Different Expression of Extracellular Matrix Genes: Primary vs. Recurrent Disc Herniation

  • Objective: Recurrent lumbar disc herniation has been reported to occur in 5% to 15% of surgically treated primary lumbar disc herniation cases. We investigated the molecular biologic characteristics of primary herniated discs and recurrent discs to see whether the recurrent discs has the similar biological features with primary herniated discs. Methods: Primary hemiated disc and recurrent disc cells were obtained by discectomy of lumbar disc patients and cells were isolated and then taken through monolayer cultures. We compared chondrogenic and osteogenic mRNA gene expression, and western blot between the two groups. Results: The mRNA gene expression of recurrent disc cells were increased 1.47* times for aggrecan, 1.38 times for type I collagen, 2.04 times for type II collagen, 1.22 times for both Sox-9 and osteocalcin, and 1.31 times for alkaline phosphatase, respectively, compared with the primary herniated lumbar disc cells (*indicates p < 0.05). Westem blot results for each aggrecan, type I collagen, type II collagen, Sox-9, osteocalcin, and alkaline phosphatase were similar between the primary herniated disc cells and recurrent disc cells. Conclusion: These results indicate that the recurrent disc cells have similar chondrogenic and osteogenic gene expression compared to primary herniated disc cells. Therefore, we assumed that the regeneration of remaining discs could fill the previous discectomy space and also it could be one of the factors for disc recurrence especially in the molecular biologic field.

Activation of Multiple Transcriptional Regulators by Growth Restriction in Pseudomonas aeruginosa

  • Growth restriction by antibiotics is a common feature that pathogenic bacteria must overcome for survival. The struggle of bacteria to escape from growth restriction eventually results in development of antibiotic-resistance through the expression of a set of genes. Here we found that some physiologically important transcriptional regulators of Pseudomonas aeruginosa including QscR, a quorum sensing (QS) receptor, SoxR, a superoxide sensor-regulator, and AntR, a regulator of anthranilate-related secondary metabolism, are activated by various growth-restricted conditions. We generated the growth-restricted conditions by various methods, such as overexpression of PA2537 and treatment with antibiotics or disinfectants. The overexpression of PA2537, encoding an acyltransferase homologue, tightly restricted the growth of P. aeruginosa and significantly activated QscR during the growth restriction. Similarly, treatments with gentamycin, tetracycline, and ethanol also activated QscR near their minimal inhibitory concentrations (MICs). Some non-QS regulators, such as AntR and SoxR, were also activated near the MICs in the same conditions. However, LasR and PqsR, other QS receptors of P. aeruginosa, were not activated, suggesting that only a specific set of transcriptional regulators is activated by growth restriction. Since paraquat, a superoxide generator, significantly activated QscR and AntR, we suggest that the oxidative stress generated by growth restriction may be partly involved in this phenomenon.

경골 파혈산동탕(破血散疼湯)이 골절 생쥐의 골 유합에 미치는 영향 (Effect of Pahyeolsandong-tang (Poxiesanteng-tang) in Tibia Fracture-induced Mice)

  • Objectives The main purpose of this study was to evaluate the bone healing effect of Pahyeolsandong-tang (PHT)(Poxiesanteng-tang) extract in tibia fracture-induced mice. Methods PHT was extracted using a solution of 35% ethanol in 60℃ for 8 hours. Mice were randomly divided into 4 groups (normal, control, PHT 50 and PHT 100). Mice of experimental groups were medicated with PHT 50 or 100 mg/kg for 7 to 21 days. To clarify the effect of bone fracture healing, relative messenger RNA (mRNA) expressions of osteocalcin (OCN), runt-related transcription factor 2 (Runx2), osterix (OSX), Sox9, collagen type II alpha 1 chain (Col2a1), receptor activator of nuclear factor kappa-B ligand (RANKL), osteoprotegerin (OPG) were examined. Results In in vitro experiment, relative mRNA expression of OCN, Runx2, Col2a1 was significantly increased in PHT treated group to compare with control differentiation group. In in vivo experiment, relative mRNA expression of OCN, Runx2, OSX, Sox9, Col2a1, RANKL, OPG was significantly increased in PHT treated group. Conclusions This study showed that PHT accelerates bone fracture healing through the activation of osteoclasts and osteoblasts. It was showed that PHT significantly promotes osteoblasts differentiation by osteoblast differentiation markers such as OCN, Runx2, Col1a2. Also it was investigated that PHT had stimulatory effect on osteoblasts function through enhancing OCN, Runx2, OSX, Sox9, Col2a1 and, osteoclasts function through enhancing RANKL and OPG markers. PHT effectively promotes bone fracture healing process through activation of osteoblasts and osteoclasts.

울산항 선박으로부터의 대기오염 배출량 산정에 관한 연구 (A studies on the Air pollutant Emission Rate calculation from vessels in the Ulsan Port)

  • 국제해사기구(IMO)에서 1997년 9월 선박으로부터의 대기오염방지를 위한 협약(MARPOL 부속서VI)을 채택하여 U년 5월 19일 협약 발효를 앞두고 있다. 따라서 향후 선박으로부터 배출되는 배기가스 중에 포함된 아황산가스(SOx)와 질소산화물(No%)등 대기오염 물질의 배출 규제와 항만이나 터미널에서 유조선으로부터 휘발성유기화합물(VOCs) 배출금지 등의 규제가 강화될 것으로 예상된다. 본 연구는 이에 대한 대비와 관리방안을 모색하고자 외국에서 이용되는 선박의 배출오염물질 산정 방법을 이용하여 선박에서 배출되는 SOx, NOx의 배출량을 산정하였다. 또한 울산항 특성을 감안하여 액체화물 적$\cdot$양하시 발생되는 휘발성유기화합물(VOCs)의 양을 산정하는 방법에 대해 고찰하고 그 배출량을 산정하였다.

  • Current desulfurization and denitrification technologies have reached a considerable level in terms of reduction efficiency. However, when compared with the simultaneous reduction technology, the individual reduction technologies have issues such as economic disadvantages due to the difficulty to scale-up apparatus, secondary pollution from wastewater/waste during the treatment process, requirement of large facilities for post-treatment, and increased installation costs. Therefore, it is necessary to enable practical application of simultaneous SOx and NOx treatment technologies to remove two or more contaminants in one process. The present study analyzes a technology capable of maintaining simultaneous treatment of SOx and NOx even at low temperatures due to the electrochemically generated strong oxidation of the wet-pulse complex system. This system also reduces unreacted residual gas and secondary products through the wet scrubbing process. It addresses common problems of the existing fuel gas treatment methods such as SDR, SCR, and activated carbon adsorption (i.e., low treatment efficiency, expensive maintenance cost, large installation area, and energy loss). Experiments were performed with varying variables such as pulse voltage, reaction temperature, chemicals and additives ratios, liquid/gas ratio, structure of the aeration cleaning nozzle, and gas inlet concentration. The performance of individual and complex processes using the wet-pulse discharge reaction were analyzed and compared.

국내의 농업기계에 의해 배출되는 대기 오염 물질 분석 (Analysis of Air Pollutant Emissions from Agricultural Machinery in South Korea)

  • From 2019 onwards, more stringent regulations (from Stage 4 to Stage 5) are to be implemented in Europe in order to reduce the air pollutant emissions. In South Korea, the government authorities started to make new regulation to meet the European regulation. As a first step, the air pollutant emissions such as CO, NOx, SOx, TSP, $PM_{10}$, $PM_{2.5}$, VOC, $NH_3$ by agricultural machinery were analyzed based on CAPSS inventory along with the analysis in the general aspect in this study. Three levels of analysis was conducted each in agricultural machinery aspect along with in the general aspect. Per agricultural tractor, all kinds of the air pollutant emissions decreased by 25, 25, 99, 25, 25, 25, 25% for the CO, NOx, SOx, TSP, $PM_{10}$, VOC, $NH_3$ emissions each from the year 2000 to the year 2014. Per combine harvester, all kinds of the air pollutant emissions decreased by 63, 63, 91, 63, 63, 63, 63% for the CO, NOx, SOx, TSP, $PM_{10}$, VOC, $NH_3$ emissions each from the year 2000 to the year 2014.

Intraperitoneal Paclitaxel Combined with S-1 Plus Oxaliplatin for Advanced Gastric Cancer with Peritoneal Metastasis: a Phase I Study

  • Purpose: We designed a new regimen by combining intraperitoneal (IP) paclitaxel (PTX) with systemic S-1 plus oxaliplatin (SOX) for the treatment of advanced gastric cancer with peritoneal metastasis. This dose-escalation study aimed to determine the maximum tolerated dose (MTD) and recommended dose (RD) of IP PTX administered weekly to patients. Materials and Methods: Eight cycles of IP PTX plus SOX regimen were administered to the patients. S-1 was administered orally twice daily at a dose of 80 mg/m2/day for 14 consecutive days, followed by 7 days of rest. Intravenous oxaliplatin was administered at a fixed dose of 100 mg/m2 on day 1, while IP PTX was administered on days 1 and 8. The initial dose of IP PTX was 40 mg/m2, and the dose escalation was set in units of 20 mg/m2 up to 80 mg/m2. Dose-limiting toxicities (DLTs) were defined as grade 3 non-hematologic toxicities, grade 4 leukopenia, grade 3 febrile neutropenia, and grade 3 thrombocytopenia. Results: Nine patients were included in the study. No DLTs were observed in any of the enrolled patients. Therefore, the MTD was not reached, and the RD of IP PTX was determined to be 80 mg/m2. Four patients (44%) showed a decreased peritoneal cancer index score on second-look laparoscopic examination. Conclusions: The present study determined the dose for further clinical trials of IP PTX to be 80 mg/m2, when combined with a systemic SOX regimen.

A Study on the Enhancement of Inventories for Precursors (NOx, SOx) Released from Open Burning of Agricultural Waste Vinyl Causing the Secondary Generation of Particulate Matters

  • Background and objective: While response measures to particulate matters in rural areas are limited due to poor inventory record keeping in the agricultural sector, it is necessary to control agricultural waste vinyl and the emission of precursors released from open burning and the secondary generation of particulate matters. Currently, the open burning emission calculation method uses the definition prescribed in CAPSS by the National Institute of Environmental Research. Methods: This study presented an open burning emission calculation formula for agricultural waste vinyl, which is included as agricultural waste. As for activity data, the open burning ratio of agricultural waste vinyl, and the annual incineration volume provided in the Status Survey by the Ministry of Agriculture, Food, and Rural Affairs were applied. The emission factor was generated through incineration tests on three agricultural plastic film samples collected by the Korea Environment Corporation. Results: Among precursors, SOx and NOx were selected and their emission features were monitored with incineration experiment infrastructure based on the EPA 5G method. The highest emission concentration by agricultural waste type was concentrated in the first and second quarters. As for emission factor of SO2, it was calculated at 98.25 g/kg for mulching-use LDPE, 52.31 g/kg for greenhouse-use LDPE, and 14.40 g/kg for HDPE. As for NOx, it was calculated at 18.21 g/kg for mulching-use LDPE, 16.49 g/kg for greenhouse-use LDPE, and 10.67 g/kg for HDPE. Conclusion: This test confirmed the incineration features of PE-based plastics, ascertained the SOx emission factor that had not been included in open burning in the past, and established that low NOx emission concentration is interfered by soil mixed with livestock excretions. The findings from this study are expected to contribute to improving the system for controlling air pollutants in rural environments.

  • 전 세계적으로 환경에 관한 관심이 높아짐에 따라 시멘트 제조 시설은 순환자원을 사용하여 원료 및 연료 대체율을 향상시켜 폐기물처리와 같은 환경문제에 기여하고 있다. 대기오염물질 중 하나인 황산화물(SOx)은 석회석을 원료로 사용하는 시멘트 제조 특성상 배출관리가 잘 이루어지고 있지만, 순환자원 사용 증대에 따른 추후 설비 공정 및 환경 변화가 어떻게 변화될지는 미지수이다. 이에 따라 시멘트 제조 시설도 탈황 관련 기술 및 연구가 필요할 것으로 판단된다. 따라서 본 연구에서는 시멘트 제조 시설에서의 탈황기술 적용 가능성 조사 및 시멘트 제조 공정에서 발생되는 부산물을 활용한 탈황기술 적용 방안을 모색하였다.

부산 북항에서의 선박 배출물질 현황과 선속제한에 의한 배출량 감소 연구 (Current Status of Ship Emissions and Reduction of Emissions According to RSZ in the Busan North Port)

  • 최근 지구 환경문제에 대한 논의가 활발해지면서 국제 운송의 큰 부분을 차지하고 있는 해상운송에서도 배출물질 규제를 위한 정책이 시행되고 있다. 이 연구에서는 선속 제한에 의한 배출량의 감축 효과를 검토하기 위하여 기관 부하율을 적용하여 선박의 배출물질을 수치계산하였다. 2017년 1월 1일부터 12월 31일까지 부산 북항의 입출항 선박을 대상으로 선속제한구역 20마일권역을 설정하고 해당 구간에서의 선종별, 선속별로 배출량을 계산하고 분석하였다. 항행, 접 이안, 정박 중일 때를 모두 포함하여 가장 많은 배출물질을 발생시키는 선박은 컨테이너선 76.1 %, 일반화물선 7.2 %, 여객선 6.8 %의 순으로 계산되었다. 항행 및 접 이안 모드일 때는 일반화물선이 여객선보다 배출물질이 적었지만 정박 모드일 때는 여객선보다 많았다. 총 배출물질은 질소산화물, 황산화물, 입자상물질, 휘발성유기화합물의 순으로 각각 49.4 %, 45 %, 4 % 1.6 %로 구성되었다. 선속 제한이 없는 경우와 선박 속도를 12노트, 10노트, 8노트로 제한시킬 때 배출물질을 비교하면 속도 12노트 제한의 경우 질소산화물 39 %, 휘발성유기화합물 40 %, 입자상물질 42 %, 황산화물 38 %의 감소효과가 있고, 10노트 제한일 때 질소산화물 52 %, 휘발성유기화합물 54 %, 황산화물 56 %, SOx 50 %의 감소효과가 있으며, 8노트 제한일 때 질소산화물 62 %, 휘발성유기화합물 64 %, 입자상물질 67%, 황산화물 59 %의 감소효과가 있었다. 이처럼 선박의 속도 감소에 따라 배출물질 역시 크게 감소되는 연구결과를 확인할 수 있었으며, 향후 항만 배출물질 감소를 위해 선박의 속도를 제한하는 방안을 적극적으로 고려할 필요가 있다.
