  • This study was done to investigate the antioxidative effect of polygoni radix in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The ethanol extract of Polygoni radix was orally administrated once a day for 7 days. The contents of Total cholesterol, triglyceride (TG) were significantly decreased in Polygoni radix treated STZ-sample group compared with to the those of STZ-control group. The hepatic cytosolic activities of glutathione-s-transferase (GST), superoxide dismutase (SOD) were significantly increased, Also the content of glutathione (GSH) was incresaed in Polygoni radix treated STZ-sample group compared with to the those of STZ-control group. but the content of malondialdehyde (MDA) and the hepatic cytosolic activity of Catalase (CAT) were decreaed but not statistical significant. These results indicated that ethanol extract of polygoni radix was effective for the antidxidative in the STZ-induced diabetic rats.

  • The purpose of this study was to investigate the effect of pretreatment with Saegchinyanghyoltang(SYT) on the serum lipid composition and atherosclerotic index in streptozotocin(STZ)-induced diabetic rats. SYT pretreatment in STZ-induced diabetic rats inhibited the rise of serum glucose concentration. Serum total lipids and triglyceride levels in the STZ -induced diabetic rats were significantly higher than those in the control group. But in the group pretreated with SYT, triglyceride and lipid levels were significantly lower compared with those of STZ -induced diabetic rat group without STZ. However, the serum phospholipid levels were not statistically different among treatment groups. In the STZ-induced diabetic group, the serum total cholesterol, VLDL-, LDL-cholesterol levels and atherosclerotic index Were higher and HDL-cholesterol level was lower compared to the control group. However, these changes were prevented by SYT pretreatment Pretreatment with SYT significantly increased the activities of serum lipase compared to the STZ-treated group.

  • In our previous study (Kim et al, 1991), GLUT 4 protein content correlated negatively with plasma glucose levels in skeletal muscles of STZ-induced diabetic rats. Thus, in this study, to confirm whether expression of GLUT 4 correlate negatively with degree of hyperglycemia, we measured levels of GLUT 4 mRNA in red and white gastrocnemius muscles in STZ-induced mild and severe diabetic rats. Rats were randomly assigned to control, mild, and severe diabetic groups, and the diabetes was induced by intraperitoneal administration of STZ. The experiment was carried out 10 days after STZ administration. Gastrocnemius red and white muscles were used fur the measurement of GLUT 4 expression. Plasma glucose levels of mild and severe diabetic rats were increased compared to control rats (control, mild, and severe diabetes; $6.4{\pm}0.32,\;9.4{\pm}0.68,\;and\;22.0{\pm}0.58$ mmol/L, respectively). Plasma insulin levels of mild and severe diabetic rats were decreased compared to control rats (control, mild, and severe diabetes; $198{\pm}37,\;l14{\pm}14,\;and\;90{\pm}15$ pmol/L, respectively). GLUT 4 mRNA levels of gastrocnemius red muscles in mild and severe diabetic rats were decreased compared to control rats ($64{\pm}1.2%\;and\;71{\pm}2.0%$ of control, respectively), but GLUT 4 mRNA levels in gastrocnemius white muscles were unaltered in diabetic rats. In summary, GLUT 4 mRNA levels were decreased in STZ-induced diabetic rats but did not correlated negatively with degree of hyperglycemia, and this result suggest that the regulatory mechanisms of decreased GLUT 4 mRNA levels are hypoinsulinemia and/or other metabolic factor but not hyperglycemia. And regulation of GLUT 4 expression in STZ-induced diabetes between red and white enriched skeletal muscles may be related to a fiber specific gene regulatory mechanism.

  • The purpose of this study was to investigate the effect of Brousson etiae fructus (BF) on the urethral nitrite level and the urethral nitric oxide synthase (NOS) activity in streptozotocin (STZ) induced diabetic rats. In vitro, the urethral NOS activity was not noted in the level of Dose of extract prepared from BF. In vivo, the urethral NOS activity was increased in normal rats and STZ induced diabetic rats by dose and term of extract prerared from BF. The the NOS activity decreased in STZ induced diabetic rats was increased as highly as norma1 group by the extract of BF The level of urethral nitrite and glutathione was increased too. But the level of urethral lipid peroxide increased in STZ induced diabetic rats was decreased as lowly as normal group by the extract of BF. In conclusion, the extract of BF can restore erectile dysfunction of STZ induced diabetic rats.

  • The purpose of this study was to examine the effect of taurine supplementation time on the activity of the enzymes metabolizing lipid peroxide in the liver of streptozotocin(STZ)-induced diabetic rats, Sprague-Dawley male rats were fed the purified diet for 7 weeks. They were supplemented with or without 1% taurine in drinking water before or after STZ injection or during all experimental procedure. In comparison to diabetic group without taurine, glucose-6-phosphatase(G6Pase) activity was decreased in diabetic group supplemented with taurine before STZ injection, and it was increased in diabetic group supplemented with taurine after STZ injection, but the difference was not significant. Glutathione S-transferase(GST) activity was significantly increased by the injection of STZ. However, the GST activities of diabetic groups exposed to taurine after STZ injection or during all experimental procedure were significantly decreased. Glutathione peroxidase(GPx) activities was significantly decreased by STZ injection. However, only in diabetic group supplemented with taurine before STZ injection, GPx activities was not decreased by the STZ injection. These results suggest that taurine supplementation may change the activities of GSH-related enzymes metabolizing lipid peroxide in the liver of streptozotocin(STZ)-induced diabetic rats and that may be helpful for the prevention of diabetic complication.

  • Objectives : This study was performed to investigate the anti-diabetic effect of the fruit extract of Lycium chinense Mill(Lycii Fructus, LF) on multiple low-dose streptozotocin-induced diabetic rats. Methods : Male Sprague-Dawley rats were divided into three groups; normal, STZ-control, Lycii Fructus extractorally administrated 300 ㎎/㎏ group (STZ-LF). Diabetes was induced in rats by consecutive injection of streptozotocin (STZ) at doses of 30 ㎎/㎏ for 5 days. After 4 weeks, all rats were sacrificed, Fasting blood glucose, total cholesterol (CHO), triglyceride (TG) and HDL-Cholesterol were measured in sera of rats. Histopathological changes of pancreas, kidney, liver and lung tissues were observed by microscope after H&E, Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured. Results : There were no differences in body, food intake and water intake in LF-administrated groups compared with STZ control group. However, LF extract significantly decreased the levels of serum glucose, CHO, TG and HDL-Cholesterol in diabetic rats. In histopathological analysis of kidney, liver and lung, LF-administrated groups showed the inhibition of morphological damage. Conclusions : These results suggest that LF have a biological action on multi low-dose STZ-induced diabetes in rats via decreasing the serum glucose, TG and TG levels and may protect the morphological changes of kidney, liver and lung.

  • The aims of this study are conducted to investigate the effect of pretreatment with Achatina fucica extract (AFE) in streptozotocin (STZ)-induced diabetic rats. Serum glucose, total lipid and triglyceride levels of administration group of AE in diabetic rats induced by STZ were significant high levels than that of control group. However, glucose, triglyceride and lipid levels were significant lower level in group of pretreatment with AFE and Atheroslerotic index decreased in the group of treatment of AFE when those levels compared with that of STZ-treated group. Serum lipase activity was inhibited in the control group STZ induced diabetic rats, in contrast lipase activity increased in the group of AFE administration. These results suggested that AFE may use to prevent the diabetes mellitus induced by STZ.

  • This study was designed to examine the effects of Allium hookeri (AH) on plasma blood glucose and fat profile levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats through injection of streptozotocin (STZ) dissolved in citrate buffer into tail veins at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were then fed for 4weeks, with the experimental groups receiving a modified diet containing 5% or 10% powder derived from AH roots. The experimental groups were divided into four groups, consisting of a control group, STZ-control group, and diabetic fed with AH 5% & 10% treated groups. Rats' body weights, blood glucose, total cholesterol, HDL-cholesterol, triglyceride (TG), and free fatty acid (FFA) values in plasma were measured along with hematocrit (Hct) values and aminotransferase activities. Body weight losses were observed in the STZ-control group, whereas the STZ-AH group of diabetic rats gained weight. There was a significantly decrease in brain weight of the STZ-AH group but no significant differences in kidney and liver weights of the STZ-AH 5% & STZ-AH 10% groups compared to the STZ-control group. Blood glucose was significantly reduced in the STZ-AH 5% & STZ-AH 10% diabetic groups. There were no significant differences in total cholesterol and TG levels among the diabetic groups. HDL-cholesterol significantly increased while FFA significantly decreased in the STZ-AH 5% & STZ-AH 10% diabetic groups. The Hct level of the STZ-AH group was lower than that of the STZ-control group. Aspartate aminotransferase activity was significantly reduced in the STZ-AH 5% & STZ-AH 10% diabetic groups. These results indicate that supplementation with Allium hookeri root may have beneficial effects on diabetic complications as a potential therapeutic candidate.

  • The present study was designed to investigate the hypoglycaemic and hypolipidaemic activities of Qurs-e-Ziabetus 16 (QZ-16) in Streptozotocin (STZ) induced diabetic rats. QZ-16, a polypharmaceutical herbomineral formulation developed on the principles of Unani medicine is used for non-insulin dependent diabetes mellitus (NIDDM). The elevated levels of fasting blood glucose, serum levels of cholesterol, triglycerides and urea observed in rats treated with STZ (55 mg/kg body wt.) were significantly reduced by the treatment of QZ-16 (240 mg/kg, p.o.) and gliclazide (30 mg/kg, p.o.). The reduced HDL cholesterol levels were also increased by the QZ-16 and gliclazide treatments in the STZ induced diabetic rats. These data show that QZ-16 has hypoglycaemic, hypolipidaemic properties in STZ induced diabetic rats.
