Terminal-restriction fragment length polymorphism (T-RFLP) analysis has been optimized by using in vitro model community composed of genomic DNAs of known bacterial strains and has been applied to assess the bacterial community structure in marine sediments. The specific fluorescence-labeled terminal restriction fragments (T-RFs) between 39 and 839 base long specifying each strain were precisely measured for known bacterial strains. The addition of a co-solvent (dimethylsulfoxide or glycerol) into PCR reactions has reduced differential PCR amplification. Comparative bacterial community structure was investigated for pristine and polluted sediments. A complex T-RFLP pattern showing complex bacterial community structure was obtained in the pristine sediment, whereas simple T-RFLP pattern (low bacterial diversity) was shown in polluted sediments where caged aquaculture has been conducted for several years. The results of T-RFLP analysis were compared with that of cloning and sequencing 16S rDNA clones from the same sediments. Sequence analysis of 16S rDNA clones (72) of the pristine sediment revealed a diverse collection of lineages, largely of the class Proteobacteria ($6\%$ alpha subdivision, $46\%$ gamma subdivision, $13\%$ delta subdivision, and $3\%$ epsilon subdivision), Nitrospina $(8\%)$, high G+C gram positive $(8\%)$, Verrucomicrobia $(7\%)$, and Planctomycetes $(6\%)$. In the contaminated sediments, 17 $(59\%)$ of the 16S rDNA clones (29) were related to Campylobacter and symbiont of Rimicaris exoculata belonging to epsilon subdivision of Proteobacteria. The results obtained indicated that T-RFLP analysis is a rapid and precise technique for comparative bacterial community analysis.
Variation in the microbial biomass and community structure found in sediment of heavily polluted bays and the adjacent unpolluted areas were examined using phospholipid fatty acid analysis. Total microbial biomass and microbial community structure were responding to environmental determinants, sediment grain size, depth of sediment, and pollution due to petroleum hydrocarbons. The marker fatty acids of microeukaryotes and prokaryotes - aerobic, anaerobic, and sulfate-reducing bacteria - were detected in sediments of the areas studied. Analysis of the fatty acid profiles revealed wide variations in the community structure in sediments, depending on the extent of pollution, sediment depth, and sediment grain size. The abundance of specific bacterial fatty acids points to the dominance of prokaryotic organisms, whose composition differed among the stations. Fatty acid distributions in sediments suggest the high contribution of aerobic bacteria. Sediments of polluted sites were significantly enriched with anaerobic bacteria in comparison with clean areas. The contribution of this bacterial group increased with the depth of sediments. Anaerobic bacteria were predominantly present in muddy sediments, as evidenced from the fatty acid profiles. Relatively high concentrations of marker fatty acids of sulfate-reducing bacteria were associated with organic pollution in this site. Specific fatty acids of microeukaryotes were more abundant in surface sediments than in deeper sediment layers. Among the microeukaryotes, diatoms were an important component. Significant amounts of bacterial biomass, the predominance of bacterial biomarker fatty acids with abundance of anaerobic and sulfate-reducing bacteria are indicative of a prokaryotic consortium responsive to organic pollution.
Analysis of one sinkhole, the Dodgeville sinkhole, developed in Ordovician dolostones in the Driftless Area of Wisconsin in the Upper Midwest'd Driftless Area reveals homogenous clayey sediment fills reflecting a range of dissolutional processes during the Quaternary or Pre-Quaternary. Granulometric analysis, graphical moments statistics, carbonate minerals, ana sand grain lithology were used to differentiate sinkhole sediment sources and modes of accumulation. Sediments in the dolostone sinkholes developed by dissolution. Sediments contain two major types of sediments : residual redish clay( autogenic sediments) and aeolian silt (allogenic sediments). The massive clay is generated from the weathered dolostone bedrocks as a in situ materials. The loessial silt is mostly derived from transportation of the surrounding surface materials, with some evidences of penetrated deposition. Unlike the collapsed sandstone sinkholes (Oh et al., 1993), dolostone sinkholes reveal homogenous, autogenic clay materials, and a geochemical composition indicative of in situ autogenic karstification. Dolostone sinkhole si1ts (26.9%) and sands (34.9%) are derived from weathered Plattevi1le-Galena dolostones, and contain high carbonate(37.5%), chert (57.2%) and lead ore (3%). Graphical moments statistics for sorting, skewness, and kurtosis indicate that sand grains from dolostones were derived entirely from local bedrock by in situ dissolution. Upper sinkhole sediments are pedagogically very young as carbonate is unleashed. Materials of the sinkhole sediment are definitely inherited from internal dolostones by dissolution and weathering, because not only a granulomatric comparison of dolostone and sandstone sediments demonstrates that they have heterogeneous paticle size distributions, but also 1ithologic analyses displays they differ completely.
This study was carried out geochemically and mineralogically to define how Kunsan shore sediments are related to their terrestrial source rocks in the region of Keum River Basin, western Korea. As a whole the chemical composition for major elements, trace elements and rare earth elements analysis from shore sediments and river bed sediments doesn't show the big difference, and especially rare earth elements chondrite normalized patterns are almost same. Heavy minerals of shore sediments are identified as hornblende, epidote, ilmenite, garnet, hematite, magnetite, sphene and rutile. Compared with Taean Area of Seo et al. (1998) and Byeonsan Area of Kwon et al. (1999), Kunsan shore sediments of this study area were origined mostly from Keum River Basin.
Analysis was done on the particle size distribution of sediments flown into Saemangeum from the Mankyung and Dongjin River. The organic pollutants and heavy metal existing in the sediments were analyzed, which was further used to study the properties of pollution in the sediments according to the particle size distribution. Conclusions shown below were made from these analyses. The particle size distribution showed a big difference between the upriver areas of Mankyung and Dongjin River. Particles under $75{\mu}m$ showed to be around 85% at Dongjin River, while it showed to be around 70% at Mankyung River. This kind of distribution in particle size concluded in greatly affecting the contamination density of the sediments. From the analysis done on the soil type of sediments, deposition in Mankyung River categorized into Silty loam and Sandy loam, where Silty loam covered most of area and deposition in Dongjin River categorized into Sand, Loamy sand, Silty loam, Sandy loam. Considering the weight ratio, the density of contamination of the sediments by particle size at Dongjin and Mankyung River has been analyzed to show that organic pollutants and heavy metals occupy more than 70% of the whole contamination in the range under the particle size of $75{\mu}m$.
determine the distribution of heavy metals in estuarine sediments, forty surface sediment samples were collected from the Youngsan estuary, and analysed for their contents of Fe, Mn, Co, Cr, Cu, Ni, Zn and Pb. Contents of most analysed metals were comparable to those in the near shore environment around Korean peninsula, and little anthropogenic effect was identified. However, there was a contrasting behavior among the metals with respect to their relationship with the grain-size of sediments. Contents of Fe, Co, Cr and Ni demonstrated a fairly linear relationship with the grain-size of sediments whereas those of Mn, Cu, Zn and Pb did not. This different behavior In these metals was Interpreted as being the result of the additional inputs. order to investigate physical characteristics and element concentrations of sediments, coastal bottom sediments were collected at 20 stations in the vicinity of Youngkwang Nuclear Power Plant. After air drying of samples in the laboratory. article size distribution was examined by Master sizer (X-350F), radio-activity by HPGe ${\gamma}$-spectrphotometer, and element concentrations by ICP-AES and AAS. According to particle size analysis , sediments are mainly composed of silt fraction weith 23% of sand, 65% of silt and 12% of clay on average. Most sediments are derived from muddy environment that silt dominates with the characteristics of 5.3${\varsigma}$ mean particle size, poorly sorted, very fine skewed and lepto-kurtic. Only two sediments are well sorted with sandy silt owing to wind, winnowing action, tide and current andits complex reactions. Element concentrations in the coastal bottom sediments are relatively high at finer sediment and show significant relationship with grain size. Index of geoaccumulation by heavy metals at every sampling station is classified as practically unpolluted. The radioactivities of the sediments were measured for 15 isotope elements, and 2 elements of K-40 and Cs-137 were detected in most sediments. The K-40 is the natural nuclide and the artificial nuclide of Cs-137 was thought to be derived from the fallout of past nuclear weapon test. The results of correlation coefficient between grain size and radioactivity shows that the activity of Cs-137 significantly increases in finer grain.
황해 남동 이질대에서 채취된 51개의 표층퇴적물 시료와 황해로 유입되는 한국과 중국의 하천퇴적물 시료 33개에 대해서 반정량 X선회절분석법을 통한 점토광물의 상대적인 함량을 토대로 주성분분석을 도입하여 해양표층퇴적물의 기원지를 연구하였다. 전체 자료의 98% 이상을 반영하는 제1주 성분과 제2주성분을 이용하여 R 프로그램을 통해 주성분분석을 수행하였다. 황해 남동 이질대 퇴적물의 점토광물 함량은 남부 지역과 북부 지역에서 거의 차이가 나지 않으며, 성분도표상에서의 해양퇴적물 분포는 중국의 황하, 양쯔강 퇴적물보다 한국의 하천퇴적물과 더욱 근접한 양상을 보였다. 이러한 통계적 분석 결과를 바탕으로 황해 남동 이질대는 거의 모두가 한국 하천퇴적물로부터 유래했다는 것을 추정할 수 있다. 또한, 황해 남동 이질대에 한국 하천에서 유출되는 양보다 더 많은 퇴적물이 축적된 이유로는 퇴적물의 침식과 재동에 의한 것으로 판단된다. 주성분분석은 한반도 주변 해역 퇴적물의 기원지 연구에 유용하게 사용될 것으로 기대된다.
본 연구는 안동지역에 소재하는 안동 임하호 저니토가 호소수에 미치는 영향을 조사하기 위하여 안동호와 임하호 6개 지역에서 채취한 저니토의 토양성분과 중금속 용출 특성을 분석하였다. 대부분의 저니토는 illite(I), kaolinite(Ka), quartz(Q) 및 feldspar(F)로 구성되어 있다. 저니토에 포함된 중금속 중 아연(Zn), 비소(As), 크롬(Cr), 구리(Cu), 납(Pb)의 농도는 임하호에 비하여 안동호에서 상대적으로 높게 나타났다. 특히 안동호의 저니토에서는 다른 중금속들에 비해 비소(As)와 아연(Zn)의 용출이 크게 나타났다. 저니토를 이용한 흡착실험에서는 아연(Zn), 카드늄(Cd) 및 구리(Cu)의 흡착은 적게 일어난 반면 납(Pb)과 크롬(Cr)은 높은 흡착력을 보여주었다.
한반도 동남해역(한국해협) 대륙붕의 표층 퇴적물의 입도 북석 자료에 대한 Q-Mode 요인 분석을 실시하였고, 그 결과는 해수면 변동에 따른 퇴적환경의 변화 를 시사하는 것으로 해석된다. 즉 외대륙붕과 붕단의 퇴적물은 분급이 양호한 사질 퇴적물이며 연근해성 패각을 많이 포함하고 있으므로 해수면이 낮았을 때 (약-150 m)집적된 잔유 퇴적물로 생각된다. 최대 수심 230m에 달하는 한국해곡(Korean Tro ugh)에 분포하는 퇴적물은 주로 원마도가 매우 양호한 역질 물질이며 이들은 해수 면이 상승하면서 재동작용을 받은 Palimpsest퇴적물로 해석된다. 수심 약 120m 이 내의 내 대륙붕에는 실트와 점토가 우세한 니질 퇴적물이 분포하며 이들은 해수면 이 거의 현 수준으로 상승한 지난 7000년 이후에 집적된 것으로서, 소위 내 대륙붕 니질대를 이루고 있다.
