Low-dose Radiation Induces Antitumor Effects and Erythrocyte System Hormesis

  • Objective: Low dose radiation may stimulate the growth and development of animals, increase life span, enhance fertility, and downgrade the incidence of tumor occurrence.The aim of this study was to investigate the antitumor effect and hormesis in an erythrocyte system induced by low-dose radiation. Methods: Kunming strain male mice were subcutaneously implanted with S180 sarcoma cells in the right inguen as an experimental in situ animal model. Six hours before implantation, the mice were given 75mGy whole body X-ray radiation. Tumor growth was observed 5 days later, and the tumor volume was calculated every other day. Fifteen days later, all mice were killed to measure the tumor weight, and to observe necrotic areas and tumor-infiltration-lymphoreticular cells (TILs). At the same time, erythrocyte immune function and the level of 2,3-diphosphoglyceric acid (2,3-DPG) were determined. Immunohistochemical staining was used to detect the expression of EPO and VEGFR of tumor tissues. Results: The mice pre-exposed to low dose radiation had a lower tumor formation rate than those without low dose radiation (P < 0.05). The tumor growth slowed down significantly in mice pre-exposed to low dose radiation; the average tumor weight in mice pre-exposed to low dose radiation was lighter too (P < 0.05). The tumor necrosis areas were larger and TILs were more in the radiation group than those of the group without radiation. The erythrocyte immune function, the level of 2,3-DPG in the low dose radiation group were higher than those of the group without radiation (P < 0.05). After irradiation the expression of EPO of tumor tissues in LDR group decreased with time. LDR-24h, LDR-48h and LDR-72h groups were all statistically significantly different from sham-irradiation group. The expression of VEGFR also decreased, and LDR-24h group was the lowest (P < 0.05). Conclusion: Low dose radiation could markedly increase the anti-tumor ability of the organism and improve the erythrocyte immune function and the ability of carrying $O_2$. Low-dose total body irradiation, within a certain period of time, can decrease the expression of hypoxia factor EPO and VEGFR, which may improve the situation of tumor hypoxia and radiosensitivity of tumor itself.

행인 제제의 독성 및 항암효과의 자연살해세포 활성에 미치는 효과 (Study on toxicity, anti-cancer and NK cell activity of Lateril oil)

  • In this experiment, we examined the the effect of Laetrile Oil to the immune system and tried to disclose the anti-cancer mechanism of this. The result is listed as below. 1. After per-os, sub-cutaneous and into the peritoneal injection of Laetrile Oil to the SPF mice, the LD50 is above 5000mg/kg at even group. 2. Mean survival days of mice treated with laetril oil, after S-180 cells transplantation into the peritoneal cavity decreases 1.5 days(-6.8%) compared with the Mean survival days of the control group(22days.) 3. Effects of laetril oil on natural killer cell activity at Effector/Target Cell Ratio with 100:1, 50:1, 10:1 into methotrexate-pretreated mice is like this.: Compared with $29.22\pm12.7\%$ Cytotoxicity of the control group, sample group's Cytotoxicity had $38.83{\pm}12.5%$ of meaningful acrease. At 100:1 Effecr/Target Cell Ratio. At 50:1 Effector/Target Cell Ratio, control group has $20.02{\pm}9.6%$ Cytotoxiciy and sample group had $31.53{\pm}13.4%$ Cytotoxicity. At 25:1 Effector/Target Cell Ratio control group has $13.60{\pm}6.6%$ Cytotoxicity and sample group had $20.81{\pm}9.8%$ Cytotoxicity According to the above results, the Laetrile Oil represents nontotoxic to a SPF mice. non-effective to transplantable Sarcoma 180 tumors, and activaion in NK cell activity.

영지버섯 생장점 단백다당체 GLB의 대식세포 활성화 효과 (Activation of Macrophages by GLB, a Protein-polysaccharide of the Growing Tips of Ganoderma Lucidum)

  • In the previous study we described the antitumor activity of GLB, a protein-polysaccharide fraction of the growing tips of Ganoderma lucidum, against sarcoma 180 solid tu mor in ICR mice. In this study we investigated the stimulatory activity of GLB on macrophages. When analyzed using a flow cytometer, GLB ($100{\mu}g/ml$) was found to increase the phagocytic activity of the BALB/c mouse peritoneal macrophages as well as chicken macrophage BM2CL cells against FITC-labeled C.albicans by 55.2% and 21.2%, respectively. GLB also increased the spreading and the expression of MHC class II molecules of BM2CL cells as well as the mouse peritoneal macrophages. From these results, it is clear that GLB is a strong stimulator to the macrophages.

죽염의 약리작용 평가 (Pharmacological Evaluation of Bamboo Salt)

  • Bamboo salt has been used for the purpose of precaution and treatment of certain diseases including cancer. Therefore, present study was carried out to ascertain the effects of bamboo salt upon anti-cancer, anti-hypertensive, and anti-diabetic activities as well. To examine the anti-cancer activity of bamboo salt, ICR mice implanted with 1$\times$l0$^{6}$ cells of sarcoma 180 intraperitoneally had been treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 60 days using adriamycin (2 mg/kg) as a positive control. Neither survival rate nor body weight had been significantly influenced by all the treatments indicating that bamboo salt A did not exert the anti-cancer effect on ICR mice. Anti-hypertensive activity was examined in spontaneously hypertensive rats (SHR) which had been administered with bamboo salt A, crude salt, and reagent-grade NaCl (0.1, 0.5, and 1.0% in drinking water) for 28 days using hydralazin (2 mg/kg) as a positive control. Blood pressure and heart rate were measured at 1, 3, and 4 weeks after the starting date. Significant anti-hypertensive activity was not observed in any treated group compared to the positive control group. In order to determine if bamboo salt had anti-diabetic activity, rats in which diabetes had been induced by streptozotocin (45 mg/kg, i.m.) were treated daily with bamboo salt A, crude salt, and reagent-grade NaCl (0.2, 1.0, and 2.0 g/kg, p.o.) for 28 days using insulin (50 U/kg, s.c..) as a positive control. Blood samples were taken and analyzed at 1,2, and 4 weeks after the starting date. Bamboo salt did not cause any decreasing effect on the blood glucose levels. These results clearly demonstrated that bamboo salt A did not exert anti-cancer, anti-hypertensive, or anti-diabetic activities in the present experimental animals.

프로폴리스 물추출물의 약효연구 (Studies on the Efficacies of Water Extract of Propolis)

  • Propolis is a lipophilic, natural product prepared by mixing the exudates collected from various plants by honeybees with beeswax for the purpose of using to seal hive walls and to strengthen the borders of combs. Because of its versatile bioactivities, propolis has been attracting many investigators'interest. But the pharmacological studies on propolis has, to date, been exclusively performed for an alcohol extract, there is few information of water extract. Therefore, in this study, we investigated the various effects of water extract of Chinese propolis. The water extract of propolis and its fractions of organic solvents showed strong antioxidative activities, especially ether and ethylacetate fractions, and reduced the lipid peroxidation of rat liver in viro. Additionally the ether fraction of propolis (10 mg/ml) inhibited the activity of hyaluronidase by 50%. In vivo, the water extract of propolis considerably decreased s-GOT, s-GPT and s-LDH activities which represent for the hepatotoxicity induced by $CCl_4$ in rats, and prolonged the MST (Medium revival tinge) and ILS (Increasing in MST over control) by 18% in mice which inoculated with sarcoma 180 ascites cells. These results suggest that the water extract of propolis has various bioactivities as well as the alcohol extract.

리포포리사카라이드의 세포성 면역반응에 미치는 스와인소닌의 영향 (Effects of Swainsonine on the Cell-mediated Immune Responses of Lipopolysaccharide)

  • Effects of swainsonine (SW: 8${\alpha}$, ${\beta}$-indolizidine-1alpha, 2${\alpha}$, 8${\beta}$-triol from Locoweed) on the cellular and nonspecific immune responses of lipopolysaccharide (LPS) wer e studied in ICR mice. Mice were divided into 4 groups (10mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7mg/kg of SW by i.p. injection twice a week for 14 days at a dose of 2mg/kg. Immune responses of the delayed-type hypersensitivity response (DTH) to sheep red blood cells (s-RBC), phagocytic activity and natural killer (NK) cell activity were evaluated. LPS treatment didn`t affect NK cell activity, phagocytic activity, DTH to s-RBC compared with those in controls, and phagocytic activity of sareoma 180 tumor bearing mice. However, circulating leukocytes were significantly decreased. Combinaton of LPS and SW increased circulating leukocytes significantly compared vath that in LPS alone, and DTH to s-RBC, NK cell activity and phagocytic activities of normal and sarcoma tumor bearing mice were not affected. These findings indicate that SW didn`t affected the cellular immune responses suppressed by LPS but significantly increased circulating leukocytes.

더덕 추출물의 항돌연변이 및 항종양 효과 (Antimutagenic and Antitumor Effects of Codonopsis lanceolata Extracts)

  • 본 연구는 더덕의 돌연변이원성, 항돌연변이원성, 세포독성, 항종양 효과를 조사하기 위해서 수행되었다. 더덕을 70% 에탄올로 추출하여 추출용매에 따라 핵산, 클로로포름, 에틸아세테이트, 부탄올과 물 층으로 분획하였다. Ames test assay, SRB assay와 sarcoma-180 세포를 이용한 항종양 실험을 실시하였다. Ames test 결과, 더덕 에탄올 추출물은 돌연변이원성을 나타내지 않았다. 더덕 에틸아세테이트분획물은(200 ${\mu}g$/plate) 4NQO에 대하여 S. Typhimurium TA98과 TA100에서 각각 72.1% 및 67.0%의 억제율을 나타내었으며, MNNG에 대한 S. Typhimurium TA100은 69.6%의 억제율을 나타내었다. 더덕 추출물 및 분획물의 암세포성장 억제효과를 살펴보기 위해 인간 자궁경부암세포(HeLa), 인간 간암세포(HepG2), 인간 유방암세포(MCF-7), 인간 폐암세포(A549) 및 인간 신장정상세포(293)를 사용하였다. 더덕 에틸아세테이트 분획물을 1 mg/mL의 농도로 처리하였을 때 각각 74.5%(HeLa), 70.7%(MCF-7) 및 80.3%(A549)의 가장 높은 억제활성을 나타내었다. 반면에 인간 정상 신장세포(293)에서는 2$\sim$31%의 세포독성을 나타내었다. In vivo에서 더덕 추출물 및 분획물의 항암 효과를 검토하기 위하여 Balb/c 마우스에 sarcoma-180 종양세포로 고형암을 유발시켰다. 그 결과 더덕 에틸아세테이트 분획물의 최고농도 50 mg/kg에서 56.4%의 고형암 성장 억제 효과를 나타내었고, 이는 다른 추출물 및 분획물 중에서 가장 높은 억제율이었다.

왕겨에 의한 신령버섯균사체 액체배양액의 생쥐 항복수암성 증가 (Enhancement of Anticarcinogenic Potentials of Submerged-Liquid Culture of Agaricus blazei Murill on Mouse Ascites Cancer by Rice Hull)

  • 왕겨가 Agaricus blazei Murill (AB: 신령버섯)균사체 액체배양 추출물의 항암성을 증가시키는지에 관한 연구를 수행하였다. AB균사체를 대두박을 기본으로 한 액체배지에 다양한 조건으로 배양하여, $\beta$-glucan 함량, Brix, 균사체를 측정하여 적정 생육조건을 선정하고, 이들의 열수추출물의 S-180 cell로 유도한 mouse 복수암에 대한 항암성을 조사하였다. AB균사체는 $25^{\circ}C$에서 5일간 배양하였을 때 최적 생육을 나타내었고, 이 배양물이 다른 조건에서 배양한 배양물보다 우수한 항암성을 나타내었다. AB균사체의 생육 및 항암성은 변온배양에 따른 효과는 없었다. 따라서 이 최적배양조건($25^{\circ}C$, 5일 배양)에서 AB균사체를 1% 왕겨분말이 함유된 액체배지에 배양하고, 이의 열수 추출물의 항암성을 검증하였다. 1% 왕겨가 함유된 액체배지에서 배양한 열수추출물의 항암성은 왕겨가 함유되지 않은 배지의 열수추출물보다 항암성이 유의성 있게 증가되었다(p<0.05). 왕겨의 첨가는 AB균사체의 생육을 오히려 촉진시켰다. 이 결과는 왕겨가 AB균사체 뿐 만 아니라 다른 버섯균사체 액체배양물의 항암성 증진을 위한 원료로 활용될 수 있을 것임을 의미한다.

영지와 잔나비걸상버섯의 원형질체 융합균주의 항암 성분에 관한 연구 (Studies on Antitumor Components of the Cultured Mycelia of Interspecific Protoplast Fusant F-2 of Ganoderma lucidum and Ganoderma applanatum)

  • 영지버섯 Ganoderma lucidum과 잔나비걸상버섯 Ganoderna applanatum의 원형질체 융합균주 5개에 대하여 항암실험을 실시하여 그 중에서 다른 것에 비해 항암력이 높은 융합균주 F-2를 선발하였다. 그 항암성분을 조사하기 위하여 융합체의 균사를 액내 배양하고 열수추출물로부터 얻은 단백 다당체를 DEAE cellulose ion exchange column chromatography와 Sepharose CL-4B gel filtration을 이용하여 5가지 분획(Fr. I-V)으로 분리, 정제하여 모균주들과의 차이점을 비교, 분석하였다. 모균주와 분리한 각 성분들을 20mg/kg/day 용량으로 마우스의 복강에 투여하였을 때 sarcoma 180 고형암에 대하여 균주보다 융합체 성분(Fr. ll)의 종양억제율이 최고 1.5배로 증가되었다. 그리고 마우스의 면역에 영향을 미치는 항암성분을 연구한 결과, 대조군에 비해 활성화된 대식세포에서 분비되는 superoxide anion의 양을 1.2배, 비장세포중의 용혈반 형성 세포수를 4.3배로 증가시켰다. 화학분석에 의하면 이 성분은 glucose, galactose, mannose, fucose와 xylose로 구성된 다당류가 85.2%이며 15종의 아미노산으로 구성된 단백질이 0.39%이었고, hexosamine이 0.39%로 구성되었으며 분자량은 $5.6{\times}10^4$ dalton이었다.

생감자의 열탕 추출물을 첨가한 감자 김치의 이화학적 특성 및 In Vivo에서의 항암 효과 (Physico-chemical Properties and In Vivo Anti-cancer Effects of Potato Kimchi Prepared by adding Hot Water Extracts of Potato)

  • In this study, potato kimchi was prepared by applying heat to raw potatoes, and then the physico-chemical properties and anti-cancer effects of the kimchi were analyzed. The texture results indicated the potato kimchi had very good hardness and springiness attributes. During th late storage period, total vitamin C content of the kimchi slowly increased. In addition, the potato kimchi had non-volatile organic acid changes that promoted early aging; however, after the complete aging period, it was comparatively similar to other types of kimchi. Using the methanol extracts of various kimchi samples, the potato kimchi(solid 100%) showed the highest anti-carcinogenic effects in terms of anti-tumor activity in tumor bearing Balb/c mice with sarcoma-180 cells. In addition, the effects of the methanol extracts on hepatic glutathione S-transferase content were $289.76\;{\mu}mol/mg$ protein/min, $250.97\;{\mu}mol/mg$ protein/min, $251.20\;{\mu}mol/mg$ protein/min, $219.53\;{\mu}mol/mg$ protein/min, $183.79\;{\mu}mol/mg$ protein/min, for control kimchi, mul kimchi, and two potato kimchis [(solid 100%) and(solid 60%+kimchi juice 40%)], respectively. The in vivo anti-cancer effects of the potato kimchi were investigated using AGS human gastric adenocarcionoma cells and HT-29 human colon adenocarcionoma cells. Overall, an MTT assay revealed that the methanol extract of the potato kimchi showed the highest anti-carcinogenic effects.
