  • 원자력발전소의 특성에 따라 서로 다른 방사성 폐기물의 핵종 재고량 평가에 필요한 척도인자와 주기적 검증방법을 개발하기 위하여 규제 대상의 핵종으로 검토되고 있는 다수의 핵종 중에서 개별분리가 요구되는 $^{55}Fe$, $^{90}FSr$$^{94}Nb$을 공존 핵종들로부터 선택적으로 분리, 회수할 수 있는 방법에 관하여 기술하였다. 고ㆍ액체 방사성 폐기물과 유사한 화학조성의 모의 방사성 폐기물 용해용액을 사용하여 이온교환수지법과 추출 크로마토그래피로 Fe, Sr, Nb 및 공존원소들의 흡ㆍ탈착거동을 조사하였다. 방사성 폐액의 발생과 분석자의 방사선 피폭을 최소화하기 위하여 한 개의 시료로부터 각 핵종을 순차적으로 회수하도록 분리조건을 최적화하였으며 확립된 분리조건에서 회수율을 측정하고 신뢰도를 평가했다.

  • For-${\alpha}$-APM을 For-Asp anhydride와 Phe-OMe를 MEK, $CH_3CN$, 물 등의 용매를 사용하여 반응시켜 효율적으로 합성하였다. For-${\alpha}$-APM과 For-${\beta}$-APM의 혼합물로부터 순수한 For-${\alpha}$-APM의 회수는 pH 4.00에서의 연속적인 분별추출로 분리 수거가 가능하였다. $H_2_O2$ 공급원으로 $H_2_O2$/THF, 과탄산소다, $H_2_O2$/HCl/MeOH 등의 다양한 산화계를 사용하여, $H_2_O2$에 의한 산화방법으로 탈포밀반응을 성공적으로 수행하여 높은 수율로 아스파르테임을 얻을 수 있었다. 이 때 HCl이나 TsOH와 같은 산이 $H_2_O2$의 분해를 억제시키고 산화능을 증대시킴으로써 탈포밀 반응의 효율을 높일 수 있었다.

Simultaneous Determination of Baicalein, Baicalin, Wogonin, and Wogonoside in Rat Plasma by LC-MS/MS for Studying the Pharmacokinetics of the Standardized Extract of Scutellariae Radix

  • A new composition of standardized Scutellariae Radix extract (HPO12) was developed for treatment of Alzheimer's disease. For the preclinical pharmacokinetic study of HPO12, a rapid, sensitive, and selective LCMS/MS method was developed and validated for the simultaneous determination of 4 bioactive compounds, baicalein, baicalin, wogonin, and wogonoside. After extraction with ethylacetate, chromatographic analysis was performed on a Thermo $C_{18}$ column ($150mm{\times}2.1mm$, $3{\mu}m$) with a mobile phase consisting of 0.1% formic acid (A) and 0.1% formic acid in 95% acetonitrile (B) by using gradient elution at a flow rate of $250{\mu}L/min$. Analytes introduced to a mass spectrometer were monitored by multiple reaction monitoring (MRM) in positive ion mode. Using $25{\mu}L$ of plasma sample, the method was validated over the following concentration ranges: 25-5000 ng/mL for baicalein, 20-40000 ng/mL for baicalin, 1-1000 ng/mL for wogonin, and 5-10000 ng/mL for wogonoside. The intra- and inter-day precision and accuracy of the quality control samples at the 4 concentrations showed $\leq$ 13.7% relative standard deviation (RSD) and 86.6-105.5% accuracy. The method was successfully applied to determine the concentrations of baicalein, baicalin, wogonin, and wogonoside in rat plasma after intraperitoneal and oral administrations of HPO12.

Development of an experimental model for radiation-induced inhibition of cranial bone regeneration

  • Background: Radiation therapy is widely employed in the treatment of head and neck cancer. Adverse effects of therapeutic irradiation include delayed bone healing after dental extraction or impaired bone regeneration at the irradiated bony defect. Development of a reliable experimental model may be beneficial to study tissue regeneration in the irradiated field. The current study aimed to develop a relevant animal model of post-radiation cranial bone defect. Methods: A lead shielding block was designed for selective external irradiation of the mouse calvaria. Critical-size calvarial defect was created 2 weeks after the irradiation. The defect was filled with a collagen scaffold, with or without incorporation of bone morphogenetic protein 2 (BMP-2) (1 ㎍/ml). The non-irradiated mice treated with or without BMP-2-included scaffold served as control. Four weeks after the surgery, the specimens were harvested and the degree of bone formation was evaluated by histological and radiographical examinations. Results: BMP-2-treated scaffold yielded significant bone regeneration in the mice calvarial defects. However, a single fraction of external irradiation was observed to eliminate the bone regeneration capacity of the BMP-2-incorporated scaffold without influencing the survival of the animals. Conclusion: The current study established an efficient model for post-radiation cranial bone regeneration and can be applied for evaluating the robust bone formation system using various chemokines or agents in unfavorable, demanding radiation-related bone defect models.

Clean and Efficient Synthesis of Furfural From Xylose by Microwave-Assisted Biphasic System using Bio-Based Heterogeneous Acid Catalysts

  • As an attempt to replacing petroleum-based chemicals with bio-based ones, synthesis of furfural from biomass-derived xylose attracts much attention in recent days. Conventionally, furfural from xylose has been produced via the utilization of highly corrosive, toxic, and environmentally unfriendly mineral acids such as sulfuric acid or hydrochloric acid. In this study, microwave-assisted biphasic reaction process in the presence of novel bio-based heterogeneous acid catalysts was developed for the eco-benign and effective synthesis of furfural from xylose. The microwave was irradiated for reaction acceleration and a biphasic system consisting of $H_2O$ : MIBK (1 : 2) was designed for continuous extraction of furfural into the organic phase in order to reduce the undesired side products formed by decomposition/condensation/oligomerization in the acidic aqueous phase. Moreover, sulfonated amorphous carbonaceous materials were prepared from wood powder, the most abundant lignocellulosic biomass. The prepared catalysts were characterized by FT-IR, XPS, BET, elemental analysis and they were used as bio-based heterogeneous acid catalysts for the dehydration of xylose into furfural more effectively. For further optimization, the effect of temperature, reaction time, water/organic solvent ratio, and substrate/catalyst ratio on the xylose conversion and furfural yield were investigated and 100% conversion of xylose and 74% yield of furfural was achieved within 5 h at $180^{\circ}C$. The bio-based heterogeneous acid catalysts could be used three times without any significant loss of activity. This greener protocol provides highly selective conversion of xylose to furfural as well as facile isolation of product and bio-based heterogeneous acid catalysts can alternate the environmentally-burdened mineral acids.

스칸듐定量을 위한 改良된 放射化分析法 (An Improved Method for the Determination of Scandium by Neutron Activation Analysis)

  • 모나자이트中의 스칸디움含量을 더욱 簡便히 決定할 수 있도록 改良한 放射化分析法을 記述하였다. 즉 熱中性子束($10^{12}n/cm^2/sec$)에 照射된 試料에 스칸듐擔體와 黃酸을 넣고 加熱融解한 後 물로 稀釋한 試液에 암모니아水를 加하여 稀土類元素를 沈澱分離하였다. 沈澱을 鹽酸에 녹힌 다음 티오시안酸암모늄을 넣어 스칸듐을 에테르로 抽出하였다. 스칸듐의 化學的 回收率은 스칸듐을 만델린酸으로 沈澱시켜서 重量分析法에 依하여 決定하였다. 本分離法에 依한 스칸듐의 分離能과 共存元素의 放射能의 妨害與否는 이온交換樹脂를 使用하여 스칸듐을 分離한 結果와 比較檢討하였다. 스칸듐의 放射能은 Gamma-scintillation spectrometry 에 依하여 計測하였고 韓國産 모나자이트에는 約 12 p.p.m.의 스칸듐의 含有되어 있음을 알았다.

  • Acyclovir (ACV) is one of the most effective and selective agents against viruses of the herpes group. Because of low solubility, bioavailability of ACV has shown below 30% with oral dosage form. In our previous study, we reported that the fabrication of solid dispersion of ACV was possible and the solid dispersion of ACV and PVP was the most useful in all samples. In this study, we examined the effect of mixture ratio of polymers (PEG and PVP) to ACV. Solubility of ACV was dramatically increased up to 25 mg/ml in $80^{\circ}C$ distilled water. So water was used as a solvent to eliminate problem of residual solvent. Spray drying method was used for the solid dispersion of ACV as solvent extraction. Different scanning calorimeter was used to check degradation of drug. Polymer carriers were PEG 6,000 and PVP. In summary, ACV-PVP (1:3) showed the best solubility in distilled water.

오리멀젼회로부터 바나듐 침출특성에 관한 연구 (Leaching Behaviour of vanadium from Orimulsion ash)

  • 중유의 대체연료로서 관심을 받고있는 오리멀젼회로부터 바나듐을 회수하기 위한 기초연구로서 오리멀젼회의 물성과 침출특성에 관한 연구를 수행하였다. 오리멀젼회는 16%의 바나듐, 4%의 니켈 그리고 9%의 황을 함유하고 있으며 $d_{50}$이 5.9$mu extrm{m}$로 미세한 분말이었다. 오리멀젼회 중의 금속성분이 황산염 형태로 존재하기 때문에 바나듐은 수 침출이 용이하고 침출시간도 10분 이하로 짧았다. 수침출 시 반응온도가 높으면 침출율이 감소하는데 이는 V(V)이 가수분해되어 $V_2$$O_{5}$로 침전되기 때문이다. 한편 황산을 첨가하면 바나듐의 침출율을 높일 수 있었다. 바나듐의 선택적 침출을 위한 알카리 침출의 경우 침출율을 높이기 위하여는 산화제의 사용이 필요하며 과산화수소가 적당하였다.

가공식품 중 아크릴아마이드 분석 (Determination of acrylamide in food products)

  • 고속액체크로마토그라피/질량분석기를 이용하여 가공 식품 중 아크릴아마이드를 분석하였다. 대상 식품은 감자칩 (6종), 프렌치후라이 (11종)으로 총 17종이었다. 시료를 균질화하여 3차 증류수로 추출하고 C18 및 혼합이온교환수지 카트리지를 이용하여 정제한 후 LC/MS/MS으로 분석하였다. 이동상으로는 0.1% 초산과 0.5% 메탄올을 함유하는 수용액을 사용하였으며, 대상물질의 특이이온을 ESI 질량분석기로 확인 및 정량하였다. 평균 회수율은 91~101%이었으며, 정량한계는 $10{\mu}g/kg$이었다. 대상식품의 유형에 따라 검출 수준에 차이를 나타냈으며, 평균 검출수준은 감자칩은 0.71 mg/kg, 프렌치후라이는 0.34 mg/kg이었다.

  • Thallium-201 (201Tl) is a medical radioisotope which emits gamma rays when it decays and used in myocardial perfusion scans in single-photon emission tomography due to its similar properties to potassium. Currently, the Korea Institute of Radiological & Medical Sciences is the only institution producing 201Tl in Korea, and optimization of 201Tl production research is necessary to meet supply compared to domestic demand. To this end, technical analysis of plating target production and chemical separation methods essential for 201Tl production research is conducted. It deals with the process of generating and separating 201Tl radioisotope and target production, It can be generated through a nuclear reaction such as natHg(p,xn)201Tl, 201Hg(p,n)201Tl, natPb(p,xn)201Bi → 201Pb → 201Tl, 205Tl(p,5n)201Pb → 201Tl, and considering impure nuclide generated simultaneously with the use of proton beam energy of 35 MeV or less, it is intended to be produced using the 203Tl(p,3n)201Pb→201Tl nuclear reaction. In particular, the chemical separation of Tl is a very important element, and the chemical separation methods that can separate it is broadly divided into four types, including solid phase extraction, liquid-liquid, electrochemical, and ion exchange membrane separation. Some chemical separations require additional separation steps, such as methods using selective adsorption. Therefore, this technical report describes four chemical separation methods and seeks to separate high-purity 201Tl using a method without additional separation steps
