Molecular Characterization of Apple stem grooving virus Isolated from Talaromyces flavus

  • Talaromyces flavus mediates the transmission of Apple stem grooving virus (ASGV) to several host plants. The ASGV-F carried by T.flavus was partially purified from the fungus. Based on sequence analysis and homology searches, this is closely related to other ASGV strains isolated from host plants. The partially purified viral coat protein (CP) was separated on a 12% SDS-polyacrylamide gel and analyzed by Western blotting with an ASGV anti-serum. A single band at 28 kDa reacted with the ASGV anti-serum. The deduced amino acid sequence of the ORF-l showed conserved domains, including an NTP-binding helicase motif, GFAGSGKT. The amino acid sequences of the helicase and CP showed strong homology to other ASGV strains (98%). All ASGV isolated from plants and fungi had salt bridges composed of the CP and the GFAGSGKT motif of the helicase, which are commonly conserved in plant viruses. These results suggest that ASGV-F is one of ASGV strains isolated from T.flavus based on sequence similarity as well as the serological analysis of CP.

  • This paper mainly focuses on the research of the monitoring data about the total nitrogen (TN), total phosphorus (TP) of ten monitoring points of Hongze Lake in 28 years. Our study adopts the technical methods such as difference comparison, correlation analysis, Mann-Kendall, etc. to analyze the statistical characteristics, regional differences, and temporal and spatial variation condition of ten monitoring points of Hongze Lake in 1986-2013 as well as the reason for the time and space variation of TN and TP. The results show that the TN and TP of Hongze Lake change dramatically during the period of 1994-2004. In all the monitoring periods, TN and TP of the HZ5, HZ6, HZ7, HZ8 monitoring points in the estuary surrounding the Huaihe River are the highest, which suggests that TN and TP of Hongze Lake should not be ignored for the pollution along the Huaihe River. The TN concentration in the core area is the highest, but that in the development control area is the lowest. In most of the years, TN concentration is higher than 2 mg/L, and the water quality is V class and poor V class water quality in China's Environmental Quality Standards for Surface Water (GB3838-2002). TP concentration of the three areas is basically close, that is lower than 2 mg/L in most of the years, and the water quality is within the V class. The TN and TP concentration of Hongze Lake decreased and intensifying trend of eutrophication is controlled after 2004.

Parallelizing 3D Frequency-domain Acoustic Wave Propagation Modeling using a Xeon Phi Coprocessor (제온 파이 보조 프로세서를 이용한 3차원 주파수 영역 음향파 파동 전파 모델링 병렬화)

  • 3D seismic data processing methods such as full waveform inversion or reverse-time migration require 3D wave propagation modeling and heavy calculations. We compared efficiency and accuracy of a Xeon Phi coprocessor to those of a high-end server CPU using 3D frequency-domain wave propagation modeling. We adopted the OpenMP parallel programming to the time-domain finite difference algorithm by considering the characteristics of the Xeon Phi coprocessors. We applied the Fourier transform using a running-integration to obtain the frequency-domain wavefield. A numerical test on frequency-domain wavefield modeling was performed using the 3D SEG/EAGE salt velocity model. Consequently, we could obtain an accurate frequency-domain wavefield and attain a 1.44x speedup using the Xeon Phi coprocessor compared to the CPU.

  • SWRO-PRO hybrid desalination technology is recently getting more attention especially in large desalination markets such as USA, Middle East, Japan, Singapore, etc. because of its promising potential to recover a considerable amount of osmotic energy from brine (a high-concentration solution of salt, 60,000 - 80,000 mg/L) and also to minimize the impact of the discharged brine into a marine ecosystem. By the research and development of the core technologies of the SWRO-PRO desalination system in a national desalination research project (Global MVP) supported by Ministry of Land, Infrastructure, and Transport (MOLIT) and Korea Agency for Infrastructure Technology Advancement (KAIA), it is anticipated that around 25% of total energy consumption rate (generally 3 to $4kWh/m^3$) of the SWRO desalination can be reduced by recovering the brine's osmotic energy utilizing wastewater treatment effluent as a PRO feed solution and an isobaric pressure exchanger (PX, ERI) as a PRO energy converter. However, there are still several challenges needed to be overcome in order to ultimately commercialize the novel SWRO-PRO process. They include system optimization and integration, development of efficient PRO membrane and module, development of PRO membrane fouling control technology, development of design and operation technology for the system scaling-up, development of diverse business models, and so on. In this paper, the current status and progress of the pilot study of the newly developed SWRO-PRO hybrid desalination technology is discussed.

  • ADSS(All Dielectirc Self-Supporting) cable installed under high voltage power cable line suffers a variety of environmental influence, rain, wind, snow fall, chemical pollution, salt fog and electrical stress. Its lifetime is required to be at least 20 years with this harsh weathering condition. The electrical stress under high voltage power line gives rise to dry band arcing and tracking, the severest damage, on the outer sheath of cable. Finally tracking might penetrate sheath and cause the break-down of ADSS cable. Tracking resistant sheath material, therefore, should be used to protect the core of ADSS from dry band arcing and to be sure long lifetime. In this work, we discuss various commercial tracking resistant material to investigate the way of track resistance and compare their mechanical, electrical, weathering and tracking properties through serial experiments. We found track resistant material is categorized into two main type : polyethylene with metal hydroxide and polyethylene with reduced carbon black. The Liquid contaminant, Inclined plane Tracking and Erosion test says the time to track of tracking resistant material with metal hydroxide has a little longer time to track in the high applied voltage than that with carbon black, but mechanical and weathering properties were inferior to.

Biocompatible Dispersion Methods for Carbon Black

  • The biological activity of particles is largely dependent on their size in biological systems. Dispersion in the aqueous phase has been both a critical impediment to and a prerequisite for particle studies. Carbon black has been used as a surrogate to investigate the biological effects of carbonaceous particles. Here, biocompatible methods were established to disperse carbon black into ultrafine and fine particles which are generally distinguished by the small size of 100 nm. Carbon black with a distinct particle size, N330 and N990 were suspended in blood plasma, cell culture media, Krebs-Ringer's solution (KR), or physiological salt solution (PSS). Large clumps were observed in all dispersion preparations; however, sonication improved dispersion - averaged particle sizes for N330 and N990 were $85.0{\pm}42.9$ and $112.4{\pm}67.9$ nm, respectively, in plasma; the corresponding sizes in culture media were $84.8{\pm}38.4$ and $164.1{\pm}77.8$ nm. However, sonication was not enough to disperse N330 less than 100 nm in either KR or PSS. Application of Tween 80 along with sonication reduced the size of N330 to less than 100 nm, and dispersed N990 larger than 100 nm ($73.6{\pm}28.8$ and $80.1{\pm}30.0$ nm for N330 and $349.5{\pm}161.8$ and $399.8{\pm}181.1$ nm for N990 in KR and PSS, respectively). In contrast, 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) exhibited little effect. Electron microscopy confirmed the typical aciniform structure of the carbon arrays; however, zeta potential measurement failed to explain the dispersibility of carbon black. The methods established in this study could disperse carbon black into ultrafine and fine particles, and may serve as a useful model for the study of particle toxicity, particularly size-related effects.

Comparison of Parallel Computation Performances for 3D Wave Propagation Modeling using a Xeon Phi x200 Processor (제온 파이 x200 프로세서를 이용한 3차원 음향 파동 전파 모델링 병렬 연산 성능 비교)

  • In this study, we simulated 3D wave propagation modeling using a Xeon Phi x200 processor and compared the parallel computation performance with that using a Xeon CPU. Unlike the 1st generation Xeon Phi coprocessor codenamed Knights Corner, the 2nd generation x200 Xeon Phi processor requires no additional communication between the internal memory and the main memory since it can run an operating system directly. The Xeon Phi x200 processor can run large-scale computation independently, with the large main memory and the high-bandwidth memory. For comparison of parallel computation, we performed the modeling using the MPI (Message Passing Interface) and OpenMP (Open Multi-Processing) libraries. Numerical examples using the SEG/EAGE salt model demonstrated that we can achieve 2.69 to 3.24 times faster modeling performance using the Xeon Phi with a large number of computational cores and high-bandwidth memory compared to that using the 12-core CPU.

Crossbreeding and parental lineage influences the diversity and community structure of rice seed endophytes

  • Seed endophytes are very remarkable groups of bacteria for their unique abilities of being vertically transmitted and conserved. As plants attain hybrid vigor and heterosis in the process of crossbreeding, this might also lead to the changes in the community structure and diversity of plant endophytes in the hybrid plants ultimately affecting the endophytes of the seeds. It would be interesting to characterize how seed endophyte composition change over time. The objective of this study is to gain insights into the influence of natural crossbreeding and parental lineage in the seed bacterial endophytic communities of two pure inbred lines exploring contributions of the two most important sources of plant endophytes - colonization from external sources and vertical transmission via seeds. Total genomic DNA was isolated from rice seeds and bacterial DNA was selectively amplified by PCR. The diversity of endophytic bacteria was studied through Terminal-Restriction Fragment Length Polymorphism (T-RFLP) analysis. Diversity between the original parents and the pure inbred line may show significant differences in terms of richness, evenness and diversity indices. Heat maps reveal astonishing contributions of both or either parents (IR29 ${\times}$ Pokkali and AT401 ${\times}$ IR31868) in the shaping of the bacterial seed endophytes of the hybrid, FL478 and IC32, respectively. Most of the T-RFs of the subsequent pure inbred line could be traced to any or both of the parents. Comparison of common and genotype-specific T-RFs of parents and their offspring reveals that majority of the T-RFs are shared suggesting higher transmission of bacterial communities common to both parents. The parents influence the bacterial community of their offspring. Unique T-RFs of the offspring also suggest external sources of colonization particularly as the seeds are cultivated in different ecogeographical locations. This study showed that host parental lines contributed greatly in the shaping of bacterial seed endophytes of their offspring. It also revealed transmission and potential conservation of core seed bacterial endophytes that generally become the dominant microbiota in the succeeding generations of plant hosts.

A Study on High-Resolution Seasonal Variations of Major Ionic Species in Recent Snow Near the Antarctic Jang Bogo Station (남극 장보고과학기지 인근에서 채취한 눈시료 내의 주요 이온성분들의 고해상도 계절변동성 연구)

  • A continuous series of 60 snow samples was collected at a 2.5-cm interval from a 1.5-m snow pit at a site on the Styx Glacier Plateau in Victoria Land, Antarctica, during the 2011/2012 austral summer season. Various chemical components (${\delta}D$, ${\delta}^{18}O$, $Na^+$, $K^+$, $Mg^{2+}$, $Ca^{2+}$, $Cl^-$, $SO_4{^2-}$, $NO_3{^-}$, $F^-$, $CH_3SO_3{^-}$, $CH_3CO_2{^-}$ and $HCO_2{^-}$) were determined to understand the highly resolved seasonal variations of these species in the coastal atmosphere near the Antarctic Jang Bogo station. Based on vertical profiles of ${\delta}^{18}O$, $NO_3{^-}$and MSA, which showed prominent seasonal changes in concentrations, the snow samples were dated to cover the time period from 2009 austral winter to 2012 austral summer with a mean accumulation rate of $226kgH_2Om^{-2}yr^{-1}$. Our snow profiles show pronounced seasonal variations for all the measured chemical species with a different pattern between different species. The distinctive feature of the occurrence patterns of the seasonal variations is clearly linked to changes in the relative strength of contributions from various natural sources (sea salt spray, volcanoes, crust-derived dust, and marine biogenic activities) during different short-term periods. The results allow us to understand the transport pathways and input mechanisms for each species and provide valuable information that will be useful for investigating long-term (decades to century scale periods) climate and environmental changes that can be deduced from an ice core to be retrieved from the Styx Glacier Plateau in the near future.

Synthesis, Structure, and Reactivity of the [Fe4S4(SR)4]2- (R = 2-, 3-, and 4-Pyridinemethane) Clusters

  • The $[Fe_4S_4]^{2+}$ clusters with 2-, 3-, and 4-pyridinemethanethiolate (S2-Pic, S3-Pic, and S4-Pic, respectively) terminal ligands have been synthesized from the ligand substitution reaction of the $(^nBu_4N)_2[Fe_4S_4Cl_4]$ (I) cluster. The new $(^nBu_4N)_2[Fe_4S_4(SR)_4]$ (R = 2-Pic; II, 3-Pic; III, 4-Pic; IV) clusters were characterized by FTIR and UV-Vis spectroscopy. Cluster II was crystallized in the monoclinic space group C2/c with a = 24.530 (5) $\AA$, b = 24.636(4) $\AA$, c = 21.762(4) $\AA$, ${\beta}=103.253(3)^{\circ}$, and Z = 8. The X-ray structure of II showed two unique 2:2 site-differentiated $[Fe_4S_4]^{2+}$ clusters due to the bidentate-mode coordination by 2-pyridinemethanethiolate ligands. Cluster III was crystallized in the same monoclinic space group C2/c with a = 26.0740(18) $\AA$, b = 23.3195(16) $\AA$, c = 22.3720(15) $\AA$, ${\beta}=100.467(2)^{\circ}$, and Z = 8. The 3-pyridinemethanethiolate ligand of III was coordinated to the $[Fe_4S_4]^{2+}$ core as a terminal mode. Cluster IV with 4-pyridinemethanethiolate ligands was found to have a similar structure to the cluster III. Fully reversible $[Fe_4S_4]^{2+}/[Fe_4S_4]^+$ redox waves were observed from all three clusters by cyclic voltammetry measurement. The electrochemical potentials for the $[Fe_4S_4]^{2+}/[Fe_4S_4]^+$ transition decreased in the order of II, III and IV, and the reduction potential changes by the ligands were explained based on the structural differences among the complexes. The complex III was reacted with sulfonium salt of $[PhMeSCH_2-p-C_6H_4CN](BF_4)$ in MeCN to test possible radical-involving reaction as a functional model of the [$Fe_4S_4$]-SAM (S-adenosylmethionine) cofactor. However, the isolated reaction products of 3-pyridinemethanethiolate-p-cyanobenzylsulfide and thioanisole suggested that the reaction followed an ionic mechanism and the products formed from the terminal ligand attack to the sulfonium.
