Objectives : The present study was carried out to investigate the beneficial effect of Coicis Semen extract(CSe) on streptozotocin(STZ)-induced diabetic nephropathy rats. CSe was given to rats with oral administration. Methods : The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with CSe treatment. Experimental diabetes was induced by the injection of STZ(60 mg/kg) to the rat via the peritoneum. The effects of CSe on STZ-induced diabetic nephropathy were observed by measuring the serum level of creatinine, BUN and uric level of glucose. Kidney level of lipid peroxidation and the activities of reduced glutathione(GSH) were also examined Results : STZ-induced increase of serum creatinine was lowered by CSe treatment, but BUN and uric level of glucose did not show significant changes. CSe oral administration showed statistical decrease of lipid peroxidation in renal cortical tissues, but it has no effect on the activities of GSH. Conclusion : CSe treatment showed protective effect on rat diabetic nephropathy model, but action mechanism of the effect was still not dear. We thought to be concerned with anti-oxidative stress.
This research was performed to investigate the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on blood lipid profiles and tissue differentiation in streptozotocin (STZ)-induced diabetic rats. The animal groups consisted of a normal-control group, a STZ-control group, three STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 combined with exercise; eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for a 10 week experimental period. The MF supplementation group showed a significant reduction in levels of serum total cholesterol and triglyceride compared to the STZ-control group. HDL-cholesterol levels were significantly increased in the MF supplementation group compared to STZ-control group. The ratio of HDL-cholesterol to total cholesterol was significantly higher in MF supplementation group compared to the STZ-control group. The Atherogenic Index (AI) values in the MF supplementation groups were found to be significantly lower than in the STZ-control group. Serum AST and ALT levels were significantly reduced in the MF-supplementation groups compared to the STZ-control group. Total cholesterol level in the liver tissue was significantly decreased in the MF-360 group and in the MF-360 + exercise group compared to the STZ-control group. In immunohistochemical staining of the pancreatic islets of the MF-supplemented groups, a significantly higher number of insulin-immunoreactive cells were observed compared to the STZ-control group. In the MF supplementation groups, Bowman's capsules were clearly observed as hypertrophy of the glomeruli was not obvious. In the MF supplementation groups, a relative reduction in the hypertrophy of the basement membrane of the glomeruli and a significant reduction in the mesangium were observed compared to the STZ-control group. The results of this study suggest that supplementation of MF has beneficial effect in improving plasma lipid and tissue metabolism in streptozotocin-induced rats.
Rosae laevigatae Fructus extract (RLF) was tested for the effects on the urethral nitric oxide synthase (NOS) activity and Antioxidation in streptozotocin (STZ) induced diabetic rats. RLF was treated firstly into samples, and then STZ induced diabetic rats were set with them. In vitro, the urethral NOS activity was not noted but the type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide were decreased in the level of Dose of extract prepared from RLF. In vivo, after the extract was administered to the animal model for fifteen days, the urethral NOS activity increased in STZ induced diabetic rats to the level of normal rats. The content of urethral nitrite and glutathione followed by RLF pre-medicating administration, increased as highly as normal group in compare with the group treated with STZ. The type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide followed by RLF pre-medicating administration, decreased as lowly as normal group in compare with the group treated with STZ. In conclusion, the extract of RLF will be able to restore erectile dysfunction of STZ induced diabetic rats.
The effect of corticosteroid on the diabetic pregnant rats and their fetuses was investigated. Streptozotocin (STB) was injected into the pregnant rats on the fifth day of pregnancy. Dexamethasone (DXM) was injected into the pregnant rats on the 17th, 18th, 19th and 20th days of pregnancy In prenatal rats, the body weight, an abortion rate, number of fetus, the ratio of lecithin/sphingomyelin (L/S) and the levels of blood glucose and phosphatidylglycerol (PG) were determined. In the postnatal rats, the body weight, the levels of blood glucose, fetal number, stillbirth rate, an organ weight and the levels of hepatic glycogen, protein and triglyceride were determined. The body weight of fetuses was lower in the DXM group and higher in the STZ group than the those of control group. Blood glucose of fetuses produced hypoglycemia in the STZ group compared with the control group. A significant increase in the abortion and stillbirth rates was observed in STZ group. The levels of glycogen, protein and triglyceride in fetus liver and the weight of pancreas were significantly increased in the 572 and STZ+DXM groups compared with the control group. The L/S ratio and the level of PG in the amniotic (quid were significantly decreased in STZ group compared with the control group, whereas those of the STZ+DXM group were similar to the control group. It has been observed that corticosteroid administration on the STZ-induced diabetic rats during final stage of pregnancy can prevent the respiratory depression syndrome of neonatal rats.
The present study was carried out to investigate the preventive effect of Epimedii Herba combined Samgijiwhang-Tang(SJTE) on streptozotocin(STZ)-induced diabetic nephropathy. SJTE was given to rats with oral administration. The experimental animals were divided into normal group of rats, control group of STZ-induced diabetic rats, and sample group with SJTE administration. Experimental diabetic nephropathy was induced by the injection of STZ(60mg/kg) to the rat via the peritoneum. The effect of SJTE on STZ-induced diabetic nephropathy was observed by measuring the serum level of insulin, glucose, creatinine and BUN. Urine secretion of albumin for 24 hours and urine level of glucose measures too. Anti-oxidative stress of STZ administration in living body was estimated by measuring lipid peroxide in cortex of kidneys. STZ induced increase of serum glucose. creatinine, urine albumin secretion and renal cortical lipid peroxidation were lowered by SJTE administration. In conclusion, the SJTE treatment showed protective effect on rat diabolic nephropathy model, and action mechanism of the effect was thought to be concerned with internal glucose metabolism.
This study was done to investigate the antidiabetic effect of Glechoma hederacea LINNAEUS in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The methanol extract of Glechoma hederacea was orally administrated once a day for 6 days. The contents of serum glucose, triglyceride (TG), total cholesterol and Atherogenic Index (Al) were significantly decreased, but high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were significantly increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic lipid peroxide and activity of catalase were decreased, but content of glutathione as well as activities of glutathione-S-transferase and superoxide dismutase were increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase, glucokinase were significantly increased, but activity of glucose-6-phosphatase was decreased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. These results indicated that methanol extract of Glechoma hederacea would have antidiabetic effect in STZ-induced diabetic rats.
This research was conducted to study the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on plasma insulin and glucose levels in streptozotocin (STZ)-induced diabetic rats. Eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for 10 weeks. The animal groups consisted of a normal-control group, a STZ-control group, 3 STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 along with exercise. In the normal-control group, glucose tolerance tests resulted in the peak blood glucose level being achieved in 15 minutes and a fasting blood glucose level being achieved in 60 minutes. In the STZ-control group, the peak blood glucose level was reached after 60 minutes and, even after 90 minutes, blood glucose shown at a significantly higher level compared to the fasting levels. In the groups supplemented with MF extracts, the blood glucose level peaked after 30 minutes of glucose challenge, and returned to the fasting level after 90 minutes; the MF-360 and MF-360+exercise groups showed the best levels of glucose tolerance. Blood glucose levels in the STZ-induced diabetic groups were significantly higher compared to the normal-control group. However, after 7 weeks of supplementation with MF extracts, a significant lowering of blood glucose levels was observed in all groups supplemented with the MF extract. The best effect was observed in the group given MF extract combined with exercise. Compared to the normal-control group, blood insulin levels were significantly lower in all STZ-induced diabetic groups; however, a significantly higher level of insulin was observed in the groups given MF extracts compared to the STZ-control group. This study shows that the supplementation of MF extracts in STZ-induced diabetic rats resulted in increased blood insulin levels and lower blood glucose levels.
Objectives : This study aimed to evaluate the effect of Atractylodis Rhizoma Alba water extract on streptozotocin (STZ)-induced diabetes in rats. Methods : Male Sprague-Dawley rats were divided into four groups ; normal, STZ-control and Atractylodis Rhizoma Alba (A) water extract-administrated group. Rats in which diabetic was induced by intraperitonal injection with STZ(60 mg/kg body weight). STZ-induced diabetic rats were orally administrated A extract daily for 5 weeks at doses of 200 or 500 mg/kg. Fasting blood glucose, total cholesterol, triglyceride and blood urea nitrogen were measured in sera of rats. Total volume of urine and urinary creatinine were also measured. Histopathological examination and immunohistochemical staining for the expression of insulin and ${\alpha}$-SMA in pancreas and kidney were performed, respectively. Results : There were no differences in body and kidney weights between STZ-control and A extract-administrated groups. However, serum triglyceride level was significantly decreased in A extract-administrated groups compared with those of STZ-control group. Histopathological analysis of pancreas and kidney revealed increased the number of islets and insulin-positive beta-cells in pancreas, and decreased morphological changes of glomerulus and ${\alpha}$-SMA expression in kidney after the administration of A extract. Conclusions : These results suggest that Atractylodis Rhizoma Alba has a biological action on STZ-induced diabetes in rats via decreasing the serum levels of total triglyceride, and suppressing the morphological changes of pancreas and kidney.
Objectives : This study aimed to compare the anti-diabetic efficacy of Lycii Radicis Cortex (Lycium chinense Mill.) and Corni Fructus (Cornus officinalis) on streptozotocin (STZ)-induced diabetes in rats. Methods : Male Sprague-Dawley rats were divided into four groups; normal, STZ-control, Lycii Radicis Cortex extract-administrated group (LRC) and Corni Fructus extract-administrated group(CF). Diabetes in rats was induced by intraperitonal injection with streptozotocin (STZ) at doses of, 30 mg/kg (body weight) for 5 days (once per a day). STZ-induced diabetic rats were orally administrated LRC and CF extract daily for 4 weeks at doses of 300 mg/kg. Fasting blood glucose, total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN) and creatinine were measured in sera of rats. Histopathological changes of kidney, liver and lung tissues were observed by microscope after H&E staining. Results : There were no differences in body and kidney weights, food intake and water intake in LRC- and CF-administrated groups compared with STZ control group. However, glucose, TC and TG levels in serum were significantly decreased in LRC-administrated groups compared with STZ-control group. In histopathological analysis of kidney, liver and lung, both LRC- and CF-administrated groups showed the inhibition of morphological damage. Conclusions : These results suggest that LRC and CF have a biological action on STZ-induced diabetes in rats via decreasing the serum TG and TG levels and may protect the morphological changes of kidney, liver and lung.

The purpose of this study was to investigate the effect of saponin in the shoot of Aralia elata on serum lipid level and glucose in streptozotocin(STZ) induced diabetic rats. Sprague Dawley normal male rats weighing 70$\pm$5g were randomly assigned to normal(control group), crude saponin(S group), and shoot of Aralia elata(D group). Experimental diets have been fed for 6 weeks. STZ induced diabetic rats were classified to diabetic control(DC group) and crude saponin(DS group). Diabetic rats were experimentally induced by intravenous injection of STZ(65mg/kg of body weight) dissolved in citrate buffer(pH 4.5). DS group has been i.p. injected with crude saponin solved in phosphate buffer(pH 7.0, 10mg/100g body weight) and DC group fed for 10 days. Body weight decreased significantly in crude saponin group. Feed intakes and feed efficiency ratio were not significantly different among C, S, and D group. The crude saponin group has indicated the lowest values of serum total cholesterol, glucose, and triglyceride. However, the values of serum glucose and triglyceride were not significant. Insulin levels among the crude saponin group, the shoot powder group, and the control group were not significantly different. When STZ induced diabetic rats have i.p. injection of crude saponin, the crude saponin has reduced the serum glucose but it is not been significant.
