Effects of Sap Collection on Trees Growth in Acer mono og Mt. Jiri (지리산(智異山) 고로쇠나무의 수액채취(樹液採取)가 임목생육(林木生育)에 미치는 영향(影響))

  • Closing processes of taping holes and diameter growth of taped trees of Acer mono were investigated to understand the effects of sap taping and number of tapping holes on tree growth and closing speed of holes. Closing processes of tapping holes were faster at trees taped of ten holes than trees tapped of one, three, five holes during the study period, and tapped holes were completely filed up within 15 months. Diameter growth by number of tapped hole were 0.54 mm for control trees, 0.60 mm for trees taped of one hole, 0.64 mm for trees taped of three holes, 0.33 mm for tapped of five holes and 0.31 mm for trees taped of ten holes, respectively. Closing processes of tapping holes by fusion materials was fast in the order of DB ointment, stirofoam, control and bamboo vinegar. Especially, tapped holes in the case of DB ointment was completely healed within 4 months. Therefore, DB ointment should be used to minimize damage on tree growth and secure resources for sap collection of A. mono.

Synergistic Inhibition by Bacteriocin and Bacteriophage against Staphylococcus aureus

  • Staphylococcus aureus is a representative pathogenic bacterium carefully controlled in the dairy industry because it causes bovine mastitis and thus, can enter the dairy chain. Furthermore, the emergence of multi-drug resistant S. aureus is a big problem. We previously isolated a Lactococcus lactis strain producing a bacteriocin that exhibited strong antimicrobial activity against S. aureus. In this study, we investigated the synergistic inhibition of S. aureus by the bacteriocin and a bacteriophage (SAP84) which is specific to the organism. The bacteriocin (12.5-100 AU/mL) inhibited the growth of S. aureus KCTC 3881 in a dose-dependent manner, as did the bacteriophage SAP84 (0.001-1 MOI; multiplicity of infection). Co-treatment with the bacteriocin (100 AU/mL) and the bacteriophage (0.1 MOI) significantly inhibited the growth of S. aureus compared to each treatment alone (bacteriocin or bacteriophage), indicating the two components showed synergistic inhibition of S. aureus. Therefore, the bacteriocin and bacteriophage combination can be used as a good strategy for controlling pathogenic bacteria.

  • Methods to measure and estimate transpiration of a forest composed of evergreen broad-leaved trees (Pasania edulis Makino) are studied. Heat pulse velocity has been measured along with soil moisture and micrometeorological factors at the Fukuoka Experimental Forest, the Research Institute of Kyushu University Forests in Fukuoka, Japan (33$^{\circ}$38'N, 130$^{\circ}$31'E, alt. 75m). Tree cutting measurement was conducted to convert the heat pulse velocity into sap flow and transpiration. A big leaf model to calculate transpiration and Interception loss is examined and the estimated values are compared with the measured values obtained from the heat pulse measurement. The results show that 1) Pasania edulis Makino posessing radial pore structure had relatively high water content and high heat pulse velocity even within the central part of the stem near the pith, 2) the heat pulse velocity was well correspond to the water uptake in the tree cutting measurement, 3) the estimation of sap flow based on the heat pulse velocity is accurate, and 4) the big leaf model using the parameters obtained from measurement of a portable photosynthesis system in one day in summer gives reasonable estimation of transpiration independent of seasons and weather.

Genome editing of hybrid poplar (Populus alba × P. glandulosa) protoplasts using Cas9/gRNA ribonucleoprotein (현사시나무 원형질체에서 리보핵산단백질을 활용한 유전자 교정 방법 연구)

  • Targeted genome editing using the CRISPR/Cas9 system is a ground-breaking technology that is being widely used to produce plants with useful traits. However, for woody plants, only a few successful attempts have been reported. These successes have used Agrobacterium-mediated transformation, which has been reported to be very efficient at producing genetically modified trees. Nonetheless, there are unresolved problems with plasmid sequences that remain in the plant genome. In this study, we demonstrated a DNA-free genome editing technique in which purified CRISPR/Cas9 ribonucleoproteins (RNPs) are delivered directly to the protoplasts of a hybrid poplar (Populus alba × Populus glandulosa). We designed three single-guide RNAs (sgRNAs) to target the stress-associated protein 1 gene (PagSAP1) in the hybrid poplar. Deep sequencing results showed that pre-assembled RNPs had a more efficient target mutagenesis insertion and deletion (indel) frequency than did non-assembled RNPs. Moreover, the RNP of sgRNA3 had a significantly higher editing efficacy than those of sgRNA1 and sgRNA2. Our results suggest that the CRISPR/Cas9 ribonucleoprotein-mediated transfection approach is useful for the production of transgene-free genome-edited tree plants.

Optimum Tapping Size and Number for Sap Collection of Acer mono (고로쇠나무 수액 채취를 위한 적정 천공 크기와 천공 수)

  • Optimum tapping size and number for sap collection were investigated to minimize damage on tree growth and secure resources for sap by sap collection of Acer mono. The amounts of sap collected by tapping size of 12 mm, 10 mm and 8 mm was 189 L, 140 L and 193 L, respectively. Fusion rates by tapping size were 100% for 8 mm, 89% for 10 mm and 85% for 12 mm, respectively. Amount of sap by size and number of tapping were much at three, two and three tappings to small, middle and large diameter tree in case of 8 mm tapping, and it was large in quantity at two tappings of 12 mm tapping to large diameter tree, respectively. Trees tapped from one to three tappings of 8 mm size for small diameter tree with treatment of DB ointment (mixture of thiophane ointment and 2% bamboo charcoal powder) were completely filled up within 6 months. Diameter growth by number of tapping of 8 mm size was 0.60 mm for one tapping, 1.12 mm for two tappings and 0.47 mm for three tappings to small, middle and large diameter tree, respectively. In case of 12 mm tapping size, diameter growth was fast in the order of large (0.55 mm), middle (0.30 mm) and small (0.23 mm) diameter tree, respectively.

Quality Characteristics of the White Birch Sap with Varying Collection Periods (자작나무수액의 유출시기별 품질특성)

  • This study evaluated the quality characteristics of white birch (Betula platyphylla var. japonica) sap with different collection times. The changes of browning index, turbidity, pH, total acidity, organic acid, free sugar, crude protein, crude ash, and mineral content were investigated. The browning index and turbidity increased from 0.076 to 0.222 and from 0.048 to 0.138, respectively, with increasing collection time. The pH decreased from 6.09 to 4.72, while total acidity increased with increasing collection time. Citric and malic acids were detected and malic acid increased with increasing collection time. Glucose and fructose as free sugars were detected and their contents were 0.364~0.433% and 0.497~0.664%, respectively. Crude protein and crude ash contents remarkably increased from 3.40 to 32.37 mg% and from 0.01% to 0.04%, respectively, with increasing collection time. Cu, Fe, Ca, Mg, Mn, and K were detected, and increased with increasing collection time. Particularly, K increased remarkably from 5.25 to 37.27 mg/L over time. These results indicate that the optimum processing method to improve the quality of white birch sap is necessary, because the quality of sap decreased as collection time increased, but nutritional value increased.

Native Environment and Growth Characteristics of Dendropanax morbifera Lev. In Korea

  • This study was carried out to obtain basic envirinmental information of the native area and growth characteristics at different age of Dendropanax morbifera LEV. In the southern Korea. The Dendropanax morbifera LEV. Was grown naturally at Wando, Cheju(Mt. Halla), Yecheon(Kermoon island),Sinan(Hong isand), Jindo(Mt. Chermchill) and Haenam(Mt. Taeroon), all between 30 to 450m above the sea level. The soil characterisitics of the native area had a pH of 5.3 to 5.4 with 8.4 to 9.8% content of organic matter and 18.2 to 21.3% soil moisture. Flowering starting in the middle of July begun when the trees were 6 years old. Six year old trees are 129 cm in stem height, 34 mm in stem diameter,15.4 in number of leaves with petioles 12.9 cm long. Xylem sap can be obtained from over 10 year old trees that grew well and have over 10 cm stems in diameter.

Estimation of Soil Loss into Sap-Gyo Reservoir Watershed using GIS and RUSLE (GIS와 RUSLE 기법을 이용한 삽교호유역의 토사 유실량 산정)

  • Prediction of exact soil loss yield has as important engineering meaning as prediction of exact flow measurement in a stream. The quantity of soil loss in a stream should be considered in planning and management of water resources and water quality such as design and maintenace of hydraulic structures : dams, weirs and seawalls, channel improvement, channel stabilization, flood control, design and operation of reservoirs and design of harbors. In this study, the soil loss of Sap-gyo reservoir watershed is simulated and estimated by RUSLE model which is generally used in the estimation of soil loss. The parameters of RUSLE model are selected and estimated using slope map, landuse map and soil map by GIS. These parameters are applied to RUSLE's estimating program. And soil loss under probability rainfall in different frequencies are estimated by recent 30 years of rainfall data of Sap-gyo reservoir watershed.

  • The purpose of this research is to suggest economic feasibility of cultivating Acer Mono by using profitability analysis to forest owners. To achieve this research objective, forest owner household survey with intensive cultivating has been conducted about a sap production on its age of tree. And input costs and sap production costs are calculated with silvicultural system from plant to regeneration cutting. Total income is the sum of its sap sales and thinning and regeneration cutting. The method of profitability analysis was used on the NPV(Net Present Value) and IRR(internal rate of return). Finally, when 3% discount rate is applied, NPV is about 59,436 thousand won and IRR value is 9.22% at this point. This result, therefore, proves that cultivating Acer Mono is economical feasibility to forest owners.

Plant Regeneration Depending on Explant Type in Chrysanthemum coronarium L.

  • Plant regeneration depending on explant type was inves-tigated with cotyledon, hypocotyl, and leaf explants of garland chrysanthemum (Chrysanthemum coronarium L.) cultured on MS basal medium supplemented with various concentrations of SAP and NAA combination. Among the three different types of explants, hypocotyl explants grown on MS medium containing $1.0{\mu}M\;NAA,\;1.0{\mu}M\;BA\;and\;6{\mu}M\;AgNO_3$ produced the highest adventitious shoots (4.67 per explant). Hypocotyl explants not only produced more vigorous shoots, which regenerated aster than the cotyledon and leaf explants. An efficient root formation was observed in MS medium containing $3\%$sucrose. The concentration of NAA did not show significant effects on root formation. Results from this experiment suggested that hypocotyl explants were efficient for the regeneration of garland chrysanthemum.
