A total of 45 scale insects belonging to 29 genera of five families were recognized on 15 kind of flowering plants on the basis of specimens collected in 2003 and specimens deposited in the Insect Collection of NIAST in Korea. Three species were recognized on Korean forsythia, four on chrysanthemum, 15 on common camellia, seven on kobus magnolia, three on rose of sharon, one on Indian lilac, six on rose, seven on rhododendrons, two on bamboo palm, three on benjamin tree, 12 on evergreen euonymus, five on Japanese yew, two on corn plant, six on orchids and two on cacti. Pulvinaria floccifera (Westwood) and Pseudaonidia paeoniae (Cockerell) on common camellia and Pulvinaria citricolar Kuwana on rose of sharon are reported for the first time from the Korean Peninsula.
The baculovirus/insect cell expression system is of great value for the large-scale production of normal and mutant mammalian passive glucose-transport proteins heterologously for structural and functional studies. In most mammalian cells that express HepG2, this transporter isoform is predominantly located at the cell surface. However, it had been reported that heterologous expression of other membrane proteins using the baculovirus system induced highly vacuolated cytoplasmic membranes. Therefore, how a cell responds to the synthesis of large amounts of a glycoprotein could be an interesting area for investigation. In order to examine the subcellular location of the human HepG2 transport proteins when expressed in insect cells, immunofluorescence studies were carried out. Insect cells were infected with the recombinant baculovirus AcNPVHIS-GT or with wild-type virus at a MOI of 5, or were not exposed to viral infection. A high level of fluorescence displayed in cells infected with the recombinant virus indicated that transporters are expressed abundantly and present on the surface of infected Sf21 cells. The evidence for the specificity of the immunostaining was strengthened by the negative results shown in the negative controls. Distribution of the transporter protein expressed in insect cells was further revealed by making a series of optical sections through an AcNPVHIS-GT-infected cell using a confocal microscope, which permits optical sectioning of cell sample. These sections displayed intense cytoplasmic immunofluorecence surrounding the region occupied by the enlarged nucleus, indicating that the expressed protein was present not only at the cell surface but also throughout the cytoplasmic membranous structures.
Insect cell cultures were performed in laboratory-scale vessels. The batch growth of insect cells was affected by such parameters as serum content, other nutrients, seeding density, and mechanical agitation. Lactate and ammonium were not likely to be environmental factors that inhibited cell growth at the concentrations observed at the end of batch cultures. In addition, redox potential was found to be a useful index in monitoring low-level dissolved oxygen during the cultivation of insect cells. Recombinant protein production by cells infected with a genetically-modified baculovirus was also demons treated. The maximum beta-galactosidase synthesis of 2800 units per reactor volume was achieved at the dilution rate of $0.006hr^{-1}$.

Scale insects heavily damaged many agricultural crops and trees in urban and forest areas. This study was conducted to carry out alternative control methods for environmentally friendly control of scale insects. Hot water extract of Quisquali indica produced high mortality against the black pine bast scale, Matsucoccus thunbergianae in previously experiment. Methanol extract obtained from fruit of Q. indica was successively fractionated using hexane, chloroform, ethyl acetate, buthanol and water which were examined by exposing for their insecticidal activity against four Cocoidea species (Eriococcus lagerstroemiae, Ceroplastes japonicus, Crisicoccus pini and Planococcus citri) in laboratory. Hexane fraction gave the highest insecticidal activity against scale insect pest. GC-mass analysis confirmed that the main compounds of Q. indica were alpha-pinene, methyl palmitate, eugenol, methyl myristate, phthalic acid mono(2-ethylhexyl)ester and palmitic acid. Among those compounds, ${\alpha}$-pinene was included 10 hexane subfractions. $LC_{50}$ of ${\alpha}$-pinene was 0.0114 ppm against 2nd instar of E. lagerstroemiae. These results suggest that Q. indica extracts might be used as an alternative control agent of scale insects.
The objective of this study was to evaluate the beneficial effects of the short-term application of insect-based diet in canine allergic dermatitis. Total 19 atopic dogs with concurrent cutaneous adverse food reactions were enrolled and classified into 3 groups. The treatment group (n = 7) was fed insect-based diet, the positive control group (n = 6) was fed salmon-based diet, and the negative control group (n = 6) was fed commercial or homemade diet for 12 weeks. The degree of skin lesions was evaluated based on canine atopic dermatitis extent and severity index (CADESI-4). Additionally, transepidermal water loss (TEWL) and pruritus visual analog scale were evaluated. All indices were evaluated every 4 weeks after the initial administration of hypoallergenic diets. In the treatment group, significant decrease in the CADESI-4 score was observed at 8 weeks compared to the baseline score (p = 0.031). There were significant differences in the CADESI-4 score between the groups at 8 weeks (p = 0.008), 12 weeks (p = 0.012), and TEWL at 12 weeks (p = 0.022). This preliminary result demonstrates the potential hypoallergenicity of an insect-based diet through features that diminish cutaneous lesions and skin barrier dysfunction.
This study was carried out to develop of macro-protocol and the biosafety guide for 'Nakdong', and 'Golden Rice' (genetically modified vitamin A rice) by large scale field trial of GM crops. Typically, when a new GM crop is created, breeders should conduct field test to make sure the GM crop is safe, and provide some information on GM crops for approval. A total of isolated 4,700 $m^2$ field for trial of GM crops were prepared, and 'Nakdong' and 'Golden rice' were cultivated by standard method of RDA (Rural Development Administration, Korea). Field studies indicated that the population densities of insect pests and natural enemies have no difference between two varieties. While insect pest density on 'Nakdong' was slightly higher than on Golden Rice, but natural enemy density on Golden Rice was a little higher. These results provided the insect diversity for risk assessment analysis of Golden Rice and suggested that the macro-protocol could be useful to detect GM plants.

Insect cell lines and the control of infection for obtaining the maximum amount of polyhedrin-Cry1Ac-polyhedrin fusion protein from Bactrus in monolayer and suspension culture systems were tested. Growth rates of the Trichoplusia ni(High-Five) cell line in both culture systems were better than the other insect cell lines, Spodoptera frugiferda(Sf-9, Sf-21), Trichoplusia ni(Tn5), and Spodoptera exigua(Se301). The expression of the fusion protein in a monolayer culture showed that Se301 cells were 2.3-4.8 times more productive on a per cell basis than the other cell lines. However, in suspension culture, only High-Five cells were productive. High-Five cells infected with Bactrus at a multiplicity of infection(MOI) of 5 and a cell density of $3.0{\times}10^5$ cells per ml were more productive than the other infection condition in a suspension culture suitable for a large-scale production of baculovirus. In conclusion, for the large-scale production of Bactrus in vitro, High-Five cells showing good growth and high productivity are suitable.
This study aimed to investigate the characteristics of 3 pre-treatment of mealworm(Tenebrio molitor), and offer basic research data through appropriate pre-treatment method based on the component analysis result. This study was preparing the control group sample No.0(not contain mealworm), and the other sample (Sample No.1,2,3) was using the mealworm prepared in each of the three methods. By using the sample, this study conducted an inspection for component analysis, mineral contents, pH, color value, total amount polyphenols, DPPH radical scavenging activity, reducing power, acceptance test. Each experiments are three times repeated and validated following the one-way ANOVA analysis to verify a significant difference. Acceptance test conducted by 50 consumers indicate consumption decision for sunsik. using the seven point scale, assessment for appearance, flavor, taste, texture, overall acceptance. The research findings showed that sample containing sauted mealworm(SE2) is higher preference and significant in antioxidant activity(total amount polyphenols, DPPH radical scavenging activity, reducing power). Therefore, it is better to use sauted. The result of this study suggest that sauted mealworm is good ingredient for consumer acceptability and functionality of sunsik. According to results of the study, it is suggested to make better use of food, and provide food developer with meaningful date for market targeting. In addition, this study also provides the information for improving awareness of edible insect.

Protaetia brevitarsis seulensis (Kolbe) is widely used in Korea, as a protein-rich, alternate, functional food with pharmacological benefits. In addition to anti-oxidant properties, the larvae of P. b. seulensis also show positive effects against hepatic disorder and diabetes; therefore, P. b. seulensis larvae are being reared on a large scale in Korea. We evaluated reared larvae of P. b. seulensis from Gyeong-gi in Korea. Using 16SrRNA PCR, electro-microscopy, and bioassay techniques, we found that the larvae harbored Spo-1, a bacterium identified as the insect pathogen Serratia marcescens. Therefore, we highlight the use of this insect as an alternate food and the need for its sanitary rearing conditions, as contamination may affect public health.

Animals use their odorant receptors to receive chemical information from the environment. Insect odorant receptors differ from the G protein-coupled odorant receptors in vertebrates and nematodes, and very little is known about their protein-protein interactions. Here, we introduce a mass spectrometric platform designed for the large-scale analysis of insect odorant receptor protein-protein interactions. Using this platform, we obtained the first Orco interactome from Drosophila melanogaster. From a total of 1,186 identified proteins, we narrowed the interaction candidates to 226, of which only two-thirds have been named. These candidates include the known olfactory proteins Or92a and Obp51a. Around 90% of the proteins having published names likely function inside the cell, and nearly half of these intracellular proteins are associated with the endomembrane system. In a basic loss-of-function electrophysiological screen, we found that the disruption of eight (i.e., Rab5, CG32795, Mpcp, Tom70, Vir-1, CG30427, Eaat1, and CG2781) of 28 randomly selected candidates affects olfactory responses in vivo. Thus, because this Orco interactome includes physiologically meaningful candidates, we anticipate that our platform will help guide further research on the molecular mechanisms of the insect odorant receptor family.
