This study was conducted to determine proper amount of media and the effect of fermented sawdust on yield of Grifola frondosa in bag culture. The yield of Grifola frondosa was 227 g for 2 kg of media compared with 103 g for 1kg, 256 g for 2.5 kg, and 312 g for 3 kg. The greatest yield of Grifola frondosa per mass of media was 114 g/kg media for 2 kg of media. Days to harvest for 2 kg of media was 95 days compared with 93 days for 1kg, 99 days for 2.5kg, and 104 days for 3kg. Addition of fermented oak sawdust increased yield from 90 to 145 g/kg bag for Hambak cultivar and from 104 to 121 g/kg bag for Yipsae1ho cultivar. Culture duration was shorten 4-7 days by treating fermented oak sawdust.
Effects of various kinds of sawdusts, supplements and culture conditions on activities of several enzymes such as protease, phenoloxidase and cellulase produced from mycelium of P. ostreatus grown on sawdust medium were studied and the results are as follows; Higher specific activity of these enzymes was observed when oak tree sawdust and poplar tree sawdust were supplemented with rice bran or wheat bran at rate of 30%, 20% and 10% in total volume respectively. Higher total activities of protease, phenoloxidase and cellulase were observed at 70% of the moisture contents of culture media, while lower activity of these enzymes was observed with 40% moisture contents of sawdust culture medium. The pH 4 and 9 of the sawdust media appeared to be optimum pH for the. production of protease while pH 5 and 7 were optimal for the production of phenoloxidase. The pH 6 of the sawdust medium was optimal for the production of cellulase. The optimum incubating temperature for the production of protease, phenoloxidase and cellulase was $25^{\circ}C$. Higher total activities of protease and phenoloxidase were observed when culture medium was added with wood vinegar at the control, and 0.5% for cellulase.
Currently, plastic bags are being used for sawdust cultivation of shiitake mushroom. However, due to serious environmental problems caused by the use of plastic bags, we studied the efficacy of bottle cultivation method to replace the sawdust bag method. Small detachable plastic bottles (400 g capacity) filled with Quercus spp. sawdust and wheat bran (4:1 w/w) media were incubated for 80 and 120 days. The weight loss (%) of the media was higher for the NIFoS 2464 strain at an approximate light intensity of 300 Lux than light intensity of 500 Lux; the light intensity was associated with the loss of sawdust medium-weight during the cultivation period. The highest yield was observed when the strain was cultivated for 80 days under dark conditions, 40 days under 500 Lux light, and air circulation fan speed of 30 rpm. When incubated for 120 days, mushroom yield in the bottle media was higher at 40 days of light exposure than 20 days of light exposure. In the bottle media incubated for 80 days under dark conditions, the mushrooms fruited due to repetitive water spraying on the top of the media and light stimulation during the fruiting period. The media could be separated from the bottles because the media shrank after the first harvest. These separated plastic bottles could be re-used for mushroom cultivation, thereby reducing the amount of plastic waste.
An isolate of Lepista nuda was cultivated on the solid media consisted of various cereal grains or those of the acacia sawdust mixed with other cereal grains. The mycelia grew well on the solid cultures made from millet cereals, but did not on those from sorghum or corn cereals. Thus, the millet and the acacia sawdust, as the solid media, were employed for the next experiments related to mycelial growths. For growth of this isolate, it was shown that the nitrogen source is needed for those of millet. Also, the rice bran or minerals were considered to be needed for the acacia sawdust. As minerals, however, the rice bran is shown as the best additive in the solid cultures of the acacia sawdust. The mycelia of L. nuda were cultivated directly through the solid cultures. Their growths were measured' by the production of carbon dioxide with gas chromatography.
The cultivation in sawdust media, mushroom productivity, and fruiting body characteristics of Lentinula edodes strains NIFoS 2778 and NIFoS 3363 were compared according to the inoculation conditions. The cultivation period was 5% shorter when liquid spawn was used. Fruiting bodies were induced after 113 days of incubation on media inoculated with liquid spawn, and the cultivation period was 119 days on media inoculated with solid spawn. Mushroom productivity of NIFoS 2778 was the highest (661.4 g) when 36 mL of liquid spawn was used. For NIFoS 3363, mushroom production was higher under liquid inoculation conditions when the same amount of liquid and solid spawns were used. The mushroom characteristics of the two strains were not significantly different, except for gill width and stipe diameter.
This study was conducted to verify the cause of suppression symptom in mycelial growth during summer and to be able to establish a countermeasure. Cultivation of Flammulina velutipes was experimented with varying elapsed time of 0, 3, 6, 9 hours after mixing the sawdust media and two kinds of water temperature (normal water, $24^{\circ}C$ and cold water, $6^{\circ}C$) for mixing sawdust media. There were trends of increased media temperature from $24^{\circ}C$ to $31^{\circ}C$ and decreased pH from 6.5 to 5.2~5.6 with varying elapsed time from mixing the media to sterilization. Bacterial density also increased with bacterial density in Medium $24^{\circ}C$ being 1.9~4.1 times higher than that in Medium 6. Growth of F. velutipes was delayed with dual culture of bacteria isolated from sawdust media. Total nitrogen content of sawdust media was lowered by elapsed time from mixing the media to sterilization. The use of normal water($24^{\circ}C$) delayed mushroom growth by 1~2 days compared with that of cold water($6^{\circ}C$). Furthermore, mycelial growth of F. velutipes in the bottle cultivation ceased 9 hours after mixing the media. Primordia formation of F. velutipes was delayed by 1~3 days by elapsed time after mixing sawdust media, while fruit-body yield decreased by 7~12% 6 hours after mixing the media. Fruit-body yield was more stabilized with the use of cold water($6^{\circ}C$) than with that of normal water($24^{\circ}C$). Results showed that it is effective to use cold water as low as $6^{\circ}C$ in mixing media for F. velutipes cultivation, especially during summer when environmental temperature is high, high pressure sterilization after bottling work can prevent bacterial propagation in the media and can stabilize media ingredient.
This study was made to investigate the difference of the shiitake productivity according to the use of sawdust, plug-shaped and liquid spawn. Contamination of medium was confirmed by eye. There was no contamination in the liquid spawn treatment. And, contamination rates in the sawdust spawn and plug-shaped spawn were 6.7%, respectively. Productivity of fruit-bodies was 725.1 g from sawdust spawn, 726.2 g from plug-shaped spawn and 573.9 g from liquid spawn until 3rd flushing of fruit-bodies. The number of fruit-bodies was 57.4 per medium of sawdust spawn, 64.3 plug-shaped spawn and 45.1 liquid spawn. At yield per medium, the percentage of media producing over 1,000 g was 13% in plug-shaped spawn and 7% in liquid spawn. However, there was no medium producing over 1,000 g of fruit-bodies in sawdust spawn. Fruit-bodies of $600{\sim}900g$ were yielded from 79% of sawdust spawn treatment, 53% plug-shaped spawn and 33% liquid spawn treatment. When compared the productivity of fruit-bodies, liquid spawn treatment appeared lower than sawdust spawn and plug-shaped spawn treatment at 1st flushing, but appeared higher than the others at 2nd and 3rd flushing.

Cauliflower mushroom (Sparassis crispa) is an edible mushroom that shows remarkably high contents of $1,3-{\beta}-D-glucan$ compared to other edible mushrooms. The mushroom is known to give high antitumor and immunologic activities, thus the mushroom is recently cultivated in Japan and Korea. However, cultivation methods are being kept in secret or under patents by some companies with complicated procedures. This study was conducted to solve the problem by providing a simple method for the cultivation of cauliflower mushroom. We could produce the mushroom using sawdust-based media of Larix leptolepis, Pinus densiflora and P. koraiensis with addition of barley flours and sugar. The ratio of the sawdust : barley flours : sugar used for this study was 80:20:3 in dry-weight base, and the water content was adjusted as 65%. The productivity was somewhat different among the species of sawdust media and the strains of Sparassis crispa for the cultivation, and the mushroom production reached up to 177g from 650g base(productivity : 27%). Thus, this medium is strongly recommended as a practical cultivation method for cauliflower mushroom. By the way, further study for the determination of adequate concentration of barley flour and the selection of proper strain for each sawdust species is needed.
This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.

Cultivation of cauliflower mushroom (Sparassis crispa) became a good way of consumption for coniferous sawdust. However, conventional method for the cultivation demanded ready-decomposed sawdust in field more than 6 months, which resulted in the spatial and temporal problems. This study was conducted to develop an efficient cultivation method to minimize the problem with steam-treated sawdust media of Larix leptolepis, Pinus densiflora and Pinus koraiensis. By the treatment, mycelial growth was stimulated by 10% compared to that of untreated sawdust with the sawdust media of L. leptoiepis and P. koraiensis, and the mushroom productivity was improved from 12.5% (50.1 g/400 g) to 16.7% (66.7 g/400 g) with the sawdust medium of P. densiflora from first harvest in case of KFRI644. Steam treatment is thought to be a good method for cultivation of cauliflower mushroom by minimizing culturing period and increasing productivity, which is an effective way of utilization for coniferous sawdusts.
