  • 효모 (Saccharomyces Cervisiae)는 단일 세포의 형태로 존재하는 진핵 세포로써 동물세포와 유사한 기작으로 분비성 단백질을 생성한다. 따라서 박테리아와 달리 효모를 이용하면 당단백질이나 이황결합을 포함하는 분비성 단백질을 경제적으로 대량 합성할 수 있다. 효모의 필수 단백질 중 하나인 단백질 이황이성질화 효소는 소포체에 위치하며 분비성 단백질에 구조적으로 안정한 이황결합을 제공하는 효소이다. 본 연구는 단백질 이황이성질화 효소 (protein disulfide isomerase)가 지니고 있는 두 개의 활성도메인 중 분비성 단백질들의 합성 효율에 직접적으로 관여하는 부위를 찾는 연구이다. 효모 유전체로부터 단백질 이황이성질화 효소의 유전자 (PDI1)을 제거하고 효소의 변이 유전자를 주입한 후 효모의 성장 속도를 측정하였다. 또한 효모의 대표적 분비성 단백질을 각 변이 효소를 지니는 효모에 과발현시켜 합성 및 이황결합 형성 효율을 측정하였다. 단백질 이황이성질화 효소내 두 개의 활성 부위 중 아미노 말단쪽에 위치한 a 도메인에 있는 활성 부위가 분비성 단백질의 활성에 중요한 역할을 한다는 것을 알 수 있었다. 이 결과는 이황결합이나 당을 포함하는 외래 단백질의 고효율 합성을 위한 새로운 효모종 개발에 중요한 정보를 제공할 것으로 기대 된다.

  • As a whole, the improvement of the weight gain ratio has rather a clear effect for the groups fed live yeast especially at 0.1% level, comparing with the control, and the nutrient digestibility is also improved, noticeably in protein digestibility. Feed intake is not, however, increased for the live yeast fed groups comparing with the control. The live yeast supplements are apparently responsible for the overall improvement in the nutrient availability and this, in turn, resulted in the weight gain of treated groups over the control.

  • Saccharomyces cervisiae has a highly ordered ring of filaments that lies just inside the cytoplasmic membrane in the region of the mother-bud neck. Mutants defective in any one of the our cell division cycle genes (CDC3, CDC10, CDC11, CDC12) fail to form these filaments and exhibit a pleiotropic phenotype that includes failure to complete cytokinesis and abnormal bud growth. However, the role of the filament is not clear. In order to find out the role of filament, the similar gene in S pombe (called cdc103$\^$+) to the CDC3 was cloned and sequenced. Here I report the sequence analysis of the cdc103$\^$+/ ) to the CDC3 was cloned and sequenced. Here I report the sequence analysis of the cdc103$\^$+/. Comparison of the predicted amino acid sequences of cdc103$\^$+/ and CDC3 revealed that they share significant similarity (43% identity and 56% identity or similarity) to each other.

  • Some properties of $\beta$-1, 3-glucan synthase system in Saccharamyces cerevisiae were investigated. By extraction with detergent and salt, the membrane preparations could be dissociated into two components, one soluble, the other still membrane bound. Both components, in addition to GTP, were necessary for the activity of $\beta$-1, 3-glucan synthase like other fungi. The protective effect of guanosine nucleotides on the soluble factor pointed to the possibility that this fraction contained a GTP-binding protein. Addition of increasing amounts of soluble factor to a constant amount of insoluble catalytic factor, vice versa, gave rise to a saturation curve. These results, including different types of evidence, indicate that the soluble factor and the catalytic factor form a complex.

  • The study was conducted to investigate per oral (PO) effects of Raspberry wine on testosterone levels in Sprague-Dawley rats. Raspberry wine of 13% alcohol concentration, was prepared from ripen fruits of Rubus coreanus fermented with Saccharomyces cervisiae. PO administration of Raspberry wine for 15 week (group A) produced dramatic increases of serum testosterone levels. Increase in the testosterone level was observed, using gamma counter with 1251 testosterone, starting from 1 week post administration. Maximum increase in testosterone level was observed at 5 week post administration, 7.486±6.482ng/mL, which was 14.6 times higher than normal and at 15 weeks post administration it was recorded as 1.84±3.516ng/mL. However, PO administration of Saccharomyces cervisiae broth (Group B) and 13% brewed alcohol (group C) for 15 weeks resulted slight increase in testosterone levels, indicating Raspberry wine as an effective phyto-testosogenic beverage of the future.

  • Possible yeast strains that could be used as feed for aquaculture were studied. It was shown that the maximum specific growth rate and the biomass yield of Kluyveromyces fragilis yeast and Candida utilis yeast under optimum pH and temperature were much higher than those of Saccharomyces cervisiae yeast which had been as established yeast diet for rotifer culture. Hence, this work was focussed on the growth characteristics of the two yeasts through flask dultures for mass production. With 5% inoculum dosage, the best values of $\mu$$_{max}$ and OD$_{max}$ were obtained with on 2.5% fructose medium and 2% YE medium for K. fragilis and C. utilis, respectively, where the values of $\mu$$_{max}$ and OD$_{max}$ were found to be 0.73 hr$^{-1}$ and 3.00 for K. fragilis and 0.59 hr$^{-1}$ and 2.80 for C. utilis. It was also found that the lag phase of the growth incresed with increasing initial zinc and NaCl concentrations and decreased with increasing inoculum dosage. Both yeasts could survive relatively well at 3.5% NaCl concentration, and only C. utilis yeast could utilize zinc.

A Study on the Antigen Characteristics of Rhodotorula rubra (Rhodotorula rubra의 항원특성에 관한 연구)

  • Antigenicity of Rhodotrula rubra isolated from pulmonary tissue of pulmonary tuberculosis patients was studied by means of agglutination reaction with R. rubra whole cell antiserum. And the serological reactivity of crude polyfac charide from R. frubra, Candida albicans, Candida tropicalis, Candida, glabrata, and Saccharomyces cerevisiae ATCC 26603 with antiserum to R. rubra whole cell was studied by means of immunodiffusion test. R. rubra showed stationary phase after 48h when it was cultured in GYEP broth. While agglutinogen titer was 1:64 at lag phase, agglutinogen titer was 1 :256 after 20h. After growth of R. rubra on different 11 media, nutritional environment showed similar agglu-tination reartivity. The agglutinogen titer of C. albicans, C. tropicalis, C. giabrata, which were isolated from patient's expectoration, to R. rubra antiserum by means of agglutination reaction were 1:16, respectively. But, Sacch. cervisiae ATCC26603 was negative. Those results were lower than that of R. rubra agglutinogen titer 1:256. As a result of immu-nodiffusion test with crude polysaccharide extracted from cell wall of R. rubra, C. albicans, C. tropicalis, C. glabrata, Sacch. cervisiae ATCC26603, precipitin line was found only with R. rubra, of which antibody titer was 8.

  • The cloned glyceraldehyde-3-phosphate dehydrogenase (GAP) gene of Saccharomyces cereviszae (Holland et al., 1983) has been characterized. Based on the communication, we have also cloned 2.1 kb CAP DNA fragment and modified this fragment as a portable promoter. Two yeast expression vectors, one is YCp type vector being maintained at low copy number (1 or 2) and the other is YEp type vector at high copy number, have been constructed with the GAP promoter and the PH05' gene as a reporter. Our plasrnids were introduc,ed into S. cerevisiae HY-1, which has been improved. The $Trp^+$ transformants expressed APase activity efficiently and showed high level of PH05' transcripts.

  • In order to improve the productivity of ethanol by sugar-alcohol-tolerant Saccharomyces cerevisiae D1, the effect of addition of $Ca^{2+}$ on the alcohol fermentation was investigated. The addition of $Ca^{2+}$led to both the improvement of ethanol productivity and the increase of viable cell concentration. The optimal concentration of $Ca^{2+}$ was 0.8 mM. The higher was initial concentration of glucose, the greater effect of $Ca^{2+}$ was observed. Ethanol inhibition of growth, specific death rate in lethal concentration of ethanol, and extracellular final pH decreased by the addition of $Ca^{2+}$. The effect of $Ca^{2+}$ supplementation was explained by the increase of its ethanol tolerance.

  • In order to prevent a markdown and improve a value added of apricot, this study was conducted to research the optimum condition for manufacture of wine using apricot. On fermentation of aprirot wine, the best yeasts were Saccharomyces cerevisiae JBS 15 and JBS 30, and optimum composition or medium was apricot juice of 50%, sugar of 24 。brix, (NH$_4$)$_2$SO$_4$ of 0.2%, Na$_2$SO$_3$ of 0.02%. The content of alcohol in this medium after fermentation of 10 days at 25$^{\circ}C$ was 12.3∼12.5%. sensory evaluation showed that color, taste and odor or apricot wine were good, and saccharomyces cerevisiae JBS 15 was not difference JBS 30.
