한국 동해안 울진군에 위치한 왕돌초 조하대에서 2012년 5월부터 2013년 8월까지 계절별로 해조상 및 군집구조에 관한 연구를 수행하였다. 연구기간 동안 녹조류 12종, 갈조류 29종, 홍조류 81종을 포함한 122종이 채집 및 동정되었으며, 38종의 해조류는 조사지역에서 연중 출현하였다. 정점별 평균 생물량은 봄 472.19-1,198.77 g wet wt. $m^{-2}$, 여름 68.99-631.14 g wet wt. $m^{-2}$, 가을 200.91-401.20 g wet wt. $m^{-2}$, 겨울 53.61-922.32 g wet wt. $m^{-2}$로 나타났다. 생물량에 근거한 왕돌초 해조류의 수직분포는 상부 개도박(Grateloupia lanceolata), 넓은분홍잎(Acrosorium venulosum), 마디잘록이(Lomentaria catenata), 중부 미역(Undaria pinnatifida), 참가시그물바탕말(Dictyopteris pacifica), 괭생이모자반(Sargassum horneri), 감태(Ecklonia cava), 하부 쇠꼬리산말(Desmaeistia viridis), 감태(E. cava), 괭생이모자반(S. horneri) 순으로 확인되었다. 왕돌초에서 출현한 해조류를 6가지 기능형군에 따라 구분한 결과 사상형 34.43%, 성긴분지형 25.41%, 엽상형 24.59%, 다육질형 9.71%, 각상형 5.74%, 유절산호말형 0.82%로 확인되었다. 해조상의 특성을 반영하여 해조군락 특성을 판단할 수 있는 C/P, R/P, (R+C)/P 값은 각각 0.41, 2.79, 3.21로 나타났다. 또한, 왕돌초 조하대 군집 안정도와 환경상태를 다양도지수(H')와 우점도지수(DI)를 기초로 확인한 결과 안정된 것으로 나타났다. 왕돌초 조하대에 서식하는 해조류 종다양성 및 풍부도를 유지하기 위하여 교란이나 부영양화 같은 인간활동으로부터 보호되어야 할 것으로 판단된다.
본 연구는 제주산 해조류의 생리활성을 대량검색하기 위해 HTS 기법을 이용한 실험과정의 제어 및 관리를 위한 자동화 시스템을 구축하고자 DPPH 라디칼을 이용하여 HTS를 이용한 자동화 시스템을 구축하고 제주산 해조류의 DPPH 라디칼 소거활성을 대량 검색하고자 하였다. 그 결과 HTS를 위한 효율적인 검색방법을 개발하였고 DPPH 라디칼 소거활성이 높은 해조류를 검색하게 되었다. 녹조류의 경우 납작파래 (20G6)의 $20^{\circ}C$ 추출물(1 mg/ml)에서 약 60%이 DPPH 라디칼 소거활성을 보였을 뿐 나머지 해조류에서는 40% 미만이 낮은 활성만을 보였다. 갈조류의 경우 꽈배기 모자반 (20B17), (70B17), 참그물바탕말 (70B1), 큰잎모자반 (70B16)과 감태 (70B26)에서 DPPH 라디칼 소거활성이 우수하였다. 홍조류 중에서는 왜떨기나무붉은실 (20R24)에서 가장 우수한 DPPH 라디칼 소거활성을 나타내었고 갈래잎 (20R17), 굵은참우뭇가사리 (20R18), 부채분홍잎 (20R23) 또한 높은 소거활성을 보였다. 따라서 우수한 효과를 나타내었던 해조류를 대상으로 용매분획 및 활성물질의 분리, 정제를 통해 해조류 유래의 기능성 신소재를 개발한다면, 해조류의 고부가가치화 및 HTS기술의 보편화에 큰 영향을 미칠 것으로 판단된다.
본 연구는 제주도 연안의 해역별 해조류 분포특성을 비교하기 위하여 2018년 5월부터 12월까지 제주도 본섬 4개 정점과 유인도서 2개 정점의 마을어장에서 수행되었다. 해조류는 총 101종으로 녹조류 13종(12.9%), 갈조류 24종(23.8%) 그리고 홍조류 64종(63.4%)이 출현하였다. 계절별로는 5월에 가장 많은 해조류가 출현하고, 10월에 가장 적게 출현하는 전형적인 온대해역의 특징을 나타냈다. 수심별 해조류 출현 종수는 5m와 8m에서는 66종과 65종으로 유사하였으며, 12m에서는 74종으로 최대였다. 해역별로는 도서해역인 우도에서 66종으로 가장 많은 해조류가 출현하였고, 본섬의 동부 표선에서 27종으로 가장 적게 출현하였다. 출현종 가운데 중요도가 높은 해조류는 감태(Ecklonia cava)와 둘레혹산호말(Corallina crassissima)로 각각 21.1%와 20.3%를 나타냈으며, 방황혹산호말(Corallina aberrans) 9.2%, 에페드라게발(Amphiroa ephedraea) 6.2% 그리고 큰열매모자반(Sargassum macrocarpum) 4.4%의 중요도를 나타냈다. 해조류 중 산호말류는 평균 11.2종이 출현하였고, 각 해역의 산호말류 중요도는 평균 32.6%로 우도에서 14.7%로 가장 낮았고, 표선에서 가장 높은 41.0%를 차지하였다. 해조류 군락의 생태학적 평가지수(EEI)는 2.1~10.0으로 표선의 5월과 6월 12m에서 가장 낮았고, 그 밖의 다른 해역에서는 평균 7.3이상으로 좋은 상태를 나타냈다. 본 조사에서 설정한 생태등급기준 I 등급은 우도와 추자도의 수심 12m이었으며, II 등급은 사계와 추자도의 수심 5 m와 8 m가 해당되었다. III 등급은 표선과 귀덕2의 수심 5m와 12m, 평대의 수심 5m와 8m가 해당되었으며, IV 등급은 귀덕2의 수심 8m가 해당되었다.
This study is intended to clarify the effects of heated effluents on intertidal benthic marine algal community in Korea. The species composition and biomass of marine algae at the discharge canal of Gori nuclear power plant on the southeastern coast of Korea were investigated seasonally from February 2001 to October 2006. As a result, 54 species (7 blue-green, 12 green, 9 brown and 26 red algae) of marine algae were found at the discharge canal during the past six years. In general, the number of species observed was abundant during winter to spring and less in autumn. Enteromorpha compressa, E. intestinalis, E. prolifera and Caulacanthus ustulatus were common species found more than 80% frequency during the study period. Seasonal fluctuations of mean biomass were 1-440 g dry wt m–2 and dominant species in biomass were Enteromorpha spp. (contribution to a total biomass proportion 28%), Sargassum horneri (14%) and Amphiroa beauvoisii (14%). It is evident from the floristic composition and biomass data that unique micro-environment of the discharge canal support different communities from those on the intake or control area. Results from the large numbers of surveys before and during plant operation showed that, in the regions influenced by thermal effluents such as the discharge canal of power plants, the process of ecological succession has been proceeded. It is assumed that the uni-directional water flow and the time of overhaul largely affect the development and succession of benthic marine algal communities of the discharge canal.
The present investigations were carried out with the purpose of making clear the fundamental features of the seasonal variations in chemical composition of the brown algae. Three species of samples, Ecklonia cava, Sargassum sagamianum and Hizikia fusiforme have been analyzed monthly for their contents of total ash, crude protein, alginic acid, mannitol, and laminarin over a year period. Three kinds of samples were collected from the same locality, situated on the southern coast of the Che-ju Island, from September 1966 to August 1967. In addition, the comparative analysis was made on fronds and stipes of the plant for their chemical composition. The results obtained are summarized as follows: 1, In general, the three species examined underwent a similar mode of seasonal variation, and no essential difference was detected among them. 2. The chemical composition of the plant exhibited a considerable difference between the species. The content of total ash in H. fusiforme was remarkably higher than those in the two other species respectively. The alginic acid content was relatively high in S. sagamianum and low in H. fusiforme. The contents of crude protein, mannitol and laminarin were appreciably high in E. cava and low in H. fusiforme in general. 3. The most outstanding feature in the seasonal variation was that, in general, the total ash, crude protein, and alginic acid contents were at a maximum in the winter months while laminarin and mannitol contents were at a minimum. The converse was true in summer. Total ash-Maximum values were observed from December to February and minimum from August till October. Crude Protein-All species exhibited maxima in January and February, and minima from August to October. Alginic acid-Maximum contents occured from January to March and minimum from September to November. Mannitol-The maximum content of mannitol was In May and lune and minimum in January, February and March. Leminerin-Maximum content was in September and October, and minimum in January and February. 4. The wide seasonal variation in chemical constitution occured in the fronds, but the stipes showed a slight seasonal variation. In the chemical composition, the stipe was high in alginic acid, low in mannitol and laminarin. The reverse was in the frond.
This study examined the characteristics and seasonal distribution patterns of subtidal marine algal communities along the east coast of Korea, from 2016 to 2017, and recorded the coverage ratio (%) by the algal type. During the study period, 134 marine algae species (16 green, 29 brown, and 89 red) were identified at the survey sites. The maximum number of species was observed at the Nagok site (70 species), and the minimum at the Gisamoon site (50 species). The total average biomass (dry weight) during the survey period was 66.22 g/m2, including green algae (2.19 g/m2), brown algae (38.10 g/m2), and red algae (25.94 g/m2). The dominant seaweeds in the vertical distribution based on coverage rates (%) were Sargassum yezoense and Dictyopteris divaricata at the upper, Symphyocladia latiuscula and Undaria pinnatifida at the middle, and Agarum clathratum subsp. yakishiriense and Plocamium telfairiae at the lower sections of the distribution. Richness, evenness, and diversity index, calculated based on the biomass of the abundant species, were estimated to be 6.36, 0.55, and 1.98, respectively, over the entire sea area. Based on the evaluation of the environmental states using community indices, the ecological evaluation index (EEI-c) of subtidal marine algal communities along the east coast of Korea was marked as 'good-moderate'.
Experimental design for ecological restoration approach was investigated from the P' company's slag dumping area with higher pH value. The degree of pH recovery was measured by the injection of carbon dioxide from the LNG exhaust gas, and the residual carbon, for example, calcium and carbonate which can be controlled by artificial seaweed beds. The degree of adaptability from 3 algaes (Undaria pinnatifida, Sargassum horneri and Ecklonia stolonifera) and uptake nutrient function of Ecklonia stolonifera chosen in the first treatment were measured in the laboratory to determine the transplanting algae in artificial seaweed beds. The higher value of pH was decreased to 7.0~8.5 by injection of LNG exhaust gas with flow rate $20\;m^3/min$. In the experiment design at laboratory, the upper part of frond of Undaria pinnatifida and Sargassum horneri began to decay, and the color changed after 10 days. However, those of Ecklonia stolonifera were after 14~20 days. The uptake rate of nutrient from Ecklonia stolonifera was higher than those of other algaes, and those was similar pattern in the control (e.g. seawater); The DIN concentration uptake of Ecklonia stolonifera was $1.88\;{\mu}g/L/hr$ avg. in leachate, but $2.19\;{\mu}g/L/hr$ avg. in seawater. However, the ${PO_4}^3$-P concentration uptake was $0.18\;{\mu}g/L/hr$ avg. in leachate, but $0.31\;{\mu}g/L/hr$ avg. in seawater. These result indicated Ecklonia stolonifera uptaked these nutrients in the leachate as well as in seawater, and it may suggested for this species to transplant for constructing artificial seaweed beds.
The agar-degrading bacterium GNUM-1 was isolated from the brown algal species Sargassum serratifolium, which was obtained from the West Sea of Korea, by using the selective artificial seawater agar plate. The cells were Gram-negative, $0.5-0.6{\mu}m$ wide and $2.0-2.5{\mu}m$ long curved rods with a single polar flagellum, forming nonpigmented, circular, smooth colonies. Cells grew at $20^{\circ}C-37^{\circ}C$, between pH 5.0 and 9.0, and at 1-10% (w/v) NaCl. The DNA G+C content of the GNUM-1 strain was 45.5 mol%. The 16S rRNA sequence of the GNUM-1 was very similar to those of Alteromonas stellipolaris LMG 21861 (99.86% sequence homology) and Alteromonas addita $R10SW13^T$(99.64% sequence homology), which led us to assign it to the genus Alteromonas. It showed positive activities for agarase, amylase, gelatinase, alkaline phosphatase, esterase (C8), lipase (C14), leucine arylamidase, valine arylamidase, ${\alpha}$-chymotrypsin, acid phosphatase, naphthol-AS-BI-phosphohydrolase, ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\beta}$-glucosidase, catalase, and urease. It can utilize citrate, malic acid, and trisodium citrate. The major fatty acids were summed feature 3 (21.5%, comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH) and C16:0 (15.04%). The 30 kDa protein with agarase activity was identified by zymography, and this report serves as the very first account of such a protein in the genus Alteromonas.
This paper examines relation between growth of adult top shell and their selectivity over different marine algae. For the study the top shell density and the algal species in different depth were surveyed by SCUBA. To test selectivity over different types of algae, five algal species including Ecklonia cava, Undaria pinnatifida, Ulva spp, Gelidium amansii and Sargassum spp were provided to top shells in indoor water tank condition. In the field survey, the wet weight of Ecklonia cava was significantly (r = 0.612, p < 0.05) correlated to the shell length (SL) of top shell. In the indoor water tank experiment conducted over 12 hours after sunrise, adult top shell (SL > SL 4.5 cm) tended to select Ecklonia cava and Undaria pinnatifida, which are common and abundant algal in the subtidal waters in Jeju Island. Our field survey and the indoor experiment suggest that E. cava and U. pinnatifida could be food preferred by adult top shell in its habitat.
An intertidal marine benthic algal vegetation and vertical distribution at Kangnung and it’s vicinity, the eastern coast of Korea was investigated to clarify the community structure and vertical distribution by quadrat method. Marine algae identified from the area were 116 species; 15 green, 31 brown and 70 red algae. The dominant species were Ulva pertusa, Codium fragile, Undaria pinnatifida, Dictyota dichotoma, Gelidium amansii, Corallina pilulifera, Carpopeltis affinis, Grateloupia filicina, Pachymeniopsis elliptica and Chondrus ocellatus at study sites. The vertical distribution of intertidal marine algae was divided into three distinct zones. They were characterized by Porphyra tenera at the upper, Enteromorpha spp. and Ulva pertusa at the middle, and Sargassum thunbergii, Hizikia fusiformis and Pachymeniopsis elliptica at the lower zones, respectively. The vertical distribution of subtidal zone algae were characterized by Laminaria spp. at 15m depth and Kjellmaniella crassifolia at 15 to 25 m depths, whereas Agarum cribrosum was dominant at the lower zone than 25 m depth.
