  • The purpose of the study is to build a general cost a counting model for university libraries, to clarify the possible areas of its a n.0, pplication by employing job cost accounting and process cost accounting methods. System analysis is performed as to the fields of acquisition, processing (cataloging & classification), and book shelving system. The existing operation processes and time required for each operation of these three systems are analyzed, from which detailed system flowcharts were drawn and job descriptions and the content of job were identified. The results of the study can be summarized as follows: (1) The processing time of one book in each systems: Oriental books. a. Acquisition system. the time required the time required in case of job cost case of job cost accounting after purchasing, 8 min. 21 sec. the time required in case of process cost accounting 15 min. 7 sec. b.   • The SecY is a central component of the protein export machinery that mediate the translocation of secretory proteins across the plasma membrane, and has been known to be rate-limiting factor of secretion in Escherichia coli. In order to study the extracellular protein secretion in Gram-positive microorganism, we have, constructed strains harboring more than one copy of the gene for SecY. Firstly, the gene, for B. subtilis SecY and its promoter region was subcloned into pDH32 and the chimeric vector was inserted into amyE locus by homologous recombination. Secondly, low copy number vector, pCED6, was also used for subcloning the secY gene and for constructing a strain which harbors several copies of secY. The KH1 cell which harbor two copies of secY on the chromosome excreted more extracellular proteins than the wild type PB2. Moreover, the KH2 cells which harbor several copies of secY in pCED6 vector excreted more extracellular proteins than the KH1 cells. Here, we found that the capacity of protein secretion is partly controlled by the number of secY and it is suggested that SecY has also an important role in protein secretion in B. subtilis, a gram positive microorganism, as like in E. coli. This will promote the use of B. subtilis as a host for the expression of useful foreign gene and excretion of precious proteins.

Seismic Analysis Methodology for Non-Nuclear Safety Piping in Nuclear Power Plants (원자력발전소 비안전등급 배관의 내진해석 방법론 연구)

  • Currently, there is no technical standard and regulation for seismic analysis of non-nuclear safety piping. Accordingly, ASME Sec.III ND, a standards applied to safety class 3 piping, is applied. However, the technical standard applied for other than seismic analysis is ASME B31, which leads to controversy. In this study, the feasibility of applying ASME B31E was confirmed by reviewing rulescomparing technical standards, and evaluating piping allowable stress margins. The evaluation revealed that applying ASME B31.1 as a technical standard is too conservative compared to ASME Sec.III ND. On the other hand, ASME B31E (issued at the request of the industry) clearly presents the technical standards for seismic analysis of ASME B31 piping, and shows a similar level of conservatism compared to ASME Sec.III ND. It is expected to reduce the controversy over technical standards for seismic analysis of non-nuclear safety piping by applying ASME B31E.

The Kinematic Analysis of Horizontal Velocity and Records from Start to Finish of the Elite Women 100m Hurdler (엘리트 여자 100m 허들선수의 스타트에서 피니쉬까지 구간별 수평속도 및 기록에 관한 운동학적 분석)

  • The purpose of this study was to observe the kinematic pattern of elite women 100m hurdler race from start to finish and analyze how the change of horizontal velocity makes an influence on the athletes' performance. The analysis was based on the performance of Korean elite 100m hurdler A and international elite hurdlers B and C. Following results were drawn from the analyzation of elite 100m hurdlers' technical characteristics; During the race, hurdler A made more than 8 m/s of horizontal velocity at the 3rd, 4th, 6th, and the 7th hurdle. The horizontal velocity peaked at the 4th hurdle with 8.23 m/s. On the other hand, hurdler B and hurdler C maintained more than 8 m/s of horizontal velocity from the 2nd hurdle through the 10th hurdle. Hurdler B's fastest horizontal velocity was 8.67 m/s from the 6th to the 7th hurdle and hurdler C's fastest horizontal velocity was 8.85 m/s from the 5th to the 8th hurdle. From the start line to the 3rd hurdle, the times achieved by hurdlers A, B, and C were 4.90 sec, 4.65 sec, and 4.52 sec. In the middle of the race, which is from the 4th hurdle to the 7th hurdle, hurdlers A, B, and C ran in 9.10 sec, 8.60 sec, and 8.38 sec. And the latter part of the race to the 10th hurdle, the times hurdlers A, B, and C hit were 12.32 sec, 11.66 sec, and 11.32 sec. To the finish line, it took 1.15 sec for hurdler A, 1.1 sec for B, and 1.06 sec for C. Hence, to set the record of sub-13 sec, hurdler A should improve her acceleration from the start line to the 1st hurdle with the speed more than 5.4 m/s and should maintain more than 8 m/s of horizontal velocity from the 2nd hurdle through the 10th hurdle. In addition, hurdler A should improve her speed endurance to minimize the deceleration of horizontal velocity from the 4th hurdle to the final hurdle. If hurdler A could shorten 0.05 sec of time in each hurdle section, she would be able to set the record under 13 seconds.

  • The purpose of this study was analysis inter relative the ball velocity and isotonic torque of shoulder joint. The subject were twelve overthrow pitcher of professional baseball player. The measurement was used Cybex 6000. The results of this study were as follows : 1. As the siting position of maximal adduction torque of dominant, A group was higher than B group. As the non-dominant, B group was higher than A group for improved angular velocity. There is no significance difference between group. 2. As the siting position of maximal abduction torque of dominant and non-dominant, B group was higher than A group for all measurement. There is no significance difference between group. 3. As the layed position of maximal internal torque of dominant, A group was higher than B group. As the non-dominant was just higher at $180_{\circ}$/sec than B group, B group was higher at $60_{\circ}$/sec and $300_{\circ}$/sec than A group. There is no significance difference between group. 4. As the layed position of maximal external torque of dominant and non-dominant, A group was more higher than B group for all measurement. There is no significance difference between group. 5. As the standing position of maximal internal torque of dominant, A group was just higher at $60_{\circ}$/sec than B group, B group was higher at $180_{\circ}$/sec and $300_{\circ}$/sec than A group. As the non-dominant, B group was higher than A group. There is no significance difference between group. 6. As the standing position of maximal external torque of dominant, A group was higher $60_{\circ}$/sec and $180_{\circ}$/sec than B group. But B group was higher $300_{\circ}$/sec than A group. As the non-dominant, B group was higher than A group. There is no significance difference between group.

  • Present study is to know the relation with throwing speed according to the uniform torque by the shoulder joint, over 20 persons of professional baseball, and were measured the uniform torque by the shoulder joint accordance with throwing speed between (A-low speed) and (B-high speed) of each group. In the present study, three components were measured, and analyzed with having an approximated output value appearing by changing angular velocity, and in $30^{\circ}/sec$, the maximum strength of muscles was measured, and in$210^{\circ}/sec$, the points of endurance strength of muscles were measured and analyzed, and the following result could be obtained. 1. In muscles strength (peak torque)of inner rotation, and outer rotation for respect to each speed of shoulder joint, though there did not appear outstanding difference between A group and B group in the $30^{\circ}/sec$, it showed that group B has priority in a little. Also in outer rotation by shoulder joint, though there appeared the same difference between A group and B group, B group is tended to be prior to A group in a little. 2 In the view of muscles strength (peak torque)of inner rotation, and outer rotation for respect to each speed of shoulder joint, though there did not appear outstanding difference between A group and B group in the $30^{\circ}/sec$, it showed that group B has priority in a little. In the concern of the peak torque for respect to the weight accordance with each velocity of shoulder joint, there did not appear outstanding difference between A group and B group in $210^{\circ}/sec$. In outer rotation by the shoulder joint, there was not appeared clear difference between A group and B group. As conclusion, there did not appear a correlation of uniform torque by shoulder joint versus the speed throwing by a pitcher of professional base ball.

Intragenic Suppressors for Expory-defective Signal Sequence Mutation of Ribose-binding Protein in Escherichia coli (대장균 리보스 결합단백질의 신호배열 변이에 대한 숙성체 부위의 회복돌연변이)

  • A mutational alteration in the signal sequence of ribose-binding protein (RBP) of Escherichia coli, rbsB103, completely blocks the export of the protein to the periplasm. Intragenic suppressors for this mutation have been selected on minimal medium with ribose as a sole carbon source. Six suppressor mutations were characterized in detail and were found to have single amino acid wubstitution in the mature portion of RBP, which resulted in the mobility shift of the proteins on SDS polyacrylamide gel. Amino acid changes of these suppressors were localized in several peptides which are packed to form the N terminal domain of typical bilobate conformation of RBP. The involvement of SecB, a molecular chaperone, was investigated in the suppression of signal sequence mutation. Translocation efficency was found to be increased by the presence of SecB for all suppressors. It is likely that the folding characteristics of RBP altered by the suppressor mutations affect the affinity of interaction between SecB and RBP.

Development of a Protein Secretion System with the Application of Sec-dependent Protein Secretion Components

  • In order to induce high levels of protein secretion, we have constructed a recombinant plasmid, designated pBP244, into which was incorporated key components of the type-II See-dependent secretion system, including LepB (signal peptidase), SecA (ATPase), and SecB (chaperone). The biological activities of the LepB, SecA, and SecB components expressed from genes harbored by pBP244 appeared to play their normal roles. In order to evaluate the protein secretion, a pspA (Streptococcus $\underline{p}neumoniae\;\underline{s}urface\;\underline{p}rotein\;\underline{A}$) gene was cloned into pBP244, resulting in pBP438. S. typhimurium harboring pBP438 grown until the stationary phase, secreted a higher level of PspA into the culture supernatants than did the strain harboring pYA3494. The strain harboring pBP438 secreted a supernatant amount 1.71-fold, a periplasmic space amount 1.47-fold, and an outer membrane amount 1.49-fold higher than that of pYA3494. S. typhimurium ${\chi}8554$ kept the $Asd^+$ plasmid pBP244 and pBP438 for 60 generations in LB broth harboring DAP, thereby indicating that pBP244 and pBP438 were quite stable in the Salmonella strain.

Minimization of Friction and Wear Damage of Marine Structures by Using the Advanced Anti-corrosive Composite Materials (첨단복합방식재를 이용한 각종 선박구조물의 마찰마모손상의 최소화)

  • The marine structures with sea water cooling system always expose to the oceanic atmosphere. Therefore, the protection of the equipments is very important. To investigate the effectiveness of advanced composite materials for the application in offshore environments, the tensile test, hardness test, undercutting property test, permeance test and the friction and wear test were carried out by using various applicable coating materials. The main results obtained can be summarized as follows; 1. The micro-hardness of the Archcoat 502B showed the highest value. 2. The coefficient of friction of the Rigspray coating at the speed of 2.21m/sec showed the lowest value, and that of the Archcoat 502B coating at 1.08m/sec and 0.18m/sec indicated the lowest values. 3. The wear mass at the speed of 0.18m/sec and 1.08m/sec in dry condition showed the smallest values. 4. The Archcoat 502B coating is fitted to the dynamic instruments in the range of low speed and middle speed. Rigspray coating is fitted to the dynamic instruments in the range of high speed. 5. The wear mass of five kinds of coating materials at the range of low speed was very small, and those of the Archcoat S02B, Archcoat 402B and Rigspray coating at high speed range were quitely smaller than those of the Modified Epoxy and Tar Epoxy.
