Cloning and Characterization of a Heterologous Gene Stimulating Antibiotic Production in Streptomyces lividans TK-24

  • Genetic determinant for the secondary metabolism was studied in heterologous expression in Streptomyces lividans TK-24 using Streptomyces griseus ATCC 10137 as a donor strain. Chromosomal DNA of S. griseus was ligated into the high-copy number Streptomyces shuttle plasmid, pWHM3, and introduced into S. lividans TK-24. A plasmid clone with 4.3-kb BamHI DNA of S. griseus (pMJJ201) was isolated by detecting for stimulatory effect on actinorhodin production by visual inspection. The 4.3-kb BamHI DNA was cloned into pWHM3 under the control of the strong constitutive ermEp promoter in both directions (pMJJ202); ermEp promoter-mediated transcription for coding sequence reading right to left: pMJJ203; ermEp promoter-mediated transcription for coding sequence reading left to right) and reintroduced into S. lividans TK-24. The production of actinorhodin was markedly stimulated due to introduction of pMJJ202 on regeneration agar. The introduction of pMJJ202 also stimulated production of actinorhodin and undecylproidigiosin in submerged culture employing the actinorhodin production medium. Introduction of pMJJ203 resulted in a marked decrease of production of the two pigments. Nucleotide sequence analysis of the 4.3-kb region revealed three coding sequences: two coding sequences reading left to right, ORF1 and ORF2, one coding sequence reading right to left, ORF3. Therefore, it was suggested that the ORF3 product was responsible for the stimulation of antibiotic production. The C-terminal region of ORF3 product showed a local alignment with Myb-related transcriptional factors, which implicated that the ORF3 product might be a novel DNA-binding protein related to the regulation of secondary metabolism in Streptomyces.

Growth, secondary metabolite production and antioxidant enzyme response of Morinda citrifolia adventitious root as affected by auxin and cytokinin

  • Morinda citrifolia adventitious roots were cultured in shake flasks using Murashige and Skoog medium with different types and concentrations of auxin and cytokinin. Root (fresh weight and dry weight) accumulation was enhanced at 5 $mg\;l^{-1}$ indole butyric acid (IBA) and at 7 and 9 $mg\;l^{-1}$ naphthalene acetic acid (NAA). On the other hand, 9 $mg\;l^{-1}$ NAA decreased the anthraquinone, phenolic and flavonoid contents more severely than 9 $mg\;l^{-1}$ IBA. When adventitious roots were treated with kinetin (0.1, 0.3 and 0.5 $mg\;l^{-1}$) and thidiazuron (TDZ; 0.1, 0.3 and 0.5 $mg\;l^{-1}$) in combination with 5 $mg\;l^{-1}$ IBA, fresh weight and dry weight decreased but secondary metabolite content increased. The secondary metabolite content (including 1,1-diphenyl-2-picrylhydrazyl activity) increased more in TDZ-treated than in kinetin-treated roots. Antioxidative enzymes such as catalase (CAT) and guaiacol peroxidase (G-POD), which play important roles in plant defense, also increased. A strong decrease in ascorbate peroxidase activity resulted in a high accumulation of hydrogen peroxide. This indicates that adventitious roots can grow under stress conditions with induced CAT and G-POD activities and higher accumulations of secondary metabolites. These results suggest that 5 $mg\;l^{-1}$ IBA supplementation is useful for growth and secondary metabolite production in adventitious roots of M. citrifolia.

Production of Mouse Anti-Quail IgY and Subsequent Labeling with Horseradish Peroxidase Using Cyanuric Chloride

  • Polyclonal antibodies labeled with a tracer have been commonly used as secondary antibodies in immunochemical assays to quantify the concentration of antibody-antigen complexes. The majority of these antibodies conjugated with a tracer are commercially available, with the exception of few untouched targets. This study focused on the production and application of mouse anti-quail IgY as an intermediate antibody to link between quail egg yolk IgY and goat anti-mouse IgG-HRP as primary and secondary antibodies, respectively. Subsequently, the produced mouse anti-quail IgY was labeled with horseradish peroxidase (HRP) and its efficiency on enzyme linked immunosorbent assay (ELISA) was compared with that of commercial rabbit anti-chicken IgY-HRP. As an intermediate antibody, mouse anti-quail IgY was successfully produced with good affinity and sensitivity (1:10,000) to the primary and secondary antibodies. Subsequently, mouse anti-quail IgY was effectively conjugated with HRP enzyme, resulting in a secondary antibody with good sensitivity (1:10,000) to quail anti-V. parahaemolyticus and V. vulnificus IgY. The detection limit was $10^5$ CFU/ml for both V. parahaemolyticus and V. vulnificus. The efficiency of the produced conjugate to detect quail IgY on ELISA was comparable to that of the commercial rabbit anti-chicken IgY-HRP, and hence the produced and labeled mouse anti-quail IgY-HRP can be used as a secondary antibody to detect any antibody produced in quail.

Vanadium Haloperoxidase의 구조와 작용 메커니즘과 해양천연물질 (Reaction Mechanism of Vanadium Haloperoxidase and Marine Natural Products)

  • 다양한 생리활성을 가지는 marine natural products는 일반적인 secondary metabolites와 유사한 구조를 가지는데, 염소, 브롬, 요오드의 할로겐 원소에 의해 수식이 되어있는 것이 일반적이다. Vanadium haloperoxidase는 이러한 marine natural products의 생산에 중요한 효소로 vanadate를 조효소로 하는 금속효소이다. 본 리뷰에서는 vanadium haloperoxidase의 분리와 단백질 구조를 살펴보고, 이 금속효소의 작용기작에 대해서 설명할 것이다. 마지막으로, vanadium haloperoxidase의 반응성과 secondary metabolites 중 indole, terpenoids, acetogenins의 생합성 예를 살펴볼 것이다.

Involvement of lymphoid inducer cells in the development of secondary and tertiary lymphoid structure

  • During development lymphoid tissue inducer (LTi) cells are the first hematopoietic cells to enter the secondary lymphoid anlagen and induce lymphoid tissue neogenesis. LTi cells induce lymphoid tissue neogensis by expressing a wide range of proteins that are associated with lymphoid organogenesis. Among these proteins, membrane-bound lymphotoxin (LT) $\alpha1\beta2$ has been identified as a critical component to this process. LT$\alpha1\beta2$ interacts with the LT$\beta$-receptor on stromal cells and this interaction induces up-regulation of adhesion molecules and production of chemokines that are necessary for the attraction, retention and organization of other cell types. Constitutive expression of LT$\alpha1\beta2$ in adult LTi cells can result in the formation of a lymphoid-like structure called tertiary lymphoid tissue. In this review, we summarize the function of fetal and adult LTi cells and their involvement in secondary and tertiary lymphoid tissue development in murine models.

포항지역 지열수에 대한 지화학적 고찰

  • To investigate the evidence for the influence of sea water on Pohang geothermal groundwater, the chemical data for geothermal groundwaters from which are pumped during 48 hours and other hot groundwaters, another groundwater on the well for the purpose of agriculture, were considered. And to predict possible the secondary mineral which are easily to make the clogging, geochemical modeling was carried out using EQ3NR equilibrium solubility code. The results are that 1.4%~3.3%(bulk composition) of sea water were mixed with geothermal groundwater. From the well logging data, when the level of groundwater is drow down, the conductivity is increased in the geothermal groundwater, the existence of transition zone are recognized in the well. The predicted possible secondary minerals are Antigorite [Mg48Si24O85(OH)62], Chrysolite [Mg3Si2O5(OH)4] , Cristobalite, Dolomite, Talc, Tremolite. The recommended cooling temperature of best condition to minimize the production of secondary minerals is same as temperature of geothermal water pumped from the well.

  • Lithium accounts for only 0.0017% of the earth crust, and it is produced in geographically limited regions such as South America, the United States, and China. Since the first half of 2017, the price of lithium has been continuously increasing, and with the rapid adoption of electric vehicles, lithium resources are expected to be depleted in the near future. In addition, economic blocs worldwide face intensifying scenarios such as competition for technological supremacy and protectionism of domestic industries. Consequently, Korea is deepening its dependence on China for core materials and is vulnerable to the influence of the United States Inflation Reduction Act. We analyze post-lithium secondary battery technologies that rely on more earth-abundant elements to replace lithium, whose production is limited to specific regions. Specifically, we focus on the technological status and issues of sodium-ion, zinc-air, and redox-flow batteries. In addition, research trends in post-lithium secondary batteries are examined. Post-lithium secondary batteries seem promising for large-capacity energy storage systems while reducing the costs of raw materials compared with existing lithium-based technologies.

Targeting the Osmotic Stress Response for Strain Improvement of an Industrial Producer of Secondary Metabolites

  • The transition from primary to secondary metabolism in antibiotic-producing Streptomyces correlates with expression of genes involved in stress responses. Consequently, regulatory pathways that regulate specific stress responses are potential targets to manipulate to increase antibiotic titers. In this study, genes encoding key proteins involved in regulation of the osmotic stress response in Streptomyces avermitilis, the industrial producer of avermectins, are investigated as targets. Disruption of either osaBSa, encoding a response regulator protein, or osaCSa, encoding a multidomain regulator of the alternative sigma factor SigB, led to increased production of both oligomycin, by up to 200%, and avermectin, by up to 37%. The mutations also conditionally affected morphological development; under osmotic stress, the mutants were unable to erect an aerial mycelium. In addition, we demonstrate the delivery of DNA into a streptomycete using biolistics. The data reveal that information on stress regulatory responses can be integrated in rational strain improvement to improve yields of bioactive secondary metabolites.

Adventitious root culture for secondary metabolite production in medicinal plants: A Review

  • Medicinal plants are high-value natural resources that have been used as precautionary drugs by many people globally. The increasing global demand for bioactive compounds from medicinal plants has led to the overexploitation of many valuable species. One widely used approach to overcome this problem is the use of adventitious root cultures as a propagation strategy. This review examines the scientific research published globally on the application of adventitious root cultures for many medicinal plants. Adventitious roots generated under aseptic environments in suitable phytohormone-augmented medium exhibit high growth rates and production of important secondary metabolites. Parameters such as medium properties and composition, growth hormone type, and elicitation strategies for in vitro grown adventitious roots of medicinal plants, are the main topics discussed in this review. We also examine current developments in bioreactor system cultivation for plant bioactive compounds using adventitious root cultures, a technology with possible commercial applications, via several studies on adventitious root culture of medicinal plants in which bioreactor systems play a role. In conclusion, the development of adventitious root cultures for medicinal plants is highly useful because of their capability for vegetative propagation and germplasm preservation.

Effect of New Improved Technology of Silkworm Bombyx mori L. Rearing on the Egg Production Capacity

  • Experiments were held in Egypt to test a new Package of practices for commercial cocoon production, including adding secondary macro and micronutrients to the mulberry garden, disease free rearing regime, low temperature rearing in young instars and natural mounting fur silkworms. This package for seed silkworm rearing lead to increase in fecundity by 67-121eggs (15.12-26.22%) and yield of standard boxes per 1 parent egg boxes by 57-58 egg boxes(48.33-51.66%) respectively in comparison th the traditional cocoon production technology.
