Detection of Genetically Modified Genes from Soybean Sprout Products (콩나물에서 발견된 유전자 변형 도입 유전자의 비의도적 혼입 조사)

  • A total of 219 polymerase chain reaction tests of genetically modified (GM) DNA sequences in soybean seeds and soybean sprouts were conducted during 2000-2001. No CM gene was found in 96 tests of soybean seeds. However, either a functional CP4EPSPS gene or the 355 promoter gene was found three times in 2000 and eight times in 2001, in between 0.01 and 0.17% of soybean spout products, in 123 tests. Since the amount of GM genes was much less than the threshold limit of 3%, none of the 11 positive soybean-sprout samples needed to be libeled GM crops. Of these, seven sprout samples were from domestic seeds and four were from seeds imported from China. To find the contamination route, the raw materials, seed surface, floor of the storage room, area around the selection machine, surface of the packaging film and corn powder used in the package were tested. The 35S promoter gene was detected in only two samples of the corn powder (0.1%). Although we could not find the cause of the GM contamination, the sprout package film is one possibility. In total,8.9% of the soybean sprout tests were GM positive, but the amounts were much less than the threshold of 3%. This means that there are frequent false-positives and these would threaten the sprout industry if GMO were decided qualitatively. Food companies should make their safety data available to the public and make an effort to address people's concerns about GM food more openly. In addition, there is a need to establish a quantitative test for GM genes in sampled water and a sampling method for raw materials.

Aquatic animal health management in stock enhancement (방류용 수산종묘의 질병 관리에 대한 고찰)

  • Ministry of Agriculture, Fisheries and Food (MAFF) and National Fisheries Research and Development Institute (NFRDI) have inspected the hatchery-reared seeds of 22 marine species and 11 freshwater species for aquatic animal diseases in stock enhancement program in 2009. Results showed that total 12 local selfgovernments have been restocking the sea with cultured juveniles. Gyeongsangnam-do, Jeollanam-do, Jejudo and Chungcheongnam-do have a preference for marine species seeds to freshwater species. On the contrary, freshwater species were released mostly in Gyeonggi-do, Jeollabuk-do and Chungcheongbuk-do. In the marine species group, abalone was the most abundant as 24.5%, and then sea cucumber (15.2%), olive flounder (11.5%), swimming crab (5.6%), black sea bream and rockfish (6.8%), rock bream (5.1%), black rockfish (4.6%) and scorpionfish (4.5%) were followed. Crucian carp was the most abundant as 19.4%, and then eel (17.0%), Korean bullhead (12.3%), melanian snail (12.0%), catfish (8.4%) were followed in the freshwater species group. The total number of inspection cases in this study were 1,080 and disqualification cases were 19 by detection of aquatic animals pathogens such as red sea bream iridovirus (RSIV), koi herpesvirus (KHV) or white spot syndrome virus (WSSV).

Studies on the Forage Production and Utilization on Paddy Field in Korea (한국에 있어서 답리작을 이용한 양질 조사료 생산기술)

  • The problems in the current domestic forage production were evaluated, and the prospective improvement was suggested in this paper. Grassland development in forest, production of high quality forages in upland and paddy land, efficient utilization of rice straw, development of new varieties of forages suitable for our environmental conditions and imported forages were described Among them, preferential production and utilization of forages using paddy field after rice harvest . should be enlarged for domestic supply of forages in Korea. Several studies were carried out to select the promising forage crops and barley cultivars for whole crop silage production, to determine productivity, nutritive value and production cost of forages produced in paddy field, and feeding effect of forages with Hanuwoo and milking cow for whole crop silage with forages produced in paddy field, 1999 to 2001, and also discussed restraint factors and activation plans for enlargement of forage production in paddy land. The promising forage crops in paddy field were rye and barley for Middle region, and rye, barley, early maturing Italian ryegrass and wheat for Southern region. The promising barley cultivars for whole crop silage in paddy field were Albori in Suwon, Keunalbori, Milyang 92, Saessalbori, and Naehanssalbori in Iksan, and Keunalbori, Albori, Naehanssalbori, and Saegangbori in Milyang, respectively. Silage production, quality and animal palatability of silage by trench and round bale were also compared. The production yields of whole crop barley silage(WBS) were 17,135kg as a fresh matter, and 6,011kg as a dry matter per ha, and the quality of WBS was 2∼3 grade, while that of rice straw silage was 4 grade as a farm basis. The production cost of WBS per kg was 83won as a fresh matter, and 238won as a dry matter. Feeding of WBS as forages on Hanwoo was very desirable for the improvement of live-weight gain, beef quality and farm income, particularly in growing stage of Hanwoo. Milk production and income were also increased, and feed cost was decreased by feeding of WBS. The daily voluntary intake of WBS in milking cow was 26.3kg as a fresh matter(DM 7.7kg) per head. Milk production when WBS was fed, was very similar to that of imported hay feeding such as Kentucky bluegrass or domestic corn silage. The issues to be solved in near future f3r stable forage production and supply in paddy land are sustainable livestock-forages policy, development & seed production of new varieties of barley, rye, Italian ryegrass and other promising forages, efficient demand & supply system of forages, solidification for mass production and utilization of forages, efficient application management of animal slurry on paddy field considering environmental agriculture/livestock industry, and break k development of bottleneck technique in production field. Domestic production & supply of high cost agricultural machine (round baler, wrapper, handler and so on), plastic wrapping film, and silage additives are also important.

The Study of Surface Plasmonic Bands Using Block Copolymer Nanopatterns (블록공중합체 나노패턴을 이용한 표면 플라즈몬 연구)

  • It is important to develop a simple method oftuning localized surface plasmon resonance(LSPR) properties, due to their numerous applications. In addition, the careful examination of the shape, size and combination of metal nanoparticles is useful for understanding the relation between the LSPR properties and metal nanostructures. This article describes the dependence of theLSPR properties on the arrays of metal nanoparticles obtained from a block copolymer(BCP) micellar thin film. Firstly, two different Au nanostructures, having a dot and ring shape, were fabricated using conventional block copolymer micelle lithography. Then, Ag was plated on the Au nanostructures through the silver mirror reaction technique to obtain Au/Ag bimetallic nanostructures. During the production of these metallic nanostructures, the processing factors, such as the pre-treatment by ethanol, silver mirror reaction time and removal or not of the BCP, were varied. Once the Au nanoparticles were synthesized, Ag was properly plated on the Au, providing two distinguishable characteristic plasmonic bands at around 525nm for Au and around 420nm for Ag, as confirmed bythe UV-vis measurements. However, when a small amount of Au seed nanoparticles, which accelerate the Ag plating speed,was formed by usinga block copolymer with a relatively highmolecular weight, all of the Au surfaces were fully covered by Ag during the silver mirror reaction, showing only the characteristic peak for Ag at around 420nm. The Ag plating technique on Au nanoparticles pre-synthesized from a block copolymer is useful to study the LSPR properties carefully.

A Phenomenological Study On the Characterization Experience of Middle-aged Woman using Mandala Arts Treatment (만다라 미술치료를 활용한 중년기 여성의 개성화 경험에 관한 현상학 연구)

  • When most middle-aged women live in a time when they have to take responsibility for their lives, they feel lonely when they realize that their experience of not expressing their feelings properly has ended up being left alone due to their own existence and loss. As the first step in conscious awareness of unconsciousness of middle-aged women through analytical psychology, we performed "Mandara" and "Nanhwa Mandara," which meet me inside. The intermediate stage can be divided into transition and work phase. The working phase, develops one's own advantages and unique strengths that meet the inner world of promoting and acting values, correctly looks at reality, corrects emotion perception and balance, and is self-contained, seed mandala, associative mandala. They performed the strong points of mandala, the free mandala, and the Western mandala. The results of the study showed that mandala art therapy was found in middle-aged women. First, Mandarin art therapy experience affects the physical, psychological and human relations aspects of middle-aged women. Second, middle-aged women's experience of individualization through mandala art therapy shows psychological reversals and affects positive thinking and self-effectiveness. This study was suitable for qualitative research that approached the essence through practical understanding and direct exploration of research participants, and it is meaningful to suggest that it is necessary to develop an art therapy program through various mediums considering the lack of art therapy research in middle-aged women and problems experienced by the physician.

Evaluating the agronomic characteristics and yield variations of 'Saemimyeon' by changing transplanting and harvesting time

  • 'Saemimyeon' a Tongil-type rice variety (Indica ${\times}$ Japonica), which contains high amylose contents is suitable for rice noodle production. Nowadays, the major parts of rice processing industry that includes products like rice flour and noodles are expected to partially replace wheat flour market. The volume of rice noodle market is getting bigger and can contribute to the rice surplus and farmer's income. This study was carried out to promote productivity and flour-making quality of 'Saemimyeon' by finding the most suitable transplanting and harvesting times. The transplanting days used were May $10^{th}$, May $17^{th}$, May $24^{th}$, May $31^{th}$, June $7^{th}$ and June $14^{th}$ and the planting distance used was 30 x 12cm. In addition, harvesting time was determined by days after heading time (40, 45, 50, 55 and 60 days). The field experiment was conducted at the experimental field in Miryang (Southern plain area of Korea) from 2015 to 2016. Our results suggest that the optimum transplanting days were from May $24^{th}$ to May $31^{th}$ which resulted to an average yield of 748~751kg/10a. Interestingly, yield was sharply decreased below 700kg/10a before May $10^{th}$ and after June $7^{th}$. The average grain filling rate before May $31^{th}$ was more than 83% but it declined to 75% after June $7^{th}$. The average temperature ranges from heading time to harvesting time was $ 21\sim25^{\circ}C$ and the estimated optimal temperature was $23.4^{\circ}C$ which is similar to May $24^{th}$ by regression equation. We found that the optimal harvest time was 45~50 days after heading time. It is hypothesized that low temperature at seed maturation time caused the lower grain filling rate therefore 'Saemimyeon' need to be transplanted before May $31^{th}$ for higher productivity. We found no statistical variation in amylose contents among experimental plots (28.2~30.4%). We conclude that the productivity of 'Saemimyeon' highly depends on temperature that is critical for grain filling stage controlled by transplanting time.

Reclamation of the Closed/Abandoned Coal Mine Overburden Using Lime wastes from Soda Ash Production (부산석회를 활용한 휴ㆍ폐 석탄광산 폐기물의 안정화 및 식생복원)

  • In Korea, over three hundreds of the coal mines were closed or abandoned due to the depression of the mining industry since the late 1980s. Many of them locate in the steep mountain valleys and the coal mine wastes had been disposed without a proper treatment From these mines, enormous amounts of coal mine overburdens have been abandoned in the slopes and the ample amounts of acid mine drainage (AMD) from either portal or overburdens have been discharging directly to the streams, causing the detrimental effects on soil and water qualities. Objectives of this research were to reclaim the coal mine overburdens using the lime waste cake from the soda ash production by stabilizing the overburden slopes, introducing the vegetation alleviate the environmental problems caused by the closed coal mines. The percentages of the grass distribution ratio (%) and the surface coverage ($\textrm{cm}^2$) in each treatment plot were determined during June to August after seed spraying grasses such as orchard grass (Dactylis glomerata L), Kentucky Bluegrass (Poa pratensis L.) and Eulalia (Miscanthus sinensis Anderss) at the end of May. The grasses covered only 15.5 % of the coal overburden plot at the early stage but the coverage was increased with time to 33% in August. Growth of such grasses was enhanced with the combined treatments of lime waste and topsoil resulting in the increased surface coverage by the grasses. The Increment of the surface coverage from June to August was higher with lime waste treatments. The distribution percentages and surface coverage were highest when the lime wastes were treated at 25 % of the lime requirement. This might be related with the high salt contents in the hire wastes. Results demonstrated that the amounts of lime wastes at 25% of the lime requirement were sufficient for neutralizing the acidic coal overburden and introducing the re-vegetation. Either layering between the coal waste and topsoil or mixing with coal overburdens could be adopted as the lime waste treatment method. The combined treatment of lime wastes and topsoil was recommended for re-vegetation in the coal overburden slopes. The lime wastes from the soda ash production might have a potential to be recycled for the reclamation of the abandoned coal mines to alleviate the environmental problems associated with coal mine waste.

Further Modifications to the Mobile Nylon Bag Technique to Determine Nutrient Digestibility for Swine

  • Previous studies conducted with swine have reported that the mobile nylon bag technique (MNBT) does not always accurately predict in vivo nutrient digestibilities. Therefore, in this study, the MNBT was modified so that nutrient digestibilities would more closely resemble those from conventional (Con) digestibility studies obtained using the indicator method. A total of 19 feeds were tested including five cereal grains, five legumes, three high protein sources and six mixed diets. The principle changes to the MNBT included the use of a fecal collection harness which minimized the number of bags lost. In addition, previous protocols involved pooling of bags within pig while in the present experiment all bags were analyzed separately to increase the precision of the test. Finally, chemical analyses were done using the entire nylon bag plus residue rather than opening.the bags and scraping out the contents. With the exception of the barley sample (p=0.01), dry matter digestibility (DMD) coefficients obtained with the MNBT were not significantly different from those obtained with the indicator method. The linear regression equation relating the MNBT to the indicator method was Con DMD=-O.77+1.02 MNBT DMD ($r^2=0.93$: p<0.0001). There was no significant (p>0.05) difference in gross energy digestibility (GED) coefficients determined using the MNBT or the indicator method for any of the 19 feeds. The regression line equation relating the MNBT to the indicator method was Con GED=-5.68+1.06 MNBT GED ($r^2=0.94$: p<0.0001). The MNBT was less effective in predicting in vivo crude protein digestibility (CPD) than it was in predicting dry matter and energy digestibility. Differences greater than five percentage units were observed for two of the legumes, Kabuli chickpeas (p=0.02) and the extruded pea-canola seed mixture (p=0.01) as well as for three of the mixed diets including the unheated hulled barley-based diet (p=0.01), the unheated hulless-barley based diet (p=0.08) and the barley-soybean meal based diet (p=0.008). The regression equation relating the MNBT to the indicator method was Con CPD=5.75 + 0.90 MNBT CPO ($r^2=0.76$; p<0.0001). This study indicates that the modified MNBT can be used for the rapid determination of dry matter and energy digestibility in a wide variety of ingredients. For the measurement of crude protein digestibility, the technique produces results similar to conventional digestibility studies for cereal grains and high protein feeds but tends to overestimate protein digestibility for legumes and mixed diets.

Development and Characterization of Tall fescue Variety 'Greenmaster2ho' (톨 페스큐 신품종 '그린마스터2호'의 품종 특성 및 수량성)

  • A new tall fescue variety (Festuca arundinacea Schreb.) named 'Greenmaster2ho' was developed by the National Institute of Animal Science, Rural Development Administration, in Cheonan, Korea from 2010 to 2014. For the synthetic seed production of this new variety, 5 superior clones were selected and polycrossed: 09XFa02, 09XFa03, 09XFa11, 09XFa13, and 09XFa14. The agronomic growth characteristics and forage production capability of the seeds were studied at Cheonan from 2010 onward, and regional trials were conducted in Cheonan, Hoengseong, Jeju, and Jinju from 2012 to 2014. Greenmaster2ho showed resistance to disease, persistence, and regrowth ability that were all enhanced when compared with Fawn. At 15,119 kg/ha, the dry matter (DM) yield of Greenmaster2ho was 29% higher than that of Fawn, but the nutritive value of both varieties as forage was similar. This study aimed to make a contribution to the vitalization of the Korean grassland industry by developing a new tall fescue variety with excellent environmental adaptability.

Growth Characteristics and Nutrient Content of Cowpea Sprouts Based on Light Conditions (광 조건에 따른 동부나물 생육특성 및 영양성분 변화)

  • We examined the effects of light colors (wavelength) and light quantities on the yield ratio and quality of cultivated cowpea sprouts. All light colors resulted in a lower cowpea sprout yield ratio compared to the untreated condition (darkness), but were similar in hard seed ratio. All light colors promoted the growth of the epicotyl and root when compared to the untreated condition, but limited the growth of the hypocotyl. White light (458 nm) significantly improved grade by increasing the lightness of the cotyledon and the hypocotyl and the yellowness of the hypocotyl. The Fe content of cowpea sprouts was higher in those grown under red light (632 nm), and the total amino acid content was higher for those grown under yellow light (560 nm), white light (458 nm), and blue light (460 nm) compared to plants grown in the untreated condition (darkness). The yield ratio of cowpea sprouts was lower in the yellow light condition (560 nm) at lower light quantity, but no differences were observed at other light colors and quantities. The lightness and yellowness of cowpea sprouts was higher in the yellow light (560 nm) and red light (632 nm) at lower light quantity, redness was lower. No significant differences were observed in the content of normal and inorganic components according to the light quantities of each light color, except that Fe content was higher in sprouts grown under red light (632 nm) as light quantity increased. Total amino acid content was slightly higher in sprouts grown under white light (458 nm) and blue light (460 nm) as light quantity increased.
