The Effects of Salvia Miltiorrhiza on Renal Function and Histopathological Changes in Streptozotocin-induced Diabetic Nephropathy Rat Model (단삼이 당뇨병성 신병증 Rat의 신기능 및 조직학적 변화에 미치는 영향)

  • Objective : Diabetic nephropathy is the most common cause of end stage renal disease. AGE, $TGF-{\beta}1$ type IV collagen, and macrophage/monocyte infiltration are the main factors of diabetic nephropathy. We investigated the effects of Salvia miltiorrhiza on renal function and histopathological changes in streptozotocin(STZ)-induced diabetic nephropathy rat model. Methods : Diabetes was induced in male Sprague-Dawley rats(290${\pm}$10g) by injecting STZ(45mg/kg) into the tail vein. Rats were divided into 3 groups(n = 6): normal, control, and salvia. After 8 weeks of administration of Salvia miltiorrhiza extract on the Salvia group, we checked 24 hrs urine, blood biochemistry and renal tissue to evaluate renal function and histopathological changes by examining parameters including albuminuria, BUN, creatinine, cholesterol, LDL, TG, macrophage/monocyte antigen(ED-1), $TGF-{\beta}1$, AGE, and type IV collagen. Results : Salvia miltiorrhiza decreased the amount of 24hrs proteinuria, and inhibited histopathological changes of diabetic nephropathy including the expression and accumulation of various factors which could promote development of diabetic nephropathy. Conclusion : These findings suggest that Salvia miltiorrhiza might protect the renal function and inhibit the development of renal injury by regulating factors including AGE, $TGF-{\beta}1$ Type IV collagen, macrophage and monocyte infiltration. So Salvia miltiorrhiza can be used for diabetic patients to prevent the progression of diabetic nephropathy.

Antidiabetic activity of Argyreia speciosa (sweet) (Burm.f.)Boj. in normoglycemic and Streptozotocin-induced diabetic rats

  • Effect of ethanol (ASE) and water (ASW) extracts of Argyreia speciosa on blood glucose and lipid profile was investigated in normoglycemic and Streptozotocin (STZ)-induced diabetic animals. In oral glucose and sucrose tolerance test, treatment with ASE and ASW (100 and 200 mg/kg) and Glidenclamide (10 mg/kg) significantly improved the glucose and sucrose tolerance in normal animals. In addition, respective treatment for fifteen-day resulted in significant percentage reduction in serum glucose (SG) ie., 30.39% (lower dose of ASE) and 33.21% (higher dose of ASW). In standardized STZ (50 mg/kg, iv)-induced diabetic rats, a single dose of ASE and ASW treatment exhibited reduction in SG levels at different time intervals compared to basal levels. Administration of both the doses of ASE and ASW for fifteen-day days exhibited greater percentage reduction in glycemia (24.6%, 24.7%, 23.9% and 21.9% respectively) and also ameliorated restored to near normal value of all tested lipid parameters. Further, treatment also exhibited significantly improved glucose tolerance over the period of 120 min compared to diabetic control group. Eventhough treatment failed to increase serum insulin levels significantly but peripheral utilization of insulin was increased as evident by insulin tolerance test. Taken together, present study supports the traditional usage of title plant in the treatment of diabetes mellitus.

Streptozotocin Diabetes Attenuates the Effects of Nondepolarizing Neuromuscular Relaxants on Rat Muscles

  • The hypothesis of this study was that diabetes-induced desensitization of rat soleus (SOL) and extensor digitorum longus (EDL) to non-depolarizing muscle relaxants (NDMRs) depends on the stage of diabetes and on the kind of NDMRs. We tested the different magnitude of resistance to vecuronium, cisatracurium, and rocuronium at different stages of streptozotocin (STZ)-induced diabetes by the EDL sciatic nerve-muscle preparations, and the SOL sciatic nerve-muscle preparations from rats after 4 and 16 weeks of STZ treatment. The concentration-twitch tension curves were significantly shifted from those of the control group to the right in the diabetic groups. Concentration giving 50% of maximal inhibition ($IC_{50}$) was larger in the diabetic groups for all the NDMRs. For rocuronium and cisatracurium in both SOL and EDL, $IC_{50}$ was significantly larger in diabetic 16 weeks group than those in the diabetic 4 weeks group. For SOL/EDL, the $IC_{50}$ ratios were significantly largest in the diabetic 16 weeks group, second largest in the diabetic 4 weeks group, and smallest for the control group. Diabetes-induced desensitization to NDMRs depended on the stage of diabetes and on the different kind of muscles observed while was independent on different kind of NDMRs. The resistance to NDMRs was stronger in the later stage of diabetes (16 versus 4 weeks after STZ treatment). Additionally, when monitoring in SOL, diabetes attenuated the actions of neuromuscular blockade more intensely than that in EDL. Nonetheless, the hyposensitivity to NDMRs in diabetes was not relevant for the kind of NDMRs.

Effects of Buckwheat on Organ Weight, Glucose and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats (메밀급여가 Streptozotocin유발 당뇨쥐의 장기무게 및 당질과 지질대사에 미치는 영향)

  • The present study was undertaken to evaluate the effects of 50% buckwheat diet on the body weight, organ weight, urine albumin, urine glucose, plasma glucose and plasma lipid in normal rats and diabetic rats treated with streptozotocin(STZ). The food intake, body weight, the level of urine glucose in diabetic buckwheat groups were not significantly different with diabetic control group. The level of urine albumin was lower in raw and steam buckwheat group than in the diabetic control group. Compared to the normal control group, liver and kidney weights were heavier in the diabetic groups. Pancreas weight was heavier diabetic buckwheat groups than in normal and diabetic control groups. Fasting plasma glucose level of diabetic buckwheat groups significantly decreased by 18-37% compared with the diabetic control group. Plasma triglyceride level of diabetic buckwheat groups significantly decreased by 34-50% compared with the diabetic control group. Plasma total cholesterol level of diabetic buckwheat groups decreased by 15-27% compared with the diabetic control group. The level of HDL-cholesterol was not affected by buckwheat diet. These results indicate that buckwheat is an effective therapeutic regimen for the control of metabolic derangements in diabetics.

Effects of Dietary Sea Tangle on Blood Glucose, Lipid and Glutathione Enzymes in Streptozotocin-Induced Diabetic Rats (다시마 분말이 당뇨 유발쥐의 혈당과 지질농도 및 항산화 효소계에 미치는 영향)

  • The Purpose of this study was to investigate the effect of dietary sea tangle in diabetic rats treated with streptozotocin(STZ). Four groups of rats (Sprague-Dawley male rats,180-200g) were normal rats fed control diet(C), diabetic rats fed control diet(CD), normal rats fed sea tangl diet(T), and diabetic ,rats fed sea tangle diet(TD), diabetes was induced by single injection of streptozotocin(60mg/kg B.W.). High density lipoprotein(HDL) of T and TD group were higher than other groups(C and T groups). And the weekly change of blood sugar was decreased in the 3th and 4th weeks. But serum triglyceride (TG) of diabetic rats fed sea tangle diet(TD) was lower than diabetic rats fed control dlet(CD). Activity of hepatic microsomal Glucose 6-phosphatase(G6Pase) was significantly increased CD and TD groups higher than C and T groups. Hepateic glutathione S-transferase(GST of T, CD and TD groups were significantly lower than C group(p<0.05), glutathione peroxidase (GPX) of T and TD groups were significantly higher than C and CD groups(p<0.05).

Antioxidant Effects of the Mixture of Mulberry Leaves and Silkworm Powder on the Plasma and Liver in Streptozotocin-Induced Diabetic Rats

  • This study was carried out to examine the antioxidant effects of a mixture of mulberry leaves and silkworm powder in plasma and liver of streptozotocin-induced diabetic rats. Sprague-Dawley male rats weighing 100$\pm$10 g were used and their diets were supplemented with $0.4\%$ (4 g/kg) of the mixtures. Experimental groups were diabetic rats without supplements (DM group) or with a combination of the supplements: $100\%$ mulberry leaves (M group), $25\%$ silkworm powder mixed with mulberry leaves (25SM group), $50\%$ silkworm powder mixed with mulberry leaves (50SM group), $75\%$ silkworm powder mixed with mulberry leaves (75SM group) or $100\%$ silkworm powder (100S group). The rats were fed experimental diets and water ad libitum. All animals were injected with streptozotocin at the $3^{rd}$ week for inducing diabetes and were sacrificed on $9^{th}$ day thereafter. Hepatic xanthine oxidase (XOD) activity significantly decreased in the mixture supplemented groups compared to the DM group. Hepatic superoxide dismutase (SOD) activity was not significantly different among any of the experimental groups, but glutathione peroxidase (GSH-px) activity increased in the mixture supplemented groups compared to the DM group. In particular, it was the highest in the 50SM group. The hepatic TBARS values were lower in all the mixture supplemented groups than in the DM group, and it was as lowest when ratio of mulberry leaves to silkworm powder was highest. Hepatic lipofuscin contents were similar with the TBARS value. In conclusion, the mixtures containing silkworm powder reduced oxidative damage by strengtbening the antioxidative system and suppressing oxidative stress in the STZ-induced diabetic rat. The 1:1 blend of silkworm powder and mulberry leaves was the most effective combination for antioxidant activity.

Antihyperglycemic of Gleditschiae Spina Extracts in Streptozotocin-Nicotinamide Induced Type 2 Diabetic Rats (Streptozotocin-Nicotinamide로 유도된 제2형 당뇨모델 쥐에서 조각자(Gleditschiae Spina) 추출물의 항당뇨효과)

  • The aim of the present study was to investigate antihyperglycemic effect of Gleditschiae Spina (GS) in streptozotocin (STZ)-nicotinamide (NA)-induced type 2 diabetic rats. The rats were divided into four groups: normal control (NC), diabetic control (DC), diabetic rats supplemented with acarbose (AC, 4 mg/kg), and with GS ethanol extracts (GSE, 50 mg/kg). Weekly fasting blood glucose (FBG) for 10 weeks and oral glucose tolerance test (OGTT) at 10th week were monitored using glucose oxidase-peroxidase reactive strips. The FBG level was significantly reduced in AC group after 8 weeks and in GSE group at the end of period. The AUCs for the glucose response from OGTT and blood glucose level after sacrifice were significantly lower in the AC and GSE groups than the DC group. GSE supplementation significantly increased plasma total radical-trapping antioxidant potential (TRAP) in STZ-NA-induced diabetic rats, compared with DC group. The present study indicates that GSE could ameliorate type 2 diabetes and be comparable to acarbose, a standard hypoglycemic drug. Also, we suggest that GSE may possess antioxidant activity against the STZ-NA-induced oxidative stress.

Effects of Dietary Oligosaccharide on the Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 혈당 및 혈중 지질조성에 미치는 Oligosaccharide의 영향)

  • This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.

Effect of 630 nm Light Emitting Diode (LED) Irradiation on Wound Healing in Streptozotocin-Induced Diabetic Rats

  • The purpose of this study was to clarify the effect of light emitting diode (LED) irradiation on healing of impaired wound and alteration of mast cells in experimental diabetic rats. Twenty-four male Sprague-Dawley rats were divided into four groups: excision (Ex), excision-LED irradiation (Ex-LED), diabetes + excision (DM) and diabetes + excision + LED irradiation (DM-LED). Diabetes was induced in rats by streptozotocin (STZ) injection (70 mg/kg, single dose) and 6 mm punch excision wounds were created on the back after shaving hair. The LED-irradiated rats were treated to a daily dose of $5\;J/cm^2$ LED (630 nm) light for 11 days after surgery, and were killed at day 1, 3, 7 and 11. The lesion and adjacent skin tissues were excised, fixed with 10% buffered formalin and embedded with paraffin. For evaluation of wound healing, hematoxylin-eosin (HE) and Masson trichrome staining were performed. Mast cells (MCs) were stained with toluidine blue (pH 0.5) and quantified using a computerized image analysis system. The proliferation activity of keratinocyte in skin tissues was analyzed on sections immunostained with proliferative cell nuclear antigen (PCNA). The results showed that wound healing rate, collagen density and neo-epidermis length, number of PCNA-positive cells, fibroblasts and mast cells were significantly higher in the LED-irradiated rats than in the DM and Ex rats throughout the periods of experiment. Exceptionally, the number of MCs was significantly lower at day 11 compared with day 7 after surgery in the all groups. These findings suggest that the LED irradiation may promote the tissue repair process by accelerating keratinocyte and fibroblast proliferation and collagen production in normal rats as well as in diabetic rats, and MCs may play an important role at an early stage of skin wound healing in normal and diabetic rats.

Microsomal Mixed Function Oxidase and Lipid Peroxidation in Liver and Lung of Sterptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 간장 및 폐조직에서의 Microsomal Mixed Function Oxidase System과 과산화지질 생성)

  • 본 연구는 streptozotocin 유발 당뇨쥐에서의 MFO계활성 변화 및 이에 지질과산화를 관찰하고자 체중이 140kg 내외의 Sprague-Dawley 종 흰쥐 숫컷을 대조군과 당뇨 유발 시험군으로 나누어 4주간 사육하였다. 당뇨군은 STZ로 당뇨를 유발시켰으며 당뇨 유발 6일후 쥐를 희생기켜 간조직 및 폐조직 microsome 중의 cytochrome $P_{450}$ 및 cytochrome $b_[5}$ 함량과 NADPH-cytochrome $P_{450}$ reductase 활성도를 측정하고 아울러 microsome내의 지질과산화물을 측정하여 다음과 같은 결과를 얻었다. 실험군간에 식이 섭취량은 차이가 없었으나, 체중 증가량과 식이 효율은 당뇨군이 STZ군에 비해 현저하게 감소하였다. 간장 및 폐조직의 무게는 대조군과 당뇨군간에 유의적인 차이는 없었다. 간조직 및 GP조직 중의 cytochrome $P_{450}$ 함량은 대조군에 비해 당뇨군이 150%, 175%씩 증가하였다. 간조직 및 폐조직 중의 cytochrome $b_{5}$은 대조군에 비해 당뇨군이 53%,116%씩 각각 증가하였다. 간조직에서의 NADPH-cytochrome $P_{450}$ reductase 활성은 당뇨군이 대조군에 비해 46% 증가하였으며, 폐조직에서는 75%증가하였다. 지질과산화물가는 당뇨군이 대조군에 비해 간족에서는 약 95% 높았으며 폐조직에서는 73%높았다. 이상의 결과에서 STZ 유발 당뇨쥐에서는 MFO system의 활성도가 대조군에 비해 현저하게 증가되고 지질과산화반응이 같으 srudgid이 촉진되었으며 폐조직에서도 간조직에서와 비슷한 경향이었다. 이러한 것은 당뇨군에서는 MFO system의 활성증가로 free radical 생성이 증가되고 그 결과 지질과산화가 촉진되었다고 볼 수 있다.
