Effect of AGI $({\alpha}-Glucosidase\;Inhibitor)-1120$, pine (Pinus densiflora) bark extract and Chaga mushroom (Inonotus obliquus) - and Chaga mushroom mycelium extracts on cellular $NF-{\kappa}B$ activation in malignant human keratinocytes (SCC-13) were evaluated to elucidate the possible correlation of $NF-{\kappa}B$ with antioxidant activity. The antioxidant activities of these natural products were examined in three different evaluation methods, i.e., lipid peroxidation value (POV) evaluation test, and 1,1diphenyl-2-picrylhydrazyl radical (DPPH) and nitric oxide (NO) scavenging test. In a cell-based $NF-{\kappa}B$ monitoring assay systern, all samples revealed the downregulatory profiles on the cellular $NF-{\kappa}B$ activity. AGI -1120 (1, 2 mg) and Chaga mushroom extract (0.05, 0.1 mg) downregulated the $NF-{\kappa}B$ activity in a dose-dependent manner. Chaga mushroom mycelium extract (5 mg) significantly inhibited the $NF-{\kappa}B$ activity (p<0.05). Although AGI-1120 and Chaga mushroom mycelium extract exhibited no antioxidant activities evaluated in pay, Chaga mushroom extract showed antioxidant in a dose-dependent manner at concentrations of $0.05{\sim}1$ mg. While AGI-1120 and Chaga mushroom extract possessed a relatively potential DPPH radical scavenging activity, the NO scavenging activity of Chaga mushroom extract $(SC_{50}:47\;{mu}g)$ was higher than the known antioxidant, vitamin C $(SC_{50}:77\;{mu}g)$. These results suggest that AGI-1120 and Chaga mushroom- and Chaga mushroom mycelium extracts may serve as an useful radical scavenging antioxidant agents with $NF-{\kappa}B$ inhibitory effect in human skin.
Objectives This study was conducted to investigate the beta-glucan, ginsenoside content, antioxidant activity and safety of modified Kyungohkgo added to Sparassis crispa and Hericium erinaceum. Methods The marker compounds contents, antioxidant activity and safety of modified Kyungohkgo were tested. The contents of beta-glucan and ginsenoside Rb1, Rg1, and Rg3 marker compounds were measured, the antioxidant activity was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and a safety test was conducted via single dose toxicity assessment. Results Analyzing the contents of marker compounds showed 351.75 mg/g of beta-glucan, 0.0327 mg/g of ginsenoside Rb1 and 0.0802 mg/g of ginsenosai Rg3. In the DPPH free radical scavenging activity, the inhibition concentration 50% of modified Kyungohkgo was 0.2880%. The scavenging activity of modified Kyungohkgo was 5.49% activity at 0.05% concentration, 89.66% activity at 0.5% concentration, 94.68% activity at 1% concentration, and 96.06% activity at 5% concentration. In the single dose toxicity test of modified Kyungohkgo, a dose of 2,000 mg/kg B.W. was set at its highest capacity and observed after oral administration to female and male rats. No toxicological findings were recognized. It was observed that the resulting lethal dose can be set to 2,000 mg/kg B.W. or higher for both females and males. Conclusions The results of the experiment on modified Kyungohkgo showed that the marker compounds contents were beta-glucan and ginsenoside Rb1 and Rg3, that antioxidant activity was observed through the DPPH free radical scavenging activity, and safety was confirmed through the single dose toxicity assessment.

Methods The marker compounds contents, antioxidant activity and safety of herbal medicine mix were tested. The contents of beta-glucan and ginsenoside Rg3 were measured, the antioxidant activity was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, anti-inflammatory and a safety test was conducted via single dose toxicity assessment. Results Analyzing the contents of marker compounds showed 362.3 mg/g of beta-glucan, and 0.4184 mg/g of ginsenoside Rg3. In the DPPH free radical scavenging activity, the IC50 of herbal medicine mix, was 0.146%. The scavenging activity of herbal medicine mix was 88.28% activity at 0.5% concentration, and 90.61% activity at 5% concentration. In the lipopolysaccharides (LPS) anti-inflammatory test, the herbal remix showed a significant decrease in tumor necrosis factor-alpha (TNF-𝛼) and interleukin-6 (IL-6) compared to the LPS-induced group. In the single dose toxicity test of herbal medicine mix, a dose of 2,000 mg/kg body weight (BW) was set at its highest capacity and observed after oral administration to female and male rats. No toxicological findings were recognized. It was observed that the resulting lethal dose can be set to 2,000 mg/kg BW or higher for both females and males. Objectives : Butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA) were well known as anti-oxidant, but they were limited to use because of toxicity. So, many studies are being done to develope natural anti-oxidant. Total phenolic and flavonoid contents along with total antioxidant capacity of the ethanolic extract of Plantaginis Herba (PH) were evaluated to explore the reliable and potential sources of novel natural antioxidants. Methods : Total polyphenol contents and total flavonoid contents in PH ethanol extract were determined by colorimetric method. And DPPH(1.1-diphenyl-2-picrylhydrazyl), ABTS(2'-azino-bis (3-ethylbenzothiazoline-6-Surfonicacid)) free radical scavenging capacity and reducing power inhibition activities of PH ethanol extract were measured at 100, 500, 1000, $5000{\mu}g/m{\ell}$ concentrations by spectrometric assay. Results : The total polyphenol contents and total flavonoid contents of the extract were 161.99 mg/g, 144.05 mg/g, respectively. Also, DPPH, ABTS free radical scavenging capacity and reducing power of PH ethanol extract in treated concentrations (100, 500, 1000, $5000{\mu}g/m{\ell}$) increased dose dependently. In particular, DPPH free radical scavenging capacity of PH ethanol extract from $500{\mu}g/m{\ell}$ was significantly increased compared to positive control (BHA). ABTS free radical scavenging capacity of PH ethanol extract from $1000{\mu}g/m{\ell}$ was significantly higher than BHA. Also, reducing power showed that PH ethanol extract from $500{\mu}g/m{\ell}$ was significantly increased compared to BHA. Conclusions : These results suggest that PH ethanol extract has effects to scavenge free radicals, thus PH has potential and applicable benefits for development of materials and products to have anti-oxidation functions.
Background: Centella asiatica is a well-known medicinal plant having a wound healing effect. In this study, the growth, asiaticoside content, and antioxidant components and activity were investigated in C. asiatica resources under different cultivation and irrigation conditions. Methods and Results: The cultivation and irrigation methods were divided as greenhouse and open field, and sprinkler and sub irrigation. respectively. Growth characteristics were measured in aerial parts of C. asiatica. Asiaticoside content was analyzed using high performance liquid chromatography. Total polyphenol, total flavonoid and 2,2-diphenyl-1picryl hydrazyl (DPPH) radical scavenging activity were analyzed for antioxidant activities. Growth was higher under greenhouse and sub irrigation than open field and sprinkler conditions. In the leaves, asiaticoside content was 53.45 mg/g (open field) and 34.38 mg/g (sub irrigation), total polyphenol was 41.14 mg/g (open field) and 25.73 mg/g (sub-irrigation), and total flavonoid was 27.26 mg/g (open field) and 23.72 mg/g (sub-irrigation). DPPH radical scavenging activity in the leaves was 85.97 mg·ascorbic acid equivalent (AAE)/g (open field) and 54.83 mg·AAE/g (sub irrigation). Conclusions: Asiaticoside and antioxidant components and activity were not accompanied with high yield, although high growth was observed under greenhouse conditions. Therefore, cultural requirements of C. asiatica should be sufficiently considered to suit each purpose.
최근 치과영역에서 아산화질소(Nitrous Oxide)를 이용한 의식하 진정과 필은 진정의 임상 적용이 증가함에 따라 수술실 또는 진료실 환경이 아산화질소로 오염될 수 있다. 비록 낮은 농도일지라도 장기간 아산화질소에 노출 시 자연유산의 증가, 기형아 출산 증가, 말초신경염 및 운동신경 장애 등과 같은 부작용을 초래하는 것으로 알려져 있다. 호흡시 흡입 공기의 구성성분의 변화를 줄 수 있는 구강 입구로부터 반경 12 inch 이내 영역인 호흡대역(Breathing zone)에서 아산화질소 농도는 치료자에게 영향을 주게된다. 소아 환자에게 주로 적용되는 깊은 진정시는 환자의 구호흡양에 따라서 호흡대역에서 아산화질소의 농도에 영향을 주게되므로, 깊은 진정시 구호흡의 증가 원인을 규명하기 위해 잉여가스 배출 방법을 달리하여 호흡대역에서 아산화질소 농도를 측정 비교하여 다음과 같은 결론을 얻었다. 1. 깊은 진정을 시행하는 경우 호흡대역에서 아산화질소의 농도는 공급 가스량 증가에 의한 비기도 저항 증가에 따라 증가하는 양상을 보였다. 호흡대역에서 아산화질소 농도 증가는 구호흡 증가에 의한 것이며 구호흡은 비기도 저항과 관계가 있다 할 수 있다. 즉 비기도 저항 증가는 구호흡의 한 요인이라 할 수 있다. 음압을 사용한 호기가스 배출장치를 사용하여도 NIOSH에서 권장하는 허용치에는 미치지 못하였고 이를 위해서는 팬이나 다른 제거 장치가 함께 사용되어야 한다. 2. 구강편도의 크기는 기도 저항이 적은 경우 즉 음압을 사용하여 호기가스 제거하는 경우 구호흡에 영향을 주었다.
과일에는 플라보노이드, 탄닌, 카테킨 등의 폴리페놀 뿐만 아니라 여러 항산화능과 관련된 성분을 많이 함유하고 있으며, 과일의 껍질에는 과육부분보다 2-9배 이상 높은 각종 기능성 성분이 많은 것으로 보고되고 있다. 국내에서 사과는 가장 많이 생산되는 과일 중에 하나이며, 사과껍질에는 quercetin의 함량이 높은 것으로 알려져 있다. 하지만 국내에서 사과껍질의 항산화능을 분석하거나 이를 이용하여 식품에 적용한 연구는 부족한 실정이다. 따라서 국내산 사과껍질 원물 균질액, 동결건조분말, 열풍건조분말 및 초미세분말의 총페놀함량(TPC), 총플라보노이드함량(TFC), ABTS radical 소거능(ABTS), DPPH radical 소거능(DPPH)을 비교하고, 이를 통해 사과껍질을 식품 소재로 활용 시 최적 분말화 및 추출조건을 알아보고자 했다. TPC는 동결건조분말, 열풍건조분말의 추출물에서 가장 높게 나타났고, TFC는 초미세분말 추출물에서 가장 높게 나타났다. ABTS 라디컬 소거능은 초미세분말에서, DPPH 라디컬 소거능은 동결건조분말에서 가장 높게 측정되었다. 이상의 결과를 통해, 동일 품종의 사과껍질을 사용한 경우 원물의 가공방법 및 추출조건에 따라 각 기능성의 차이가 나타났다. 이중, 초미세분말에서 TFC와 ABTS 라디컬 소거능이 가장 높은 결과를 나타낸 것을 볼 때, 분말화 방법 등 가공조건의 차이에 의해 개발된 중간소재의 여러 기능성에서 차이가 날 수 있다는 결과를 얻었다. 따라서 본 연구를 통해 기능성이 향상된 분말화된 식품중간소재 개발 시 여러 가공조건에 의해 영향을 받을 수 있으므로, 이를 고려하는 것이 중요하다는 것을 알 수 있었다.
Background: The extract of Abies holophylla is used as an ingredient in cosmetics. This study assessed the antioxidant and antibacterial activities of the material remaining after the extract is used. Methods and Results: The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azinobis (3-ethyl benzothiazoline)-6-sulfonic acid (ABTS) radical scavenging abilities were assessed to determined the free radical scavenging activity. The total phenol and flavonoid contents were determined to measure the antioxidant activity. The DPPH and ABTS radical scavenging activities of the resudual extract were higher (95.61 - 99.42% and 74.26 - 77.98% in water extract respectively) than those of the positive control. In 50% EtOH extract, the total phenol content was $389.84mg{\cdot}GAE/m{\ell}$, and the total flavonoid was $0.15mg{\cdot}QE/m{\ell}$. The minimum inhibition concentration degree for antibacterial activity against Staphylococcus aureus was < 8 to < $125{\mu}g/m{\ell}$ compared to that of the positive control in all extracts. The clear zone against S. aureus was found to be $12.2{\pm}3.8mm$. Conclusions: The A. holophylla byproducts were found to have antioxidant and antibacterial activities. Therefore, the materials remaining after the A. holophylla extract is used in cosmetics has potential functional uses.
Objectives The purpose of this study was to investigate the Anti-oxidation and anti-inflammatory effects of ethanol extract from asiasari radix (AR) on lipopolysaccharide (LPS)-induced in RAW 264.7 Cells Methods Anti-oxidative effects of AR were measured by scavenging activities of 1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and production of reactive oxygen species (ROS) in RAW 264.7 cells. Anti-inflammatory effects of AR were measured by mediators including nitric oxide(NO), interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), tumor necosis factors-${\alpha}$ (TNF-${\alpha}$) and iNOS, IL-$1{\beta}$, IL-6, TNF-${\alpha}$ mRNA expression in RAW 264.7 cells. Results Total phenolic content was expressed $28.77{\pm}1.67$. DPPH radical Scavenging was increased depend on AR ethanol extract. ABAT radical Scavenging was increased depend on AR ethanol extract. Production of ROS was significantly decreased by AR ethanol extract on concentration of 100 (${\mu}g/ml$). Production of NO was significantly decreased by AR ethanol extract on concentration of $100({\mu}g/ml)$. Production of IL-$1{\beta}$, interleukin-6 and TNF-${\alpha}$ were increased depend on AR ethanol extract. And Production of interleukin-6, TNF-${\alpha}$ were significantly decreased AR ethanol extract. iNOS, IL-$1{\beta}$, IL-6, TNF-${\alpha}$ mRNA expression of RAW 264.7 cells was increased depend on AR ethanol extract. Conclusions According to this study, AR ethanol extract has anti-oxidative and anti-inflammatoy effects.
Objectives: To investigate the effect of fermented extract of Artemisiae Iwayomogii Herba, Curcumae Longae, Crataegi Fructus and Salviae Miltiorrhizae Radix (FMH) on anti-inflammation associated with dyslipidemia and anti-oxidation in RAW264.7 and HUVEC cells. Methods: The total polyphenols, total flavonoids, DPPH radical scavenging activity, ABTS radical scavenging activity, and cytotoxicity of FMH were measured. RAW264.7 cells treated with FMH were tested for production of NO, and for cytokine and LTB4 levels and HUVEC cells treated with FMH were examined for production of cDNA of genes related to inflammation. Results: 1. FMH contained polyphenols and flavonoids. The DPPH and ABTS radical scavenging activity of FMH increased in a concentration-dependent manner. 2. FMH treatment inhibited the production of nitric oxide (NO), cytokines, and LTB4 in RAW264.7 cell when compared to the untreated control group. 3. FMH decreased the transcription of pro-inflammatory genes, whereas it increased transcription of anti-inflammatory genes, in HUVEC cells. Conclusion: FMH is effective as an antioxidant and for treatment and prevention of dyslipidemia, atherosclerosis, ischemic heart disease, stroke, and other cardiocerebrovascular diseases.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
