Transformation of Gourd through Leaf Explant Regeneration (잎 절편의 재분화에 의한 참박 형질전환)

  • In order to develop a disease-resistant root stock for the growth of watermelon, an efficient regeneration system of the gourd(Lagenaria leucantha Duch.) inbred line GO701-2 via organogenesis was established in this experiment. Using proximal parts of cotyledon explant excised from germinated seedling in vitro, maximum adventitious shoot formation (39%) was achieved on MS medium where cytokinin (BA) and auxin (IAA) were added at a concentration of 3mg/L and 0.1mg/L, respectively. Roots of the elongated shoots were successfully formed on MS medium without adding any plant growth regulators. The cucumber CsGolS1 gene known as a resistance gene against biotic and abiotic stresses, was constructed into the binary vector pBI121 under the control of CaMV 35S promoter. When the gene was introduced into the genome of gourd by Agrobacterium-mediated transformation, putative transgenic plants were obtained with the transformation efficiency of approximately 20 percent.

Effect of 1,8-NA and CGA 123'407 on Reducing Herbicide Injury of Rice Plants (1, 8-NA와 CGA 123'407 처리(處理)가 수도용(水稻用) 제초제(除草劑)의 약해경감(藥害輕減)에 미치는 영향(影響))

  • Since herbicide damage has been occurred in subirrigated rice seedbed in Korea, the use of crop safener is required to reduce crop injury. Field studies were conducted to evaluate safening effect of 1,8-naphthalic anhydride (NA) and CGA 123'407 (4, 6-dichloro-2-phenyl-pyrimidine) against four kinds of paddy herbicides applied to subirrigated vinyl-covered rice seedbed. NA as a seed dressing at 0.5% (W/W) was effective in reducing rice injury from butachlor, pretilachlor, chlormethoxynil and benthiocarb, and increased number of seedling established, plant height, and dry weight of rice seedlings, while growth inhibition and stand loss were observed at 1% NA. Pretilachlor slightly decreased number of seedlings established and inhibited growth of rice seedlings. But pre-mix combination of pretilachlor (2%) and CGA 123'407 (0.5%) reduced rice injury caused by pretilachlor alone and increased number of seedlings established and dry weight of rice seedlings. The results indicate that NA and CGA 123'407 would permit the safe application of butachlor, pretilachlor, chlormethoxynil, and benthiocarb in subirrigated rice seedbed.

  • Fusarium proliferatum KGL0401 was previously isolated from Physalis alkekengi var. francheti plant roots and exhibited higher GA productivity than wild type Gibberella fujikuroi. The :tim of this work was to find out an optimal culture condition for GA production. Various carbon(fructose, glucose, lactose, maltose, sucrose) and nitrogen($KNO_3$, urea, glycine, $NaNO_3,\;NH_4Cl$) sources were used for this study. GAs activities were analysed by gas chromatography and mass spectrometry(GC-MS). The highest yield of $GA_3$ was found in the growth medium supplemented with sucrose as carbon source and $NH_4Cl$ as nitrogen source. The optimum carbon-nitrogen concentration for $GA_3$ production was found to be 0.5 M:0.17 M. Supernatant was prepared from the culture fluid of F. proliferatum KGL0401 cultured for 7 days at 3 0'E and the 10 ul supernatant was treated with 2 leaf-rice seedling.

Effects of Simulated Acid Rain on Growth and Contents of Chemical Substances in Needles of Pinus koraiensis Seedlings and on Chemical Properties of the Tested Soil (인공산성우(人工酸性雨)가 잣나무 유묘(幼苗)의 생장(生長), 엽내함유성분(葉內含有成分) 및 토양(土壤)의 화학적(化學的) 성질(性質)에 미치는 영향(影響))

  • Simulated acid rain (pH 4.0, pH 2.0) containing sulfuric and nitric acid in the ratio of 60:40 (chemical equivalent basis) diluted with underground water, and underground water (pH 6.5) as control were treated on potted Pines koraiensis seeds during the growing season (May 1 to August 31) in 1985. The regime of artificial acid rain, in terms of spray frequency and amount per plot, was simulated on the basis of climatological data averaged for 30 years of records. The seedling growth, contents of chemical substances in needles and chemical properties of the tested soil were compared among the various pH levels of acid rain on October 31, 1985. Following results were obtained. 1. With decreasing pH levels of acid rain, S and $K_2O$ contents in leaf tissue were increased, but MgO and $P_2O_5$ contents were decreased. 2. Soil pH was dropped, and exchangeable aluminum content in the tested soil was dramatically increased as the pH levels of acid rain decreased. 3. Exchangeable calcium, magnesium, potassium contents, and base saturation degree of the soil were significantly decreased with decreasing pH levels of acid rain. 4. Sulfate concentrations in the soil were significantly increased as rain pH decreased, but total nitrogen and available phosphate contents were not influenced by acid rain.

Methodological Consideration for Estimating Growing Stock of Young Forests based on Early Growth Characteristics of Standing Trees in Korea (우리나라 입목의 초기 생장 특성에 따른 유령림의 임목축적 산출방안 고찰)

  • The growing stocks of young forests that are less than10 years of age have been excluded from the Korean forest resource statistics, despite the existence of standing trees; however, sustainable forest management and carbon removals in the forestry section require complete information regarding forest resources. This study developed a method to estimate the growing stocks for young forests from National Forest Inventory (NFI) data. After reviewing previous research on growth characteristics for young forests, we conducted stem analysis of major species, and examined stand characteristics by site index, based on real yield tables. Our statistical analysis results showed that there were few standing trees with diameters at breast height (DBH) above 6 cm in young stands, and that it would have taken 12 years, on average, to reach 6 cm DBH. This suggests that mean tree height by diameter should be assessed at the root, in order to assess growing stocks for young stands through the NFI. Moreover, the database system should be improved to differentiate tree species, since diverse shrubs, including trees, have been surveyed.

Effect of Pinching Times on Growth and Yield of Okra (Abelmoschus esculentus L.) in Rain Shielding Vinyl Houses (비가림 재배 시 오크라의 적심처리 시기가 생육 및 수량에 미치는 영향)

  • The experiment was carried out to examine the effect of pinching time on growth and marketable yield of okra in rain shielding vinyl houses. Okra (Abelmoschus esculentus L., 'Green sod') seedling was transplanted in May, 2013. Pinching times of okra were in each 30th, 45th, 60th day after transplanting of okra. Shoot branch and top fresh weight were significantly different between each pinching times. Top fresh weight were highest at 60th day pinching time after transplanting investigated, and were lowest at 30th day pinching time after transplanting. There was no significant difference in plant height and stem length among pinching times. According to pinching times, marketable yield and number of okra fruits were varied. Marketable yields were highest at 60th day after transplanting, whereas were lowest at 30th day after transplanting. Mineral concentrations of okra fruits were not significantly different among pinching times. Therefore pinching times 60th day after transplanting was effective to increase yield of okra fruit in rain shielding vinyl houses.

Effects of Germination Condition, Nursery Media and Nutrient Concentration on Seedling Growth Characteristics of Pak-choi and Lettuce in Plant Factory (발아조건과 배지종류 및 양액 농도에 따른 식물공장 재배 청경채와 상추의 묘 생육특성)

  • This experiment was conducted to study the effects of germination condition, nursery media and nutrient concentration in plant factory on the growth characteristics of pak-choi and lettuce seedlings. The results showed that temperature (15, 20, $25^{\circ}C$) and light conditions (lighting and no lighting) had no significant effect on germination rate of pak-choi and lettuce, whereas the interaction of growing media (rockwool and urethane sponge) and moisture management before and after sowing had significant effect on germination percentage. In case of no moisture management before and after sowing, germination percentage was much higher in rockwool medium than urethane sponge medium. But germination percentage of urethane sponge was higher than that of rockwool with applying moisture before and after sowing. At the nutrient concentration of EC $1.0dS{\cdot}m^{-1}$ the fresh weight of pak-choi was heavier than in EC 1.5 and $0.5dS{\cdot}m^{-1}$ by 1.8 and 6.0 times, respectively. At the nutrient concentration of EC $1.0dS{\cdot}m^{-1}$ the fresh weight of lettuce was also heavier than in EC 1.5 and $0.5dS{\cdot}m^{-1}$ by 1.7 and 3.5 times, respectively.

Effect of Cold Water Irrigation on the Growth and Yield Characters of Rice Varieties at Mid-mountains Area (중산간지에서 냉수처리가 벼 품종의 생육과 수량에 미치는 영향)

  • This study was conducted to obtain the basic information on rice cold damage and relationship to be connected with cold tolerance. The 20 rice varieties were screened to cold tolerance under low water temperature condition. The water gradient were controlled to $17^{\circ}C$ at the inlet and $21^{\circ}C$ at outlet of screening field. In the relationship between cold water irrigation and growth of rice varieties, effect on medium maturing cultivars were higher than early maturing cultivars. In relationship of shorting of culm length and panicle exsertion, culm length and panicle exsertion showed positive correlation. Yield of cultivars and culm length and percent of fertile grain showed positive correlation. However, heading date presented negative correlation. In the early maturing cultivars, except 'Kumobyeo', all cultivars range of discoloration value (1-3) was resistant. In the medium maturing cultivars, 'Donghaebyeo' was resistant in both the seedling stage whereas 'Hwajinbyeo' was resistant only in tiller stage. Cold water irrigation reduced spikelet number per panicle and percent of filled grain. Unhulled rice yield was increased according to water temperature gradient from inlet to outlet. There was varietal differences in head rice recovery by cold water treatment.

Screening of Pesticidal Active Compounds from Various Domestic Wild Plants (국내 자생 식물자원 중 농약활성물질 탐색)

  • For the selection of plants contained pesticidal active conpounds, 31 families 59 species of native plants were collected and biological activites of their crude extracts against brown planthopper (Nilaparvata lugens), Xanthomonas oryzae pv. oryzae and allelopathy were examined. Among the screened plants, the crude extracts from the leaves of Ricinus communis and Sophora angustifolia showed 100% and 82% of mortality on brown planthopper at the concentration of 1% (w/v) respectively. Mixed crude extracts of Sophora angustifolia root and Melia azedarach seed exhibited 128${\sim}$155% of synergistic effects on the mortality of brown planthopper. In case of fungicidal activity, the crude extracts from the leaves of 8 plants including Chrysanthemum indicum were inhibitory of the growth of Xanthomonas oryzae pv. oryzae at 1%(w/v) soluction. Seed germination of radish (Raphanus sativas) and barnyardgrass (Echinochloa crus-galli) was inhibited by 0.5%(w/v) leaf extracts of Pinus densiflora and Quercus acutissima. MeOH extracts of Pinus denislora leaves gave 100% inhibition in seed germintion of radish at 2%(w/v) soluction and showed a complete inhibition in seedling growth of barnyardgrass as well as radish at 5% solution.

Proteomic Analyses of Chinese Cabbage(Brassica campestris L. pekinensis) Affected by High Temperature Stresses in Highland Cultivation During Summer in Korea (Proteomics를 이용한 고랭지 배추의 고온장해 해석)

  • High temperature stresses have caused growth inhibition and delayed heading in highland cultivation Chinese cabbage during summer in Korea. We have studied high temperature stress responses in the terms of changes of inorganic components and proteins by proteomic analyses. Insufficiencies of nitrogen and phosphorus have affected growth rate and calcium deficiency has caused blunted heading. Proteins extracted from Brassica seedling grown at the altitude of 600m and 900m in the Mount Jilun were extracted and analysed by 2-dimentional polyacrylamide gel electrophoresis. Profiles of protein expression was then analyzed by 2-dimentional gel analyses. Protein spots showing different expression level were picked using the spot handling workstation and subjected to MALDI-TOF MS. Total 48 protein spots were analyzed by MALDI-TOF MS and 30 proteins spots out of 48 were identified by peptide mass fingerprinting analyses. Fourteen proteins were up-regulated in extracts from the altitude of 900m and they were identified as oxygen-evolving proteins, rubisco activase and ATPase etc. Sixteen proteins were up-regulated in extracts from the altitude of 600m and they were identified as glutathione S-transferase(1, 28kD cold induced- and 24 kD auxin-binding proteins) and salt-stress induced protein etc. These stress-induced proteins were related to the mediated protective mechanism against oxidative damage during various stresses. The results indicated that physiological phenomenon in response to high temperature stresses might be resulted by complex and multiple array of responses with drought, heat, oxidative, salt, and cold by high temperature.
