AFLP 방법을 이용한 담배 버어리종 특이 프라이머의 개발 (Identification of tobacco Burley species specific marker in several tobacco species by AFLP)

  • AFLP(Amplified Fragment Length Polymorphism) analysis was conducted to cultivars of tobacco, Nicotiana tabacum in order to select the cultivar-specific markers. AFLP results using 12 primer sets revealed genetic diversity among 12 field grown tobacco cultivars. Polymorphic fragments amplified by PCR was purified and cloned to identify their nucleotide sequences. From the sequences of them, 40 primer sets were designed to select cultivar-specific markers. When genomic DNA isolated from tobacco were used as PCR template, a set of primers, BrSF/BrSR showed Burley-specific band patterns. The results indicate that AFLP technique used in this experiments is useful for identifying tobacco cultivars in a rapid manner.

Novel Systemic Therapies for Advanced Gastric Cancer

  • Gastric cancer (GC) is the second leading cause of cancer mortality and the fourth most commonly diagnosed malignant diseases. While continued efforts have been focused on GC treatment, the introduction of trastuzumab marked the beginning of a new era of target-specific treatments. Considering the diversity of mutations in GC, satisfactory results obtained from various target-specific therapies were expected, yet most of them were unsuccessful in controlled clinical trials. There are several possible reasons underlying the failures, including the absence of patient selection depending on validated predictive biomarkers, the inappropriate combination of drugs, and tumor heterogeneity. In contrast to targeted agents, immuno-oncologic agents are designed to regulate and boost immunity, are not target-specific, and may overcome tumor heterogeneity. With the successful establishment of predictive biomarkers, including Epstein-Barr virus pattern, microsatellite instability status, and programmed death-ligand 1 (PD-L1) expression, as well as ideal combination regimens, a new frontier in the immuno-oncology of GC treatment is on the horizon. Since the field of immuno-oncology has witnessed innovative, practice-changing successes in other cancer types, several trials on GC are ongoing. Among immuno-oncologic therapies, immune checkpoint inhibitors are the mainstay of clinical trials performed on GC. In this article, we review target-specific agents currently used in clinics or are undergoing clinical trials, and highlight the future clinical application of immuno-oncologic agents in inoperable GC.

Molecular markers based on chloroplast and nuclear ribosomal DNA regions which distinguish Korean-specific ecotypes of the medicinal plant Cudrania tricuspidata Bureau

  • Cudrania tricuspidata Bureau is a widely-used, medicinal, perennial and woody plant. Obtaining information about the genetic diversity of plant populations is highly important with regard toconservation and germplasm utilization. Although C. tricuspidata is an important medicinal plant species registered in South Korea, no molecular markers are currently available to distinguish Korean-specific ecotypes from other ecotypes from different countries. In this study, we developed single nucleotide polymorphism (SNP) markers derived from the chloroplast and nuclear genomic sequences, which serve to to identify distinct Korean-specific ecotypes of C. tricuspidata via amplification refractory mutation system (ARMS)-PCR and high resolution melting (HRM) curve analyses. We performed molecular authentication of twelve C. tricuspidata ecotypes from different regions using DNA sequences in the maturaseK (MatK) chloroplast intergenic region and nuclear ribosomal DNA internal transcribed spacer (ITS) regions. The SNP markers developed in this study are useful for rapidly identifying specific C. tricuspidata ecotypes from different regions.

조류 다양성 확보를 위한 비오톱 평가지표개발 (Development of Biotope Evaluation Indexes for Improving Bird Diversity)

  • 토지이용 강도가 높은 도시비오톱을 대상으로 조류다양성 확보를 위한 비오톱 면적지수의 활용방안에 대한 연구를 수행하였다. 이 연구의 목적은 비오톱 평가지표들사이의 상관관계를 살펴보고 조류다양성을 보호하기 위한 비오톱 평가지표를 개발하여 생태적으로 건전한 도시비오톱의 효과적인 복원방법을 계획하는 데 있다. 도시 비오톱에서 조류 다양성에 영향을 주는 평가지수의 분석을 위해서, 각각의 사이트에 대해 비오톱 면적, 곤충종수, 인접도로와의 거리, 비오톱 면적지수를 조사하였다. 결론적으로, 비오톱평가 지수들 사이의 상관관계는 다음과 같은 결과가 나타났다. 조류종수와 곤층종수 > 조류종수와 비오톱 면적지수 > 곤충종수와 비오톱 면적지수 다시말해, 조류다양성은 실질적으로 곤충종수와 비오톱 면적지수에 의해 영향을 받는다. 결과로부터 얻어진 등식은 조류종수는 6.124 $\times$ 비오톱면적지수 + 0.095 $\times$ 곤충종수 - 1.197 이다. 그러므로, 비오톱면적지수와 곤충을 위한 수직적 경관다양성이 유지된다면, 작은 면적의 비오톱이라 할지라도 도시지 역에서 조류다양성을 증진시키는 데 도움을 줄 수 있을 것이다.

당귀 종판별을 위한 엽록체 기반 SSR 마커 개발 (Development of Chloroplast DNA-Based Simple Sequence Repeat Markers for Angelica Species Differentiation)

  • Background: In the herbal medicine market, Angelica gigas, Angelica sinensis, and Angelica acutiloba are all called "Danggui" and used confusingly. We aimed to assess the genetic diversity and relationships among 14 Angelica species collected from different global seed companies. Toward this aim we developed DNA markers to differentiate the Angelica species. Methods and Results: A total of 14 Angelica species, A. gigas, A. acutiloba, A. sinensis, A. pachycarpa, A. hendersonii, A. arguta, A. keiskei, A. atropurpurea, A. dahurica, A. genuflexa, A. tenuissima, A. archangelica, A. taiwaniana, and A. hispanica were collected. The genetic diversity of all 14 species was analyzed by using five chloroplast DNA-based simple sequence repeat (SSR) markers and employing the DNA fragment analysis method. Each primer amplified 3 - 12 bands, with an average of 6.6 bands. Based on the genetic diversity analysis, these species were classified into specific species groups. The cluster dendrogram showed that the similarity coefficients ranged from 0.77 to 1.00. Conclusions: These findings could be used for further research on cultivar development by using molecular breeding techniques and for conservation of the genetic diversity of Angelica species. The analysis of polymorphic SSRs could provide an important experimental tool for examining a range of issues in plant genetics.

Genetic Diversity and Population Structure of Korean Soybean Collection Using 75 Microsatellite Markers

  • Soybean (Glycine max L.) is crucial legume crop as source of high quality vegetable protein and oil, and Korea is regarded as a part of center of soybean origin. To expand the information of conserved genetic diversity, we analyzed the genetic variability of soybean collection mainly introduced Korean accessions using 75 microsatellite markers. A total of 1,503 alleles with an average value of 20.0 alleles were detected among 644 accessions. Korean collection revealed average allele number of 13.4 while Chinese, Japanese and Southeast Asian accessions showed 9.0, 5.4 and 6.5 mean alleles, respectively. Especially, Korean accessions showed more number of private allele per locus as 3.4 contrary to other geographical groups. The mean expected heterozygosity and polymorphic information content was 0.654 and 0.616, respectively, and expected heterozygosity values were not significantly distinguished according to the geographical groups. The phylogenetic dendrogram and deduced population structure based on DNA profiles of 75 SSR loci showed Korean accessions formed distinct gene pool against Chinese accessions, and could be divided into five subpopulations. Korean soybean accessions have specific genetic diversity and might be serve the valuable alleles for bio-industry as a part of the center of soybean origin.

상향링크 협력기지국 시스템을 위한 사용자 간 준동기 OFDMA 기법 (Inter-user Quasi-synchronous OFDMA for Cooperative Base Stations Systems)

  • 본 논문에서는 상향링크 협력 기지국 통신 시스템에서 사용자 간 시간 동기오차에 강한 OFDMA 기법을 제안한다. 상향링크에서는 하향링크와 달리 각 사용자 신호들의 신호가 각각 다른 타이밍으로 수신되어 간섭이 발생하는 문제점이 있다. 이러한 문제점을 해결하기 위해 각 사용자들의 신호의 송신 타이밍을 제어하는 과정이 필요하다. 하지만, 협력 기지국 통신에서는 각 기지국들에 대한 각 사용자의 상대적 위치가 각각 다르기 때문에 동기화 과정이 제대로 수행되기 어렵다. 즉, 한 기지국에 맞춰 각 사용자들의 동기화 과정을 수행하더라도 다른 기지국에서는 동기오차가 발생할 수밖에 없다. 본 논문에서는 한 칩의 시간 동기 오차가 존재하더라도 영상관도를 유지하는 ZCZ 부호를 시간축으로 스프레딩 하는 기법을 적용함으로써 상향링크 협력 시스템에서 모든 기지국에 대해 완벽한 동기화가 가능하게 한다. 제안하는 방식의 성능이 시간 동기 오차가 존재하지 않는 기존의 협력 기지국 OFDMA 기법의 성능에 도달함을 실험결과를 통해 보인다.

Lipase Diversity in Glacier Soil Based on Analysis of Metagenomic DNA Fragments and Cell Culture

  • Lipase diversity in glacier soil was assessed by culture-independent metagenomic DNA fragment screening and confirmed by cell culture experiments. A set of degenerate PCR primers specific for lipases of the hormone-sensitive lipase family was designed based on conserved motifs and used to directly PCR amplify metagenomic DNA from glacier soil. These products were used to construct a lipase fragment clone library. Among the 300 clones sequenced for the analysis, 201 clones encoding partiallipases shared 51-82% identity to known lipases in GenBank. Based on a phylogenetic analysis, five divergent clusters were established, one of which may represent a previously unidentified lipase subfamily. In the culture study, 11 lipase-producing bacteria were selectively isolated and characterized by 16S rDNA sequences. Using the above-mentioned degenerate primers, seven lipase gene fragments were cloned, but not all of them could be accounted for by the clones in the library. Two full-length lipase genes obtained by TAIL-PCR were expressed in Pichia pastoris and characterized. Both were authentic lipases with optimum temperatures of ${\le}40^{\circ}C$. Our study indicates the abundant lipase diversity in glacier soil as well as the feasibility of sequence-based screening in discovering new lipase genes from complex environmental samples.

  • 본 연구는 동아시아 지역이 유교 문화권으로 일컬어져 온 지역이지만 유교 문화권 안에서 발달했던 가족 중심의 문화, 전통 문화, 대인 관계 등의 인간관계와 생활습관 문화에서 차이가 있음을 전제하고, 앞으로 국가간 교류의 활성화와 원활한 의사소통, 인간 관계 형성을 위해서는 동아시아 지역의 문화 다양성을 인식하고 이해하는 교육으로의 전환이 필요함을 제시하였다. 그리고 지리수업에서 동아시아 문화 다양성 교육을 위한 내용을 구성하여 제시하였다. 교과와의 연계성을 높이기 위하여 2009개정 교육과정 지리 영역 단원과 단원별 성취 기준을 분석하였고 분석에 의거하여 초, 중, 고등학교 수준에 적합한 학습 내용을 구성하여 제시하였다.

Composition and Diversity of Tree Species in Kamalachari Natural Forest of Chittagong South Forest Division, Bangladesh

  • Information on plant diversity and community structure are required to chalk out necessary actions for conservation management. The present study assessed the composition and diversity of tree species in Kamalachari Natural Forest of Chittagong South Forest Division, Bangladesh, during April 2010 to November 2011. A total of 107 tree species belonging to 72 genera and 37 families were recorded, where Moraceae family was represented by maximum (11) species. Density, Basal area and volume of tree species were $418{\pm}20.09stem/ha$, $21.10{\pm}2.62m^2/ha$ and $417.4{\pm}79.8m^3/ha$ respectively. Diameter and height class distribution of tree species revealed an almost reverse J-shaped curve. Both the number of species and percentage of tree individuals were maximum in the lower DBH and height ranges. Anthropogenic disturbances like illegal tree cutting, over extraction, settlement inside forest area etc. were noticed during the study, which are supposed to cause gradual decrease of both tree species and individuals in the higher DBH and height classes. However, Artocarpus chama was found dominant showing maximum IVI followed by Schima wallichii, Aporosa wallichii, and Lithocarpus acuminata. The quantitative structure of the tree species of Kamalachari natural forest is comparable to other tree species rich tropical natural forests. The findings of the study may help in monitoring future plant population changes of the identified species and adopting species specific conservation programs in Kamalachari natural forest.
