Effects of Storage Period and Germination Condition on Germination Rate of Vicia tetrasperma and V. hirsuta (피복식물용 자생잡초 얼치기완두와 새완두 종자의 저장방법이 발아에 미치는 영향)

  • This study was conducted to evaluate germination rate of Vicia tetrasperma and V. hirsuta as affected by storage duration, temperature, and light. All seeds of both species were gathered from Dangjin, Chungnam in the late June of 2004 and 2005, stored for a certain period of time, and then exposed for 20 days to the alternating temperatures ($15/^{\circ}C,\;20/10^{\circ}C,\;25/15^{\circ}C$, and $30/20^{\circ}C$) for evaluating the germination rate. Both 2004 and 2005, two species stored at room temperature for 3 months had germination rates less then 30% with applying various alternating temperatures for 20 days although the highest germination rate was obtained with $25/15^{\circ}C$ treatment. V. tetrasperma at 12 month storage after seed gathering in 2005 had 82% of germination rate with $25/15^{\circ}C$ treatment and showed the increased rate as time passed. V. hirsuta at 6, 9, and 12 month storage after seed gathering in 2005 ranged, however, from 33 to 53% of the germination rate at alternating temperature treatments for 20 days and did not seem to be affected by the elapsed time of the storage. Storage at room temperature was more effective for increasing germination rate of both species compared to those of the storage at low temperature. The germination rate of both species was not significantly affected by either exposure of light or short period of high temperature.

Cryopreseryation and Germination of Native Aquilegia Species Seeds by Predehydration Treatment (건조 전처리에 의한 자생 Aquilegia속 식물 종자의 초저온 저장과 발아)

  • Predehydration effects for cryopreservation in -196$^{\circ}C$ liquid nitrogen were studied in Korean native Aquilegia buergeriana var. oxysepala Kitamura and A. flabellata var. pumila Kudo seeds. Aquilegia species seeds were adjusted to moisture contents between 3.2 and 9.7% by air dry treatments. Seeds were placed in paper envelopes after submerged in liquid nitrogen and rewarming in 38 $^{\circ}C$ water. Seeds moisture contents by duration of drying were identified as controlling factors in the survival of Aquilegia species seeds for cryopreservation. Aquilegia species seeds having approximate 5% moisture content were able to withstand cooling to -l96$^{\circ}C$. Undehydrated seeds of Aquilegia buergeriana var. oxysepala Kitamura after being cryopreservated in liquid nitrogen have a 10.9% of moisture content and show 52.5% in germination. But, Aquilegia buergeriana var. oxysepala Kitamura seeds dehydrated by drying for 60 min. to have 6.0% of moisture content before cryopreservation show 84.7% in germination test. Properly dehydrated seeds after being stored in liquid nitrogen showed over 60% in germination rate and also shows an uniform sprouting time,11~13 days in average. Any morphologically deformity in germinating beds has not been observed. Results from this study suggest that Aquilegia species seeds can cryopreservation in liquid nitrogen if the seed moisture content is controled by a proper amount of dehydration.

Germination and Seedling Growth Response of Sprouts and Leafy Vegetables after Applying Oxygen Nanobubble Water

  • Background and objective: The nanobubbles remain stable in water, and it increased dissolved oxygen (DO) in the water that promotes the seed germination and the plant growth. We evaluated the seed germination and growth of sprouts (radish, wheat, and barley) and leafy vegetables (red mustard and pak choi) when irrigated with various DO of nanobubble water (NB). Methods: The oxygen NB was generated by surface friction and treated in 4 levels: NB 0% (control, DO 9.21 mg·L-1), NB 20% (DO 15.40 mg·L-1), NB 33% (DO 20.93 mg·L-1), and NB 100% (DO 39.29 mg·L-1). Results: The root length of radish and wheat increased more in NB 33% than the control plot. The fresh weight increased in NB 33% compared to the control plot in radish and wheat, and both fresh and dry weight increased more in NB 20%, NB 33%, and NB 100% than the control plot of barley. The leaf length and width of red mustard decreased more in NB 33% and NB 100% than the control plot and NB 20%, which indicated the leaf compactness. The fresh and dry weight of shoot and root increased more in NB 100% than the control plot in red mustard. In pak choi, the shoot fresh weight increased more in NB 100% than the control plot. In leafy vegetables, the germination rate of red mustard in NB 100% was higher than the control plot, however, it was not significantly different between oxygen NB plots in sprout vegetables. Conclusion: The results showed that the root growth and biomass increased after applying NB 33% in sprout vegetables. The leaf growth properties as the number of leaves and leaf size were not significantly different or decreased in NB treatments compared to control plots, but NB 100% (DO 39.29 mg·L-1) effectively increased the root growth and plant biomass in leafy vegetables.

Effects of Plant Growth Regulator Treatments During the Grain Filling Period on Seed and Viviparous Germination in Rice (벼 등숙기의 생장조절제처리가 종실 및 수발아에 미치는 영향)

  • Experiments were conducted to find out the method of reducing viviparous germination of rice seeds using growth regulators during grain filling period. Among the recommended varieties, early Japonica type showed higher rate of viviparous germination, while late Japonica type and Tongil type varieties did not show viviparous germination in field condition. Grain showed high germination ability from 30 days of grain filling period in Daeseongbyeo of Japonica type and 35 days in Taebackbyeo of Tongil type. Growth regulators, NAA, CMH and Reglone were effective for reducing viviparous germination to the two rice varieties and three treatment stages in this experiment. NAA and CMH were most effective at 100 ppm and Reglone was at 0.4-0.6% concentration. Germination rates of rice seed were not different from non-treatment when NAA CMH were treated, while Reglone lowered germination rate. NAA and CMH had no effects on grain filling rate for both varieties, which showed high grain filling rate as the non-treatment, but Reglone results low grain filling rate. Grain color was not changes when NAA was applied, while CMH and Reglone made a lot of grain to dark brown as the concentration became higher. When grain were hulled, NAA had no difference with the non-treatment whereas CMH and Reglone had lower rate of perfect brown rice when they were applied at 20-30 days after heading.

Effects of Germination Rate of Grafting-Stock Gourd Seeds as Affected by Durations of Ripening and After-ripening of Fruits (성숙기간과 후숙기간을 달리한 대목용 박(Lagenaria siceraria Standl.) 종자의 발아율)

  • This study was conducted to elucidate the effect of after-ripening of gourd fruits before seed collection. Five varieties of commercial gourd varieties were cultivated and the fruits were picked after 24, 31, 50, and 70 days from pollination. For after-ripening treatments the fruits were placed for five days in a cool, air-circulated place while a number of fruits were placed for longer periods to 85 days including growing period, 24 + 62 days, 31 + 55 days, 50 + 35 days, and 70 + 15 days. The seeds collected from fruits harvested on 24th day were not emerged at all, but the seeds from after-ripening treatment for 62 days showed 78% emergence or higher. The growth of seedlings showed the effects of after-ripening treatment of the fruits before seed collection; the seeds collected after ripening treatment showed better hypocotyl growth and larger embryo size than the non-treated seeds.

Effect of Phytogenic Feed Additives in Soybean Meal on In vitro Swine Fermentation for Odor Reduction and Bacterial Community Comparison

  • The effect of different phytogenic feed additives on reducing odorous compounds in swine was investigated using in vitro fermentation and analyzed their microbial communities. Soybean meal (1%) added with 0.1% different phytogenic feed additives (FA) were in vitro fermented using swine fecal slurries and anaerobically incubated for 12 and 24 h. The phytogenic FAs used were red ginseng barn powder (Panax ginseng C. A. Meyer, FA1), persimmon leaf powder (Diospyros virginiana L., FA2), ginkgo leaf powder (Ginkgo biloba L., FA3), and oregano lippia seed oil extract (Lippia graveolens Kunth, OL, FA4). Total gas production, pH, ammonianitrogen ($NH_3$-N), hydrogen sulfide ($H_2S$), nitrite-nitrogen ($NO_2{^-}$-N), nitrate-nitrogen ($NO_3{^-}$-N), sulfate (${SO_4}^{--}$), volatile fatty acids (VFA) and other metabolites concentration were determined. Microbial communities were also analyzed using 16S rRNA DGGE. Results showed that the pH values on all treatments increased as incubation time became longer except for FA4 where it decreased. Moreover, FA4 incubated for 12 and 24 h was not detected in $NH_3$-N and $H_2S$. Addition of FAs decreased (p<0.05) propionate production but increased (p<0.05) the total VFA production. Ten 16S rRNA DGGE bands were identified which ranged from 96 to 100% identity which were mostly isolated from the intestine. Similarity index showed three clearly different clusters: I (FA2 and FA3), II (Con and FA1), and III (FA4). Dominant bands which were identified closest to Eubacterium limosum (ATCC 8486T), Uncultured bacterium clone PF6641 and Streptococcus lutetiensis (CIP 106849T) were present only in the FA4 treatment group and were not found in other groups. FA4 had a different bacterial diversity compared to control and other treatments and thus explains having lowest odorous compounds. Addition of FA4 to an enriched protein feed source for growing swine may effectively reduce odorous compounds which are typically associated with swine production.

Germination of Gourd Seed Affected by Drying and Light Quality Treatments during its Period (건조방법과 건조중 광질처리가 박 종자의 발아에 미치는 영향)

  • Seeds treated before sowing should be dried for their quality control on their sale or storage. The experiments were done to clarify the effect of drying, temperature and light quality treated during drying and germination on germination of gourd (Lagenaria siceraria Standl.) seeds. Drying of treated seeds was done at $35^{\circ}C$. After treated seeds were dried. their germination test was done at 10, 20 and $30^{\circ}C$ on the condition of darkness, but with blue, red, far-red light and darkness as light quality treatments. FR-yongjadaemok and FR-kunghap were used as test cultivars and their daily germination rates were measured at treatment levels. Its presowing hydrated seeds were dried 10 hours at $35^{\circ}C$ until reached below their original moisture content, and germination rate between the above dried seeds and the conventional dry-heated ones was not significantly different. At $30^{\circ}C$ germination temperature, the treated seeds showed higher initial germination rate when dried under red light illumination than under darkness although their germination rates were nearly same at each temperature of 10 and $20^{\circ}C$ being relatively low. Effect of the red light treatment during their drying continued to more than 3 months. Germination rate of the red light treated seeds during drying was reduced by far-red light treated during their germination. The initial rate to the second day after sowing were higher under red light and darkness treated during germination than under the two other lights.

Effect of Light, $GA_3$, $KNO_3$ Treatments on the Germination of Chilean Wild Flower (칠레 야생화의 발아율에 미치는 광조건 그리고 $GA_3$, $KNO_3$ 처리 효과)

  • Chilean wild flower seeds showed low germination rate and lacked germination uniformity. This experiment was conducted to find out the effects of light conditions, $GA_3$ (50 mg/L, 100 mg/L, 200 mg/L) and $KNO_3$ (0.1%, 0.2%, 0.4%) treatments on germination of five species of Chilean wild flower seeds. The seeds of $Schizanthus$ $candidus$, $Schizanthus$ $hookerii$, and $Schizanthus$ $litoralis$ did not germinated in light condition. The others did not show a significant different germination rate under light and dark conditions. The germination rate of $Mimulus$ $luteus$, $Schizanthus$ $candidus$, $Schizanthus$ $hookerii$ increased 2.3, 5.6, and 3.2 times compared with control, respectively in $GA_3$-200 mg/L treatment, but that of $Rhodophiala$ $ananuca$ enhanced slightly in $GA_3$-50 mg/L treatment. Those results suggested that the germination rate of Chilean wild flower seeds enhanced by $GA_3$ treatment that is very efficient in breaking seed dormancy.

Effects of Aeration Period and Temperature after Imbibition on Growth of Mungbean Sprouts (침종 이후의 Aeration 기간과 온도에 따른 숙주나물의 생장)

  • Lateral roots formed on mungbean sprouts should lower their quality. The study was carried out to clarify the effects of aeration periods (AP; 1, 2, 3, and 4 hours) and temperature (AT; 20, 30, and $40^{\circ}C$) after 5 hour seed imbibition into 50 ppm benzyladenopurine (BA) solution on growth and morphological characters of mungbean (cv. Keumseongnogdu, Owoolnogdu and Zhong Lu 1) sprouts. On the 6th day, the mungbean sprouts were sorted by 4 categories on the base of hypocotyl lengths; > 7cm, 4 to 7cm, < 4cm, and non-germination to calculate their composition rates, number of lateral roots, lengths of hypocotyl and root, diameters at middle and upper parts of hypocotyl, fraction fresh and dry weights were measured. AP more affected growth of the cultivars than AT showing little effect on them. In the composition rate of the above 4 categories, cv. Zhong Lu 1 had the highest rate in longer than 7cm but nearly the same rate in AP treatments. Rates of longer than 4cm hypocotyls in cv. Keumseongnogdu, Owoolnogdu were increased with longer AP but their rates of shorter than 4cm showed reverse response to the former. Formation rate and number of lateral roots per sprout were decreased with longer AP, showing more severe decrement when delayed 3 to 4 hour AP. Upper part of hypocotyls and roots were more thickened and shortened in longer AP, respectively. Total fresh weights had no significant difference between AP treatments while hypocotyl fresh weights were increased with longer AP. It was concluded that in mungbean sprout culture aeration from BA treatment to the first watering permitted at least 4 hours.

Factors Affecting Dark Germination of Oenothera lamarckiana Seeds (큰달맞이꽃 종자(種子)의 암발아(暗発芽)에 영향(影響)을 미치는 요인(要因))

  • Effect of various physical and chemical treatments on dark germination of Oenothera lamarckiana seeds were primarily investigated to find out factors affecting germination. Germination of seeds which did not germinate in the constant temperature and darkness was induced by several physical treatments such as sonification, wetting and drying, freezing and thawing, and removal of seed coat. Pretreatment of chilling ($4^{\circ}C$), freezing ($-10^{\circ}C$) and incubation at high temperature ($80^{\circ}C$) induced dark germination of seeds which did not germinate in the constant temperature ranges of 15 to $40^{\circ}C$ under darkness. Alternating temperature also had a stimulatory effect on dark germination of Oenothera lamarckiana seeds. Sensing of seeds to alternating temperature appeared to be completed during the first two days after imbibition. The minimum difference of temperature required for dark germination was $5^{\circ}C$ in the range of $15-25^{\circ}C$. A thiourea (1.0%) treatment induced dark germination, but GA, IAA, BA and Ethrel failed to do so.
