This study was undertaken to measure the degree of browning and mutagenicity by Ames test using Salmonella typhimurium TA 98 and TA 100 strains for roasted barley and sesame seeds used as food materials. The degree of browning of roasted barley for barley tea on the market showed a wide variation; barley for restaurant-use was heavily roasted (5 times) in comparison with homeuse barley. Sesame seeds for oil extraction were more roasted (4 times) than those for seasoning. Water-, ethanol- and ether-soluble fractions from roasted barley and sesame seeds did not show any signs of mutagenicity, even at the extremely high concentrations of the extracts.

The methanol extracts of perilla leaves reduced the mutagenicities mediated by of aflatoxin B$_1$(AFB$_1$), 3-amino-l-methyl-5H-pyrido (4,3-b) indole (Trp-P-2) and Benzo(a)pyrene (B(a)p) in Salmonella typhimurium TA98 and TA100. The methanol extracts were more fractionated, and the fractions of hexane and butanol revealed the antimutagenic activities against AFB$_1$ and B(a)p. The production of malondialdehyde (MDA) was decreased when the methanol extracts of perilla leaves were added to the system. The significantly higher antioxidative activity was observed in the butanol fraction. 2-Propyl furan, ethanedioic acid, dibutyl ester, benzaldehyde, 2-methyl-2-ethyl-3-hydroxy-propanoic acid and octahydro-3a-methyl-2H-inden-2-one were identified tentatively as major compounds from the butanol fraction.
Gamma irradiation was applied to Angelicae gigantis radix and Aloe to improve their hygienic quality. The effective dose of irradiation was 7 kGy in Angelicae gigantis radlx and 5 kGy in Aloe for the sterilization of all contaminated microorganisms tested. After 8 months of storage at room temperature, no growth of microorganisms was observed in the irradiated products. The safety of these products were evaluated by Salmonella typhimurium reversion assay and in vivo micronucleus assay using mouse bone marrow cells. They were negative in the bacterial reversion assay with S. typhimurium TA 98, TA100, TA1535 and TA1537. In the in vivo mouse micronucleus assay, they did not show any clastogenic effect at all doses tested. These results indicate that the gamma irradiation of Angelicae gigantis radix at 12 kGy and of Aloe at 10 kGy have no genotoxic effects under these experimental conditions.

The antimutagenic effects of juices from green pepper(GP), red pepper leaf(RPL), red pepper(RP) and sweet pepper(SP) were examined by the Ames method using Salmonella typhimurium TA100. The juice supernatants of GP, RPL and RP showed antimutagenic activities against afltoxin B1(AFB1) in Salmonella typhimurium TA 100. The juice supermatants of GP and RPL also exhibited the inhibitory effects(p<0.05) to the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitro-quinoline-1-oxide(4-NQO). The juice of RP showed antimutagenic activities against indirect mutagen of AFB1, however, the activity was reduced at higher concentration(5.0%), furthermore, as the adding concentration of sample increased to 5.0%, it exhibited slight comutagenicith on direct mutagen of MNNG. Theantimutagenic activities of GP and RPL juices were reduced significantly after heating at 100℃ for 20min, supposing that the antimutagenic compound(s) in the juices were heat labile.
On the mutagenicity induced by 3-amino-l,4-dimethyl-5-H-pyrido[4,3-b]indol (Trp-P-1) and 2-aminotluorene (2-AF), the antimutagenic effects of edible muntain herb juices were examined by the Ames assay using Salmonella typhimurium TA98 and TA100. Juices prepared from aralia bud, small water dropwort, mugwort, roots of belltlower and sedum didn't have mutagenicity. Most of sample juices showed the antimutagenicity. Especially, juices prepared from aralia bud, small water dropwort and mugwort were found to possess strong antimutagenic effects. Sedum was moderatly effective and root of belltlower had little effect on mutagenicity caused by Trp-P1 and 2-AF. The experimental results with TA98 were similar to those with TA100 in the antimutagenicity test of edible mountain herb juices. In this study, antimutagenicity on Trp-P-1 was more effective than that on 2-AF.

This study was carried out to determine the antimutagenic effect of soybean sprouts cultured in water containing germanium by Ames test and SOS chromotest. Germanium significantly inhibited the mutagenicity induced by aflatoxin B$_1$ (AFB$_1$) in Salmonella typhimurium TA100 by Ames test, and 4-nitroquinoline-N-oxide (4-NQO) in SOS chromotest. Juice from germanium treated soybean sprouts (GTS) inhibited 57∼75% mutagenicity induced by AFB$_1$, N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-NQO compared with 20∼48% inhibition rate of control soybean sprouts (germanium non-treated soybean sprouts, GNTS) in the Ames test. Also, methanol extracts from GTS inhibited 65% mutagenicity induced by AFB$_1$ in Salmonella typhimurium TA100, and 51% mutagenicity by 4-NQO in SOS chromotest. Therefore, it suggests that GTS has strong potential antimutagenic effect.
To evaluate mutagenicity of Phosphinotricin Acetyltransferase(PAT) which is expressed by the glufosinate-resistance gene pat, in vitro reverse mutation test using Salmonella typhimurium, chromosome aberration test using chinese hamster lung(CHL) cells and in vivo micronucleus test of mice were performed. In the reverse mutation, the PAT did not induce mutagenicity in Salmonella typhimurium TA 98, TA 100, TA 1535, TA 1537 with and without metabolic activation at $5000{\mu}g/plate$. In the chromosome aberration test, the results showed no incidence of increased structural and numerical chromosome aberrations at any doses tested(100, 10, $1{\mu}g/mL$). In micronucleus test, the ratio of micronuclei was measured in polychromatic erythrocytes of bone marrow of male ICR mice intraperitoneally administrated with PAT(1250, 625, and 313 mg/kg), the results showed no incidence of increased micronucleated polychromatic erythrocytes (MNPCE). These results indicate that PAT might not have mutagenic potential in vitro and vivo systems.
Mutagenic actions of aflatoxin B$_1$ (AFB$_1$) in the presence of various concentrations of L-ascorbIc acid (AA) in Salmonella typhimurium strains TA 100 and TA98 were studied. Spontaneous revertants per plate of the tester strains TA100 and TA98 were 121-125 and 25-30 with or without S9 mix, respectively. The negative controls used in the study did not show any mutagenesis in the tester strains. AFB$_1$ revealed strong mutagenicity at the dose levels of 0.05, 0.1 and 0.25 ${\mu}$g/plate with metabolic activation system in both strains. However, it showed a toxic effect when the levels were more than 0.5 ${\mu}$g/plate. When lower concentrations of AA (5-20 ${\mu}$g/plate) were added to AFB$_1$ in the Ames assay system with S9 mix the mutagenic action of AFB$_1$ decreased in both strains. About 70-90% of mutagenicity of AFB$_1$ disappeared in strain TA100 when 20${\mu}$g of AA was added to 0.05 ${\mu}$g of AFB$_1$. The inhibitory effect was greatly increased by the addition of higher concentrations of AA to AFB$_1$ in TA100 strain. The mutagenicity of AFB$_1$ was completely inhibited when 100 ${\mu}$g and 500 ${\mu}$g of AA were added to 0.05 ${\mu}$g and 0.1 ${\mu}$g of AFB$_1$, respectively, However, this protective effect of AA on AFB$_1$ mediated mutagenesis was less effective in TA98 strain than that in TA100.

Dietary intake of whole grains, vegetable and fruit is known to reduce the degenerative chronic diseases, such as cancer and cardiovascular diseases. Antioxidative and antimutagenic effects of the ethanol extract of Korean Millet, Buckwheat, Sorghum and Job's tears were examined by inhibition against iron-induced linoleate per-oxidation, DPPH (1,l-diphenyl-2-picrylhydrazyl) radical generation and MDA-BSA (malondialdehyde-bovine serum albumin) conjugation, and Ames test using Salmonella. Buckwheat showed the strongest antioxidative effect in three different systems among these four grains, but it showed the lowest antimutagenic effect. Sorghum was the second to Buckwheat in iron-induced linoleate peroxidation inhibition activity and DPPH radical scavenging activity, and showed very good direct-antimutagenic effect in 2-Nitrofluorene treated Salmonella Typhimurium TA98 and indirect-antimutagenic effect in 2-Anthramine treated Salmonella Typhimurium TA98 and TA100 with hepatic S9 mixture. Millet showed the strongest antimutagenic effect in Salmonella Typhimurium TA98 and TA 100 with or without S9. Buckwheat contained the highest total flavonoids and polyphenols, 1.14 mg/g and 3.71 mg/g, respectively. Total flavonoid content in these four grains was negatively correlated with $IC_{50}$/ for DPPH radical scavenging antioxidative effect significantly (r=-0.9924, p=0.0076), but not with antimutagenic effect.
This study was carried out to determine the antimutagenic and anticancer effects of fermented soybean using Ames test and cytotoxicity, respectively. The ethyl acetate fraction (200 g/plate) of fermented soybean in the Salmonella typhimurium TA100 strain showed 86.6% of inhibition rate against the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). In addition, the suppression of ethyl acetate fraction with same concentration of fermented soybean in the Salmonella typhimurium TA98 and TAI00 strains showed 82.4% and 90.8% inhibition against 3-amino-l,4-dimethyl-5H-pyrido-(4,3-b)indol (Trp-P-l), respectively. The cytotoxicity effects of fermented soybean against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL ethyl acetate fraction of fermented soybean showed strong cytotoxicities of 71.6%, 91.5% and 80.7% against A549, AGS and MCF-7, respectively.
