Purification, and Biochemical and Biophysical Characterization of Cellobiohydrolase I from Trichoderma harzianum IOC 3844

  • Because of its elevated cellulolytic activity, the filamentous fungus Trichoderma harzianum has a considerable potential in biomass hydrolysis applications. Trichoderma harzianum cellobiohydrolase I (ThCBHI), an exoglucanase, is an important enzyme in the process of cellulose degradation. Here, we report an easy single-step ion-exchange chromatographic method for purification of ThCBHI and its initial biophysical and biochemical characterization. The ThCBHI produced by induction with microcrystalline cellulose under submerged fermentation was purified on DEAE-Sephadex A-50 media and its identity was confirmed by mass spectrometry. The ThCBHI biochemical characterization showed that the protein has a molecular mass of 66 kDa and pI of 5.23. As confirmed by smallangle X-ray scattering (SAXS), both full-length ThCBHI and its catalytic core domain (CCD) obtained by digestion with papain are monomeric in solution. Secondary structure analysis of ThCBHI by circular dichroism revealed ${\alpha}$- helices and ${\beta}$-strands contents in the 28% and 38% range, respectively. The intrinsic fluorescence emission maximum of 337 nm was accounted for as different degrees of exposure of ThCBHI tryptophan residues to water. Moreover, ThCBHI displayed maximum activity at pH 5.0 and temperature of $50^{\circ}C$ with specific activities against Avicel and p-nitrophenyl-${\beta}$-D-cellobioside of 1.25 U/mg and 1.53 U/mg, respectively.

Cubic zirconia single crystal growth using shell by skull melting method (스컬용융법에 의한 패각을 이용한 큐빅지르코니아 단결정 성장)

  • In this research, cubic zirconia is synthesized with a refined CaO from shells as a stabilizer through Skull melting method. The proper process time and concentration are defined by Hydration reaction to produce the refined CaO after two different treatments using 0.1 mol% of HCl respectively with Cockle shell. The highest purity of CaO is reached when the shell is immersed in 1 mol% HCl. In Hydration reaction step, the pure $Ca(OH)_2$ is produced at $45^{\circ}C$ for 24 hours. The highest purity of CaO is measured when the $Ca(OH)_2$ is treated by heat at $1200^{\circ}C$ for 5 hours. The single crystals are grown through Skull melting method by adding the different contents of the refined CaO from 10 mol% to 30 mol% into $ZrO_2$. The frequency of High-frequency oscillator used for Skull melting method is 3.4 MHz. The descending speed of the single crystal is 3 mm/hour. The grown length of the single crystal is 4 cm. As a result of this study, 15 mol% of CaO has the best crystallinity.

  • This paper describes a l0b CMOS A/D converter (ADC) for HDTV applications. The proposed ADC adopts a typical multi-step pipelined architecture. The proposed circuit design techniques are as fo1lows: A selective channel-length adjustment technique for a bias circuit minimizes the mismatch of the bias current due to the short channel effect by supply voltage variations. A power reduction technique for a high-speed two-stage operational amplifier decreases the power consumption of amplifiers with wide bandwidths by turning on and off bias currents in the suggested sequence. A typical capacitor scaling technique optimizes the chip area and power dissipation of the ADC. The proposed ADC is designed and fabricated in s 0.8 um double-poly double-metal n-well CMOS technology. The measured differential and integral nonlinearities of the prototype ADC show less than ${\pm}0.6LSB\;and\;{\pm}2.0LSB$, respectively. The typical ADC power consumption is 119 mW at 3 V with a 40 MHz sampling rate, and 320 mW at 5 V with a 50 MHz sampling rate.

Clinical Study of the Floating-Sinking Pulse Quantification Analysis on Ages, Left/Right, and Palpation Positions (20/60대 여성을 중심으로 살펴본 좌우 촌관척 부/침맥 정량화 임상연구)

  • Pulse diagnosis is a central diagnosis method used in traditional Oriental medicine. To standardize and modernize the pulse diagnosis method, it is essential to develop an instrument-based reinterpretation of the clinically used pulse images in terms of the physical quantities such as the strength, period, width, length, and depth of the pulse. As a step towards such standardization, we conducted a clinical study on the floating/sinking pulses based on an automated palpation instrument (3D-MAC, Daeyo Medi, Korea) for 213 female subjects in their 20s and 174 female subjects in their 60s. The floating/sinking pulses are the two representative pulse images depending only on the depth of the pulse, and can be conveniently scaled by the coefficient of the floating-sinking pulse ($C_{fs}{\in}(0,1)$), which represents how strong one should apply the hold-down pressure to obtain the maximal pulse strength. As a result, primarily we found that it tends to appear more floating-like pulse ($C_{fs}{\rightarrow}0$) at Gwan and more sinking-like pulse ($C_{fs}{\rightarrow}1$) at Cheek, at both age groups and at both wrists. This result is consistent with a previous study on the geometrical structure of the blood vessel by an ultrasonograph. Second, the pulse tends to be more sinking-like in the age group of 60s than 20s. Finally, the pulses at the right palpation positions were found to be more sinking-like than the left, at both age groups.

Characteristics of the media under a self-propelled compost turner in button mushroom cultivation (양송이버섯 재배시 자주식 배지교반기 활용 배지의 특성 및 수량성)

  • This study was conducted to investigate the characteristics of the medium used on the composting step, comparing the excavator agitator with the self-propelled turner. The temperature of the outdoor composting medium tended to increase rapidly after flipping in the turner. The late composting medium temperature was maintained at the excavator treatment area (farm practice), and the late composting effect progressed. During the field composting stage, various microorganisms such as Bacillus spp., Actinomycetes, fluorescent Pseudomonas spp., and filamentous fungi were distributed in the medium, and the density of aerobic bacteria involved in the decomposition of the medium was increased. Under high-temperature composting conditions, blue fungi, and mesophilic actinomycetes were inhibited or killed. Thermophilic actinomycetes, which play an important role in decomposing organic matter, showed higher densities than those observed in farm practices in the self-propelled turner process. The length of rice straw was slightly shorter when the self-propelled turner was used, and the water content did not show any significant difference between treatments. The a and b values tended to increase as the inverter was turned over. The CN ratio of the composting broth was lowered from 23.1 to 16.2 for the 5th turnover in the context of farming practices, and from 23.3 to 16.9 in the context of the self-propelled turner. The yield of each treatment was increased by 20% in 1 period, 28% in 2 periods, and 26% in 3 periods; the overall yield was 23%.

Wheel tread defect detection for high-speed trains using FBG-based online monitoring techniques

  • The problem of wheel tread defects has become a major challenge for the health management of high-speed rail as a wheel defect with small radius deviation may suffice to give rise to severe damage on both the train bogie components and the track structure when a train runs at high speeds. It is thus highly desirable to detect the defects soon after their occurrences and then conduct wheel turning for the defective wheelsets. Online wheel condition monitoring using wheel impact load detector (WILD) can be an effective solution, since it can assess the wheel condition and detect potential defects during train passage. This study aims to develop an FBG-based track-side wheel condition monitoring method for the detection of wheel tread defects. The track-side sensing system uses two FBG strain gauge arrays mounted on the rail foot, measuring the dynamic strains of the paired rails excited by passing wheelsets. Each FBG array has a length of about 3 m, slightly longer than the wheel circumference to ensure a full coverage for the detection of any potential defect on the tread. A defect detection algorithm is developed for using the online-monitored rail responses to identify the potential wheel tread defects. This algorithm consists of three steps: 1) strain data pre-processing by using a data smoothing technique to remove the trends; 2) diagnosis of novel responses by outlier analysis for the normalized data; and 3) local defect identification by a refined analysis on the novel responses extracted in Step 2. To verify the proposed method, a field test was conducted using a test train incorporating defective wheels. The train ran at different speeds on an instrumented track with the purpose of wheel condition monitoring. By using the proposed method to process the monitoring data, all the defects were identified and the results agreed well with those from the static inspection of the wheelsets in the depot. A comparison is also drawn for the detection accuracy under different running speeds of the test train, and the results show that the proposed method can achieve a satisfactory accuracy in wheel defect detection when the train runs at a speed higher than 30 kph. Some minor defects with a depth of 0.05 mm~0.06 mm are also successfully detected.

Development of Organ Culture Medium for Long Term Culture of Human Hair Follicle (인체 두피 모낭의 장기간 배양을 위한 기관 배양 배지의 개발)

  • We successfully isolated human anagen hair follicles from human scalp skin by microdissection and tried to culture them under various conditions. First we confirmed negative effect of serum on human hair follicle organ culture. As a next step serum-free medium compositions, Philpott medium, IMDM, and DHGM (Dongguk hair growth medium) were tried. Philpott medium is a general medium for hair organ culture based on Williams' E medium and DHGM is a special self-developed medium containing high amino acids and vitamins (B group) composition. As results, hair follicle in Philpott medium and IMDM showed anagen phase morphological structure, but rapid loss of hair elongation, low alkaline phosphatase expression, and very low expression of CK19. It is thought these hair follicles rapidly regressed from apoptosis. However, hair follicles in DHGM showed long term anagen phase morphological structure, continuous hair elongation, high alkaline phosphatase, and CK19 expression. These results demonstrate that high amino acids and vitamins (B group) composition are essential to in vitro long term human hair follicle organ culture and this culture medium will be useful in basic study of hair biology or application study to the development of alopecia treatment drugs.

Study on the Numerical Method of Classifying Treeforms : A case study of Zelkova serrata, Pinus strobus, and Magnolia denudata (주요조경수목의 수형분류방법에 관한 연구: 느티나무, 스트로브잣나무, 백목련을 대상으로)

  • Predicting treeform should be very important, eventhough it is difficult, step for landscape designers. This study was carried out to develop a method to classify the treeforms of landscape plants by numerical data. Twenty five treeforms are selected to be compared with 41 Zelkova serrata MAKINO, 26 Pinus strobus L., and 44 Magnolia denudata DESROUX. Statistical judgement was made by using X$^2$-Test. The results are summerized as follows. 1. Zelkova serrata was classified as B3 or C3. This species showed a tendency of changing from A type to B or C type, and the CL/CH ratio(canopy length/canopy height) is thought to be increased as aged. 2. pinus strobus was classified as D4-D5 and the CL/CH ratio tends to be increased as aged. 3. Magnolia denudata was classified as D4 and the change by aging was not observed clearly.

Analytical Study of Ultimate Behavior of Steel Cable-stayed Bridges (완성계 강사장교의 극한 거동의 해석적 연구)

  • This paper presents an investigation on the ultimate behavior of steel cable-stayed bridges using nonlinear finite element analysis method. Cable-stayed bridges exhibit various geometric nonlinearities as well as material nonlinearities, so rational nonlinear finite element analysis should be performed for investigation of the ultimate behavior. In this study, ultimate behavior of steel cable-stayed bridges was studied using rational ultimate analysis method. Nonlinear equivalent truss element and nonlinear frame element were used for modeling the cable, girder and mast. Moreover, refined plastic hinge method was adopted for considering the material nonlinearity of steel members. In this study, the 2-step analysis method was used. Before live load analysis, initial shape analysis was performed in order to consider the dead load condition. For investigation of the ultimate behavior of steel cable-stayed bridges, analysis models which span length is 920.0 m were used. Radiating type and fan type were considered as the cable-arrangement types. With various quantitative evidences such as load-displacement curves, deformed shapes, locations of the yield point or region, bending moment distribution and so on, the ultimate behavior of steel cable-stayed bridges was investigated and described in this paper.

Molecular Cloning and Expression of a Novel Protease-resistant GH-36 $\alpha$-Galactosidase from Rhizopus sp. F78 ACCC 30795

  • A 2,172-bp full-length gene (aga-F78), encoding a protease-resistant $\alpha$-galactosidase, was cloned from Rhizopus sp. F78 and expressed in Escherichia coli. The deduced amino acid sequence shared highest identity (45.0%) with an $\alpha$-galactosidase of glycoside hydrolase family 36 from Absidia corymbifera. After one-step purification with a Ni-NTA chelating column, the recombinant Aga-F78 migrated as a single band of ~82 and ~210 kDa on SDS-PAGE and nondenaturing gradient PAGE, respectively, indicating that the native structure of the recombinant Aga-F78 was a trimer. Exhibiting the similar properties as the authentic protein, purified recombinant Aga-F78 was optimally active at $50^{\circ}C$ and pH 4.8, highly pH stable over the pH range 5.0-10.0, more resistant to some cations and proteases, and had wide substrate specificity (pNPG, melidiose, raffinose, and stachyose). The recombinant enzyme also showed good hydrolytic ability to soybean meal, releasing galactose of $415.58\;{\mu}g/g$ soybean meal. When combined with trypsin, the enzyme retained over 90% degradability to soybean meal. These favorable properties make Aga-F78 a potential candidate for applications in the food and feed industries.
