  • This study was carried out to investigate the effect of high pressure low temperature (HPLT) on the inactivation rates of S. cerevisiae in 0.9% saline solution depending on the pressurization time and temperature. S. cerevisiae was inoculated with UHT milk and submitted to HPLT of 200 MPa at 4, 12 and $20^{\circ}C$, respectively Inactivation increased with pressurization time and HPLT of S. cerevisiae at 200 MPa was time dependent at any temperature. The morphological changes of yeast cells observed with a SEM after HPLT.

  • Chitin, a polymer of N-acetylglucosamine ($\beta$-1,4 linked 2-acetamido-D-glucose), is a cellulose-like biopolymer present richly in the exoskeleton of crustaceans and in cell walls of fungi, insects and yeast. Chitosan is derived from chitin by deacetylation, to different degrees, in the presence of alkali. [l]. Recent studies for chitin and chitosan have been concentrated in bioactivities such as antitumor activity, immuno-enhancing effects, enhancing protective effects against infection with some pathogens in mice, and antimicrobial activeity [2]. • The biological production of a potent anticancer agent, epothilone, by Sorangium cellulosum was carried out using flask and fermentor cultures. Soluble starch was selected as the main carbon source and the concentrations of lactose and yeast extract were optimized at 4 and 0 g/L, respectively, when using the flask cultures. In the fermentor cultures, the cells were cultivated at a high DO level of more than 80% of air saturation in the growth stage and then the DO level was controlled at about 50, 20 or 1-2% when the carbon source was exhausted. The epothilone production increased with decreasing DO level after the exhaustion of the carbon source, and the maximum concentration of epothilone was 5.4 mg/L. It was found that the DO level had significant regulation effects on the epothilone production.

  • Pholiota nameko is one of the four major profitable mushrooms along with oak mushroom, winter mushroom, and oyster mushroom. It contains abundant proteins, carbohydrates, organic acids and vitamins. Its unique taste and flavor as well as its nutritional features make it widely favoured. Mushroom complete medium was the optimal medium for mycelial growth of Pholiota nameko. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and 5.0, respectively. The best carbon sources for mycelial growth were glucose and mannose, and the best nitrogen sources were yeast extract, peptone, asparagine, etc. The 8:2 ratio mix of oak sawdust and wheat bran was the best for the bottle cultivation. The best mushroom was yielded after 30 days incubation. The best yield was produced with 850g of medium weight in a PP bag and bottle.

  • Yeasts are common inhabitants of the phyllosphere, but our knowledge of their diversity in various fruits and blossoms are limited. We collected different kinds of blossoms and unripened fruits from Hanbat arboretum, Daejeon, Korea at the year of 2013. Yeasts were isolated by plating of suspensions prepared for collected samples onto YPD medium containing antibiotics. BLAST searches were subsequently performed for the comparison of the partially determined sequences of D1/D2 domain of 26S rDNA. As a result, we isolated 57 yeast strains of 31 species from 29 different kinds of blossoms and 6 kinds of fruits samples. We found huge differences in yeast flora depending on the sample collection season.

  • An X-linked skeletal disorder, SEDT (spondyloepiphyseal dysplasia tarda) is a genetic disease characterized by a disproportionately short trunk and short stature caused by mutations in the SEDL gene. This gene is evolutionarily conserved from yeast to human. The yeast SEDL protein ortholog, Trs20p, has been isolated as a member of a large multi-protein complex called the transport protein particle (TRAPP), which is involved in endoplasmic reticulum (ER)-to-Golgi transport. The interaction between SEDL and partner proteins is important in order to understand the molecular mechanism of SEDL functions. We isolated several SEDL-binding proteins derived from rat cells by an immobilized GST-fusion method. Furthermore, the SEDL-homologue proteins were identified using motif based methods. Common motifs between SEDL-binding proteins and SEDL-homologue proteins were classified into seven types and 78 common motifs were revealed. Sequence similarities were contracted to seven types using phylogenetic trees. In general, types I-III and VI were classified as having the function of acetyl-CoA carboxylase, glycogen phosphorylase, isocitrate dehydrogenase, and enolase, respectively, and type IV was found to be functionally related to the GST protein. Types V and VII were found to contribute to TRAPP vesicle trafficking.

  • Six unrecorded yeasts, Cryptococcus festucosus 41-3, Cryptococcus heveanensis 56-4, Debaryomyces nepalensis 95-4, Issatchenkia occidentalis 142-1, Dioszegia zsoltii 39-1, and Kwoniella europala 47-2 were screened from 108 yeasts isolated from flowers and fruits in orchards of Yesan-gun, Chungcheongnam-do, Korea. The morphological and cultural characteristics of these unrecorded yeasts were investigated. They had various shapes, including ellipsoidal, globose, and oval, and also had budding mode in vegetable reproduction, except I. occidentalis 142-1 (fission mode). K. europaea 47-2 only formed pseudomycelium. D. zsoliti 39-1 did not grow in yeast extract-malt extract medium, potato dextrose medium, and vitamin-free medium. C. festucosus 41-3 grew well in 5% NaCl-containing yeast extract-peptone-dextrose medium and had a growth pH range of 7.0~10.0. Three unrecorded yeasts Ogataea polymorpha HB45-1, Rhodotonula hinnulla HB62-2, and Cryptococcus rajasthanensis HB80-4 were screened from 51 yeasts isolated from flowers in Hanbat arboretum in Daejeon city, Korea. They were globose in shape and did not form pseudomycelium. In addition, O. polymorpha HB45-1 and C. rajasthanensis HB80-4 had budding mode in vegetable reproduction. All of them grew well in vitamin-free medium and C. rajasthanesis HB80-4 also grew in 50% glucose and 5% NaCl-containing YPD medium.

  • Sixty one yeast strains of 21 species were isolated from wild flowers in Gogunsanyeoldo, including Seonyudo of Jeollabuk-do, Korea, and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Among them, Cryptococcus sp. including C. aureus SY1-4 were found to be dominant, and Metschnikowia sp. including M. reukaufii SY20-1 and Rhodotnula sp. such as R. ingeniosa SY1-1 were also abundantly isolated. Some physiological functionalities of the culture broth and cell-free extracts from 61 yeast strains were determined. Supernatant from Metschnilowia reukaufii SY44-6 showed anti-gout xanthine oxidase inhibitory activity of 49.6% and whitening tyrosinase inhibitory activity of 38.4%, respectively.

  • A response surface method based on a central composite design experiment was used to determine the optimum conditions for pretreatment of persimmon peel. It was mathematically predicted that the maximum amount of reducing sugars would be obtained at an $H_2SO_4$ concentration of 1.77% (w/v) and a heat treatment time of 26.4 min. A reducing sugar concentration of 63.23 g/l was obtained under the optimum pretreatment conditions determined by RSM. Under anaerobic growth conditions, Saccharomyces cerevisiae NK28 produced 15.52 g/l of ethanol with a yield of 0.34 g ethanol/g glucose from pretreated persimmon peel, which corresponded to 14% and 26% enhancements in ethanol productivity and ethanol yield, respectively, compared with those obtained in aerobic growth conditions. This study suggests that persimmon peel might be a useful substrate for bioethanol production by yeast fermentation.
