Complete Genome Analysis of Spodoptera exigua Nucleopolyhedrovirus Isolated in Korea (한국에서 분리된 파밤나방 핵다각체병 바이러스의 전체 유전체 분석)

  • The morphology and whole genome sequence of Spodoptera exigua nucleopolyhedrovirus K1 (SeNPV-K1) isolated in Korea were analyzed for the use as an eco-friendly control source against S. exigua. The polyhedra of SeNPV-K1 was amorphous with a size of 0.6-1.8 ㎛, and there was no external difference with the previously reported SeNPV. As a result of analyzing the nucleotide sequence of the whole genome, it was composed of 135,756 bp, which is 145 bp more than that of the previously reported SeNPV. The G+CG+C content was 44% and there were 6 homologous repeated sequences, so there was no significant difference from the previous report. As a result of ORF analysis, SeNPV-K1 had 137, two fewer than those previously reported, and 4 ORFs present only in SeNPV-K1 were confirmed. These 4 ORFs are non-essential genes and were not considered to have a significant influence on the characteristics of the SeNPV. The genome vista analysis showed that the overall sequence similarity between SeNPV-K1 and the previously reported SeNPV was very high. The whole genome of SeNPV-K1 analyzed for the first time in Korea was found to be similar to the previously reported SeNPV, but it was confirmed that it was a novel resource in Korea with different isolate.

  • Biochemical characteristics of Spodoptera uigua nuclear polyhedrosis virus (SeNPV) isolated in Jinju were studied. SeNPV contained a number of nucleocapsids within a viral envelope embeded in polyhedra. The polyhedral protein of SeNPV was composed of a single polypeptide with a M. W. of 30kd. Double-immunodiffusion test showed that the polyhedral protein of SeNPV had common antigenic determinants with SINPV and BmNPV. Virion proteins of SeNPV were resolved into 49 polypeptides by silver staining after SDS-PAGE. The approximate genome size of SeNPV by restriction endonuclease analysis was 1l0kb.

  • To study the usefulness of Se301 cells, which is originated from Spodoptera exigua and has susceptibility to the Autographa californica NPV (AcNPV), as a host for the AcNPV-based expression vector system, we compared the characteristics of AcNPV in Se301 and Sf-21 cells. The symptom by viral infection was similar in both of cells, but the ratio of polyhedra released from the cell was higher in Se301 cells than in Sf-21 cells. The overall PIB productivity of AcNPV was similar in both cells but the size of polyhedra was larger in Se301 cells. While the polyhedrin expression efficiency was about 2.4 times higher in Se301 cells than in Sf-21 cells, the viral growth was higher in Sf-21 cells. These results suggested that Se301 cell is very useful in the AcNPV-based expression system as a host.

  • The role of polyhedrin in the polyhedra production in baculovirus Autograha californica Nucelopolyhedro-sisvirus (AcNPV) was studied by over-expression of AcNPV polyhedrin or heterologous polyhedrin from Bombyx mori (Bm) NPV or Spodoptera exigua (Se) NPV. The transfer vectors containing additional polyhedrin from AcNPV, BmNPV, or SeNPV were constructed and cotransfected with bacmid bApGOZA into Sf9 cells. The resulting recombinants, designated as vApAcPol, vApBmPol, and vApSePol were tonstructed, and the polyhedra production of the recombinant was characterized. All of the recombinants produced polyhedra in the nucleus, and the polyhedrin was over-expressed. Among three recombinants, vApAcPol and vApBmPol were discriminated by their larger polyhedra size than that of wild type AcNPV, and vApSePol also produced larger polyhedra than wild type SeNPV polyhedra.

Characterization of Spodoptera exigua Nuclear Polyhedrosis Virus Polyhedrin Gene Structure (파밤나방 핵다각체병 바이러스의 다각체 단백질 유전자 구조)

  • To develope the baculovirus expression vector system (BEVS) using Spodoptera exigua nuclear polyhedrosis virus (SeNPV), we characterized the polyhedrin of SeNPV. The SeNPV polyhedra was irregular and composed of the major protein molecular weight of 30 kDa determined by electronmicroscopy and SDS-AGE analysis, respectively. The nucleotid suquences of 876 bases including the coding region of polyhedrin gene was determined and it was revealed that the polyhedrin gene is located within Xho I 3.0Kb and Nco I 6.0 Kb by Southern blot analysis, respectively. Also, the Xho I 3.0 Kb and the Nco I 6.0 Kb fragments were cloned and restriction enzyme map of these clones were determined.

  • To develop the baculovirus expression vector system (BEVS) adopting p10 gene promoter of Spodoptera exigua nucleopolyhedrovirus (SeNPV), we characterized the p10 gene of SeNPV. The nucleotide sequence of 545 bases including the coding region of p10 gene was determined. Compared with the previously reported SeNPV p10 gene (Zuidema et al., 1993), 4 bases were different in the 5' and 3' flanking region but no difference was found in the coding region. The p10 gene was located within Xho I 1.5 Kb, Sph 1 2.4 Kb and Cla I 4.0 Kb fragments by Southern hybridization analysis. Also, the Sph I 2.4 Kb and the Cla I 4.0 Kb fragments were cloned and their restriction enzyme maps were determined.

Effects of Different Temperatures on Pathogenicity of Spodoptera exigua Nucleopolyhedrovirus (SeNPV) (온도조건에 따른 파밤나방핵다각체병바이러스(SeNPV)의 병원 활성)

  • This experiment was conducted to investigate pathogenicity of Spodoptera exigua nucleopolyhedrovirus (SeNPV) with different temperatures for mass production. In laboratory condition, $LC_{50}$ values of SeNPV were $9.797{\times}10^{3}PIBs/mL$ at $20^{\circ}C$ and $3.351{\times}10^{2}PIBs/mL$ at $32^{\circ}C$ in 2nd instar larvae. $LC_{50}$ of the other larval stage were similar to that of 2nd instar. $LT_{50}$ values of SeNPV was 9.0 days in $1.0{\times}10^{3}PIBs/mL$ but 6.9 to 3.5 days in $1.0{\times}10^{4-6}PIBs/mL$ against 3rd instar of Spodoptera exigua. $LT_{50}$ Values of $1.0{\times}10^{4}PIBs/mL$ were 5.7, 5.5 and 4.9 days in 24, 28 and $32^{\circ}C$, respectively. As a results, $LT_{50}$ was shortened with increase of temperatures up to $32^{\circ}C$ and also dependent on viral concentration and larval instars.

  • To produce Spodoptera exigua nuclear polyhedrosis virus (SeNPV) using S. exigua larvae, the efficiency of the in vivo production was analysed by larval instar, inoculum and mortality. The results revealed that the mortality of 4th instar larvae inoculated with 1.OX 10' EIBs per ml was 86.7% and the yields of SeNPV was maximal, demonstrating that 4th instar larvae inoculated with 1 . 0l~o6 PIBs per ml were effective to mass production of SeNPV.

  • Suppressor of cytokine signaling (SOCS) is known to playa key role as a negative feedback regulator in JAK/STAT signaling cascade in innate immunity. Our laboratory has recently been interested in elucidating the interactions between Spodoptera exigua (Se) and SeNPV. This context leads us to clone and characterize SeSOCS that may have important functions in response to SeNPV infection. Using the RT-PCR and TA cloning approach, we found a partial fragment (416 bp) of SeSOCS. Blast search and multiple alignment data showed that it has a homology to various insects such as Anopheles gambiae (78%), Aedes aegypti (75%), Drosophila melanogastar (77%), Mus musculus (69%), and Homo sapiens (69%). Temporal induction patterns of SeSOCS were analysed after being immune-challenged with either NPV or laminarin. It showed that the level of SeSOCS mRNA was strongly induced in a biphasic manner in response to SeNPV and laminarin, respectively. It seems that SOCS, a negative regulator of JAK/STAT signaling system is also present in S. exigua and may playa role in innate immunity albeit its precise role should be further elucidated at the molecular and cellular level in the early phase of SeNPV infection in larvae.

Control Efficacy of Entomopathogenic Microagent against Spodoptera exigua on Organic Chinese Cabbage (유기재배 배추포장 내 파밤나방에 대한 곤충병원성 미생물의 방제 효과)

  • This study was carried out to identify the control effect of entomopathogenic microagent against Spodoptera exigua on organic chinese cabbage. In laboratory condition, insecticidal activity of 4 commercial BT pesticides against S. exigua were lower than 10% against second instar S. exigua. The insecticidal activity of entomopathogenic nematode were 33.3%, 83.3% and 100% at the concentration of $1{\times}10^2$, $3{\times}10^2$, $1{\times}10^3nematodes/ml$, respectively. Mixture of BT and nematode showed growth inhibition against S. exigua larvae. S. exigua nucleopolyhedrovirus (SeNPV) of $10^5PIBs/ml$ showed more than 70% insecticial activity. The yield of SeNPV was increased as in higher initial inoculation concentration of NPV, food supply, and growth temperature increased. In greenhouse experiment, the control value of BT and nematode mixture treatment was higher than BT and nematode treatment alone against S. exigua. In treatment of $10^7PIBs/ml$ of SeNPV, S. exigua was controlled completely. In farm condition, mixture of microbial agent and organic agricultural material showed higher control value against lepidopteran pest including S. exigua than BT single treatment.
