This study was conducted to investigate antibiotic resistance among Salmonella Typhimurium isolates from diseased pigs in Gyeongbuk province during the period 1998~2011. One hundred forty one isolates were tested for antibiotic resistance using the standard disk diffusion method and were examined for presence of resistance gene by PCR method. S. Typhimurium showed high drug resistance rates to tetracycline (95.7%), streptomycin (93.6%), ampicillin (86.5%), cephalothin (80.1%), gentamicin (79.4%), and trimethoprim/sulfamethoxazole (72.3%). Resistance gene, blaTEM, blaPSE1, tetA, tetB, tetG, sul1, sul2, aadA, strA, grm, and temA were detected among the antibiotic resistance isolates and temB, tetC, aadB gene were not detected. One hundred twenty one (89.6%) tetA, two (1.5%) tetB and one (0.7%) tetG gene were detected in the 135 tetracycline resistant isolates. Two (1.6%) temA gene were detected in one hundred twenty two ampicillin resistance isolates and temB was not detected.
Ten strains of Salmonella were isolated from feces and lymph nodes of swine slaughtered at Daegu slaughter-house and the rate of isolation was 6.0 percent. Nine strains of Salmonella were isolated by enrichment in selenite F broth and one strain by direct culture on SS agar, but none of Salmonella were isolated from MacConkey ager and in SS broth. Among Salmonella isolated, Salmonella typhimurium occupied over half (6 strains) and the importance of Salmonella in swine for the incidence of food poisoning in man was discussed.

This study was performed to develop a rapid test kit for pathogenic Salmonella in various samples. The rapid detection kit has been fabricated based on nitrocellulose lateral-flow strip. Colloidal gold and biotin conjugated Salmonella antibodies were used as a tag and a receptor, respectively. Manually spotted Salmonella antibody and Neutravidin on nitrocellulose membrane were used as test and control lines, respectively. Feasibility of the rapid kit to detect Salmonella typhimurium in samples were evaluated. The intensity of the color of the test line started to increase with the samples in which higher concentration of the cells were contained. The sensitivity of the sensor was $10^6$ cfu/mL Salmonella spiked in PBS. Also, the rapid test kit could detect $10^6$ cfu/mL of Salmonella in chicken meat extract.
The mutagenicity of 22 food and cosmetic dyes had been evaluated. Two different short-term mutagenicity tests were used: (1) Salmonella typhimurium preincubation assay (Ames test) (2) Chromosome aberration test with cultured Chinese hamster lung (CHL) fibroblast cells. Orange No. 203 was mutagenic in Salmonella typhimurium with and without rat liver microsomal activation, and Red No. 204 was mutagenic in Salmonelhl typhimurium with rat liver microsomal activation. Red No. 104-1 and Red No. 215 showed slight increase of chromosomal aberration in CHL cells.

The objective of this study was to evaluate the effects of various organic acids on microbial characteristics and Salmonella typhimurium in pork loins. Fresh pork loins were sprayed with various organic acids such as lactic acid, citric acid and acetic acid at various concentrations (0.5, 1, 1.5 and 2%). After spraying, the samples were packaged by HDPE film under air and stored at $4^{\circ}C$ for 14 days, and analyzed. Microbial deterioration of pork loins during the aerobic cold storage was delayed by organic acid spray. The bactericidal effect of acids increased with the increasing concentration. However, the inhibitory activity of organic acids during the storage varied with the kinds and concentrations of the acids. As for total plate counts, acetic acid was found to have the highest bactericidal activity, whereas citric acid was found to be the most inhibitory for coliform and S. typhimurium.
Salmonella (S.) Typhimurium is highly contagious, and its infection may rapidly spread within pig populations of herd. According to the survey (1,191 pigs) from 2003 to 2012, 155 pigs (13.0%) were diagnosed as salmonellosis in Jeju. Major porcine salmonellosis cases (88.4%) were concentrated in 4- to 12-week-old weaned pigs, but 6 pigs (3.9%) under 4 weeks old were also diagnosed. Based on the histopathologic examinations, ulcerative enteritis (63.9%) in the large intestine and/or paratyphoid nodules formation (57.4%) in the liver were most prevalent lesions in porcine salmonellosis. Single infection of S. Typhimurium and mixed infection with more than 2 pathogens were detected in 38 (24.5%) and 117 (75.5%) in pigs, respectively. Co-infections of Porcine reproductive and respiratory syndrome virus and Porcine circovirus type 2 were very common in porcine salmonellosis in Jeju and detected in 84 (54.2%) and 59 (38.1%) pigs, respectively. Based on the serotyping tests using 41 bacterial isolates, S. Typhimurium and S. Rissen were confirmed in 39 (95.1%) and 2 (4.9%) cases, respectively.
Salmonella typhimurium, causing mouse typhoid, infects hosts such as macrophage cells, and proliferates in intracellular vacuoles causing infected cells to trigger numerous genes to respond against the infection. In this study, we tried to identify such genes in RAW264.7 cells by using the PCR screening method with degenerate primers. Fourteen genes were found to be differentially expressed after a 4 h infection in which the expression of 8 genes increased while expression of the others decreased. Most of the genes were involved in proinflammatory responses such as cytokines production and cell death. The mutation in msbB gene encoding the myristoyl transferase in lipid A of lipopolysaccharide (LPS) resulted in much lower toxicity to the inoculated animals. We compared the expression of the identified genes in wild-type and msbB-mutated S. typhimurium infections and found that Lyzs encoding lysozyme type M was differentially expressed. This gene is quite likely to be related to bacterial survival in the host cells.
New regulatory, loci which participate in the regulation of anaerobic inducible gene expression in Salmonella typhimurium were identified. We observed the regulatory network of new regulator mutations to various anaerobic inducible gene (1). Some anaerobic inducible lac fusions were also induced at low pH condition which was severe environment to withstand for its virulence at the place like phagolysosome. Sic oxygen-regulated regulatory mutants (oxr) isolated by Tn10 mutagenesis were divided into two groups. Five of them were found to show negative effect on the regulation of anaerobic gene expression, while on e showed positive effect on the regulation. Genetic loci of four oxr were identified with 54 Mud-P22 lysogens covering the whole chromosome of S. typhimurium, in the nearby region of map unit 87 min (oxr101), 63 min (oxr104), 97 min (oxr 105), and 57 min (oxr 106), respectively. Two oxr mutants were subjected to two-dimensional polyacrylamide electrophoretic analysis of anaerobic inducible proteins for searching the control circuitry of our oxr mutants.

Fifty and forty two Salmonella enterica subspecies enterica serovar 52 Typhimurium (ST) strains were isolated from chicken and pigs, respectively, collected from markets throughout Korea from 2008 to 2011. The isolates were investigated for the presence of antimicrobial resistance and multi-drug patterns. All 50 ST isolates from chicken and 42 ST isolates from pigs were resistant to at least one of 13 antibiotics used in this study, 92.0% of ST isolates from chicken and 88.1% of ST isolates from pigs were resistant to three or more antimicrobials. As many as 3 isolates of ST isolates from chicken were resistant to 11 of 13 antimicrobials tested in this study. Only one isolate of ST isolates from pigs was resistant to 10 of 13 antimicrobials. The ACSSuT resistance phenotype was observed in 34% of the 50 isolates and 23.8% of the 42 isolates. Especially, 1 isolate from pigs had the ACSSuTAu. The high rate of antimicrobial-resist and multi-drug resistant (MDR) ST isolation may give rise to crucial public health problems. Therefore, control of antimicrobial use, and continuous monitoring of antimicrobial resistance and MDR patterns among Salmonella isolates in chicken and pig farms is necessary to ensure public health.
Antimutagenic effect of chitosan hydrolysates was investigated using Salmonella typhimurium reversion assay and SOS chromotest against 3-amino-1-methyl-5H-pyrido[4,3-b]lindole(Trp-P-2), aflatoxin $B_1$, 2-nitrofluorene and 4-nitroquinoline oxide. After partial acid hydrolysis of chitosan, six fraction of different molecular size were obtained by ultrafiltration. Chitosan hydrolysates showed antimutagenic effect of $0{\sim}78%$ on Trp-P-2, $0{\sim}92%$ on aflatoxin $B_1$ and $0{\sim}51%$ on 2-nitrofluorene in Salmonella typhimurium reversion assay. Inhibitory effect in Salmonella typhimurium reversion assay showed the highest at 5% concentration of fraction 6 in Trp-P-2, 10% concentration of fraction 5 in aflatoxin $B_1$ and 5% concentration of fraction 6 on 2-nitrofluorene. In SOS chromotest, chitosan hydrolysates showed anitimutagenic effect of $0{\sim}54%$ on Trp-P-2 and $0{\sim}77%$ on 4-nitroquinoline oxide, These results suggest that high molecular weight fraction of chitosan hydrolysates (MW>30,000) in most effective to inhibit mutagenicity of tested mutagens.
