참다래를 가해하는 해충과 우점종인 열매꼭지나방의 가해특성 (Investigation of Insect Pests and Injury Characteristics of Stathmopoda auriferella (Walker) on Kiwi Fruit Tree)

  • 1991~1993년에 참다래 집단 재배지역인 전남 남부해안지방을 중심으로 참다래를 가해하는 해충과 우점종인 열매꼭지나방의 가해특성에 관하여 연구한 결과 해충은 총 12과 13종이 조사되었으며 그중 열매꼭지나방과 뽕나무깍지벌레가 우점종이었다. 열매꼭지나방 의 태별크기는 난이 0.12mm, 종령유충이 9.8mm, 용이 5.9mm 그리고 성충의 날개편 길이는 12.3mm이었다. 열매꼭지나방에 의한 피해과율은 평균 45.9%이었으며 지역에 따라 큰 차이를 인정할 수 없었다. 성충은 연 2회발생하며 1회는 5월하순~7월중순 , 2회는 8월중순~9월상순으로 최성기는 각각 6월상중순, 8월하순이었다. 시기별 유충과 용의 비율은 7월상순에서 100:0이었다. 중순은 96.1:3.9, 하순은 64.9:35.1 이었고 8월 상순에는 19.8:80.2, 중순은 0:100이었다. 또한 피해과는 7월상순에 4.6%로 처음 발견되었으며 7월중순 이후에는 40%이상으로서 이 시기의 피해가 당년의 피해정도를 결정하였고 가해하는 부위는 과정부가 70%, 과경부가 11.1%이었으며 품종별로는 국내 주 지배품종인 헤이워드 32.2%, 몬티 22.2% 향록 13.9%, 아보트 9.3%. 브로노 6.0% 이었다.

도로교통소음에 대한 도입 연출음의 마스킹 효과 (The Masking Effects of Introducing Sounds about Road Traffic Noise)

  • This paper describes a experimental study determining the effects of the type and loudness of introducing sounds on the amenity and the masking degree of background noise. 132 relative test sounds with 11 types of introducing sounds and 3 steps of road traffic noise were presented to 29 subjects through headphone in the laboratory experiment. Seven scale of polar adjective such as 'Strong Weak', 'Pleasant Unpleasant', 'Clear Unclear', 'Suitable Unsuitable', 'Comfortable-Discomfortable' was employed for the evaluation of amenity. And Seven scale of mono polar adjective such as 'Very-Not at all' was employed for the evaluation of masking degree against road traffic noise. The results are as follows; Green music was evaluated the highest amenity among test sounds and murmurs of a stream, signal music, song of a bird, crying of insect was showed the amenity rank of order. The softer and weaker introducing sound was evaluated more suitable and comfortable under the road traffic noise. So adequate level of introducing sounds was considered for the desirable urban soundscape. Also the louder sounds compared to background noises had greater masking effects. But similar levels of introducing sounds were recommended considering both good amenity and masking effects.

A Study on the Inhibition of 2-deoxy-D-Glucose Transport of the Endogenous Glucose Transporters in Spodoptera frugiperda Clone 21-AE Cells by Using Hexoses

  • The baculovirus/insect cell expression system is of great value in the study of structure-function relationships in mammalian glucose-transport proteins by site-directed mutagenesis and for the large-scale production of these proteins for mechanistic and biochemical studies. Spodoptera frugiperda Clone 21 (Sf2l) cells grow well on TC-100 medium that contains $0.1\%$ D-glucose as the major carbon source, strongly suggesting the presence of endogenous glucose transporters. However, very little is known about the properties of the endogenous sugar transporter(s) in Sf2l cells, although a saturable transport system for hexose uptake has been previously revealed in the Sf cells. In order to further examine the substrate and inhibitor recognition properties of the Sf2l cell transporter, the ability of hexoses to inhibit 2-deoxy-D-glucose (2dGlc) transport was investigated by measuring inhibition constants $(K_i)$. The $K_i's$ for reversible inhibitors were determined from plots of uptake versus inhibitor concentration. Transport was effectively inhibited by D-mannose and D-glucose. Of the hexoses tested, L-glucose had the least effect on 2dGlc transport in the Sf2l cells, indicating that the transport is stereoselective. Unlike the human HepG2 type glucose transport system, D-mannose had a somewhat greater affinity for the Sf2l cell transporter than D-glucose, implying that the hydroxyl group at the C-2 position is not necessary for strong binding. However, epimerization at the C-4 position of D-glucose (D-galactose) resulted in a dramatic decrease in affinity of the hexose for the Sf2l cell transporter. Such a lowering of affinity might be the result of the involvement of the C-4 hydroxyl in hydrogen bonding. It is therefore suggested that Sf2l cells were found to contain an endogenous sugar transport activity that in several aspects resembles the human HepG2 type glucose transporter, although the insect and human transporters do differ in their affinity for cytochalasin B.

Expression of porcine circovirus type 2 capsid protein fused with partial polyhedrin using baculovirus

  • Porcine circovirus type 2 (PCV2) is an important infectious swine virus causing postweaning multisystemic wasting syndrome (PMWS). PCV2 capsid protein, encoded by ORF2 has type-specific epitopes, is very immunogenic, and is associated with the induction of neutralizing antibodies. For the efficient production of capsid protein, recombinant Autographa californica nucleopolyhedroviruses were generated to express ORF2 fused with two forms of a partial polyhedrin. Recombinant capsid protein was produced successfully with the partial polyhedrin fusion form and the yield was high, as was shown by SDS-PAGE. Production of recombinant capsid proteins in insect cells was confirmed by Western blot analysis using anti-His monoclonal antibody, anti-ORF2 monoclonal antibody, and anti-PCV2 porcine serum. Fusion expression with amino acids 19 to 110 of the polyhedrin increased the production of recombinant capsid protein, but fusion with amino acids 32 to 85 did not. Additionally, PCV2 capsid protein is a glycoprotein; however, the glycosylation of recombinant protein was not observed. The results of an Enzyme-linked immunosorbent assay (ELISA) showed that recombinant capsid proteins could be utilized as antigens for fast, large-scale diagnosis of PCV2-infected pigs. Our results suggest that the fusion expression of partial polyhedrin is able to increase the production of recombinant PCV2 capsid protein in insect cells.

  • The aim of the present investigation is to determine the nutritional composition of various insects and their potential uses as alternative protein sources in animal diets. The feeding industry requires production systems that use accessible resources, such as feed resources, and concentrates on the potential impacts on production yield and nutritional quality. Invertebrate insects, such as black soldier flies, grasshoppers, mealworms, housefly larvae, and crickets, have been used as human food and as feed for nonruminants and aqua culture while for ruminants their use has been limited. Insects can be mass-produced, participating in a circular economy that minimizes or eliminates food- and feed-waste through bioconversion. Although the model for formula-scale production of insects as feed for domestic animals has been explored for a number of years, significant production and transformation to being a conventional protein resource remains to be deeply investigated. This review will focus on the nutritional composition of various insects and their potential use as alternative protein sources, as well as their potential use to promote and support sustainable animal production. Furthermore, nutritional compositions, such as high protein, lauric acid omega 6, and omega 3, and bioactive compounds, such as chitin, are of great potential use for animal feeding.

Characterization of a Septobasidium sp. Associated with Felt Disease of Schisandra chinensis

  • Extensive disease surveys performed during the summers of 2013 and 2014 in Schisandra chinensis orchards resulted in the finding of a Septobasidium sp. associated with felt disease. The fungus was characterized to be symbiotic with a scale insect (Pseudaulacaspis cockerelli). Morphological and molecular characteristics of the Septobasidium isolates were investigated. The isolates were morphologically and phylogenetically close to S. bogoriense. We tentatively describe this isolate as a Septobasidium sp., mainly because of the limited amount of information available on the internal transcribed spacer region of the ribosomal DNA of Septobasidium spp.

Flower Habitat Supplementation can Conserve Pollinators and Natural Enemies in Agricultural Ecosystem: Case Study in the Pepper Field

  • Pollinators play important roles for crop production as well as maintenance of wild plants' reproduction. Pollinator dependency in Korean agriculture have increased, but the degradation of habitat quality pose significant threats to pollination in agroecosystem. Provisioning flower habitat is one of the major activities to increase pollinator abundance and richness. We tested the effect of flower habitat supplementation on the pollinator, pest and natural enemy abundance in pepper field. Results showed that Hymenopteran pollinators were more abundant in the pepper plots close to the flower habitat. Among natural enemies, the similar pattern from pollinators was observed to parasitoids in the family of Eulophidae, but not to those of Ichneumonidae. There were no statistically significant differences of insect pests like aphids and thrips, as well as pepper production among plots in the pepper field. Even limited spatial scale, this study showed supplementing the flowering plant habitat to the agricultural landscape could conserve and boost pollinators and natural enemies, and possibly resulting better fruit production by pollination service.

2007년과 2008년 경북 상주지역 감 과원의 과원 관리실태 조사 (Survey of Actual Condition of Management of Persimmon Orchards in Sangju, Gyeongbuk in 2007 and 2008)

  • 우리나라 최대의 곶감 및 감 생산지인 경북 상주지역 감 재배농가의 병해충 관리 실태와 과원관리 실태를 알아보기 위하여 2007년과 2008년 감 재배 농가를 대상으로 설문 조사를 수행하였다. 감 경작자의 91.1%가 50대 이상이었으며 재배농가의 94.7%가 '상주둥시'를 재배하고 있었다. 58.4%의 경작자가 병에 의한 수확량 감소가 가장 심각하며 68.1%가 탄저병(Colletotrichum gloeosporioides)에 의한 피해가 가장 심하다고 하였다. 해충은 깍지벌레류의 피해가 심하다고 응답한 농가가 60.2%로 가장 많았다. 병해충 관리에 관한 지식은 경험에 의존하는 비율이 37.2%였으며 약제 선정은 농약 판매상의 권유가 48.7%로 가장 많았다. 년 평군 방제 횟수는 $3{\sim}5$회가 54%로 가장 많았으며 방제력을 기록하는 농가는 17.7%에 불과하였다. 병해충 관리의 가장 큰 문제점은 병해충 지식의 부족이라는 응답자가 60.2%로 가장 많았으며 과원 관리시 병해충 방제 비용이 가장 많이 들어간다는 농가가 52.2%로 가장 많았다. 감 재배농가의 66.4%가 타 작물과 혼작을 하고 있었으며 콩과 고추를 혼작하는 농가가 가장 많았다. 감 재배와 곶감 가공을 겸하는 농가가 44.2%였으며 곶감 가공 과정중 가장 큰 애로사항은 인력부족이 54.5%로 가장 많았다.

Effect of a Bombyx mori Protein Disulfide Isomerase on Production of Recombinant Antibacterial Peptides

  • The insect baculovirus expression vector system (BEVS) is useful for producing biologically active recombinant proteins. However, the overexpression of heterologous proteins using this system often results in misfolded proteins and the formation of protein aggregates. To overcome this limitation, we developed a versatile baculovirus expression and secretion system using Bombyx mori protein disulfide isomerase (bPDI) as a fusion partner. bPDI gene fusion was found to improve the secretions and antibacterial activities of recombinant nuecin and enbocin proteins. Thus, we conclude that bPDI gene fusion is a useful addition to BEVS for the large-scale production of bioactive recombinant proteins.

Genomic Insights into Nematicidal Activity of a Bacterial Endophyte, Raoultella ornithinolytica MG against Pine Wilt Nematode

  • Pine wilt disease, caused by the nematode Bursaphelenchus xylophilus, is one of the most devastating conifer diseases decimating several species of pine trees on a global scale. Here, we report the draft genome of Raoultella ornithinolytica MG, which is isolated from mountain-cultivated ginseng plant as an bacterial endophyte and shows nematicidal activity against B. xylophilus. Our analysis of R. ornithinolytica MG genome showed that it possesses many genes encoding potential nematicidal factors in addition to some secondary metabolite biosynthetic gene clusters that may contribute to the observed nematicidal activity of the strain. Furthermore, the genome was lacking key components of avermectin gene cluster, suggesting that nematicidal activity of the bacterium is not likely due to the famous anthelmintic agent of wide-spread use, avermectin. This genomic information of R. ornithinolytica will provide basis for identification and engineering of genes and their products toward control of pine wilt disease.
