Extraction of Geometric Primitives from Point Cloud Data

  • Object detection and parameter estimation in point cloud data is a relevant subject to robotics, reverse engineering, computer vision, and sport mechanics. In this paper a software is presented for fully-automatic object detection and parameter estimation in unordered, incomplete and error-contaminated point cloud with a large number of data points. The software consists of three algorithmic modules each for object identification, point segmentation, and model fitting. The newly developed algorithms for orthogonal distance fitting (ODF) play a fundamental role in each of the three modules. The ODF algorithms estimate the model parameters by minimizing the square sum of the shortest distances between the model feature and the measurement points. Curvature analysis of the local quadric surfaces fitted to small patches of point cloud provides the necessary seed information for automatic model selection, point segmentation, and model fitting. The performance of the software on a variety of point cloud data will be demonstrated live.

Characteristics of F$_2$Hybrids from Crosses between Korean Cultivars and Canadian Cultivars in Buckwheat(Fagopyrum esculentum Moench.)

  • Korean cultivars of buckwheat(Fagopyrum esculentum Moench.) was crossed with Canadian cultivars in order to improve seed yield as well as leaf production of buckwheat for using as food and medicine. The agronomic characteristics and rutin contents of F$_2$ hybrids are investigated for further selection of superior lines. Dry weight per plant was the highest in a line 1110(6.71g) and leaf weight per plant was the highest in a line 1110(1.91g). Hybrid seeds were 0.55 - 0.70cm long and 0.37 - 0.47cm wide on average. 100 seeds weight ranged from 2.57g to 3.58g. Line 1076 produced the longest seeds(0.70cm) and line 1186 was the longest in seed width. Line 1196 showed the highest 100 seeds weight(3.58g). The highest frequency of the LWR(length/width rate) was 0.66~0.70, indicating that seed shape of the hybrids was mostly oval. Line 1087 showed the highest contents of rutin(77.26ppm). Lines 1090 and 1181 contained respectively rutin of 54.76ppm and 54.35ppm in the seeds. From the yield and rutin point of view, the most superior lines was line 1087 among the lines used for this study.

Development of genetic transformation method of Korean soybean (국내콩 형질전환 기술개발)

  • Current status of soybean transformation method in Korera was reviewed with recent publications. Most frequently used method for genetic transformation was Agrobacterium-mediated transformation on cotyledonary node which is most popular method used in foreign country. In addition to this, various methods such as sonicationmediated transformation, in planta transformation, and transformation on meristem tissue of germinating seed, have been tried in Korea, even though their efficiencies on repeatability and stability were relatively low. Based on the promising results developed recently by reviewer, several important considerations for successful soybean transformations were suggested. They are 1) proper genotype screening, 2) targeting transformation on exact point, 3) multiple shoot formation, 4) efficient selection pressure, 5) successful shoot elongation, 6) efficient root formation. These are the basic requirements for stable and highly efficient soybean transformation of Korean soybean.

Breeding of Lilium Asiatic Hybrids 'Point' for Cut Flower with Orange Red Colored Petals (절화용 진한 주황색 아시아틱나리 신품종 '포인트' 육성)

  • A new asiatic lily cultivar 'Point' was derived from a cross between $Lilium$ $concolor$ var. $partheneion$ and 97-5 with orange colored petals made in 2000. The selection and the survey for the flower characteristics were conducted from 2005 to 2008 by using vinyl tunnels and greenhouse at Highland Agriculture Research Center, NICS, RDA. Based on flower color, 'Point' cultivar belongs to orange red group (O-R N30C). There was lots of spots in petal. The flowers had brown stigma and dark brown pollen. The stem color was dark purple. The flowering date was July 8. The plant height was 88.2 cm and flower numbers were 5.4 ea. The length and width of petal were 6.3 cm and 3.2 cm, respectively. The flower size was smaller than that of 'Prato' cultivar. The number of leaves was 12.2 ea. The length and width of leaves were 9.2 cm and 1.1 cm, respectively. The weight and circumference of bulb were about 24.6 g and 11.6 cm, respectively. 'Point' cultivar had resistance to bulb rot disease. The flowering duration was 16.1 days. The preference was good to 4.1 on 5 grades. 'Point' cultivar was registered as a new variety with No. 3446 in Korea Seed and Variety Service on April 2011.

A Study on the Hyperspectral Image Classification with the Iterative Self-Organizing Unsupervised Spectral Angle Classification (반복최적화 무감독 분광각 분류 기법을 이용한 하이퍼스펙트럴 영상 분류에 관한 연구)

  • The classification using spectral angle is a new approach based on the fact that the spectra of the same type of surface objects in RS data are approximately linearly scaled variations of one another due to atmospheric and topographic effects. There are many researches on the unsupervised classification using spectral angle recently. Nevertheless, there are only a few which consider the characteristics of Hyperspectral data. On this study, we propose the ISOMUSAC(Iterative Self-Organizing Modified Unsupervised Spectral Angle Classification) which can supplement the defects of previous unsupervised spectral angle classification. ISOMUSAC uses the Angle Division for the selection of seed points and calculates the center of clusters using spectral angle. In addition, ISOMUSAC perform the iterative merging and splitting clusters. As a result, the proposed algorithm can reduce the time of processing and generate better classification result than previous unsupervised classification algorithms by visual and quantitative analysis. For the comparison with previous unsupervised spectral angle classification by quantitative analysis, we propose Validity Index using spectral angle.

A Novel Medium for the Enhanced Production of Cyclosporin A by Tolypocladium inflatum MTCC 557 Using Solid State Fermentation

  • Cyclosporin A (CyA) produced by Tolypocladium inflatum is a promising drug owing to its immunosuppressive and antifungal activities. From an industrial point of view, the necessity to obtain a suitable and economic medium for higher production of CyA was the aim of this work. The present study evaluated the effect of different fermentation parameters in solid state fermentation, such as selection of solid substrate, hydrolysis of substrates, initial moisture content, supplementation of salts, additional carbon, and nitrogen sources, as well as the inoculum age and size, on production of CyA by Tolypocladium inflatum MTCC 557. The fermentation was carried out at $25{\pm}2^{\circ}C$ for 9 days. A combination of hydrolyzed wheat bran flour and coconut oil cake (1:1) at 70% initial moisture content supported a maximum production of $3,872{\pm}156\;mg$ CyA/kg substrate as compared with $792{\pm}33\;mg/kg$ substrate before optimization. Furthermore, supplementation of salts, glycerol (1% w/w), and ammonium sulfate (1% w/w) increased the production of CyA to $5,454{\pm}75\;mg/kg$ substrate. Inoculation of 5 g of solid substrate with 6 ml of 72-h-old seed culture resulted in a maximum production of $6,480{\pm}95\;mg$ CyA/kg substrate.

Effect of Callus Type and Antioxidants on Plant Regeneration and Transformation of Tall Fescue (캘러스의 형태와 항산화물질 첨가가 톨 페스큐의 식물체 재분화와 형질전환효율에 미치는 영향)

  • An efficient transformation system for the production of transgenic plants has been developed for tall fescue (Festuca arundinacea Schreb.) via Agrobacterium-mediated transformation of seed-derived callus. From the point of morphogenetic capacity, three types of callus were selected. High frequency of plant regeneration was obtained by selection of type II callus, and the plant regeneration frequency was 52.6% when embryogenic callus were cultured on the regeneration medium. Supplementation of the media with 10 mg/L $AgNO_3$ and 40 mg/L cysteine enhanced frequencies of plant regeneration up to 65.3%. The highest transformation efficiency was also obtained when type II callus were inoculated with Agrobacterium. Southern blot analysis of PCR products of transgenic plants demonstrated that transgenes were successfully integrated into the genome of tall fescue. Efficient regeneration system and transformation established in this study will be useful for molecular breeding of tall fescue through genetic transformation.

Development of Selectable Marker of High Oleate Trait in Peanut (Arachis hypogaea L.) (땅콩에서 고 올레인산 형질관련 분자마커의 선발)

  • Peanut(Arachis hypogaea L.) is one of the major oilseed crops. The peanut oil consists of palmitic, oleic and linoleic acids, which are present at levels of 10%, 36-67% and 15-43%, respectively. High oleate mutant of peanut F435 contains 80% oleate and as little as 2% linoleate in seed oil. Previous study indicated that delta 12 fatty acid desaturase is a major enzyme controlling the oleate content in seeds of oilseed crops. F435 sequence alignment of their coding regions disclosed that an extra A(adenine) was inserted at the position +2,823 bp of delta 12 fatty acid desaturase gene. This study was to develop molecular marker (SNP marker) co-segregating with the high oleate trait. Chopyeong ${\times}$ F435 $F_2$ 41 population were investigated using molecular marker and fatty acid assay (NIR and gas chromatography). Finally, this marker segregates Chopyeong type 26 lines, heterotype 9 lines and F435 type 6 lines. These results in our study suggested that SNP marker conform fatty acid assay.
