오이풀 첨가 쿠키의 특성연구 (The Quality Characteristics of Cookies containing of Powder Sanguisorba officinalis L.)

  • This study was to investigate the physiological activity of cookies with Sanguisorba officinalis L. powder. Our results showed that the pH of the cookies ranged from 6.34 to 6.29, and the density ranged from 4.12 to 4.40. Increasing the amount of Sanguisorba officinalis L. powder in the cookies decreased the spread factor of the cookies and tended to decrease lightness (L) in the Hunter color value with yellowness (b) and redness (a) increased. For the textural characteristics, the addition of Sanguisorba officinalis L. powder increased hardness. The sensory evaluation showed that the cookies with Sanguisorba officinalis L. powder were preferred more than those without in color, odor, texture, taste and overall preference. Based on the results above, the cookies that contained 3% of Sanguisorba officinalis L. powder presented the best quality.

지유 분획의 Candida albicans에 대한 항균효과 (Antifungal Effect of Sanguisorba officinalis L. fractions on Candida albicans)

  • We have studied the antifungal effect of 19 medicinal plants with paper disc diffusion method against candida albicans. As a result, Sanguisorba officinalis L., Cinnamomum cassia, Rheum coreanum, Perilla frutescens and Eugenia caryophyllata have been found to be effective against C. albicans. Among these, Sanguisorba officinalis L. was most effective at 24 hours and 48 hours. Its clear zone diameter was 17 mm for 24 hours and 16 mm for 48 hours. The antifungal activity of the solvent fraction of Sanguisorba officinalis L. by n-hexane, chloroform, ethyl acetate and n-butanol was the best for the chloroform fraction of 28 mm for 24 hours and 18 mm for 48 hours. The MIC concentration of the chloroform fraction was $80{\mu}g/50{\mu}l$ at 24 hours and $240{\mu}g/50{\mu}l$ at 48 hours.

지유(地楡)의 피부 노화(老化)에 대한 연구(硏究) (Study on Anti-Skin Aging Effect of Sanguisorba officinalis L.)

  • To develop a new anti-skin aging cosmetics or functional foods by using antioxidative activity and collagenase inhibitor, a potent collagenase or elastase inhibitor was screened from various extracts of medicinal plants and its optimal extraction condition was investigated. And antioxidative activity, antimicrobial activity and inhibitory of effect against collagenase activity were investigated. In the these results, we selected the Sanguisorba (Sanguisorba officinalis L.) that presents a potential biological activities. Sanguisorba which is very rich in triterpenoid saponin and tannins was recently reported its anti-oxidant activities and phytoestogenic activities in vivo test and many clinical studies. The experiments were carried out in vitro to determine anti-oxidant activities of Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, Elastase and collagenase activity assay. It show that the Sanguisorba extracts have the most significant anti-oxidant on free radical scavenging activity assay, and also inhibited significantly activities of elastase, collagenase. Further, Sanguisorba extracts are activated Type I collagen protein expression in CCD-986sk cells. These result suggest that the Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, elastase and collagenase activity assay, Type I collagen protein expression in CCD-986sk cells effected could be developed cosmetic ingredients for anti-aging.

당뇨를 유도한 동물모델에서 지유 추출물의 항산화 활성 (Antioxidative Activities of Sanguisorba officinalis L. in Diabetic Rats)

  • Background: Sanguisorba officinalis has been used in traditional Asian medicine owing to its beneficial effects on various diseases. The purpose of this study was to evaluate the effect of S. officinalis on the antioxidant system of Streptozotocin (STZ) and Alloxan (ALL) induced diabetic rats. Methods and Results: Triglyceride and Low-Density Lipoprotein (LDL)-cholesterol levels decreased in the STZ-induced diabetic groups treated with S. officinalis extract (SOE) compared to the corresponding levels in the control groups. Moreover, in the ALL-induced diabetic groups, SOE reduced triglyceride, LDL-cholesterol, and High-Density Lipoprotein (HDL)-cholesterol levels. Malondialdehyde (MDA) levels decreased significantly in the STZ and ALL-induced groups treated with SOE compared to the corresponding levels in the control group. Further, Glutathione (GSH) levels increased but did not reach statistical significance. The levels of Superoxide Dismutase (SOD) and Glutathione-S-Transferase (GST) showed a tendency to recover with SOE treatment in the STZ and ALL-induced diabetic groups. In addition, Catalase (CAT) levels in the SOE treatment group decreased significantly compared to those in the control group. Conclusions: These results suggest that SOE might be an effective agent in attenuating oxidative stress in diabetic patients by improving blood lipid profiles and inducing the anti-oxidative enzyme systems.

피부세포에서 아토피 피부염의 유발과 관련된 PAR-2 및 사이토카인의 발현을 감소시키는 한약재 탐색 (Screening of Herbal Extracts to Reduce PAR-2 and Cytokine Expression Related to Atopic Dermatitis in Keratocytes)

  • The prevalence of atopic dermatitis has markedly increased in recent years but the mechanism has not been clearly revealed. Recent study exhibited that atopic dermatitis was exacerbated by the increase of proteinase-activated receptor (PAR)-2 expression, which activated $I{\kappa}B$ kinase --> nuclear factor kappa B. Therefore, we determined whether the allergens of dust mites induced the expression of PAR-2, intercellular adhesion molecule-1 (ICAM-1, adehision molecule), interleukins (IL)-6 in HaCaT keratocytes and which herbal 1,3-butylene glycol extracts (Mori Cortex Radicis, Sanguisorba officinalis L., Arctium lappa Linne, Torilis japonica DC, Melia azedarach Linne var. japoinca Makino) suppressed their expression. Dust mite allergen increased PAR-2, ICAM-1 and IL-6 expression in HaCaT cells in a dose-dependent manner up to $3{\mu}g/mL$ but their expression reached the plateau over the dosages. The allergen ($3{\mu}g/mL$) also secreted more cytokines such as tumor necrosis factor (TNF)-${\alpha}$ and IL-6 into the media. Among five different herbal extracts ($50{\mu}g/mL$), Mori Cortex Radicis and Sanguisorba officinalis L. suppressed the PAR-2, ICAM-1 and IL-6 expression in HaCaT cells, which was activated by dust mite allergen ($3{\mu}g/mL$) and they also reduced the secretion of TNF-${\alpha}$ and IL-6 into the media. In conclusion, Mori Cortex Radicis and Sanguisorba officinalis L. can effectively reduce the prevalence and progression of atopic dermatitis by dute mite allergen.

Sanguisorba officinalis Inhibits Immediate-type Allergic Reactions

  • The effect of aqueous extract of Sanguisorba officinalis L. (Rosaceae) root (SOAE) on the immediate-type allergic reactions by anal therapy was investigated. SOAE (0.01 to 1 g/kg) dose-dependently inhibited systemic anaphylaxis induced by compound 48/80 in mice. When SOAE was pretreated at the same concentrations with systemic anaphylaxis, the plasma histamine levels were reduced in a dose-dependent manner. SOAE (0.1 and 1 g/kg) also significantly inhibited local anaphylaxis activated by anti-DNP IgE. SOAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. The level of cyclic AMP (cAMP) in RPMC, when SOAE (1 mg/ml) was added, transiently and significantly increased compared with that of basal cells. These results provide evidence that anal therapy of SOAE may be beneficial in the treatment of allergic diseases.

지유 추출물 및 분획물의 항산화 활성과 식중독 원인균에 대한 항균활성 (Antioxidant Activity and Antimicrobial Effect for Foodborne Pathogens from Extract and Fractions of Sanguisorba officinalis L.)

  • Background: This study aimed to investigate the antioxidat and antimicrobial activities of the methanol extract and its fractions prepared from the roots of Sanguisorba officinalis L. Methods and Results: The antioxidant activities were compared by evaluating the DPPH radical and nitric oxide (NO) scavenging ability. Measurement of DPPH radical scavenging ability showed that the $SC_{50}$ values of the ethyl acetate fraction was $3.85{\mu}g/m{\ell}$. The ethyl acetate fraction exhibited the most effective DPPH radical scavenging ability compared with the other samples. As for the NO scavenging ability, at all tested concentrations, the ethyl acetate fraction showed a higher scavenging activity than that of the extract and other fractions. These results are related to the total phenolic compound and flavonoid contents of the ethyl acetate fraction. Antimicrobial activity against foodborne pathogens was investigated using the disc diffusion assay. The ethyl acetate fraction showed the highest antimicrobial activity against gram-positive Staphylococcus aureus and Bacillus cereus. However, the chloroform fraction had a higher antimicrobial activity against gram-negative Vibrio vulnificus than that of the extract and other fractions. Conclusions: The results show that the ethyl acetate fraction had a higher antioxidant as well as antimicrobial activity, than did the other samples. Therefore, the ethyl acetate fraction has potential application in the food industry.

지유 에탄올 추출물의 식품 위해성 세균에 대한 항균 활성 및 Salmonella serotype Typhimurium TA100에 대한 항돌연변이 활성 효과 (Antimicrobial Activity against Food Hazardous Microorganisms and Antimutagenicity against Salmonella serotype Typhimurium TA100 of an Ethanol Extract from Sanguisorba officinalis L.)

  • 본 연구는 지유 에탄올 추출물의 항균 및 항돌연변이 활성을 측정하여 새로운 기능성 소재로서의 기초자료를 얻고자 실시하였다. E. coli, E. coli O157:H7, P. aeruginosa, S. Typhimurium, L. monocytogenes, B. cereus, S. aureus의 7가지 주요 식품위해성 세균에 대한 항균활성을 검증하기 위해 paper disc diffusion assay 및 최소저해농도를 측정하였으며 24시간 동안 균의 생육억제도를 측정하였다. Paper disc assay 결과 E. coli와 E. coli O157:H7을 제외한 5가지 균에 대한 지유 에탄올 추출물의 뚜렷한 clear zone이 확인되었다. Micro-well dilution법으로 확인한 최소저해농도의 범위는 E. coli와 E. coli O157:H7을 제외한 5가지 균에 대해서 0.625~2.5 mg/mL로 나타났으며 생육억제도 실험결과도 최소 저해농도 결과와 일치하였다. Salmonella Typhimurium TA100을 이용한 Ames test 결과, 5 mg/plate의 농도에서 직접 돌연변이원인 sodium azide와 4-NQO에 대한 지유 에탄올 추출물의 항돌연변이 활성이 각각 72.42와 89.85%로 나타나 지유 에탄올 추출물의 우수한 항돌연변이 활성을 확인하였다. 본 연구를 통해 지유 에탄올 추출물이 주요 식품위해성 세균들에 대한 우수한 항균성과 직접돌연변이원에 대한 높은 항돌연변이 활성을 나타내어 천연물 유래 생리활성 소재로 이용될 수 있는 가능성을 보여주었다.

수종 한약재 추출물의 Staphylococcus aureus에 대한 항균활성 (Antibacterial activity from medicinal plant extracts on the Staphylococcus aureus)

  • 수종의 한약계를 70% 메탄올로 추출하여 식중독 세균인 Staphylococcus aureus에 대한 항균활성으로 생균수의 측정, 생육저해환의 측정, 최소저해농도와 최소사멸농도를 조사한 결과 이들 추출물의 Staphylococcus aureus에 대한 항균활성의 크기는 Rubus coreanus > Sanguisorba officinalis > Eriobotrya japonica > Prunus mume > Crataegus pinnatifida로 나타났다. 생육저해환은 복분자가 16.5 mm, 지유 14.3 mm, 오매 14.0 mm, 산사 12.7 mm로 측정되었다. MIC는 지유가 5mg / ml로 가장 낮았으며, MBC는 복분자가 10mg / ml로 나타났다. 실험에 사용된 한약재 추출물중 복분자, 비파엽, 산사, 오매, 지유는 Staphylococcus aureus에 대해서 강한 항균활성을 나타내었다. 따라서 이들 추출물을 이용한 천연보존료를 개발하여 식품에 적용한다면 그 기대효과가 상당히 클 것으로 생각된다.

지유가 흰쥐의 급성위염 및 소화성 궤양에 미치는 영향 (The Effects of Ethylacetate Fraction of Sanguisorba officinalis L. on Experimentally-induced Acute Gastritis and Peptic Ulcers in Rats)

  • 본 실험에서 흰쥐에 실험적으로 급성위염, 위궤양 및 십이지장궤양을 유발하고 지유 에틸아세테이트분획을 투여하여 항위염 및 항궤양에 미치는 영향을 검토하였던 바 다음과 같은 결과를 얻었다. 지유 에탄올 추출물을 n-헥산, 클로르포룸, 에틸아세테이트 및 n-부타놀과 물로 계통분획하여 농축하고 in vitro에서 항산화력을 측정한 결과 에틸아세테이트분획이 가장 우수하였다. 지유 에틸아세테이트분획 (SOL & SOH)의 HCI-EtOH급성위염에 대한 억제율은 각각 $27.7\%$, $31.7\%$로 대조약물로 사용된 cimetidine(CMT, $47.2\%$)의 억제효과에는 미치지 못했으나 CON에 비해 우수하였다. Indomethacin유발 위궤양에서, SOL & SOH의 위궤양억제율은 각각 $48.28\%$, $51.08\%$로 대조약물로 사용된 CMT($58.26\%$)의 억제효과에는 미치지 못했으나 CON에 비하며 유의 성있는 억제효과를 나타냈다. Shay 위궤양에서, 지유 에틸아세테이트분획을 십이지장에서 위내로 투여하였을 때 궤양 억제율은 SOL이 $42.86\%$로 CON에 비하여 억제되었으나 유의성 있는 효과는 아니었으며, 용량을 증량하여 투여한 SOH는 $47.71\%$로 대조약물인 omeprazol(OMP)의 저해효과($62.00\%$)에 미치지는 못했으나 CON에 비해 유의적민 효과를 나타내었다. 위액량 및 총위산분비량에서, 지유 에틸아세데이트분획 투여로 CON에 비하여 $32.11\%$$47.69\%$, $24.28\%$$34.69\%$의 감소로 유의성 있는 효과를 나타냈으며 OMP($55.69\%$, $41.62\%$)에 근접하게 감소되었다. pepsin 활성은 지유 에틸아세테이트분획 투여로 CON에 비해 각각 $19.55\%$, $\%21.45$가 저하되었으나 유의성있는 변화는 아니었다. 수침 $\cdot$ 구속 스트레스에 의해 유발된 궤양의 경우, SOL는 궤양억제율이 $9.23\%$ 로 CON와 유사하게 출혈을 동반한 깊은 궤양이 관찰되었으나 용량을 증량하여 투여한 SOH는 $48.45\%$로 CON에 비해 궤양의 깊이도 얕아졌고 출혈을 동반한 궤양도 감소되어 유의적인 억제율을 나타내었다. Cysteamine 유발 십이지장궤양에서, CMT는 유발된 십이지장 궤양을 CON에 비해 $54\%$ 정도 억제하여 우수한 항궤양효과를 나타냈고 시료투여군인 SOL 및 SOH는 CMT보다 우수하지는 않았으나 약 $35\∼42\%$ 정도의 궤양억제율을 나타냈다. 이상의 실험결과에서 지유의 항위염 및 항궤양효과는 항산화물질에 의한 결과로 판단되어지며, 앞으로 위염 및 위궤양 개선을 위한 건강 기능성 식품으로 개발이 기대된다
