Simulation of (R)- and (S)- Ketoprofen Separation in Simulated Moving Bed (SMB) ((R)-케토프로펜과 (S)- 케토프로펜 분리를 위한 유사 이동층 크로마토그래피의 전산모사)

  • A simulation study for finding purity changes of extract and raffinate as well as the best purity of (S)-ketoprofen in simulated moving bed (SMB) was performed with changing parameters of $m_2$ and $m_3$ from triangle theory. Aspen simulator allowed separation process simulation of (R)- and (S)-ketoprofen in SMB and compared 4-bed SMB and 8-bed SMB based on the same Henry constant and mass transfer coefficient. The 4-bed SMB consisted of 4 columns (200 mm of length, 10 mm of diameter) and the 8-bed SMB constructed by 8 columns (100 mm of length, 10 mm of diameter), and therefore total column length was made the same as 800 mm. Considering purities of both (R)-and (S)-ketoprofen, both 4-bed SMB and 8-bed SMB had the best purity when $m_2$ and $m_3$ were on 12.0 and 13.0 in the center of triangle. Taking only (S)-ketoprofen into account, 4-bed SMB as well as 8-bed SMB had the best purity when $m_2$ and $m_3$ were on 10.9 and 12.6 in the left outside triangle, and their purities were 93.3 % for 4-bed SMB and 96.9 % for 8-bed SMB.

  • SMB(simulated moving bed) chromatography system has been developed to realize continuous separation and save solvent consumption for binary mixture such as chiral compounds in especial. The parameters of SMB chromatography system can be calculated from mass balance equations of true moving bed chromatography, and they are used in design of 6-column SMB chromatography. We can separate 1'R-25 and 1'S-2S enantiomers as a raffinate product in 95% of purity using assembled SMB chromatography system.

  • IgY (Immunoglobulin Yolk) is a specific antibody in egg yolk, and it protects human body from virus and antigen. There are a lot of egg yolk components such as lipoprotein and protein. To separate IgY, HPLC (High Performance Liquid Chromatography) and precipitation were used in a batch mode and SMB (Simulated Moving Bed) was adopted for continuous purification of yolk proteins. IgY and other proteins in yolk were separated by using three-zone and four-zone SMB chromatography. Before performing SMB experiments, batch chromatography simulation parameters and adsorption isotherms were obtained. The parameters of batch chromatography were used to simulate SMB using Aspen chromatography. To compare three-zone and four-zone SMB chromatography, simulations in $m_2-m_3$ plane on the triangle theory were carried out. In terms of concentration and purity of both IgY and other lipoproteins, 3-zone SMB process is considered as ideal at the vertex of triangle ($m_2$, $m_3$=0.1, 1.1). 4-zone SMB yields the highest IgY purity at the coordinate ($m_2$, $m_3$=0.06, 0.5), which is the pure raffinate region. In 3-zone SMB without recycle, other lipoproteins in extract are largely affected in purity by small shift from the vertex of triangle ($m_2$, $m_3$=0.1, 1.1).

  • Currently, researches on recycling and reuse of waste energetic materials have recently gained a great attention from advanced countries due to ever tightening environmental regulations. In this study, as a part of a recycling technology, the experiments and dynamic simulation of simulated moving bed (SMB) process were performed to efficiently separate TNT and RDX from their mixture, which are main components of ammunition. In order to determine the operation zone of SMB process, the retention times of TNT and RDX were measured using HPLC at different flow rates and the adsorption equilibrium of each component was obtained by using a moment method. According to the adsorption equilibrium and the triangle theory of SMB process, four operation points were determined and separation experiments were carried out by the SMB process using the solvent consisting of acetonitrile and water. Two different mixing ratios (6:4 and 1:1) of acetonitrile and water were chosen for the experiment due to the great impact of mixing ratio of the solvent on separation. The performance of SMB process was evaluated by purity, recovery, productivity and solvent consumption. Pure TNT and RDX were successfully obtained from the SMB process and the dynamic simulation for the SMB process agreed well with the experimental results. Therefore, the dynamic model could be applied for predicting the dynamic behavior of the SMB process and designing a large scale SMB process.

  • The effect of sodium malate buffer(SMB) on the fermentation of Kimchi was investigated. Chemical and microbial changes were investigated during fermentation of Kimchi containing 0,2,4 and 6% salt at $25^{\circ}C$. The fermentaton of lower salted Kimchi was taster than that of higher salted Kimchi. One percent addition of SMB, as pH adjuster, showed the retardation of Kimchi fermentation about 36 hours at $25^{\circ}C$. Due to the buffer action of SMB, the acidity was inclosed during the fermentation as concentration of SMB increased. The buffer action of SMB was increased during fermentation of Kimchi.

  • Simulated moving bed (SMB) chromatography has been investigated in order to separate chiral compounds for pharmaceutical use. SMB utilizes the principle of true moving bed (TMB) chromatography, and has the advantages of low solvent usage, flexible configuration of columns and hardwares, and high productivity of the chiral compounds over the TMB. Large scale separation of xylene isomers and saccharides has been conducted since 1960s. However, the application of SMB in the fine chemical industries is still in the infant stage. The study of SMB for the chiral compounds production was initiated in the mid 1990s and further researches are actively undergoing. This review summarizes the principle of SMB as well as the chemistry of chiral stationary phase and chiral compounds.

A novel combination of sodium metabisulfite and a chemical mixture based on sodium benzoate, potassium sorbate, and sodium nitrite for aerobic preservation of fruit and vegetable discards and lactic acid fermentation in a total mixed ration for ruminants

  • Objective: Our recent findings confirmed the effectiveness of sodium metabisulfite (SMB) in controlling the growth of undesirable microorganisms in fruit and vegetable discards (FVD); however, lactic acid bacteria (LAB) are susceptible to its antibacterial effects. Two series of experiments were conducted to enable the survivability of LAB during silage fermentation of a total mixed ration (TMR) containing SMB-treated FVD. Methods: In Exp. 1, the objective was to isolate a strain of LAB tolerable to the toxic effect of SMB. In Exp. 2, the SMB load was minimized through its partial replacement with a chemical mixture (CM) based on sodium benzoate (57%), potassium sorbate (29%), and sodium nitrite (14%). FVD was treated with SMB + CM (2 g each/kg biomass) and added to the TMR at varying levels (0%, 10%, or 20%), with or without KU18 inoculation. Results: The KU18 was screened as a presumptive LAB strain showing superior tolerance to SMB in broth medium, and was identified at the molecular level using 16S rRNA gene sequence analysis as Lactobacillus plantarum. Inoculation of KU18 in TMR containing SMB was not successful for the LAB development, biomass acidification, and organoleptic properties of the resultant silage. In Exp. 2, based on the effectiveness and economic considerations, an equal proportion of SMB and CM (2 g each/kg FVD) was selected as the optimal loads for the subsequent silage fermentation experiment. Slight differences were determined in LAB development, biomass acidification, and sensorial characteristics among the experimental silages, suggesting the low toxicity of the preservatives on LAB growth. Conclusion: Although KU18 strain was not able to efficiently develop in silage mass containing SMB-treated FVD, the partial substitution of SMB load with the CM effectively alleviated the toxic effect of SMB and allowed LAB development during the fermentation of SMB + CM-treated FVD in TMR.

  • Simulated moving bed (SMB) process is a continuous chromatographic technology used to separate a large amount of petrochemicals, fine chemicals, pharmaceuticals, and so on, drawing a great attraction of related industries. With the recent development of biotechnology, the SMB process has been adopted for the separation of various useful bio-products. Attempts to understand the separation mechanism of the SMB process in many aspects are reported in many publications. These researches have dealt with the improvement of SMB for easier operation and solving problem in process. The feed mixture fed into the SMB process may be of different concentration batch by batch rather than in uniform concentration. Retention behaviors of feed (psicose (A) and fructose (B) mixture) existing in the SMB unit in dynamic steady-state and feed (psicose (C) and fructose (D) mixture) newly injected into the SMB were analyzed. It was observed that the existing components, (A) and (B), were eluted relatively faster as the injection time of new feed was earlier during the port-switching period. In addition, the components (C) and (D) were eluted earlier as fresh feed was injected earlier in a port-switching time.

  • The South Korean Government had supported the IT implementation of 30,000 SMB(Small and Medium-sized) enterprises from '2000 to '2003 for the purpose of the competitiveness of SMB enterprises. However, the utilization rate of If package is very low. This paper recommanded the improved suggestion of the existed policies of the following service of the SMB industry's information technology implementation. There are industry-university cooperation program, efficient on-offline integration system, the development of SMB industry's IT performance measure evaluation index and so on.
