AcrAB-TolC, a major efflux pump in Gram negative bacteria: toward understanding its operation mechanism

  • Antibiotic resistance (AR) is a silent pandemic that kills millions worldwide. Although the development of new therapeutic agents against antibiotic resistance is in urgent demand, this has presented a great challenge, especially for Gram-negative bacteria that have inherent drug-resistance mediated by impermeable outer membranes and multidrug efflux pumps that actively extrude various drugs from the bacteria. For the last two decades, multidrug efflux pumps, including AcrAB-TolC, the most clinically important efflux pump in Gram-negative bacteria, have drawn great attention as strategic targets for re-sensitizing bacteria to the existing antibiotics. This article aims to provide a concise overview of the AcrAB-TolC operational mechanism, reviewing its architecture and substrate specificity, as well as the recent development of AcrAB-TolC inhibitors.

  • 신규 살충제인 KH-502의 안전성을 조사하기 위하여 다양한 조건하에서 광(光) 분해(分解) 실험을 수행하였다. 이 실험에서 KH-502를 acetone 또는 acetonitrile 용액에 첨가한 뒤 태양광선 또는 자외선 $(300{\sim}350nm)$을 조사(照射)하였고, acetone 또는 acetonitrile 용액에는 수분함량, 산소함량이 달랐으며, 부식산,rosebengal 또는 tryptophan이 첨가 되었다. KH-502의 안전성 및 분해경향에 대한 결과를 요약하면 다음과 같다. 1. KH-502의 광분해(光分解)는 자외선을 조사(照射)한 경우와 그렇치 않은 경우에 매우 유의성있는 차이를 보여 주었다. KH-502는 acetone 용매에서 광관여효과에 의해 광분해(光分解)되었으나, acetonitrile 용매 하(下)에서는 광분해되지 않았다. 2. KH-502의 광분해 경향은 열 분해 경향과 판이하게 달랐으며, 광분해산물은 KH-502 oxo형, S-ethyl KH-502, PTMHP 및 몇가지 미지물질 등이었다. 3. acetone 또는 acetonitrile에 첨가된 부식산, rosebengal, tryptophan은 KH-502의 광분해에 광관여효과를 나타내지 않았다. acetone 용매의 용존산소는 acetone과 함께 경쟁적으로 광관여효과를 나타내어 KH-502의 분해를 촉진시켰다. 4. acetone 용매를 부식산 또는 논물로 처리했을때, KH-502의 광분해는 감소하였다.

  • 근육의 수축 및 사후강직은 myosin 과 actin 그리고 ATP 와의 상호작용에 의한 것임은 널리 알려진 정설(定說)이다. 최근 myosin 과 actin 및 ATP 의 상호작용이 근조절단백질의 지배를 받고 Ca ion 이 관여하고 있다는 것이 알려졌다. 그런데 이들 조절단백질은 수용성단백질로써의 성질을 가지고 있음에도 불구하고 염용성단백질 구분에 들어 있다. 본 연구의 목적은 염용성단백질 구분에 들어 있는 이들 조절단백질의 새로운 분리정제방법을 연구하는 데 있었다. 이를 위하여 본 연구에서는 새로운 정제방법의 flow sheet 를 작성하였다. 이 제안된 새로운 정제방법은 근원섬유중의 조절단백질의 함량을 정량적으로 추적할 수 있는 장점을 가지고 있다는데 그 특색이 있다.

Tuberculin에 관한 연구(硏究) - (2) 항원감작적혈구응집반응(抗原感作赤血球凝集反應)에 의한 결핵사균면역(結核死菌免疫) Guinea pig 혈제(血濟)의 적혈구응집(赤血球凝集) 항체가(抗體價) 소장(消長) (Studies on the Tuberculin - 2) Serological observation of hemagglutinating antibody in guinea pigs sensitized with heat killed tubercle bacilli)

  • In order to assay one lot of the potency of tuberculin for intradermic use, four guinea pigs sensitized with heat killed M. tuberculosis were used by the method originally of the Agriculture Research Service Center(A. R. S.) and Bureau of Animal. Industry(B. A. I.) in the United. States of America in which guinea pigs were used on 21st day after sensitization. There is doubt whether the guinea pigs are sensitized or not. Therefore, this study has taken the method of Middle Brook-Dubos hemagglutination test, and observed the humoral hemagglutinating antibody in the blood. The results obtained are as follows. 1. The maximum titer of hemagglutinating antibody was demonstrated 30days after sensitizing the guinea pigs with killed tubercle bacilli. The maximum titer was continued for a period of 45 days and thereafter declined gradually. 2. The swelling and redness of the region of the sensitized guinea pigs was shown to be reached to the maximum size within 24 hours after intradermal inoculation with HCSM tuberculin and gradually reduced 96 hours after inoculation. The swelling and redness could not be detected by macro-observation after 14 days.

은도금 중공미세구를 이용한 경량 전파흡수체의 제조 (Fabrication of Light Weighted Microwave Absorbers Using Silver-Coated Hollow Microspheres)

  • Conductive microspheres with a density of 0.2 g/cc were fabricated by electroless silver plating for application to the light-weighted microwave absorbers. The silver plating was conducted with the variation of plating conditions (sensitizing condition, $AgNO_3$, concentration, amount of reducing agent). Specimens have very low electro-resistivity. Under an optimum processing condition, conductive microspheres with uniform silver plating layer can be produced. Rubber-sphere composites were fabricated and their microwave absorbing properties were measured by HP8722D Network Analyzer. It was found that the lower the electrical resistance of microsphere, the better the microwave absorbing properties. Feasibility of microwave absorbers using this microspheres can be demonstrated with the result of microwave reflection loss of -15 dB and thickness of 1.44 mm.

Octylmethoxycinnamate의 SPF 지수와 피부자극지수와의 상관성 연구 (Studies on the Correlation between SPF index and Primary Irritation Index of Octylmethoxycinnamte)

  • In recent years, the safety of sunscreens have been challenged based on the reports of its adverse effects on users; dermatitis, allergic contact dermatitis, photoallergic contact dermatitis. In the toxicological aspect, the need to investigate the safety of sunscreens is steadily increasing. However, there were few studies on the correlation between sun protection factor (SPF) and the safety of sunscreens. The objective of this study was to assess whether there was a correlation between SPF and the skin irritation index of sunscreens. Octyl methoxycinnamate (OMC) was selected, which was the most frequently used (about 92% in korea and about 90% in the world) in sunscreens. In vitro SPF test was performed using the standard samples recommended by KFDA guideline. We examined primary and cumulative skin irritation and eye irritation of OMC standard sample as well as homosalate. As in vitro SPF was increased, skin irritation index was increased in primary skin irritation test ($r^2$=0.986) and cumulative skin irritation test ($r^2$=0.939). In addition, OMC was non-irritating and non-sensitizing. In this results, there was a good correlation between SPF index and skin irritation index.

  • This review summarizes the cause of allergy and control by lactic acid bacteria. Atopic diseases such as asthma, rhinitis, eczema and food allergy have increased in most industrialized countries of the world during the last 20 years. The reasons for this increase are not clear and different hypotheses have been assessed including increased exposure to sensitizing allergens or decreased stimulation of the immune system during critical periods of development. Probiotic bacteria, which beneficially affect the host by improving its microbial balance, may mediate anti-allergenic effects by immune stimulation. Although more clinical evidences are required, the possible role of specific LAB strains in the prevention of allergic diseases has become more evident. since the role of functional food is important for prevention, it is expected for the more anti-allergy fermented milk products to be on the market.

Long Term Therapeutic Plan for Patients with Non-Small Cell Lung Cancer Harboring EGFR Mutation

  • Non-small cell lung cancer harboring epidermal growth factor receptor (EGFR) sensitizing mutations has a distinct disease entity. Patients with this cancer have better prognosis, and frequently achieve long-term survival. EGFR-tyrosine kinase inhibitor (TKI) is the drug of choice for this cancer; but the disease inevitably progresses, after durable response. The tumor is a mixture of EGFR-TKI sensitive clones and resistant clones, regardless of their molecular mechanisms. EGFR-TKI sensitive clones are very susceptible to this drug, but rarely eradicated; so, withdrawal of the drug permits rapid regrowth of drug sensitive clones, possibly causing "disease flare." Re-administration or continuation of EGFR-TKI can effectively suppress the expansion of drug sensitive clones, even when the total tumor volume continuously increases. Chemotherapy can definitely prolong the survival of patients experiencing EGFR-TKI failure. Prospective clinical trials are warranted to compare efficacies of chemotherapeutic agents. A few retrospective studies suggested that a taxanebased regimen may be superior to others. Here, we reviewed therapeutic options and clinical evidence about this unique disease entity.

Phospholipase D and Its Essential Role in Cancer

  • The role of phospholipase D (PLD) in cancer development and management has been a major area of interest for researchers. The purpose of this mini-review is to explore PLD and its distinct role during chemotherapy including anti-apoptotic function. PLD is an enzyme that belongs to the phospholipase super family and is found in a broad range of organisms such as viruses, yeast, bacteria, animals, and plants. The function and activity of PLD are widely dependent on and regulated by neurotransmitters, hormones, small monomeric GTPases, and lipids. A growing body of research has shown that PLD activity is significantly increased in cancer tissues and cells, indicating that it plays a critical role in signal transduction, cell proliferation, and anti-apoptotic processes. In addition, recent studies show that PLD is a downstream transcriptional target of proteins that contribute to inflammation and carcinogenesis such as Sp1, $NF{\kappa}B$, TCF4, ATF-2, NFATc2, and EWS-Fli. Thus, compounds that inhibit expression or activity of PLD in cells can be potentially useful in reducing inflammation and sensitizing resistant cancers during chemotherapy.

고려인삼과 당뇨병: 세포와 동물 및 인체실험을 통한 고려인삼의 당뇨병에 대한 효능 (Korean Ginseng and Diabetes: An Insight into Antidiabetic Effects of Korean Ginseng (Panax ginseng C. A. Meyer) in Cultured Cells, Animal Models and Human Studies)

  • Diabetes mellitus, commonly known as diabetes, is a group of metabolic disorders characterized by high blood sugar levels over a prolonged period. Diabetes has been found to show many acute complications such as cardiovascular disease, stroke, chronic kidney disease, foot ulcer and damage to eyes. Korean ginseng (Panax ginseng) has been traditionally known to normalize the functional deficiencies of the lung, spleen and stomach, and thus improve the secretion of body fluids, thereby quenching thirst, suggesting it to be effective in the treatment of diabetes. Experimental studies (in vitro and in vivo) have recently shown that Korean ginseng and its extracts exhibit antidiabetic effects, and also insulin secretion and sensitizing effects related to blood glucose control. Moreover, clinical trials on antidiabetic effects of Korean ginseng have been reported to show blood glucose control, improvement of insulin resistance, reduction of postprandial blood glucose level and improvement of serum lipids (TG, TC, LDL-C). These will be critically examined by means of in vitro studies, cell experiment, animal models and human trials with a focus on understanding of molecular mechanisms.
