Physiological Response of the Cultured Olive Flounder (Paralichthys olivaceus) to the Sharp changes of Water Temperature (급격한 수온변화에 따른 양식 넙치(Paralichthys olivaceus)의 생리적 반응)

  • The effects of sparp changes of water temperature (WT) on the stree response and physiological change of the cultured olive flounder (Paralichthys olivaceus) were examined by manipulating WT (3 patterns) in a running seawater culture system. In the first group (Exp. I), the WT was decreased from 18$^{\circ}C$ to 11$^{\circ}C$ within 6 hours and increased back to the original WT quickly. WT was decreased from 2$0^{\circ}C$ to 11$^{\circ}C$ within 5 hours and main-tained at 11$^{\circ}C$ for 10 hours. and then increased to 2$0^{\circ}C$ in the second group (Exp. II). In the third group(Exp. III) WT was decreased to 11$^{\circ}C$ within 5 hours (type A) or 10 hrs. (type B). In Exp. I and III, the level of serum cortisol was increased from 2.5$\pm$0.3 ng/ml and 2.6$\pm$0.9 ng/ml to 13.6$\pm$3.0 ng/ml and 12.4$\pm$3.2ng/ml, respectively, with WT decrease. However, no consistent tendency in the change of serum glucose level was shown according to WT decrease. In Exp. III, the glucose level of fish in type A was decreased until 5 hours and increased at 7 hours, then decreased until 12 hours where as the glucose level in type B was decreased until 5 hours and stayed at the level of 15.7 mg/dl. The serum osmolality was reduced with WT decrease and the response of serum electrolytes in this experiment conflicted, and a tendency in total protein, AST and ALT was not found following WT decrease. In conclusion, olive flounder responded to the stress caused by WT decrease and acclimated to this condition when the lower temperature was maintained. But there was no stress response in the blood of olive flounder when WT was increased. On the other hand, the degree of stress response in olive flounder was various according to the range and gradient of WT change.

First record of a marine microalgal species, Micractinium singularis (Trebouxiophyceae) isolated from Janghang Harbor, Korea

  • A eukaryotic microalga was isolated from seawater in Janghang Harbor, Korea and its morphological, molecular, and physiological characteristics were investigated. Due to its simple morphology, no distinctive characters were found by morphological observation, such as light microscope or scanning/transmission electron microscopy (S/TEM). However, molecular phylogenetic evidence inferred from the concatenated small subunit (SSU) 18S rRNA and internal transcribed spacer (ITS) sequence data indicated that the isolate belonged to the newly described Micractinium singularis. Furthermore, it was clustered with Antarctic Micractinium strains and it also showed a psychrotolerant property, surviving at temperatures as low as 5℃. However, its optimal growth temperatures range from 15℃ to 25℃, indicating that this microalga is a mesophile. Additionally, gas chromatography-mass spectrometry (GC/MS) analysis showed that the isolate was rich in nutritionally important omega-3 polyunsaturated fatty acid, and high-performance liquid chromatography analysis (HPLC) revealed that the high-value antioxidant lutein was biosynthesized as an accessory pigment by this microalga, with glucose as the major monosaccharide. Therefore, in this study, a Korean marine M. singularis species was discovered, characterized, and described. It was subsequently added to the national culture collections.

Characterization of Pigment-Producing Pseudoalteromonas spp. from Marine Habitats and Their Optimal Conditions for Pigment Production (해양환경에서 분리한 Pseudoalteromonas 속 균주들의 특징 및 색소 생성 최적 조건)

  • Three marine bacteria producing pigments were isolated from seawater of Jeju-Do and local solar saltern in Korea. Based on phenotypic characteristics and 16S rRNA sequence analysis, the strains were identified as Pseudoalteromonas spp., which produced red (Ju11-1), yellow (Ju14), and orange (TA20) pigments. The pigments showed UV absorption maxima at 537, 378 and 387 nm, respectively. The strains were growing well on Marine broth 2216 culture medium. The productivity of pigments reached the maximum value after 28 hours (Ju11-1, Ju14) and 24 hours (TA20) at $30^{\circ}C$, 2% NaCl and pH 6-7. The best pigment production of strains were supported by 1% of lactose (Ju11-1) and maltose (Ju14, TA20) as a carbon source and 1% of beef extract as a nitrogen source.

Disinfection of Culture Water Supply by Ozonization I. Susceptibility of Some Fish-Pathogenic Bacteria Isolated From Cultured Marine Fish (오존처리법에 의한 양어용수 살균에 대하여 I. 해산어류 병원세균의 오존 감수성)

  • The disinfectant effects of total residual oxidants (TROs) produced by ozonization of natural sea water were investigated against fish pathogenic bacteria isolated from flounder and red seabream. The concentration of 0.1 mg TROs/liter was stable for 20 min in filtered natural seawater, and those of 0.3 and 0.5 mg TROs/liter were also stable for more 1 hr. Disinfectant effects of TRO against Edwardsiella tarda, Vibrio sp., Streptococcus sp. and Staphylococcus sp. were observed with a concentration of 0.1 mg/liter for 180 sec, and the treatment killed more than 99.9% of bacterial cells. With TROs of 0.3 to 0.5 mg/ liter, the viable cells of the bacteria were reduced by more than 99.99% in 60 sec.

Effect of the concentrated-diatom of Caloneis schroederi and Rhaphoneis sp. for seedling production of Haliotis discus hannai (북방전복 종묘생산을 위한 농축 규조류 Caloneis schroederi와 Rhaphoneis sp.의 효과)

  • Even though seedling production of Haliotis discus hannai has fully developed, the culture of benthic diatom as a live food for larvae is still a barrier to solve in commercial hatchery. The farmer depends on mixed microalgae which are non-selectively attached on the plate by flowing of natural seawater. The adequate diatom on the plate for the larvae in terms of quality and quantity is always significant bottleneck in the hatchery. In this study, two benthic diatom species, Caloneis schroederi and Rhaphoneis sp. were separately cultured in mass and inoculated to four tons' settlement tank of the larvae. And the larvae and the spats were cultured for nineteen and nine weeks, respectively. The result on seedling production of H. discus hannai with this method was compared to that of the farmer's traditional method as a control. With regard to variation of species composition of benthic diatom on the plate, C. schroederi and Rhaphoneis sp. were dominant for first three weeks after inoculation. But the diverse diatoms mainly, Navicula, Amphora, Cylindrotheca, Licmophora, Pleurosigma began to attache on the plate from the 4th week. The larvae attached 2.5 times more in C. schroederi tank than in the control tank. The final total biomass of the seeds in Rhaphoneis sp. tank was 3.2 times more than that of the control tank. The retared-spats in the seedling production also showed significantly higher growth and survival in the spat fed Rhaphoneis sp. or C. schroederi than those in the control group. We suggest that C. schroederi is proper for settlement of the larvae and Rhaphoneis sp. is appropriate for the growth of the larvae and spats in the commercial hatchery of H. discus hannai.

Seawater Temperature Variation at Aquafarms off Wando in the Southwest Coast of Korea (완도 양식장 해역의 수온변동)

  • Abalone culture is one of important coastal fisheries off Wando in the southeast coast of Korea. Since cage culture for abalones was popularized, Understanding of temperature variation, which is important to raise abalones and to prevent their mass mortalities, is necessary. We analyzed temperature data from 2005 to 2009 obtained at Sinji-do and Cheongsan-do off Wando. Sinji-do, which is relatively close to land, had yearly wide range of temperature and rate of temperature variation. It is likely to be caused by heating of solar radiation in summer and cooling in winter at the shallow area. Rate of temperature variation in autumn was higher than that in spring. In summer short term variation of temperature corresponding tidal period was distinguished clearly. Diurnal temperature range, abrupt temperature change, was larger open sea. Comparison between temperatures of two stations and favorable raising conditions of abalones showed that Cheongsan-do, located out to sea, appears to be more appropriate than Sinji-do.

Changes in Water Quality and Bacterial Compositions in Culture Water of an Ozonated Flounder Farm (오존 처리한 넙치 양식장 사육수의 수질과 미생물 변동)

  • This study assessed the effect of ozone to control pathogenic bacteria in inlet water flowing to flounder farms, establishing operational parameters of ozonation at seawater conditions. Hydraulic retention time in a reaction pipeline after ozonation was fixed at 3 minutes in a flow through system. Concentrations of total residual oxidant (TRO) by ozonation were measured according to different ozonation intensities. The oxidant reduction potential (ORP), which is indirect but enables real-time measurement, was measured in relation to TRO values. TRO values were $0.01{\pm}0.01mg\;L^{-1}$ at an ORP range of 320-410 mV, $0.07{\pm}0.02mg\;L^{-1}$ at 600 mV, and $0.16{\pm}0.03mg\;L^{-1}$ at 700 mV. A heterotrophic marine bacteria colony was reduced by 80.6-97.9%, showing the suppression effect of ozonation on total bacteria in inlet water. At an ORP range of 400-500 mV, colonies of heterotrophic marine bacteria, Vibrio spp., and gram negative bacteria were significantly reduced in outlet water from a culture tank with ongrowing flounder (230 g) at a stocking density of $8kg\;m^{-2}$. Especially, Vibrio spp. and gram negative bacteria were seldom found at 400-500 mV. The daily feeding rate was from over 0.7% to total body weight at 300-500 mV, showing better performance than that in the control. The pathogenic bacteria entering the flounder farm were effectively removed when the ORP range to 400 mV or less.

Studies on Bacterial Characteristics and Cd Accumulation of Vibrio sp. S-1-2, Isolated from Eutrophic Coastal Area (부영양화 해역에서 분리된 Vibrio sp. S-1-2균의 특성과 Cd축적에 관한 연구)

  • Vibrio sp. S-1-2 was isolated from seawater in the Masan bay and its bacterial characteristics and bioaccumulation of $CdCl_2$ in the cell were investigated. As the result of microscopic and biochemical test, the S-1-2 strain was identified to Vibrio sp. and this strain can be tolerated even in 200 ppm $CdCl_2$ media, however its growth was inhibited. In 25 ppm $ZnCl_2$ media, the growth of Cd resistant Vibrio sp. S-1-2 was promoted at later stage of growth. The growth of Vibrio sp. S-1-2 was inhibited on 25 ppm $CuCl_2$ and $PbCl_2$ media and was not able to grow in 25 ppm $HgCl_2$ media at all. The uptake of cadium in the cells was increased exponentially with increasing concentration of $CdCl_2$ in media. But the uptake rate of cadmium was suddenly inhibited at 50 ppm $CdCl_2$ media. Optimal pH and NaCl concentration for bioaccumulation of $CdCl_2$ were 8-9 and 1-2%, respectively. In the case of pH, maximum dpm value was found at pH 7 after 96 hours culture and in the case of NaCl concentration, it was detected in 2% NaCl media after 36 hours culture.

Effects of Environmental Factors on the Zoospore Release and Germling Growth of the Green Macroalga Chaetomorpha linum (녹조류 실염주말의 유주자 방출과 배아생장에 미치는 환경요인의 영향)

  • Effects of environmental factors on the zoospore release and germling growth of Chaetomorpha linum were examined. Zoospore release and germling growth experiments were carried out under a combination of temperatures ${\times}$ irradiances, and a single factor of irradiances, salinities, daylength and nutrients (nitrogen, phosphorus). Zoospore release was maximal under condition of $20^{\circ}C{\times}100{\mu}mol\;photons\;m^{-2}s^{-1}$ in a factorial experiment (temperatures ${\times}$ irradiances), at $100{\mu}mol\;photons\;m^{-2}s^{-1}$ in irradiance experiment and at 34 psu in salinity conditions. After 24 days in culture, germling growth was maximal at $25^{\circ}C{\times}100{\mu}mol\;photons\;m^{-2}s^{-1}$ in temperatures ${\times}$ irradiances and at normal seawater (34 psu) in salinity. Germling growth is faster at higher irradiance under same temperature condition and it increased with increasing daylengths (8-16 h). Chaetomorpha linum grew in wide range of N and P concentrations. The growth of germling was maximal at $50{\mu}M\;{PO_4}^{3-}$ and $40{\mu}M\;{NH_4}^+$. Germling growth was more effective under nitrogen addition than phosphorus addition in culture. In conclusion, optimal environmental conditions for zoospore release were $20^{\circ}C$, $100{\mu}mol\;photons\;m^{-2}s^{-1}$, and 34 psu. In addition, the optimal germling growth was observed at $25^{\circ}C$, $100{\mu}mol\;photons\;m^{-2}s^{-1}$, 34 psu, 16 h, $50{\mu}M\;({PO_4}^{3-})$ and $40{\mu}M\;{(NH_4}^+)$.

Identification of a New Agar-hydrolyzing Bacterium Vibrio sp. S4 from the Seawater of Jeju Island and the Biochemical Characterization of Thermostable Agarose (제주도 연안 해양에서 분리한 한천분해 미생물 Vibrio sp. S4의 동정 및 내열성 agarase의 생화학적 특성)

  • Agar-hydrolyzing bacteria were isolated from the coastal sea water of Jeju Island. One isolate, designated as S4, was selected for further study. The S4 cells were Gram-negative and rod-shaped with smooth beige surfaces and single polar flagellum. Cells were grown at $15-42^{\circ}C$, 0.5-5% (w/v) NaCl, between pH 6.0 and 9.0, and in media containing 0.5-5% (w/v) NaCl. The G+C content was 49.93 mol%. The major fatty acids (>15%) were $C_{18:1}{\omega}7c$, $C_{16:0}$ and Summed feature 3 (comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH). Based on 16S rRNA sequencing and biochemical and chemotaxonomic characteristics, the strain was designated as Vibrio sp. S4. In liquid culture supplemented with 0.1% agar the cell density and agarase activity reached a maximum level in 72 h, while agarase activity in the culture without agar was negligible, implying agarose expression is induced by agar. The optimum pH and temperature for the extracellular crude agarase of S4 were 7.0 and $45^{\circ}C$, respectively. However, it also exhibited 98.6% and 87.6% at $40^{\circ}C$ and $50^{\circ}C$, respectively, of the maximum activity seen at $45^{\circ}C$. The crude agarase hydrolyzed agarose into (neo)agarotetraose and (neo)agarohexaose.
