Novel SSF Process for Ethanol Production from Microcrystalline Cellulose Using the $\delta$-Integrated Recombinant Yeast, Saccharomyces cerevisiae L2612$\delta$GC

  • A novel simultaneous saccharification and fermentation (SSF) process from the microcrystalline cellulose to ethanol was developed by using $\delta$-integrated recombinant cellulolytic Saccharomyces cerevisiae L2612$L2612\deltaGC$, which can utilize cellulose as carbon and energy sources. The optimum amount of enzymes needed for the efficient conversion of cellulose to ethanol at $30^{\circ}C$ was determined with commercial cellulolytic enzymes. By fed-batch cultivation, the heterologous cellulolytic enzymes were accumulated up to 42.67% of the total cellulase and 29% of the $\beta$-glucosidase needed for the efficient SSF process. When this $\delta$-integrated recombinant yeast was applied to the successive SSF step for ethanol production, 20.35 g/l of ethanol was produced after 12 h from 50 g/l of microcrystalline cellulose. By using this novel SSF process, a considerable amount of commercial enzymes was reduced.

Bioethanol Production from Seaweed Kappaphycus alvarezii by Simultaneous Saccharification and Fermentation (홍조류(Kappaphycus alvarezii)의 동시 당화 발효를 이용한 바이오에탄올의 생산)

  • Thermal acid hydrolysis pretreatment of Kappaphycus alvarezii was carried out with 12% (w/v) seaweed slurry and 180 mM H2SO4 at 140°C for 5 min. Utility of the thermotolerant yeast Kluyveromyces marxianus KCTC7150 was evaluated with respect to cell growth and ethanol fermentation at 40°C was close to optimal for enzymatic hydrolysis. This could lead to the integration of both the saccharification and fermentation processes. The levels of ethanol production by simultaneous saccharification and fermentation (SSF) with non-adapted and adapted K. marxianus KCTC7150 were 9.1 g/l with an ethanol yield (YEtOH) of 0.24 and 10.2 g/l with an ethanol yield (YEtOH) of 0.27 at 156 h, respectively. The two-phase SSF process was employed in this study to improve the efficiency of ethanol fermentation. Adapted K. marxianus KCTC7150 using the two-phase SSF process produced 13.5 g/l with an ethanol yield (YEtOH) of 0.35 at 96 h. Development of the two-phase SSF process could enhance the overall ethanol fermentation yields of the seaweed K. alvarezii.

  • Recently, many automobile parts produced by semi-solid forming(SSF) process has been applied for improved weight reduction, better environmental protection and energy savings. SSF process was well developed for high volume production of light weight aluminum components. In this paper, knuckle has been manufactured with A357 by SSF and thor investigated for microstructures and mechanical properties followed by various heat-treatment conditions. It was found that the examined microstructure was equiaxed at the whole cross-section area and as a result, the mechanical properties were satisfied by 100MPa YS, 260MPa UTS and $14\%$ elongation.

  • Modeling and simulation for simultaneous saccharification and fermentation (SSF) process in bioconversion of paper mill sludge to lactic acid was carried out. The SSF process combined the enzymatic hydrolysis of paper mill sludge into glucose and the fermentation of glucose into lactic acid in one reactor. A mathematical modeling for cellulose hydrolysis was developed, based on the proposed mechanism of cellulase adsorption deactivation. Another model for simple lactic acid fermentation was also made. A whole mathematical model for SSF was developed by combining the above two models for cellulose hydrolysis and lactic acid fermentation. The characteristics of the SSF process were investigated using the mathematical model.

  • Semi-solid forging(SSF) process of A356 aluminium alloy has been studied to assess the effect of process variables on the component integrity. Semi-solid material(SSM) was fabricated by mechanical and electro-magnetic stirring process. The fabricated SSM by using mechanical stirring process has been carried out on cooling rate of 0.022.deg. C/sec 0.0094.deg. C/sec and stirring speed n=600, 1000 rpm, respectively. The fabricated SSM by using electro-magnetic stirring process is supplied by Pechiney. The holding time and temperature in the semi-solid state before forging also affects the globular microstructure of alloy. Therefore, the influence of these two parameters is discussed in terms of the microstructure of alloy. The SSF process has been conducted with three different die temperatures($T_{die}$=250.deg. C, 300.deg. C, 350.deg. C) and two kinds of gate types(straight gate and orifice gate). This paper is to investigate the influence of gate shapes of die on filling phenomena in SSF process more deeply. The mechanical properties of forged components were also investigated for variation of process conditions such as die temperature, gate shape and SSM.

  • The technology of Semi-Solid Forging (SSF) has been actively developed to fabricate near-net- shape products using light and hardly formable materials. Generally, the SSF process is composed of slug heating, forming, compression holding and ejecting step. After forming step in SSF, the slug is compressed during a certain holding time in order to be completely filled in the die cavity and be accelerated in solidification rate. This paper presents the analysis of temperature, solid fraction and shrinkage at compression holding step for a cylindrical slug, then predicts the solidification time to obtain the final shaped part. Enthalpy-based finite element analysis is performed to solve the heat transfer problem considering phase change in solidification.

SSF: Sentence Similar Function Based on word2vector Similar Elements

  • In this paper, to improve the accuracy of long sentence similarity calculation, we proposed a sentence similarity calculation method based on a system similarity function. The algorithm uses word2vector as the system elements to calculate the sentence similarity. The higher accuracy of our algorithm is derived from two characteristics: one is the negative effect of penalty item, and the other is that sentence similar function (SSF) based on word2vector similar elements doesn't satisfy the exchange rule. In later studies, we found the time complexity of our algorithm depends on the process of calculating similar elements, so we build an index of potentially similar elements when training the word vector process. Finally, the experimental results show that our algorithm has higher accuracy than the word mover's distance (WMD), and has the least query time of three calculation methods of SSF.

  • As a process to utilize agricultural residues, simultaneous hydrolysis-fermentation (SSF) was compared with fermentation of enzymic hydrolyzate using koji cultures of Trichoderma sp. KI 7-2 and a thermotolerant yeast Saccharomyces cerevisiae NCYC 716. Cellobiose was not detected in SSF broth whilst 15 mg/$m\ell$ of the disaccharide was found in enzymic hydrolysate of rice straw using the same enzyme source. It was found that converting glucose to ethanol in SSF process reactivated the cellobiase activity, which is inhibited by the accumulation of glucose in enzymic hydrolysis process. Cutting milled rice straw was fermented as effectively as ball milled one in SSF process. From tile results discussions are made on the product inhibition mechanism of cllulolytic enzyme system.

  • As the manufacturing processes of automotive engine piston, gravity die-casting, squeeze casting, hot forging and powder forging process are generally used for the various specifications. As the semi-solid forming(SSF) is compared with conventional casting such as gravity die-casting and squeeze casting for the characteristics of its process, the product without inner defects such as gas porosity and segregation can be obtained and its microstructure is globular grain. In SSF process, the materials are heated up to the temperature between the solvus and liquidus line at which the materials exists in the form of liquid-solid mixture. In this time, Discussion is given about reheating process of row material and results are presented regarding accurate temperature and process variables controlling for right solid fractions.
