Scientific Analysis of the Formulation Theory of Chungpesagan-tang; In Vitro Cytotoxicity of Chungpesagan-tang

  • To analyse scientifically the fundamental formulation theory and drug interaction of Chungpegagan-tang, the extraction level of puerarin and daidzin, the transforming activity of puerarin and daidzin to daidzein by human intestinal bacteria and in vitro cytotoxicity against tumor cell lines of Chungpesagan-tang were investigated. When Puerariae Radix was extracted with Chungpesagan-tang composing herbal medicines, the puerarin extraction level from these polyprescriptions was decreased by the extraction with Raphani Semen or Cimicifugae Rhizoma, but the other herbal medicines increased it. The activity transforming puerarin and daidzin to daidzein by human intestinal bacteria was increased by Raphani Semen, Cimicifugae Rhizoma and Angelicae Tenuissimae Radix, but decreased by Scutellariae Radix and Rhei Rhizoma. Puerariae Radix did not showed in vitro cytotoxicity against tumor cell lines. However, by its anaerobic incubation with human intestinal bacteria, it showed a potent cytotoxicity. When the main components, puerarin and daidzin, of Puerariae Radix were incubated with human intestinal bacteria, the main metabolites were daidzein and calycosin. These metabolites had the most potent cytotoxicity, compared to those of puerarin and daidzin. Raphani Semen, Rhei Rhizoma and Chungpesagan-tang had also the potent cytotoxicity against tumor cell lines by the anaerobic incubation with human intestinal bacteria.

Effects of Several Medicinal Plants on the Activity of GABA-metabolizing Enzymes (수종 생약재의 GABA 대사 관련 효소의 활성에 미치는 영향)

  • The effect of seventy kinds of medicinal plants on the activities of GABA-metabolizing enzymes as glutamate dehydrogenase I (GDH I), glutamate dehydrogenase II (GDH II), GABA transaminase (GABA-T), succinic semialdehyde dehydrogenase (SSADH) and succinic semialdehyde reductase (SSAR) were estimated. The following plants extracts from Acori graminei Rhizoma, Longnae Arillus, Gastrodiae Herba, Lycii Fructus, Ligusticum officinale, Ferula assafoetida, Corydalis Tuber, Eucommiae Cortex, Zizyphi spinosi Semen activated the activity of GDH I to more than 35%, and the following ones from Visci Ramulus, Ligusticum officinale, Myristicae Semen, Ferulae Resina, Scolopendrae Corpus, Corydalis Tuber, Eucommiae Cortex, Zizyphi spinosi Semen did that of GDH II. The plant extracts from Cynanchi Radix, Astragali Semen, Angelicae dahuricae Radix, Biotae orientalis Folium, Uncariae Ramulus et Uncus, Polygalae Radix, Cynomorii Herba inhibited that of GABA-T to 35% and over, and the following ones from Hyoscyamus niger, Cynanchi Radix, Acori graminei, Caesalpiniae Lignum, Cannabis Semen, Sedum aizoon, Sedum kamtschaticum, Schisandrae Fructus, Lilii Bulbus, Biotae orientalis Folium, Uncariae Ramulus et Uncus, Myristicae Semen, Akebiae Fructus, Cynomorii Herba, Buddleiae Flos, Mucunae Caulis, Zizyphi Fructus, Paeoniae Radix rubra did that of SSADH to 70% and over; the following ones from, Caesalpiniae Lignum, Sedum kamtschaticum, Schisandrae Fructus, Astragali Semen, Angelicae dahuricae Radix, Dioscorea nipponica, Myristicae Semen, Akebiae Fructus, Cynomorii Herba, Scutellariae Radix did that of SSAR.

Monitoring and Risk Assessment of Benzo(a)pyrene Content in Medicinal Herbs (유통 한약재 중 벤조피렌 함량 모니터링 및 안전성 평가)

  • In this study, to investigate of safety for Benzo(a)pyrene in medicinal herb, 8 kinds of selected commercial herbal medicines (Rehmanniae Radix, Rehmanniae Radix Preparata, Amomi Tsao-Ko Fructus, Cimicifugae Rhizoma, Cyperi Rhizoma, Magnoliae Cortex, Scutellariae Radix, Scrophulariae Radix) were analysed using the high performance liquid chromatography with fluorescence detector and assessed the health risk. The levels of benzo(a)pyrene were from non-detection to $28.1{\mu}g/kg$, and the average was $3.6{\mu}g/kg$. Based on a nationwide survey of the consumption of medicinal herb by the Korean population, we estimated the potential risk from the ingestion of benzo(a)pyrene. The estimated daily intake of benzo(a)pyrene was 1.6 ng/kg b.w./day for group only know the daily average intake of medicinal herb. The MOE (margin of exposure) of benzo(a)pyrene for estimate of health risk was $1.93{\times}10^5$. Therefore, health risk from benzo(a)pyrene through intake of herbal medicine was considered negligible.

Review of Clinical Research about Herbal Medicine Treatment on Chronic Pancreatitis - Research on CNKI (만성 췌장염의 한약치료에 대한 최신 임상연구 동향 - CNKI검색을 중심으로)

  • Objectives: This study was performed to review the recent clinical research trends of the effects of herbal medicine treatment on chronic pancreatitis in China. Methods: We searched for clinical studies about chronic pancreatitis from the China Academic Journal (CAJ) of the China National Knowledge Infrastructure (CNKI) from January 1, 2005, to August 1, 2016. Results: Thirty-seven papers were selected from 126 studies. The most commonly used herbal prescriptions included Bupleuri Radix (柴胡), Paeoniae Radix Alba (白芍藥), Glycyrrhizae Radix (甘草), Scutellariae Radix (黃芩), Aucklandiae Radix (木香), and Rhei Rhizoma (大黃). Qingyi-tang (淸胰湯) was the most frequently used herbal prescription. Clinical symptoms, physicochemical examination, radiological examination, and relapse were used to evaluate the treatments. The effective treatment rate was 77.5-100%, and effectiveness in the experimental group was significantly higher than in the control group in 15 papers (p<0.05). Conclusions: Herbal medicine treatment has been shown as an effective treatment for chronic pancreatitis. In the future, more scientifically designed clinical studies should be performed to prove the effectiveness of herbal medicine treatment on chronic pancreatitis. These research results can be utilized in other clinical studies and in the treatment of chronic pancreatitis.

Analysis of Korean Herbal Medicine Used in Case Studies of Treating Urticaria (두드러기 증례 연구에서 사용된 한약 처방 분석)

  • Objectives : The purpose of this study is to analyze the use of herbal medicine for the treatment of urticaria in case studies and to find the general tendency of herbal medicine treatment and to establish the primary treatment direction of urticaria. Methods : In the domestic databases, Oriental medicine Advanced Searching Integrated System (OASIS), Korean Traditional Knowledge Portal (KTKP), National Discovery for Science Leader (NDSL), Research Information Sharing Service (RISS), we selected among the case study papers published in the last 20 years using search terms related to "urticaria". Reports related to Sasang Constitutional medicine were excluded and total of 15 papers were finally selected. Results : 23 prescriptions were retrieved from 15 papers. The most commonly used prescription was Seungmagalgeun-tang (升麻葛根湯), and Bangpoongtongsung-san (防風通聖散) and Pyeongwi-san (平胃散) were presented twice. Commonly used herbs are Glycyrrhizae Radix et Rhizoma (甘草), Zingiberis Rhizoma Recens (生薑), and Scutellariae Radix (黃芩). 18 out of 78 herbs are belonging to heat-clearing medicinal (淸熱藥) and 16 out of 78 herbs are belonging to exterior-releasing medicinal (解表藥). Concurrent therapies such as acupuncture, pharmacoacupuncture, moxibustion, cupping therapy, ice pack applying therapy were performed and several evaluation methods were used, patient's subjective remission rate was the most used. Conclusions : Through this study, we were able to discover the tendency of herbal medicine prescription for the treatment of urticaria.

A study on the literal research kimi - theory (기미론에 대한 문헌적 연구)

  • Kimi(氣味) means five tastes(五味), sour, bitterness, sweetness, hot taste and astringency, and four conditions(四氣), cold, hot, warm and cool, in the oriental medicine. This is defined based on yn-yang 5 evolutive phasis(陰陽五行), and 5 evolutive phasis on the change of season. Four seasons, spring, summer, autumm, winter are clearly different but, the rainy season(長夏) is not. In the thee of Ki (氣), not worm not hot(平) is included in addition to the four conditions that is because the rainy season is not differentiated clearly. KImi have realations to the four seasons, that is, warm and hot taste is considered as spring, hot and astringency as summer, not worm not hot and sweetness as rainy season, cool and sour as autumm, and cold and bitterness as winter. 4 conditions can be classified more detail, because the changes of the seasons are continuous. In the action mechanisms, Gardeniae Fructus reduces Heat of Insufficency Type(虛熱) of the upper class of medicinal herbs(上焦), and Rhel Rhizoma reduces Exessive Heat(實熱) of the under class of them(下焦). The assay methods for four groups medicines can be developed in three ways according to the indicators as follows. First, by the indicator which defines cold-acting medicine(寒性藥) such as Rhei Rhizoma, Coptidis Rhizoma, Scutellariae Radix, Gardeniae Fructus and is differentiated clearly from Hot-acting medicine(熱性藥) at the same time. Second, when the medicines are classified into another four groups as Drugs for Dispelling Internal Cold(溫裏藥), Drugs for Relieving Exterior Syndrome(解表藥), Drugs for Dispelling Phlegm(祛痰藥), Drugs for Regulatings Ki Flow(理氣藥), by the indicator which satisfies each group and is differentiated from other groups, at the same time. Third, by the indicator which has to be defined for each medicinal herb for four classification, individually.

Analysis of Prescriptions from Somunsunmyungronbang and Nansilbijang (소문선명논방(素問宣明論方)과 난실비장(蘭室秘藏)의 본초구성 비교 분석)

  • Objectives : To quantitatively analyze the academic characteristics of Yuhagan and Idongwon and compare it to existing qualitative analysis. Methods : The prescriptions listed in Somunsunmyungronbang and Nansilbijang were entered into a database. Commonly used combination of medical herbs were researched throughout the literature. Results : In Nansilbijang, among the combinations of 8 medical herbs, Glycyrrhizae Radix, Cimicifugae Rhizoma, Bupleuri Radix, Ginseng Radix, Citri Pericarpium, Atractyodis Rhizoma, Astragali Radix, Phellodendri Cortex combination was used 11 times out of the 277 prescriptions. In Somunsunmyungronbang, combinations of 7 medical herbs: (Glycyrrhizae Radix, Platycodi Radix, Rhei Radix et Rhizoma, Menthae Herba, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Forsythiae Fructus), (Glycyrrhizae Radix, Angelicae gigantis Radix, Paeniae Radix Alba, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Cnidii Rhizoma, Scutellariae Radix), (Glycyrrhizae Radix, Platycodi Radix, Angelicae gigantis Radix, Rhei Radix et Rhizoma, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Schizonepete Herba) were each used 7 times out of the 350 prescriptions. Conclusions : Nansilbijang shows more prescriptions applying Cimicifugae Rhizoma, Bupleuri Radix, Ginseng Radix, Citri Pericarpium, Atractyodis Rhizoma, Astragali Radix, Phellodendri Cortex. Prescriptions aimed to tonify ql with Ginseng Radix and Astragali Radix, to upraise the middle qi with Bupleuri Radix and Cimicifugae Rhizoma, and to regulate qi with Citri Pericarpium and Atractyodis Rhizoma were common. Somunsunmyungronbang preferred the use of Angelicae gigantis Radix, Platycodi Radix, Rhel Radix et Rhizoma, Gypsum Fibrosum, Schizonepete Herba, Forsythiae Fructus and Menthae Herba so its focus was on the aggressive management of febrile diseases.

Kagam-bojungikgitang Inhibits LPS-induced Inflammatory Mediators in RAW 264.7 Macrophages (가감보중익기탕의 LPS 유도 염증성 매개물에 대한 억제 효과)

  • Kagam-bojungikgitang is the water extracts prepared from Ginseng Radix, Astragali Radix. Angelicae gigantis Radix, Astractylodis Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisiae iwayomogii Herba, and Scutellariae Radix. This is a modified prescription of Bojungikgitang, which has been used for the treatment of indigestion, and immunological disease in oriental countries. In this study, the effects of Kagam-bojungikgitang and Bojungikgitang on the production of prostaglandin E₂ (PGE₂) and the expression of cyclooxygenase-2 (COX-2) were examined using RAW 264.7 macrophages activated with lipopolysaccharide (LPS). Both prescriptions dose-dependently reduced the release of PGE2 and expression of COX-2 caused by stimulation of LPS without cytotoxic effect. Kagam-bojungikgitang's inhibitory effects were better than Bojungikgitang in PGE2 production and COX-2 expression. Moreover, Kagam-bojungikgitang also attenuated markedly the production of tumor necrosis factor (TNF)-α, and IL-6 than Bojungikgitang in LPS-stimulated RAW 264.7 macrophages. These results suggest that Kagam-bojungikgitang decreases PGE2 and pro-inflammatory cytokine production in macrophages and these properties may contribute to the anti-inflammatory activity of Kagam-bojungikgitang.

Literature Review on Safety of Pharmacopuncture according to Sasang Constitutional Medicine (동의수세보원(東醫壽世保元) 신정(新定) 처방(處方) 약재(藥材)의 약침 안전성 문헌 조사)

  • 1. Objectives: Pharmacopuncture according to Sasang Constitutional Medicine is new field of constitutional medicine. We investigated safety of pharmacopunctures to develop this treatment. 2. Methods: We searched for papers in fifteen Korean oriental medical journals and four online searching portal articles. Hepatotoxicity, nephrotoxicity and general toxicity tests were investigated about one hundred thirty kinds of pharmacopunctures. 3. Results: Toxicity test about 36 kinds of 130 pharmacopunctures were investigated. 1) Hepatotoxicity and nephrotoxicity test of pharmacopunctures were searched in 11 of 38 Soeumin herbs, 8 of 36 Soyangin herbs, 5 of 44 Taeeumin herbs and 1 of 12 Taeyangin herbs. From among these herbs, toxicity was detected in Radix aconiti lateralis preparata(附子), Fructus alpiniae oxyphyllae(益智仁), Semen crotonis(巴豆), Radix bupleuri(柴胡) and Semen armeniacae amarum(杏仁) pharmacopunctures. 2) General toxicity test of pharmacopunctures were searched in 8 of 38 Soeumin herbs, 9 of 36 Soyangin herbs, 6 of 44 Taeeumin herbs and 2 of 12 Taeyangin herbs. from among thes herbs, toxicity was detected in Radix aconiti lateralis preparata(附子), Radix ginseng(人蔘), Semen crotonis(巴豆), Rhizoma seu radix notopterygii(羌活), Radix aralia cordatae(獨活), Semen armeniacae amarum(杏仁) and Radix scutellariae(黃芩) and Cortex acanthopanacis(五加皮) pharmacopunctures. 4. Conclusions: From these results, we must decide extraction method, concentration and dose carefully to use the toxic pharmacopuncture. More toxicity test and further study is needed about pharmacopuncture treatment in Sasang Constitutional Medicine.

  • Chrysin and chlorogenic acid are natural products derived from Scutellariae Radix and Lonicerae Flos, respectively. We examined whether chrysin and chlorogenic acid affect airway mucin production induced by TNF-${\alpha}$ in NCI-H292 cells. Cells were pretreated with each agent for 30 min and then stimulated with TNF-${\alpha}$ for 24 h. Of the two compounds, chrysin suppressed airway MUC5AC mucin production. Also, chrysin suppressed MUC5AC mucin gene expression and translocation of NF-${\kappa}B$ p65 induced by TNF-${\alpha}$. This result suggests that chrysin can regulate the production and gene expression of mucin induced by TNF-${\alpha}$ through the inactivation of NF-${\kappa}B$ in airway epithelial cells.
