Genetic and Functional Analyses of the DKxanthene Biosynthetic Gene Cluster from Myxococcus stipitatus DSM 14675

  • DKxanthenes are a class of yellow secondary metabolites produced by myxobacterial genera Myxococcus and Stigmatella. We identified a putative 49.5 kb DKxanthene biosynthetic gene cluster from Myxococcus stipitatus DSM 14675 by genomic sequence and mutational analyses. The cluster consisted of 15 genes (MYSTI_06004-MYSTI_06018) encoding polyketide synthases, non-ribosomal peptide synthases, and proteins with unknown functions. Disruption of the genes by plasmid insertion resulted in defects in the production of yellow pigments. High-performance liquid chromatography and liquid chromatography-tandem mass spectrometry analyses indicated that the yellow pigments produced by M. stipitatus DSM 14675 might be novel DKxanthene derivatives. M. stipitatus did not require DKxanthenes for the formation of heat-resistant viable spores, unlike Myxococcus xanthus. Furthermore, DKxanthenes showed growth inhibitory activity against the fungi Aspergillus niger, Candida albicans, and Rhizopus stolonifer.

  • An orthotropic steel deck system is widely adapted form for a long-span bridge. It has many advantages, such as the big reduction of dead weight, the simplicity for erection, and the reduction of the construction period. However, an orthotropic steel deck system requires a lot of welding work, which may result in defects and deformation of connection. Therefore, the research for the general behavior and fatigue strength of the several details in orthotropic steel deck bridge is necessary. The fatigue failure with distortion results from secondary stress by out-of-plane deformation; these kinds of cracks are very difficult to measure, and can not be precisely calculated through finite element analysis. This stress concentration phenomenon generates the fatigue failure around the lower scallop of the transverse rib. This paper presents improved details of the intersection between the longitudinal rib and the transverse rib of an orthotropic steel deck bridge by the third dimensional hit size test, and the finite element method, which can minimize local stress through parametric study.

  • In the case of long secondary (applied in long distance), as it is established horizontal, the defects of droop arises resulting damage of entire system. Actually, it is difficult to expect reasonable characteristics and desirable operating in that case. This paper is about numerical analysis of secondary droop, non-symmetric and position displacement problems. In the base of this paper, the correction of mechanical difficulties in practical manufacturing and optimal design of TLIM considering these aspects can be progressed successively.

  • Characteristics of doped MgO layer deposited under hydrogen atmosphere were investigated. Hydrogen gas was introduced during e-beam evaporation of doped MgO and its effects on microstructure, cathodoluminescence, discharge voltages and effective yield of secondary electron emission were examined. The results indicated that the hydrogen influences and doped impurities the concentration and energy levels of defects in MgO layer and that affects the luminance efficiency and discharge delays of the panels significantly.

  • ZnWO4 single crystals were grown by Czochralski method. And the orientation of grown crystals were determined by Laue back reflection, and the crystals were siliced at (100), (010), (001) face before polishing. The morphologys and distribution of etch pits on each face were observed by optical microscopy. In the present study, we understood that dislocation distributjioon rely on shape of solid-liquid interface, and secondary phase acts on the dislocation source. We also observed dislocation trace(etch pits) of (100) slip plane on (010) cleavage plane.

On Power Allocation Schemes for Bi-directional Communication in a Spectrum Sharing-based Cognitive Radio System

  • This paper presents the results of an investigation into bi-directional communication in spectrum sharing-based cognitive radio (Bi-CR) systems. A Bi-CR system can increase the spectral efficiency significantly by sharing the spectrum and through the bi-directional use of spatial resources for two-way communication. On the other hand, the primary user experiences more interference from the secondary users in a Bi-CR system. Satisfying the interference constraint by simply reducing the transmission power results in performance degradation for secondary users. In addition, secondary users also experience self-interference from echo channels due to full duplexing. These imperfections may weaken the potential benefits of the Bi-CR system. Therefore, a new way to overcome these defects in the Bi-CR system is needed. To address this need, this paper proposes some novel power allocation schemes for the Bi-CR system. This contribution is based on two major analytic environments, i.e., noise-limited and interference-limited environments, for providing useful analysis. This paper first proposes an optimal power allocation (OPA) scheme in a noise-limited environment and then analyzes the achievable sum rates. This OPA scheme has an effect in the noise-limited environment. In addition, a power allocation scheme for the Bi-CR system in an interference-limited environment was also investigated. The numerical results showed that the proposed schemes can achieve the full duplexing gain available from the bi-directional use of spatial resources.

  • The natural graphite particles A and heat-treated graphite particles B at $1800\;^{\circ}C$ after pitch-coating were used as the anode base materials for lithium ion secondary battery. In order to improve the performance of anode materials, the base anode materials were treated with various acids. With the acid treatments of 62% $HNO_3$ and 95% $H_2SO_4$ aqueous solution, the specific surface area and electrical conductivity of base anode materials were increased, and the initial charge-discharge capacity and cycle performance were improved due to the elimination of structural defects.

Effects of Risering Design and Chemical Composition on Formation of Shrinkage Cavity in Gray Cast Iron (회주철의 수축결함생성에 미치는 주조방안 및 화학조성의 영향)

  • The effects of risering design and alloying element on the formation of defects such as external depression, primary and secondary shrinkage cavity in gray cast iron were investigated. Two types of risering design for the cylindrically step-wise specimen, No. 1(progressive solidification) and No. 2(directional solidification) risering designs, were prepared and five different alloy compositions were casted. In the No. 1 risering design, external depression or primary shrinkage cavity due to liquid contraction was observed in all the specimens from ISO 150 to ISO 350. The primary shrinkage cavity was located right under the top surface or connected to the top surface, and was characterized by smooth surface. Its size increased with an increase in ISO number. However, neither secondary shrinkage cavity nor swollen surface was observed in all the castings. In the No.2 risering design, neither primary shrinkage cavity nor secondary shrinkage cavity was observed in all the specimens due to proper risering design. A swollen surface was also not observed in all the castings with the application of pep-set mold.

  • In this work the friction and wear characteristics of the gray cast iron surface processed by broaching method, which is widely used in the machinery industry, were investigated. The broaching process is mainly used for mass production because it has high dimensional accuracy and processing speed, but the defects on surface can be easily generated. In order to improve the tribological characteristics, the approach was to reduce the roughness and hardness of the surface by adding a machining process to the broaching specimen. The secondary machining process using abrasive grains produces low roughness and hardness than broaching because it has high tool accuracy and removes the work hardened surface. The friction coefficient and the wear rate were assessed using a reciprocating-type tribotester to analyze the effects of surface finishing on the tribological properties. The friction tests were conducted under dry and lubricated conditions. The test results showed that the reduction of surface roughness and hardness through secondary machining process in lubricated condition improved the friction and wear characteristics. The reason why the same results did not appear in a dry condition was that wear occurred more rapidly than in lubricated condition. Thus, the positive effect of roughness and hardness of the surface obtained through the secondary machining process was not observed.
