Acremonium chrysogenum was immobilized in ionotropic gel beads to develop semi-continuous production of cephalosporin C (CPC). Barium alginate beads were more stable than calcium alginate or strontium alginate beads in chemically defined media. The gel stability of Ba-alginate was further increased by cross-linking with polyethyleneimine (PEI). The presence of carboxymethyl cellulose inside Ba-alginate beads did not reduce mass transfer resistance. Ba-alginate microbeads that had little diffusion limitation increased CPC production rate 1.6 fold higher than that of normal beads. CPC fermentation with immobilized cells in Ba-alginate microbeads was performed continuously for 40 days by way of repeated fed-batch operations. Mathematical modeling was developed to describe the repeated fed-batch fermentation system. Results of the computer simulation agreed well with the experimental data, which made it possible to predict an optimal feeding rate that could maximize total CPC productions.

클래스라벨에 대한 정보를 가진 데이터가 매우 적을 때 클래스라벨 정보가 없는 데이터로부터의 정보를 학습에 활용하는 반감독학습(semi-supervised learning) 방법들 중에서 데이터 샘플들 간의 유사도를 나타내는 그래프를 이용하는 방법이 잘 알려져 있다. 본 논문에서는 그래프 기반 반감독 학습에서 배치 모드 능동적 학습을 위한 방법을 제안하고 실제 데이터를 이용한 실험결과를 통해 제안된 방법의 성능을 입증한다.
The effect of operating parameters (reaction temperature and time) and reaction modes (batch and semi-batch) on the behavior of amino acid production from hydrothermal decomposition of fish-derived wastes was investigated. The amino acids obtained in batch experiments at temperature of 250$^{\circ}C$ were mainly alanine (Ala) and glycine (Gly) at maximum yield of 65 and 28mg/g-dry fish, respectively. At relatively lower temperature of 200$^{\circ}C$, the yield of high-molecular-weight amino acids such as aspartic acid (Asp) and serine (Ser) is high, but decreases as temperature increases. It is likely that high-molecular-weight amino acids decompose faster than low-molecular ones. Semi-batch mode of reaction suppressed decomposition of amino acids into organic acids (or volatile materials) by continuously removing the products from the reaction zone as soon as they are formed. Thus, large amount of high-molecular-weight amino acids such as Asp and Ser at this reaction mode was observed.

In this paper, I propose a response modeling with a Semi-Supervised Support Vector Regression (SS-SVR) algorithm. In order to increase the accuracy and profit of response modeling, unlabeled data in the customer dataset are used with the labeled data during training. The proposed SS-SVR algorithm is designed to be a batch learning to reduce the training complexity. The label distributions of unlabeled data are estimated in order to consider the uncertainty of labeling. Then, multiple training data are generated from the unlabeled data and their estimated label distributions with oversampling to construct the training dataset with the labeled data. Finally, a data selection algorithm, Expected Margin based Pattern Selection (EMPS), is employed to reduce the training complexity. The experimental results conducted on a real-world marketing dataset showed that the proposed response modeling method trained efficiently, and improved the accuracy and the expected profit.
Several culture systems including batch, two-stage CSTR, semi-fed batch, and two-stage cyclic fed-batch were investigated for the efficient production of the Fab fraction of PDC-E2 specific human monoclonal antibody using high cell density recombinant E. coli. A two-phase batch system and a two-stage continuous system were examined to overcome plasmid instability problems, by separating the growth and the production stages. The cell density and productivity of the two-stage continuous culture was better than that of the two-phase batch fermentation. In the two-stage continuous culture system with DO-stat, the cell growth and the productivity were superior to those of the system without the DO control. Also, almost total plasmid stability was maintained in the two-stage continuous culture system. Modified M9 medium was selected as an optimum feeding medium for the fed-batch process, and the optimum C/N ratio determined to be 2:3. The optimum feeding rate was $0.6g/\ell/hr$ for a constant feeding strategy in semi-fed batch system. When the feeding medium was fed by pulsing, it was observed that more frequent pulsing resulted in improved cell growth. The linear feeding method was the most efficient of the various feeding methods tested. Finally, high cell density culture using a two-stage cyclic fed batch system with pH-stat was tried because the linear feeding method showed limitations in terms of obtaining high cell densities, and a cell density of $54 g/\ell$ was achieved. It was concluded that the two-stage cyclic fed batch system was the most efficient system for high cell density culture of the systems tested. However, productivity improvements were lower than expected due to the extremely high accumulations of acetate, although the low levels of residual glucose were maintained.
The feasibility of municipal sewage sludge digestion was investigated by using thermophilic anaerobic sequencing batch reactor(ASBR). One-day settle time was enough for the high performance of solid-liquid separation. The conversion of semi-continuous mode to sequencing batch mode is easily achieved without any adverse effects, although the large amount of sludge equal to the volume ratio of 0.3~06 to reactor volume was added in the feed step of the start-up. The ASBRs had higher conversion capability of organics to biogas than the control reactor. Gas yields of the ASBRs were increased by the average of 50% over the control reactor across a range of hydraulic retention time(HRT)s from 10days to 5days. The thermophilic reactors showed higher gas production than mesophilic reactor. Removal efficiencies of organic matter exceeded 80% on the basis of supernatants, except that at the reactor. Solid-liquid separation was essential in the performance of the ASBR, especially, at the lower HFT. The ASBRs were highly efficient in the retention of activated biomass within the reactor. thus compensating for increased equivalent organic loading rate through increased solids retention times followed by the increased solids, while maintaining shorter HRTs.

Four activated carbons were produced by two-stage process as followings; semi-carbonization of indigenous biomass waste, i.e. cotton stalks, followed by chemical activation with KOH under various activation temperatures and chemical ratios of KOH to semi-carbonized cotton stalks (CCS). The surface area, total pore volume and average pore diameter were evaluated by $N_2$-adsorption at 77 K. The surface morphology and oxygen functional groups were determined by SEM and FTIR, respectively. Batch equilibrium and kinetic studies were carried out by using a basic dye, methylene blue as a probe molecule to evaluate the adsorption capacity and mechanism over the produced carbons. The obtained activated carbon (CCS-1K800) exhibited highly microporous structure with high surface area of 950 $m^2/g$, total pore volume of 0.423 $cm^3/g$ and average pore diameter of 17.8 ${\AA}$. The isotherm data fitted well to the Langmuir isotherm with monolayer adsorption capacity of 222 mg/g for CCS-1K800. The kinetic data obtained at different concentrations were analyzed using a pseudo-first-order, pseudo-second-order and intraparticle diffusion equations. The pseudo-second-order model fitted better for kinetic removal of MB dye. The results indicate that such laboratory carbons could be employed as low cost alternative to commercial carbons in wastewater treatment.
A lipid-enriched strain of Botryococcus braunii (UTEX 572) was cultivated in a semi-batch aeration tank to enhance biomass as well as to develop intracellular lipids and fatty acids. A 30 day period of incubation produced 1.39 g/L of biomass and 0.31 g/L of total lipids in the biomass. The grown biomass was pre-treated using several methods to extract the total lipid content efficiently: ultrasonication was found to yield the highest percentage of lipids-namely 19.8% per biomass. Direct heating of biomass in an autoclave also showed better performance than when using only conventional solvent extraction. To enhance the biomass harvest and lipid extraction efficiency, coagulation and flocculation steps were added to the extraction process. It is noteworthy that not only the solvent type but also the solvent/biomass ratio greatly affected efficiency. In addition, the moisture content of the harvested(wet) biomass affected the efficiency significantly. This study elucidated the need for future research on optimizing this extraction process.

The test did for the determine the optimized ratio of cation to anion in mixed ion exchange demineralizers. Binary, ternary, quaternary, and quinary cation and anion adsorption was performed to develop a comprehensive experimental data set from small-volume batch tests to obtain the selectivity coefficients of many cations and anions. The quantitative run time might be estimated by such ion exchange models as semi-empirical mass action and surface complexation models. The demineralizer can be used longer by increasing the ratios of cation to anion exchange resins in the bed.
Lovastatin is a cholesterol-lowering agent, which plays a role of an inhibitor of 3-hydroxy-3- methylglutaryl coenzyme A reductase (HMG-CoA). When thiamine was supplemented in 3L batch fermentation, the production of lovastatin was improved. At the same time, the levels of pyruvic acid and NAD(P)H were estimated in the course of the fermentation of A. terreus. For the high level production of lovastatin, semi fed-batch fermentation was performed. And the thiamine level was maintained to a concentration of 20 mg/L and glucose was supplied. The final dry cell weight was lowered by 30 % and final lovastatin concentration was increased by 33 %. Final lovastatin concentration of 3.3 g/L was achieved in 8 days.
