We investigated the effect of moisturizing and anti-oxidation of Astragalus membranaceus (Astragali Radix) root extract with respective to growing districts and extract methods for the purpose of development of cosmetic ingredients. Astragalii Radix was collected in Jecheon, Jeongseon, Taebaek, and Yeongju in Korea and China as growing districts. Formononetin was determinated by HPLC method as one of the various active agents in Astragalus membranaceus root extract. The 75% ethanol extract demonstrated to be more effective than $H_2O$-extracted one for a scavenging activities to DPPH radicals and reactive oxygens. The 75% ethanol extract showed $IC_{50}$ (50% scavenging concentration) of 2.162 mg/mL and 2.981 mg/mL in case of free radical scavenging activity test and reactive oxygen scavenging activity test, respectively Especially, free radical scavenging activity of isoflavonoids isolated from ethylacetate fraction was similar to scavenging activity of genistein. Astragalus membranaceus root extract of Jecheon district by sonicating extraction method was more effective in skin hydration compared with others.

Background: A growing interest in health has increased the need for the development of potent antioxidant materials known to play a role in various physiological activities. Currently research and development of non-toxic natural antioxidants with high activity is ongoing. Methods and Results: In this study, we measured 2,2'-azinobis- (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability of 10 plant essential oils, selecting samples of Dendranthema indicum, Dendranthema zawadskii, and Citrus sunki essential oils. The samples were analyzed using liquid chromatography (LC) and the radical scavenging activity on LC-based systems with the same conditions. In the LC-mass spectroscopy (MS)/MS analysis of the active compound peak, 2-methoxy-4-vinylphenol with a molecular weight of 150.1 g/mol was identified in C. sunki essential oils. Eugenol or isoeugenol with a molecular weight of 164.1 g/mol as identified in D. indicum and D. zawadskii essential oils as radical scavenging active compounds. Conclusions: In the LC-based measurement system, the active ingredient can be identified by simultaneously conducting profile analysis and the radical scavenging activity of essential oil samples. In addition, LC-MS/MS analysis of the active compound peaks can be performed under the same separation conditions to obtain data that can identify the active compounds in the sample.
An antioxidant peptide derived from Pinctada fucata meat using an Alcalase2.4L enzymatic hydrolysis method (named AOP) and identified by LC-TOF-MS has promising clinical potential for generating cosmetic products that protect skin from sunshine. To date, there have been few published studies investigating the structure-activity relationship in these peptides. To prepare antioxidant peptides better and improve their stability, the design and expression of an antioxidant peptide from Pinctada fucata (named DSAOP) was studied. The peptide contains a common precursor of an expression vector containing an ${\alpha}$-helix tandemly linked according to the BamHI restriction sites. The DNA fragments encoding DSAOP were synthesized and subcloned into the expression vector pET-30a (+), and the peptide was expressed mostly as soluble protein in recombinant Escherichia coli. Meanwhile, the DPPH radical scavenging activity, superoxide radical scavenging activity, and hydroxyl radical scavenging activity of DSAOP $IC_{50}$ values were $0.136{\pm}0.006$, $0.625{\pm}0.025$, and $0.306{\pm}0.015mg/ml$, respectively, with 2-fold higher DPPH radical scavenging activity compared with chemosynthesized AOP (p < 0.05), as well as higher superoxide radical scavenging activity compared with natural AOP (p < 0.05). This preparation method was at the international advanced level. Furthermore, pilot-scale production results showed that DSAOP was expressed successfully in fermenter cultures, which indicated that the design strategy and expression methods would be useful for obtaining substantial amounts of stable peptides at low costs. These results showed that DSAOP produced with recombinant Escherichia coli could be useful in cosmetic skin care products, health foods, and pharmaceuticals.

Background: This study aimed to investigate the antioxidat and antimicrobial activities of the methanol extract and its fractions prepared from the roots of Sanguisorba officinalis L. Methods and Results: The antioxidant activities were compared by evaluating the DPPH radical and nitric oxide (NO) scavenging ability. Measurement of DPPH radical scavenging ability showed that the $SC_{50}$ values of the ethyl acetate fraction was $3.85{\mu}g/m{\ell}$. The ethyl acetate fraction exhibited the most effective DPPH radical scavenging ability compared with the other samples. As for the NO scavenging ability, at all tested concentrations, the ethyl acetate fraction showed a higher scavenging activity than that of the extract and other fractions. These results are related to the total phenolic compound and flavonoid contents of the ethyl acetate fraction. Antimicrobial activity against foodborne pathogens was investigated using the disc diffusion assay. The ethyl acetate fraction showed the highest antimicrobial activity against gram-positive Staphylococcus aureus and Bacillus cereus. However, the chloroform fraction had a higher antimicrobial activity against gram-negative Vibrio vulnificus than that of the extract and other fractions. Conclusions: The results show that the ethyl acetate fraction had a higher antioxidant as well as antimicrobial activity, than did the other samples. Therefore, the ethyl acetate fraction has potential application in the food industry.

Objective: For designing an optimal countermeasure against ageing and ageing related disease, it is necessary to understand the ageing mechanism and other reactive species. Free radicals are associated with the mechanisam of biological ageing and age-related diseases. This study was performed to identifiy the scavenging effect of Radix Cynanchi Wilfordii Herbal-Acupunture Solution(RCWHA) on Nitric Oxide(NO), 1, 1-diphenyl-2-picryl-hydrazyl(DPPH) and Interleukin-4(IL -4). Methods: The activity to scavenge DPPH radicals and NO was observed in vitro, We evaluated the antioxidant activity of RCWHA by using DPPH or NO free radical method. Also the inhibitory effect on IL-4 in RCWHA was observed in vitro, Results: The followings are the summary of the results; (1) There is a significant scavenging effect of RCWHA on NO in 0.1mg/ml and 10mg/ml group after 24hrs. (2) There is no significant scavenging effect of RCWHA on DPPH. (3) There is a significant scavenging effect of RCWHA on IL-4 in $10{\mu}g/m{\ell}$ and $100{\mu}g/m{\ell}$ group. Conclusions: These findings indicate that RCWHA can be used as antioxidant or antiimflamatory drug. But further study is needed about the effect of RCWHA.

Objectives: Our previous studies have clearly demonstrated that the scavenging activity of reactive oxygen species (ROS), protective effect of lipid peroxidation (LPO), and inhibition of cytochrome P450 isozymes (CYPs) from the Circium japonicum aqua-acupuncture solution (CJAS). But, Circium japonicum water extracted solution (CJWS) was weakly reported in cardiovascular diseases such as oxidative stress-mediated atherosclerosis or its value evaluated. Methods: CJWS was assessed to determine the mechanism of its scavenging activity of ROS and inhibitory effect of CYP 2E1. Results: CJWS exhibited a concentration-dependent scavenger of DPPH and superoxide anions radicals using different assay systems. In addition, CJWS showed dose-dependent free radical scavenging activity, including hydroxyl radicals, peroxynitrite, and nitric oxide. The CJWS was also found to be effective in protecting rat liver homogenate against LPO. Futhermore, the CJWS showed significant inhibition of CYP 2E1 induced by pyrazol in a rat liver microsome. Conclusion : ROS and CYPs may play a role in several diseases, such as cardiovascular disease and heart failure. Our study demonstrated that the CJWS has excellent scavenging activity of ROS. Hence, it is worthwhile to investigate the potential effectiveness of CJWS in preventing oxidative stress-mediated cardiovascular diseases.
Laver is one of the most consumed edible red algae seaweeds in the genus Porphyra. Laver is primarily prepared in the form of dried, roasted, and seasoned products. We investigated the total polyphenol and flavonoid contents of laver products, and evaluated the in vitro antioxidant properties of solvent extracts from commercially processed laver products. Significant differences in the concentration of phenolic compounds were found among differently processed laver. The total phenolic content for laver extracts ranged from 10.81 mg gallic acid equivalent (GAE)/g extract to 32.14 mg GAE/g extract, depending on extraction solvent and temperature. Laver extracts contained very few flavonoids (0.55 mg catechin equivalent/g extracts to 1.75 mg catechin equivalent/g extracts). 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), hydroxyl radical, and superoxide anion scavenging assays were used to determine the radical scavenging capacities of laver extracts. These assays revealed that the processing method and extraction condition affected the antioxidant potentials of laver. Antioxidant activity of dried laver, roasted laver, and seasoned laver increased in a concentration-dependent manner ($100{\sim}1,000{\mu}g/mL$). The radical scavenging activities of $37^{\circ}C$ and $100^{\circ}C$ water extracts were lower than that of a $37^{\circ}C$ 70% ethanol extract. The highest radical scavenging capacity was observed in the $37^{\circ}C$ 70% ethanol extracts of dried laver, roasted laver, and seasoned laver. Overall, these results support that notion that laver contains bioactive compounds, such as polyphenols and flavonoids, which may have a positive effect on health.
Objectives : The objective of this study was to compare the antioxidant effects among cultivated wild ginseng and ginseng extracts. Methods : In vitro antioxidant activities were examined by superoxide and hydroxyl radical scavenging activities of ginseng and cultivated wild ginseng extracts. Results : 1. In the superoxide radical scavenging activities of ginseng and cultivated wild ginseng extracts, antioxidant activities of cultivated wild ginseng extracts was showed higher than cultivated ginseng in the concentration of 0.25 and $0.50mg/m{\ell}$. 2. In the hydroxyl radical scavenging activities of ginseng and cultivated wild ginseng extracts, antioxidant activities of cultivated wild ginseng extracts was showed higher than cultivated ginseng in the concentration of 1.0, 2.5, and $5.0mg/m{\ell}$. Conclusions : In summary, the results of this study demonstrate that cultivated wild ginseng extracts had higher antioxidant activities to cultivated ginseng.
Water and methanolic extracts of Ecklonia cava, a marine brown alga, were prepared by ultrasonic extraction (UE) and conventional extraction (CE) methods. The radical-scavenging activity and the inhibitory effects against hydrogen peroxide ($H_2O_2$)-induced DNA damage of the extracts were investigated. All extracts prepared by CE exhibited higher total polyphenolic content than that in the extracts prepared by UE. Extraction yield and total phenolic content increased as the UE time increased. The radical-scavenging activities increased as the UE time increased. All extracts prepared by CE exhibited higher 1,1-diphenyl-2-pricrylhydrazyl (DPPH) and hydroxyl radical-scavenging activities than did those prepared by UE. Extracts prepared by UE showed stronger scavenging activities on alkyl radical and $H_2O_2$ than those prepared by CE did. Methanolic extract with UE 12 h (100MEU-12h) and methanolic extract with CE 24 h (100MEC-24h) were selected and evaluated by comet assay for their inhibitory effect against $H_2O_2$-induced DNA damage. 100MEU-12h showed slightly greater protective effect against $H_2O_2$-induced DNA damage than 100MEC-24h. Thus, UE can be effectively used as a seaweed extraction technique, and there is potential for scale-up of the extraction process.
Objectives : This study was performed to investigate the antioxidant activity of water extracts from Lycopus lucidus Turcz. ex Benth. leaves, stems and roots at the $100{\mu}g/m{\ell}$ concentration. Methods : The different part of Lycopus lucidus Turcz. ex Benth. extract was prepared using water. The antioxidant activities of polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity, SOD like activity, hydroxyl radical, ABTS(2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), $Fe^{2+}$ chelating, and nitrite scavenging activity. Results : The total polyphenol and total flavonoid content of leaves were the highest at $221.85{\mu}g/mg$ and $794.13{\mu}g/mg$, respectively. Electron donating ability was the 79.68% in the water extract from leaves. The ABTS radical scavenging activity of the hot extracts, leaves ${\gg}$ roots > stems was higher in the order. It was shown the highest at 94.53% in the water extract from leaves, which showed a value equal to 94.7% of ascorbic acid. The hydroxyl radical scavenging activity was the highest at 8.07% in the water extract from leaves. SOD like activity and $Fe^{2+}$chelating activity were leaves of 12.3% and 27%, respectively, which were much higher than those of any other parts. The nitrite scavenging ability of extracts was increased at pH 2.5, and those was the highest in leaves of 83.03%. Its were more than twice the 41.61% of BHT. Conclusion : The results suggest that Lycopus lucidus Turcz. ex Benth. can be used as nutraceutical foods and natural antioxidant.
