Micropropagation from root segments to improve seedling quality in Chinese foxglove crops

  • This is the first study to establish a complete protocol for micropropagation of Rehmannia glutinosa from root segments. The study involved investigating the effect of plant growth regulators on in vitro shoot regeneration and rooting and identifying substrates supporting survival and growth performance of ex vitro seedlings. A Murashige and Skoog (MS) medium containing 30 g/L sucrose for shoot induction and 0.2 mg/L indole-3-acetic acid (IAA), 1 mg/L 6-benzylaminopurine (BAP), and 1 g/L polyvinylpyrrolidone (PVP) for shoot multiplication resulted in the highest number of shoots per explant and shoot height. Applying a medium containing 0.5 mg/L IAA and 1 g/L PVP yielded optimal rooting of the shoots grown in vitro. Compost enriched with microbial inoculants and perlite enhanced seedling growth better than that with organic biofertilizer-free substrates (soil and sand). We recommend the continuous production of micropropagated R. glutinosa seedlings from root segments under the aforementioned conditions as a possible propagation technique for crops of this species.

Enhancing Effect of Shimizuomyces paradoxus on Seed Germination and Seedling Growth of Canola, Plant Growth of Cucumber, and Harvest of Tomato

  • Shimizuomyces paradoxus showed no inhibitory effect against plant pathogen fungi, such as Fusarium oxysporum f. sp. lycopersici and Alternaria solani. The S. paradoxus culture filtrate showed higher seed germination and seedling growth rates in canola than distilled water and potato-dextrose broth. A conidial suspension of $1.0{\times}10^4/mL$ resulted in the highest growth stimulating effects on total plant length, and fresh and dry weight of shoots and roots in cucumber, when compared to the highest suspension concentration. Total plant length and shoot weight increased with the foliar spray treatment, and root length and root weight increased by simultaneous treatments of soil drenching and foliar spray in cucumber. Lower concentrations of the S. paradoxus conidial suspension increased the harvest of tomato fruit.

  • 고품질의 경쟁력이 있는 상품을 지속적으로 생산하기 위해서는 각 작물별 적합한 배양토(토양조건) 선정과 과학적인 영양(비배)관리가 절실히 요구된다. 이미 선진국에서는 분화용 또는 재배용 배양토를 기능적으로 전문화하여 산업에 활용하고 있으며, 이 분야에 대해서 가축분뇨 등 폐자원을 이용한 배양토 생산이 실용화되고 있다. 국내에서 공정육묘용 상토와 화분용, 식물재배용 배양토에 사용되는 원료(피트모스, 펄라이트, 암면 등)는 거의 대부분 외국으로부터 수입에 의존하고 있으며, 국내의 대규모 기업형 축사 또는 돈사에서 발생되는 축분은 환경오염의 근원이 되나 충분히 부숙 또는 특수가공처리를 한다면 필요한 배양토 조제에 효율적이고 기능적인 자원으로 활용할 수 있을 것으로 평가되고 있다. Early Ontogeny of Vascular Cambium in the Seedling Roots of Acer saccharinum L. (은단풍(Acer saccharinum L.) 유식물의 뿌리에서 유관속 형성층의 초기발생)

  • The origin of cambial initials from procambium was studied in the seedling root of Acer saccharinum. In transverse view, the first periclinal divisions of procambial cells occurred just outside of each early metaxylem and resulted in meristematic strips. As root development progressed, the division activities appeared subsequently outside of each late metaxylem and then in pericycle cells opposite the four protoxylem poles. Eventually, such meristematic strips were connected completely each other. Thus, a nearly rectangular shaped meristematic layer in outline was formed outside the xylem in a whole root transectioned. In tangential section, early procambium showed a homogeneous structure consisted of uniform short cells with transverse end walls. However, some of the procambial cells did elongate, whereas others divided transversely. The former become more elongate, tapered, and vacuolated. Finally, they differentiated into fusiform initials. Short cells consisting axial strands divided continuousely in transverse plane and became ray initials, while some short cells elongated and transformed into long cells. The early ontogeny of vascular cambium in Acer saccharinum root was interpreted to be established by a gradual process.

  • The allelopathic effects of volatile substances from Chamaecyparis obtusa (S. et Z.) Endl. were examined on the germination and seedling growth of some plant species, and on the population growth of some microorganisms. The germination and seedling growth of the receptor plants were suppressed more severely by leaf and fruit essential oils than by those of other parts. Colonial growth of fungi was severely inhibited by essential oils extracted from leaves and fruits. The development of root hairs of the receptor plants was also severely inhibited by the essential oils. The cortical cells at the root tips of Lactuca sativa L. treated with essential oils showed contraction of the cytoplasm, resulting in plasma membranes becoming detached from the cell walls and the cells metamorphosing irregularly. Accumulation of lipid granules inside the contracted cytoplasm and degeneration of mitochondrial cristae were also observed.

  • Introduction Cellulose microfibril is a major cell wall polymer that plays an important role in the growth and development of plants. The gene cellulose synthase A (CesA), encoding cellulose synthases, is involved in the synthesis of cellulose microfibrils. However, the regulatory mechanism of CesA gene expression is not well understood, especially during the early developmental stages. Objective To identify factor(s) that regulate the expression of CesA genes and ultimately control seedling growth and development. Methods The presence of cis-elements in the promoter region of the eight CesA genes identified in flax (Linum usitatissimum L. 'Nike') seedlings was verified, and three kinds of ethylene-responsive cis-elements were identified in the promoters. Therefore, the effect of ethylene on the expression of four selected CesA genes classified into Clades 1 and 6 after treatment with $10^{-4}$ and $10^{-3}M$ 1-aminocyclopropane-1-carboxylic acid (ACC) was examined in the hypocotyl of 4-6-day-old flax seedlings. Results ACC-induced ethylene either up- or down-regulated the expression of the CesA genes depending on the clade to which these genes belonged, age of seedlings, part of the hypocotyl, and concentration of ACC. Conclusion Ethylene might be one of the factors regulating the expression of CesA genes in flax seedlings.

Effects of LED Light Quality of Urban Agricultural Plant Factories on the Growth of Daughter Plants of 'Seolhyang' Strawberry

  • This study was conducted to examine the influence of Light-Emitting Diode (LED) light quality in urban agricultural plant factories on the growth and development of Seolhyang strawberry daughter plants in order to improve the efficiency of daughter plant growth and urban agriculture. LED light quality by demonstrated that above-ground growth and development were greatest for daughter plant 2. Daughter plant 1 showed the next highest growth and development, followed by daughter plant 3. Among the different qualities of LED light, the stem was thickest and growth rate of leaves was highest for R + B III (LED quality: red 660 nm + blue 450 nm/photosynthetic photon flux density (PPFD): $241-243{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and lowest for R (red $660nm/115-117{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). Plant height, leaf width, petiole length, and the leaf growth rate were highest for W (white fluorescent lamp/$241-243{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and lowest for R + B I (red 660nm+blue 450nm/$80-82{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). For above-ground growth and development, as the plants surpassed the seedling age, mixed light (red + blue), rather than monochromatic light (red or blue), and higher PPFD values tended to increase development. Regarding the quality of the LED light, daughter plant 2 showed the highest chlorophyll content, followed by daughter plant 1, and daughter plant 3 showed the least chlorophyll content. When the wavelength was monochromatic, chlorophyll content increased, compared to that when PPFD values were increased. Mixed light vitality was highest in daughter plant 2, followed by 1, and 3, showed increased photosynthesis when PPFD values were high with mixed light, in contrast to the results observed for chlorophyll content.

  • To develop techniques for the production of artificial seedling of Corbicula japonica, protocols for induction of spawning and larval development were developed. During the assumed spawning period of August to mid-September, attempts were first made to induce spawning by gonadal incision and UV-irradiation but all in vain. At the end of August, elevated thermal induction evoked 90 % positive response in animals maintained at $3\textperthousand$ salinity. Immersion in (1/1000~3/1000 N) ammonium hydroxide ($NH_4$OH) also induced spawning in 15~45%) of the treated animals at $3\textperthousand$ salinity. Fertilized eggs measured 86$\mum. At 23.0~24.5$^{\circ}C$, the fertilized egos developed into 4-cell stage embryos within 2 hours, trochophores 15 hours, D-shaped larvae 2 days, umbo 9 days and fully grown veligers, ready to infiltrate into the sediment, within 16 days.
