibly due to the expansion of agricultural areas toward highland. On the other hand, nitrate concentrations in the Specially Designated Groundwater Quality Protection Zones show the temporally decreasing trend, which implies the efficiency of groundwater management actions in Jeju. Proper measures for sustainable groundwater quality management are suggested in this study.

Diversity and Plant Growth-Promotion of Endophytic Fungi Isolated from the Roots of Plants in Dokdo Islands (독도의 자생식물 뿌리에서 분리한 내생진균의 다양성과 생장촉진활성)

  • Endophytic fungi were isolated from the roots of plants growing naturally on the island of Dokdo. Plant samples, such as Miscanthus sinensis, Achyranthus japonica and Echinochloa crusgali were isolated from Dongdo, and those such as Honkenya peploides and Artemsia koidzumii were isolated from Seodo. Twenty one strains of endophytic fungi were isolated from these plants. To identify the strains, PCR (polymerase chain reaction) amplification of the partial ITS (Internal Transcribed Spacer) regions was done with universal primers ITS-1 and ITS-4 to determine the nucleotide sequence of the ITS regions. Of the strains isolated from Miscanthus sinensis, 75% were Penicillium sp. and 25% were Aspergillus sp. Fifty five percent of strains isolated from Achyranthus japonica were Penicillium sp., 30% were Aspergillus sp. and 15% were Zygorhynchus sp. Strains isolated from Echinochloa crusgali were Penicillium sp. (50%), Aspergillus sp. (12%), Giberella sp. (13%), Talaromyces sp. (9%) and Umbelopsis sp. (8%). Of the strains isolated from Honkenya peploides, 76% were Penicillium sp. and 24% were Pestalotiopsis sp. Strains isolated from Artemisia koidzumii were Penicillium sp. (81%) and Mucor sp. (19%). As a result of bioassay, Ec-3-1 strain isolated from Echinochloa crusgalli showed plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp. was the most abundantly distributed fungal strain in all plants used in this study.
