Effect of Osmotic Priming and Solid Matrix Priming to Improved Seed Vigor and Early Growth of Pepper and Tomato Seeds (고추와 토마토 종자의 발아력 증진과 초기생육에 미치는 Osmotic Priming 및 Solid Matrix Priming 처리 효과)

  • Osmotic and solid matrix priming treatments enhanced germination performance. We compared osmotic with solid matrix priming to determine the more effective treatment for improving seed germination in pepper and tomato. Seed hydration was immediately observed after osmotic priming and solid matrix priming treatment. The moisture content of solid matrix primed seeds was lower than that of osmotic primed seeds in the two vegetable crops. Osmotic priming and solid matrix priming did not increased percent germination, but showed shorter number of days to 50% of the final germination percentage ($T_{50}$) compared with untreated seeds, regardless of germination temperature. $T_{50}$ value was reduced in osmotic or solid matrix primed pepper seeds about 6.0, 5.0, 4.6 and 4.0 days compared with untreated seeds at 15, 20, 25, and $30^{\circ}C$, respectively. While, that in tomato seeds was reduced about 3.3, 5.0, 4.6 and 4.0 days compared with untreated seeds at 15, 20, 25, and $30^{\circ}C$, respectively. The effectiveness of osmotic priming or solid matrix priming in reducing the $T_{50}$ was greater when the seeds were germinated at $15^{\circ}C$ than at temperature of higher than $20^{\circ}C$. Solid matrix primed seeds germinated faster than osmotic primed seeds at all temperature in pepper. However, there was no significant difference on the percentage germination between solid matrix and osmotic primed seeds in tomato. After priming, dried-bark seeds showed faster germination than surface-dried seeds in pepper. On the other hand, there was no significant difference in tomato. Emergence of pepper and tomato seeds was markedly enhanced by osmotic priming or SMP treatment although the final emergence percentage was not significantly influenced. On the other hand, early growth was not significantly influenced by osmotic priming or SMP treatment of pepper and tomato seeds.

Characteristics and Germination of Xanthoceras sorbifolia Bunge Seeds Originated from Inner Mongolia and Liaoning, China

  • Seeds of Xanthoceras sorbifolia Bunge were collected from two plantations and two superior trees in Inner Mongolia: and one plantation and one superior tree in Liaoning, China in late August, 2011. Yellowhorn or goldenhorn is an important tree species, from the aspects of source of edible oil and biodiesel and pioneering capacity of degraded and desert land. Characteristics investigated were seed length, width, and weight: weight and volume of 1,000 seeds: and weight and volume of one-liter seeds. The seeds of Qingsonglingxiang No. 1, growing alone in an open space, showed the highest values in seed length (16.08 mm), width (14.48 mm) and weight (1.40 g), while those of Tree No. 160 in Ar Khorqin Banner were the lowest ones: that is, 11.48mm for length, 11.81 mm for width, and 0.73 g for weight, respectively. Traits of seeds varied quite much between trees and among areas; for example, Tree No. 38 and No. 160 produced quite different seeds in several traits, although they are adjacent to each other in the same farm. Weight of 1,000 seeds varied from 718.0 g to 1,010.1 g and volume from 0.76 L to 1.52 L. Weight of one-liter seeds were 522.3 g to 688.2 g, while the number of seeds were 603 to 935. Seeds which were soaked in the water at $4^{\circ}C$ for 2 days showed the highest germination rate (89%) in a 30-day test, which was about 10% to 40% higher than those of non-treatment and dipping treatment at $36^{\circ}C$ followed by keeping under room temperature for 2 days. 81% of seeds in the wet sand at room temperature germinated, while 23% of seeds deprived of seed coat germinated. It is necessary to understand seed traits to select superior clones or provenances for the increased, unfluctuating production of seed.

  • When germination beds of carrot seeds were treated with either 0.01M or 0.05M concentrations of Ca(NO3)2, CaSO4, MgSO4, K2SO4 and KH2PO4, an acceleration in the germination rate was observed in the groups treated with 0.01M KH2PO4 and 0.05M MgSO4 and 0.05M Ca(NO2)3. In earlier work by the author with acetone a similar result was observed and reported. The pH range in these experiments was maintained between 5.0 and 6.0. It was found that the groups treated with 0.05M K2SO4, 0.05M Ca(NO3)2, 0.05M Ca(NO3)2, 0.05M MgSO4, 0.01M KH2PO4, 0.01M Ca(NO3)2 germinated earlier than the control group. The acceleration of the germintion rate varied with the inorganic compounds used in the following descending order; 0.01M KH2PO4, 0.05M Ca(NO3)2, 0.05M K2SO4, 0.05M CaSO4 and 0.05M KH2PO4. As a result of these expriments, it occurs to the author that in the germination of carrot seeds some inorganic compounds appear to activate the osmotic function of carrot seeds causing acceleration in the germination rate.

Non-deep physiological dormancy in seeds of Euphorbia jolkinii Boiss. native to Korea

  • Background: Euphorbia jolkinii Boiss. is a perennial species native to Jeju Island and the southern coastal area of Korea. Particularly on Jeju Island, the yellow flowers of E. jolkinii Boiss. have a high ornamental value because of their contrast with basalt. This study was conducted to investigate the effects of different temperatures (5, 15, 20, and 25 ℃) and gibberellic acid (GA3) concentrations (0, 10, 100, or 1000 mg/L) on seed dormancy and germination of E. jolkinii. In addition, we classified the seed dormancy type and compared types with those of other species in the same genus. Results: The number of seeds with viable embryos and endosperms was approximately 66%. The final germination percentages at 5, 15, 20, and 25 ℃ were 51.7%, 83.5%, 2.6%, and 0.0%, respectively. In GA3 concentration experiments, the final germination percentages of 0, 10, 100, and 1000 mg/L were 83.5%, 91.7%, 79.1%, and 83.4%, respectively, at 15 ℃ conditions, and 0.0%, 6.9%, 13.2%, and 27.3%, respectively, at 25 ℃. Conclusions: Germination improved at temperatures of 15 ℃ or lower. Furthermore, GA3 treatment effectively reduced germination times. Thus, the seeds of E. jolkinni were classified as having non-deep physiological dormancy.

Seed Dormancy and Germination Characteristics of Endemic Elder Species (Sambucus racemosa subsp. pendula) and Common Elder Species (S. williamsii) in Korea

  • This study was conducted to determine the seed dormancy types of Sambucus racemosa subsp. pendula Nakai and S. williamsii Hance. Low temperature stratification (1, 2, 3, 4 months) and GA3 treatment (1,000 mg/L) were performed on seeds to determine the type of seed dormancy. After the treatment, seeds were placed on a petri dish at 25℃ under light conditions. The germination rate and mean germination time were investigated. Results showed that cold stratification was effective in breaking the dormant state of the seed in both species. In the low temperature stratification treatment, the seed germination performance was improved as the treatment period was prolonged. Gibberellin treatment was effective in breaking the dormant state of S. racemosa subsp. pendula without low temperature stratification. However, S. williamsii did not break the dormant state of the seed by gibberellin treatment without low temperature stratification treatment. In the gibberellin treatment, germination performance was improved according to the low temperature stratification treatment period. As a result of this study, the seeds of S. williamsii have both an intermediate complex and a deep complex morphophysiological dormancy (MPD). In comparison, it was found that the S. racemosa subsp. pendula had intermediate composite MPD.

Germination and Physical Characteristics of Pelleted Sesame Seed with Different Materials (참깨종자의 과위화 재료에 따른 발아 및 물리적 특성)

  • A seed pelleting technique was developed for easy handling of small sesame seeds (Chinbaeckkae) and for direct seeding in field. The mixture of pelleting material, binder(sodium alginate) and seeds were dropped in 100mM CaCl$_2$ solution. Charcoal was appeared as a best material among all to make pellets with the characters of hardness, breakdown, destruction rate and germination percentage. Zeolite formed the hardest pellet seeds among the materials and 1~3 seeds contained in a pellet on the treatment of 87g of sesame seeds mixed in 1$\ell$ CaCl$_2$ solution was estimated optimum rate of seed mixure with few appearance of seedless pellet. The moisture absorption rate was greater in the order of peat+charcoal > peat > charcoal > zeolite. Germination percentage of pelleted seeds with charcoal was the best and similar to that of common non-treated seeds. Germination percentage of pelleted seeds by sowing depth of exposing a quarter of pellet seeds on the surface of vermiculite germination test box and were 92% in charcoal treatment and was the best among all released materials.

Desiccation Tolerance and Storage Behavior of Sarcandra glabra Seeds for Use in ex-situ Conservation (현지외 보존을 위한 죽절초 종자의 건조 내성과 저장성 평가)

  • This study sought to determine the desiccation tolerance and storage behavior of Sarcandra glabra seeds to assist in the establishment of an ex-situ conservation strategy for the species. The basic germination characteristics of S. glabra seeds were investigated. Subsequently, the seeds were dried to various desiccation levels to allow for analysis of their germination characteristics and seed integrity. In addition, the seeds were subjected to germination tests after being stored for different durations. The initial seed moisture content following the removal of the fleshy fruit was 32.8%, and the fresh seeds maintained their seed viability even after a decrease in the seed moisture content to 4.8%, while the germination percentage was 81.0%. The germination percentage of the seeds decreased to 55.0% and 30.0% when they were desiccated to moisture contents of 2.3% and 2.2%, respectively. By contrast, when the stored seeds were desiccated, they maintained their seed viability and vigor at a moisture content of 18.6%; however, when the moisture content decreased below 5.0%, both the seed viability and vigor significantly declined and the germination percentage fell below 20.0%. Furthermore, analysis of the seed leachate revealed that the decrease in the seed viability and vigor was associated with both increased electrical conductivity and increased concentrations of inorganic compounds such as potassium and calcium. In conclusion, the findings of this study show S. glabra seeds to possess desiccation tolerance that falls somewhere between the recalcitrant and intermediate seed types, with the observed variations depending on the degree of deterioration.

Effect of Germination Temperature and Light Quality on Germination of Pretreated Gourd Seeds (발아온도 및 발아중 광질 처리가 전처리된 박 종자의 발아에 미치는 영향)

  • Seed germinability might be highly related to seedling establishment. The experiments were carried out to evaluate the effect of presowing treatments of aging, priming, $GA_3$ , prechilling and water imbibition, different temperatures and light quality during germination on germination of gourd (Lagenaria siceraria Standl.) seeds. Aging treatment with different temperatures and periods was done by the accelerated aging method. Priming using KNO$_3$ and $GA_3$ treatment for 24 hours were done at 100 mM and at 0.01 mM before a week prechilling. The germination tests using a week prechilled seeds were done at 10, 20 and $30^{\circ}C$ on the condition of darkness, and also with blue, red, far-red light and darkness as light quality treatments. FR-yongjadaemok and FR-kunghap were used as test cultivars and their daily germination rates were measured at treatment levels. Germination rates were reduced in the seeds aged at $45^{\circ}C$ for 6 days. Prechilling showed the highest and fastest rate among the four presowing treatments but the two other presowing treatments had similar or less rates than water imbibition. Prechilling done before sowing enhanced the low vigor seeds, and accelerated the germination of cv. FR-kunghap at 20 to $30^{\circ}C$ and with blue, red light or darkness during germination although far-red light inhibited their germination.

Variation of Sugar Content in Different Parts of Seedling during Germination in Phaseolus vulgaris (강낭콩 발아중 유묘 부위별 당함량의 변화)

  • The experiment was carried out to the find variation of the sugar contents in the tip and basal part of the cotyledon and remaining portion of embryos in Phaseolus vulgahs seeds during germination with HPLC analysis method. Water content in cotyledon of kidney bean seed was about 6.4~6.5 of fresh weight and incresed to 45.8~71.2% during germination showing that tip part of cotyledon has more water content by 1.1~3.8% compared to the lower part of it. Higher water onten was observed in the rest parts of the seed except cotyledon such as plumule, radicle and hypocotyl showing that it increased to 72.2~93.3% depending on the different tissue organs. Main important sugars in kidney seeds during germination stages are; raffinose, sucrose, glucose and fructose, and the amount are differed with the kinds of embryo in kidney bean seed organs and stages of germination. Raffinose amount in kidney bean is increasing repeatly when seeds become wet but disappear it soon after seed have germinated especially in growing embryo parts. Raffinose in basal parts of cotyledons were still presented some an amount after germination. Sucrose is synthesized highly in plumule embryo at the beginning stage of germination but disappeared it from 5 days after seeding stages. Amount of sucrose in cotyledon of kidney seeds and seedlings increased continuously after germination. The amount of glucose and fructose in the cotyledons of kidney seeds during germination varied 5~10% or 5~15% but in the germinating and growing organs, plumule, they increased continuously after germination.

Inhibition of Seed Germination and Induction of Systemic Disease Resistance by Pseudomonas chlororaphis O6 Requires Phenazine Production Regulated by the Global Regulator, GacS

  • Seed coating by a phenazine-producing bacterium, Pseudomonas chlororaphis O6, induced dose-dependent inhibition of germination in wheat and barley seeds, but did not inhibit germination of rice or cucumber seeds. In wheat seedlings grown from inoculated seeds, phenazine production levels near the seed were higher than in the roots. Deletion of the gacS gene reduced transcription from the genes required for phenazine synthesis, the regulatory phzI gene and the biosynthetic phzA gene. The inhibition of seed germination and the induction of systemic disease resistance against a bacterial soft-rot pathogen, Erwinia carotovora subsp. carotovora, were impaired in the gacS and phzA mutants of P chlororaphis O6. Culture filtrates of the gacS and phzA mutants of P. chlororaphis O6 did not inhibit seed germination of wheat, whereas that of the wild-type was inhibitory. Our results showed that the production of phenazines by P. chlororaphis O6 was correlated with reduced germination of barley and wheat seeds, and the level of systemic resistance in tobacco against E. carotovora.
