Monitoring of Feed-Nutritional Components, Toxic Heavy Metals and Pesticide Residues in Mushroom Substrates According to Bottle Type and Vinyl Bag Type Cultivation (버섯의 봉지재배 및 병재배 시 재배단계별 배지의 사료영양적 성분, 독성중금속 및 잔류농약 모니터링)

  • This study was carried out to monitor feed-nutritional components, toxic heavy metals (Cd, Pb and As) and pesticide residues through three cultivation stages (1st initial culture stage, 2nd mycelial growth stage, and 3rd fruit body-harvested stage) of king oyster mushroom (Pleurotus eryngii) produced by bottle type cultivation and oyster mushroom (Pleurotus osteratus) produced by vinyl bag type cultivation. For both cultivation types, compared with the initial culture, the weight reduction rate in spent mushroom substrates (SMS) after fruit body harvest was 29% for total wet mass, 21~25% for dry and organic matters and 19 ~22% for neutral detergent fiber. Two thirds to 3/4 of organic matter degraded and utilized by mycelia and fruit bodies was originated from fiber, of which the primary source (50~70%) was hemicellulose. The effect of mycelial growth stage on chemical compositional change in culture was little (P>0.05) for bottle type cultivation of king oyster mushroom but considerable (P<0.05) for vinyl type cultivation of oyster mushroom. Culture nutrients uptake by fruit bodies was very active for the bottle type cultivation. Compared with SMS, harvested fruit bodies (mushrooms) contained higher (P<0.05) crude protein, non-fibrous carbohydrate, and crude ash and lower (P<0.05) neutral detergent fiber. Regardless of stages, no culture samples were contaminated with toxic heavy metals and pesticide residues. In conclusion, the increase of fiber (neutral and acid detergent fibers) and indigestible protein contents and the decrease of true protein content in SMS indicated that the feed-nutritional value of SMS was significantly reduced compared with that of the initial culture and they were safe from toxic heavy metals and pesticide residues.

Dry Matter Yields of the Hybrids Associated with Festulolium braunii (Festulolium braunii 교잡종의 재배특성, 사료가치 및 수량성)

  • This experiment was carried out in order to examine the winterhardiness, heading date, fresh and dry matter yeild of Festulolium braunii, a hybrid of Festuca pratensis and Lolium multiflum, which is newly introduced in Korea. This experiment was performed at National Livestock Research Institute in Suwon and Namwon from September 2004 to July 2005. The results are as follows. 'Fleurial', 'Boxer', and 'Perun' showed stronger winterhardiness than 'Hwasan 101', which is one of the strongest in the winterhardiness hybrids. Therefore, we do not see there is any problem in winterhardiness when these hybrids are cultivated in Korea. The hybrids are turned out to be mid and late maturing hybrid. The average heading dates of the three hybrids were May 25 in Namwon and May 27 in Suwon. These dates are lower($1{\sim}3$ days) than that of Hwasan 101. They showed $91{\sim}96cm$ in leaf length at 1st cutting times which are comparatively long. This experiment also disclosed that their lodging and regrowth are strong and excellent. We also found out that there are significant difference between regions and hybrids in the total fresh yield per ha. The average fresh yield of Suwon was 83,222 kg/ha and that of Namwon was 69,139 kg/ha. We could not see any difference in the dry matter yield affected by region, however, there were significant difference in hybrids(p<0.05). The crude protein contents of the three hybrids indicated very high figures. the crude protein cotents of Perun, Fleurial and Boxer are 18.0% 17.4% and 16.7% respectively. However the ADF (Acid Detergent Fiber) shows $28.1{\sim}29.2%$ which is less contents than 33.2% of Hwasan 101. We found out from this experiment that these three hybrids have excellent feed quality because the NDF (Neutral Detergent Fiber) is decreased by $7{\sim}3.8%$ comparing that the NDF of Hwasan 101 is 58.3%. Also each hybrids contains more crude fat by 1%, a little more chemical composition and less ash contents. Thus we can tern that chemical composition contents of these hybrids are also fittable.

Evolutionary Explanation for Beauveria bassiana Being a Potent Biological Control Agent Against Agricultural Pests

  • Beauveria bassiana (Cordycipitaceae, Hypocreales, Ascomycota) is an anamorphic fungus having a potential to be used as a biological control agent because it parasitizes a wide range of arthropod hosts including termites, aphids, beetles and many other insects. A number of bioactive secondary metabolites (SMs) have been isolated from B. bassiana and functionally verified. Among them, beauvericin and bassianolide are cyclic depsipeptides with antibiotic and insecticidal effects belonging to the enniatin family. Non-ribosomal peptide synthetases (NRPSs) play a crucial role in the synthesis of these secondary metabolites. NRPSs are modularly organized multienzyme complexes in which each module is responsible for the elongation of proteinogenic and non-protein amino acids, as well as carboxyl and hydroxyacids. A minimum of three domains are necessary for one NRPS elongation module: an adenylation (A) domain for substrate recognition and activation; a tholation (T) domain that tethers the growing peptide chain and the incoming aminoacyl unit; and a condensation (C) domain to catalyze peptide bond formation. Some of the optional domains include epimerization (E), heterocyclization (Cy) and oxidation (Ox) domains, which may modify the enzyme-bound precursors or intermediates. In the present study, we analyzed genomes of B. bassiana and its allied species in Hypocreales to verify the distribution of NRPS-encoding genes involving biosynthesis of beauvericin and bassianolide, and to unveil the evolutionary processes of the gene clusters. Initially, we retrieved completely or partially assembled genomic sequences of fungal species belonging to Hypocreales from public databases. SM biosynthesizing genes were predicted from the selected genomes using antiSMASH program. Adenylation (A) domains were extracted from the predicted NRPS, NRPS-like and NRPS-PKS hybrid genes, and used them to construct a phylogenetic tree. Based on the preliminary results of SM biosynthetic gene prediction in B. bassiana, we analyzed the conserved gene orders of beauvericin and bassianolide biosynthetic gene clusters among the hypocrealean fungi. Reciprocal best blast hit (RBH) approach was performed to identify the regions orthologous to the biosynthetic gene cluster in the selected fungal genomes. A clear recombination pattern was recognized in the inferred A-domain tree in which A-domains in the 1st and 2nd modules of beauvericin and bassianolide synthetases were grouped in CYCLO and EAS clades, respectively, suggesting that two modules of each synthetase have evolved independently. In addition, inferred topologies were congruent with the species phylogeny of Cordycipitaceae, indicating that the gene fusion event have occurred before the species divergence. Beauvericin and bassianolide synthetases turned out to possess identical domain organization as C-A-T-C-A-NM-T-T-C. We also predicted precursors of beauvericin and bassianolide synthetases based on the extracted signature residues in A-domain core motifs. The result showed that the A-domains in the 1st module of both synthetases select D-2-hydroxyisovalerate (D-Hiv), while A-domains in the 2nd modules specifically activate L-phenylalanine (Phe) in beauvericin synthetase and leucine (Leu) in bassianolide synthetase. antiSMASH ver. 2.0 predicted 15 genes in the beauvericin biosynthetic gene cluster of the B. bassiana genome dispersed across a total length of approximately 50kb. The beauvericin biosynthetic gene cluster contains beauvericin synthetase as well as kivr gene encoding NADPH-dependent ketoisovalerate reductase which is necessary to convert 2-ketoisovalarate to D-Hiv and a gene encoding a putative Gal4-like transcriptional regulator. Our syntenic comparison showed that species in Cordycipitaceae have almost conserved beauvericin biosynthetic gene cluster although the gene order and direction were sometimes variable. It is intriguing that there is no region orthologous to beauvericin synthetase gene in Cordyceps militaris genome. It is likely that beauvericin synthetase was present in common ancestor of Cordycipitaceae but selective gene loss has occurred in several species including C. militaris. Putative bassianolide biosynthetic gene cluster consisted of 16 genes including bassianolide synthetase, cytochrome P450 monooxygenase, and putative Gal4-like transcriptional regulator genes. Our synteny analysis found that only B. bassiana possessed a bassianolide synthetase gene among the studied fungi. This result is consistent with the groupings in A-domain tree in which bassianolide synthetase gene found in B. bassiana was not grouped with NRPS genes predicted in other species. We hypothesized that bassianolide biosynthesizing cluster genes in B. bassiana are possibly acquired by horizontal gene transfer (HGT) from distantly related fungi. The present study showed that B. bassiana is the only species capable of producing both beauvericin and bassianolide. This property led to B. bassiana infect multiple hosts and to be a potential biological control agent against agricultural pests.

  • Object : Cigarette smoking is a major cause of mucus hypersecretion, which is a pathophysiological feature of many inflammatory airway diseases. Mucins, which are an important part of the airway mucus, are synthesized from the Muc gene in airway epithelial cells. However, the signaling pathways for cigarette smoke-induced mucin synthesis are unknown. The aim of this study was to determine the signal pathway for smoking induced Muc5ac gene expression. Methods : A549 cells were cultured and transiently transfected with the Muc5ac promoter fragment. These cells were stimulated with 5% cigarette smoke extract (CSE) alone or with CSE after a pretreatment with various signal transduction pathway inhibitors (AG1478, PD98059 and SB203580). The Muc5ac promoter activity was examined using the luciferase reporter system, and the level of phosphorylated EGFR, ERK1/2, p38 MAPK and JNK were all examined using Western blot analysis. Muc5ac mRNA expression was also examined using reverse transcriptase polymerase chain reactions (RT-PCR). Results : 1. The peak level of luciferase activity of the Muc5ac promoter was observed at 5% concentration and after 3 hours of incubation with the CSE. The level of EGFR phosphorylation and the luciferase activity of the transfected cells caused by the CSE were significantly suppressed by AG1478 or PD98059 (P<0.01). 2. CSE phosphorylated ERK1/2 or p38 MAPK but not JNK. The Muc5ac mRNA expression level was increased by the CSE but that was suppressed by PD98059 or AG1478. 3. The CSE-induced phosphorylation of ERK1/2 was blocked by PD98059 and that of p38 MAPK was blocked by either PD98059 or SB203580. Either PD98059 or SB203580 suppressed the luciferase activity of the transfected cells (P<0.0001). Conclusion : The Muc5ac mRNA expression level was increased by the CSE. The increased CSE-induced transcriptional activity was mediated via EGF receptor activation, which led to ERK1/2 and p38 MAPK phosphorylation.

Socioeconomic Factors Relating to Obesity and Inadequate Nutrient Intake in Women in Low Income Families Residing in Seoul (서울지역 저소득층 여성의 비만 및 영양소 섭취 부족과 관련된 사회경제적 요인)

  • This study was performed to investigate socioeconomic factors relating to obesity and inadequate nutrient intake in women in low income families residing in Seoul. The subjects were 125 women (aged 41-87 y) recruited from the local health center for free health examination for low income families. The socioeconomic status such as educational level, income level, and housing condition were poor. For subjects aged less than 65 years, the prevalence of obesity was 44% based on BMI (${\geq}\;25\;kg/m^2$), 48% based on WHR (${\geq}\;0.85$), and 50% based on waist circumference (${\geq}\;80\;cm$) and for those more than 65 years, these were greater and 57%, 81%, and 79%, respectively. The main food sources of daily diet were vegetables, grains, and fruits. Energy and other nutrient intake was not adequate for all subjects and the inadequacy was more profound in the elderly, showing percentages of subjects whose intake was less than EAR were greater than 50% for all nutrients except for iron and below RI were also greater than 50% for all nutrients. The prevalence of obesity and nutrient inadequacy were not associated with socioeconomic status in subjects aged less than 65 years, however, obesity was associated with household income and nutrient inadequacy was related to education (vitamin A) and housing status (protein, phosphate, and iron) in the elderly. After adjustment for ages, in the elderly, OR for obesity (BMI ${\geq}$ 25, OR = 12.601; 95% CI = 2.338-67.911) and central obesity (WC ${\geq}$ 80 cm, OR = 4.778; 95% CI = 1.103-20.696) were greater in subjects who earned less than 500 thousand Won per month than who earned more. For inadequate nutrient intake, the OR for inadequate intake of Vitamin A (OR = 4.555; 95% CI = 1.491-13.914) was greater in subjects with no education than those educated. Subjects without her own house had greater risk for inadequate intake for protein (OR = 3.660; 95% CI = 1.118-11.981), phosphate (OR = 3.428; 95% CI = 1.157-10.158), and iron (OR = 3.765; 95% CI = 1.205-11.766) than subjects possessing her house. In elderly females in low income families, the socioeconomic status was associated with the risk for obesity (income level) and inadequate nutrient intake (education level and housing status). More attention on these groups should be given for prevention of obesity and inadequate nutrient intake.

Neuroprotective Effects of Modified Yuldahanso-tang (MYH) in a Parkinson's Disease Mouse Model (MPTP로 유도된 Parkinson's disease 동물 모델에서 열다한소탕 가감방 (MYH)의 신경 세포 보호 효과)

  • Objectives To evaluate the neuroprotective effects of modified Yuldahanso-tang (MYH) in a Parkinson's disease mouse model. Methods 1) Four groups (each of 8 rats per group) were used in this study. 2) The neuroprotective effect of MYH was examined in a Parkinson's disease mouse model. C57BL/6 mice treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP, 30 mg/kg/day), intraperitoneal (i.p.) for 5 days. 3) The brains of 2 mice per group were removed and frozen at $-20^{\circ}C$, and the striatum-substantia nigra part was seperated. The protein volume was measured by Bradford method following Bio-Rad protein analyzing kit. Using mouse/Rat Dopamine ELISA Assay Kit. 4) The brains of 2 mice per group were separated and removed. TH-immunohistochemical was examined in the MPTP-induced Parkinson's disease mice to evaluate the neuroprotective effects of MYH on ST and SNpc. 5) Two mice out of each group were anesthetized and skulls were opened from occipital to frontal direction to take out the brains. The brains added TTC solution for 20 minutes for staining. 6) The water tank used for morris water maze test was filled with $28^{\circ}C$ water, and a round platform of 10cm in diameter was installed for mice to step on. The study was carried out once a day within 30 seconds, keep exercising to step on the platform in the pool. 7) The brains of two mice out of each group were fixed in 10% formaldehyde solution and paraphillin substance was infiltrated. They were fragmented by microtome, and observed under an optical microscope after Hematoxylin & Eosin staining. 8) A round acrylic cylinder with its upper side open was filled with clean water and depressive mouse models were forced to swim for 15 minutes. After 24 hours the animals were put in the same equipment for 5 minutes and were forced to swim. 9) The convenient, simple, and accurate high-performance liquid chromatography (HPLC) method was established for simultaneous determination of Neurotransmitters in MPTP-MYH group. Results 1) MYH possess Dopamine cell protective effect on MPTP-induced injury in striatum and substantia nigra pars compacta. 2) MYH inhibits the loss of tyrosine hydroxylase-immunoreacitive (TH-IR) cells in the striatum and substantia nigra pars compacta on MPTP-induced injury in C57BL/6 mice. 3) MYH possesses improvement effect on MPTP-induced memory deterioration in C57BL/6 mice through the reduction of prolongated Sort of lost time by MPTP injection using the Morris water maze test. 4) MYH possesses hippocampal neuron protective effect on MPTP-induced injury in C57BL/6 mice. 5) MYH possesses improvement effect on MPTP-induced motor behaviour deficits and depression in C57BL/6 mice through the reduction of prolongated losing motion by MPTP injection using the Forced swimming test. 6) MYH increases serotonin product amount on MPTP-induced injury in C57BL/6 mice. Conclusions This experiment suggests that the neuroprotective effect of MYH is mediated by the increase in Dopamin, TH-ir cell, Hippocampus and Serotonin. Furthermore, MYH essential oil may serve as a potential preventive or therapeutic agent regarding Parkinson's disease.

Growth and Development of the Academic Societies and Animal Production in Korea, China and Asia over the Last 50 Years

  • The Korean Society of Animal Science (KSAS) was officially born on October 8, 1956 under the leadership of Professor Sang W. Yun of Seoul National University, Seoul, Korea a few years after the end of the Korean War. At that time, there were 0.9 million Korean native cattle, 1.3 million pigs and 8.9 million chickens in Korea. Per capita income for Korea (US$ 66) or China (US$ 59) was about 10% of Asian's average income (US$ 513) in 1956. Korea produced less than 0.2 million M/T of formula feed and consumed 6.1 kg/person/year of animal products. One could say that Korea was an example of an under-developed country in the world. Although the first issue of the Proceedings of the KSAS was published on October 28, 1958, regular quarterly journals of the KSAS were not published until March 1, 1969. Major activities other than publishing its journal were: holding an annual meeting and/or scientific forum at national or international level. The Asian-Australasian Association of Animal Production Societies (AAAP) was founded on September 1, 1980 at Kuala Lumpur, Malaysia with founding members from Australia, Indonesia, Japan, Korea, Malaysia, NZ, Philippines and Thailand. Thirteen AAAP Animal Science Congresses have been held in its 28 year history. Hosting countries were Malaysia (1980), Philippines (1982), Korea (1985), NZ (1987), Taiwan (1990), Thailand (1992), Indonesia (1994), Japan (1996), Australia (2000), India (2002), Malaysia (2004), Korea (2006) and Vietnam(2008). In 1988, significant progress of the AAAP was made by creating an official English journal of the AAAP entitled "Asian-Australasian Journal of Animal Sciences (AJAS)" under the initiative of the KSAS. This journal is now published monthly and distributed to more than 50 countries in the AAAP region and the world. It should be mentioned that the KSAS was able to successfully host the $3^{rd}$ AAAP Animal Science Congress in 1985 and the 12th in 2006, as well as the $8^{th}$ WCAP in 1998. During the last 50 years of KSAS history, per capita income of Korea increased to US$ 17,690 (268 fold), formula feed production increased to 15 million M/T (97 fold) and consumption of animal products increased to 105 kg/person/year (17 fold). Cattle, pig and chicken numbers also increased to 2.5 million (2.8 fold), 9 million (7.4 fold) and 119 million (13 fold). This trend was also found for China and Asia, even if the rate of growth was slightly lower than that of Korea. It is expected that a similar rate of growth in economics, animal numbers, formula feed production and animal protein intake will likely be achieved by other Asian countries in the $21^{st}$ century with somewhat lower rate of development than that of Korea.

Evaluation of Productivity and Silage Quality for Domestically Developed Forage Crops in Korea (국내 육성 사료작물의 생산성 및 사일리지 품질 평가)

  • This experiment was conducted to evaluate the productivity and silage quality of domestically developed forage crops in Korea. A variety of rye ("Gogu"), oat ("Samhan"), three Italian ryegrass ("Kogreen", "Kospeed" and "Kowinearly") and two barley ("Yuyeon" and "Youngyang") were evaluated at an experimental field of Grassland and Forage Division, National Institute of Animal Science, RDA in Korea. Gogu showed the highest dry matter yield among the forage crop but the lowest was Samhan. Crude protein (CP) was higher in the oat and Italian ryegrass varieties. The total digestible nutrient (TDN) content levels were similar with the except for the rye. The acidity (pH) of the oat silage was the lowest among forage crop species. Higher lactic acid content levels were found in the Kogreen and Samhan compared to the other varieties (p<0.05). According to Flieg's score, all of the silages were assigned a $1^{st}$ quality grade with the exception of the Gogu. These results suggest that domestically developed forage crop varieties show high performance in terms of productivity and silage quality, and are therefore recommendable forage crop varieties for Korea.

Distributional Shape of Food Intake and Nutrition Data for Adults and Children (성인과 어린이의 식품섭취와 영양소 섭취량의 분포에 대한 연구)

  • Food intake data from 228 persons (96 male adult ranging in age from 19 to 54, 27 female adult ranging in age from 20 to 46, 54 boys ranging in age from 9 to 11, and 51 girls ranging in age from 8 to II) were studied with respect to the shape of the underlying probablity distributions. For each menu items distributional shapes of food intake were different. Most of distributions for food intakes from normaJ distributions. From food intake data of 2 meals nutrition intake data are calculated. For each meal, energy, protein, fat, carbohydrate, fiber, calcium, iron, vitamin A, thiamin, ribofavin, niacin and vitamin C were computed and thier distributions were compared with normal distributions. Distributions for adult female showed normal distributions for some food items. For nutrient intake data from male adults, distributions for vitamin C from 1st meal and calcium from 2nd meal were marginal and the remains were differed from normal distributions. For adult female and childern, distiributions for some nutients were differed from normal distributions. It is hard to find special patterns for each nutrient distributions. Therefore the normal distributions assumptions should be verified prior to applying parametric techniques to thier data. If those assumptions are not valid, non-parametric techniques should be used to analyze their data.

  • A survey of nutrition and incidence of parasites was conducted from January 23 to 31 and February 14 to 18, 1981, to determine the nutritional status of young children under the age of 6 years, in one of the rural family health project areas: Whaseoung-gun, Kyunggi-do, Korea. This study covered a total of 233 children. The results are summarised as follows: Approximately one-half of these families had an annual income between \1,000,000 and \2,500,000. Nearly 84% of the households were involved in farming. Energy intake of children met 70.6% of the requirement: carhohydrates provided 77%, protein provided 11%, and fat provided 12% of the total energy intake. Low intake of iron; calcium; and vitamins, excluding thiamin, was also found. Mean hemoglobin value was $10.6{\pm}1.0gm/100ml.;\;42%$ of subjects were categorized as low group, 22% were in the deficient group, and only 36% were at an acceptable level as indicated by O'Neal. Thus, 22% of the subjects were proven to be anemic by the hemoglobin criteria. Mean hematocrit value was $30.1{\pm}2.8%$, and 80 of 208 children, or 38.4% of subjects, were below 30% of the standard established by O'Neal. The positive prevalence of parasitic infection was 20% , significantly higher than that observed in the Yowido compound (5.5%) in 1980. Hematologic findings had no significant correlation to parasitic infection. The majority, 95.2% of the subjects, met or surpassed 90% of the KlST's standard of height:whtile 28. 6% of the subjects were below 90% of the KIST's standard of weight. The ratio of weight to height for boys 4 to 6 years old was 15.8 to 17.5, in contrast to 16.3 to 16.9 for boys of KHDI project areas, and also in contrast to 17.7 to 17.9 for boys of the same age group from the Yowido apartment compound in Seoul. According to the results of the assessments, the authors found a clear difference in growth values, hematologic findings and dietary patterns between rural areas and advantaged urban areas of Seoul. Our results suggest the need to develop the nutrition and parasite control training of village women for better nutritional care of their young children.
